Description

【発明の詳細な説明】[Detailed description of the invention]

本発明は、油脂、或いは油脂含有食品などに添
加してすぐれた抗酸化性を示す天然源抗酸化成分
の製法に関する。 更に詳しくは、ローズマリー及び/又はセージ
の抗酸化成分含有アルコール溶液を、アルコール
濃度約80重量%以下のアルカリ性含水アルコール
溶液となし、該溶液を多孔性樹脂吸着剤で処理
し、得られた処理液を酸性にして形成される固型
分を採取することを特徴とする天然源抗酸化成分
の製造方法に関する。 従来、ハーブ系香辛料が油脂類に対して酸化防
止効果のあることは知られており、殊に強い抗酸
化性を有するローズマリー及びセージについて
は、多くの研究報告が成されている。 例えばローズマリー及びセージのそれぞれの粉
末、該粉末の石油エーテル抽出液、及びその石油
エーテル抽出残渣のエタノール抽出液を、それぞ
れ、ラードに添加した場合、何れも著しい抗酸化
性が認められ〔Food Res.17、46(1952)〕、また
ローズマリー或いはセージを、夫字、水抽出、或
いはエタノール抽出すると、エタノール抽出液は
ラードに対してすぐれた抗酸化性を示し、且つ香
辛料に含有されている抗酸化性成分の大部分はエ
タノールによつて抽出されると言われている〔栄
養と食糧27、(4)、181(1974)〕。 しかしながら上記の如き方法によつて得られた
抽出物、或いはヘキサン、アセトン、メタノー
ル、エタノール、メチレンクロライド、エチレン
ジクロライドなどの有機溶媒で抽出後、抽出液か
ら溶媒を除去して得られるオレオレジン類、或い
は水蒸気蒸留などの手段によつて得られる精油類
など何れも、原料ローズマリー及び/又はセージ
に由来する香辛料特有の香味を有するために、酸
化防止の目的でこれらを飲食品に添加すると、該
飲食品の風味に悪影響を及ぼす。従つて、その添
加量は著しく制限されるので、該飲食品に対する
酸化防止効果が不充分となつてしまう。 かゝる欠点を改善するためにローズマリー或い
はセージの溶媒抽出物から香味成分及び着色成分
を除去する方法が種々提案されている。 例えばシソ科植物又はセリ科植物をPH7〜10の
塩基性水溶液で処理して酸化防止剤物質を抽出す
る方法(特公昭53−9395)、或いはローズマリー
及び/又はセージを40〜60%濃度の含水メタノー
ル、もしくは含水エタノールで処理して抗酸化性
成分を抽出し、次いで得られた抽出液に水を加え
て非水溶性の抗酸化性成分を析出させ、更に活性
炭を加えて撹拌した後過して非水溶性抗酸化性
成分と活性炭の混合物を得、この混合物をメタノ
ール或いはエタノールで処理して非水溶性抗酸化
性成分を抽出し、この抽出液から溶媒を留去して
非水溶性抗酸化性成分濃縮物を取得し、一方、上
記非水溶性抗酸化性成分と活性炭との混合物を
過して得た液から溶媒を留去して、水溶性抗酸
化剤を取得する方法(特開昭55−18435)などが
提案されているが、何れの方法によつて得られた
抗酸化性物質も、香辛料特有の香味を有してお
り、到底満足すべきものではなかつた。 かゝる欠点を更に改善するために、脱臭工程を
付加して無味、無臭の抗酸化性物質を得る方法も
種々試みられている。 例えばローズマリー、或いはセージの粉末を低
沸点の有機溶媒で抽出し、得られた抽出液を濃縮
後、濃縮物を熱水で洗浄し、次いで該濃縮物をメ
タノールなどの低沸点溶剤に溶解しそれに活性
炭、活性白土、硅酸などの吸着剤を加えて脱色処
理を行ない、次いで該処理液を濃縮して得た濃縮
物を油脂類などに溶解乃至懸濁させ、これを水蒸
気蒸留、或いは分子蒸留などの蒸留処理して香気
成分を除去し、無味、無臭の抗酸化性物質を製造
する方法〔USP.3950266(1976)〕、或いはローズ
マリー、セージ、もしくはバジルの粉末を動植物
性油脂と共に約80〜約180℃で約30分乃至3時間
加熱後不溶固形物を除去して得た動植物油層を減
圧下で約120〜220℃で約10〜約60分間加熱処理し
て香気成分を留去させて抗酸化性物質を製造する
方法〔USP.3732111(1973)〕、更には酸化抑制物
質を含む植物材料粉砕物、或いはその低沸点溶媒
抽出物にモノグリセリドなどの蒸留媒体及び油脂
を添加して、分子蒸留し、酸化抑制物質を含有す
る該蒸留媒体を採取する方法(特開昭55−
48281)、ハーブ系香辛料、その精油採取残渣、有
機溶媒抽出して得たオレオレジン、或いはオレオ
レジン抽出残渣などの原料をエタノールなどの極
性溶媒で抽出処理して抽出液を得た後、これに活
性炭、珪藻土、酸性白土などの吸着剤を加えて脱
色処理し、次いでこの吸着剤を分離して得た溶液
部を濃縮した後水蒸気蒸留処理し、その水蒸気蒸
留残渣中の不溶部を採取して保存剤を製造する方
法(特開昭55−102508)、ハーブ系香辛料、その
精油採取残渣、ハーブ系香辛料のオレオレジンな
どの原材料を極性溶媒と非極性溶媒との混合溶媒
(容量比で2:98〜50:50)で抽出処理して得た
抽出液を活性炭、珪藻土、酸性白土などの吸着剤
を加え脱色処理し、次いでこの吸着剤を分離して
得た溶液部に水を加えて非極性溶媒層、含水極性
溶媒層及び沈澱物層の3層に分離させた後、各層
より抗酸化性物質を採取し、必要により水蒸気蒸
留処理を行なつて保存剤を製造する方法(特開昭
56−144078)など、数多くの提案が成されてい
る。 しかしながら、上記の如き方法は、何れも工業
的製法としては煩雑であり且つかかる方法によつ
て得られた抗酸化性物質は香味の面でやはり満足
すべきものではなかつた。 本発明者らは、ローズマリー及び/又はセージ
の抗酸成分含有アルコール溶液を、アルコール濃
度約80重量%以下のアルカリ性含水アルコール溶
液として、色素成分を沈澱させることにより、上
記溶液を脱色することができ、次いで該溶液を多
孔性スチレン系樹脂吸着剤で処理することにより
香気成分を除去でき、得られた処理液を酸性にす
ることにより、無色、無味、無臭の天然源抗酸化
成分を得ることができることを発見し、本発明を
完成するに至つた。 従つて本発明の目的は、ローズマリー及び/又
はセージの抗酸化成分含有材料より、無色、無
味、無臭で且つ抗酸化力の強い天然源抗酸化成分
を高収率で、工業的に有利に製造する方法を提供
するにある。 本発明の上記目的並びに更に多くの他の目的及
び利点は以下の記載から一層明らかとなるであろ
う。 本発明で用いるローズマリー及び/又はセージ
の抗酸化成分含有アルコール溶液は、ローズマリ
ー及び/又はセージの抗酸化成分含有材料をアル
コール類で抽出処理することにより得ることがで
きる。 上記ローズマリー及び/又はセージの抗酸化成
分含有材料としては例えば、ローズマリー及び/
又はセージの微粉砕物;該香辛料を水蒸気蒸留な
どして精油を採取した残りの精油採取残渣;或い
はこれらの材料をn−ペンタン、イソペンタン、
n−ヘキサン、石油エーテル、リグロイン、アセ
トン、酢酸メチル、酢酸エチル、メタノール、エ
タノール、n−プロパノール、イソプロパノー
ル、n−ブタノール或いはこれらの複数種の混合
物などの有機溶媒で抽出し、抽出液から溶媒を除
去して得られるオレオレジンを好ましく例示でき
る。 かゝるローズマリー及び/又はセージの抗酸化
成分含有材料を抽出処理するために用いるアルコ
ール類としては、例えばメタノール、エタノー
ル、プロパノール、、イソプロパノール、プロピ
レングリコール、グリセリン、これらのアルコー
ル類の複数種の混合物、或いはこれらの含水物な
どを例示でき、殊にエタノールを好ましく例示す
ることができる。 上記の如きローズマリー及び/又はセージの抗
酸化成分含有材料をアルコール類で処理するため
の抽出条件は、適宜選択することができ、例えば
該材料に約0.2〜約50倍重量のアルコール類を添
加後室温乃至使用溶媒の沸点温度で常圧下約5分
乃至約24時間静置、もしくは撹拌して抽出するこ
とができる。 また他の抽出処理態様としては、例えば上記抗
酸化成分含有材料、殊に好ましくはローズマリー
及び/又はセージの粉砕物、もしくはその精油採
取残渣の粉砕物乃至細断物を抽出カラムに詰め
て、該カラムの上部もしくは下部よりアルコール
類を連続的に送入してカラム抽出する方法を例示
することもできる。 更にまた他の好ましい抽出処理態様として、上
記の如き抗酸化成分含有材料にアルコール類を加
えて、加圧条件下で約90゜〜約180℃で加熱処理す
る方法を挙げることができる。 抽出終了後、不溶性固形物を分離除去すること
によりローズマリー及び/又はセージの抗酸化成
分含有アルコール溶液を得ることができ、また必
要ならば分離した不溶性固形物残渣にアルコール
類を加えて上記と同様な方法で抽出処理をくり返
しても良い。 本発明においては、上記の如き何れの抽出処理
法を採用しても差し支えないが、更にはローズマ
リー及び/又はセージの抗酸化成分含有材料に活
性炭、酸性白土、シリカ、多孔性スチレン系樹
脂、活性白土などの如き固体吸着剤を添加後、上
記の如くしてアルコール類で抽出処理しても良
く、かゝる方法によつて脱色された抗酸化成分含
有アルコール溶液を得ることができる。 本発明で用いるローズマリー及び/又はセージ
の抗酸化成分含有アルコール溶液は、また上記の
如き抗酸化成分含有材料をアルコール類以外の有
機溶媒で抽出して得られた抽出液から有機溶媒を
除去し、これをアルコール類に溶解することによ
り得ることができ、例えば、該材料をn−ペンタ
ン、イソペンタン、石油エーテル、n−ヘキサ
ン、リグロイン、メチレンクロライド、エチレン
ジクロライドなどの非極性溶媒、エチルエーテ
ル、アセトン、酢酸メチル、酢酸エチルなどの極
性溶媒、あるいはかかる非極性溶媒と極性溶媒と
の混合溶媒を用いて上記アルコール類による抽出
処理法と同様にして抽出処理し、次いで得られた
抽出液を常圧乃至減圧下で蒸留して溶媒を留去し
て濃縮物を得、これに約0.5乃至300倍重量のメタ
ノール、エタノール、n−プロパノール、イソプ
ロパノール、プロピレングリコール、グリセリ
ン、これらの混合物、或いはこれらの含水物など
の如きアルコール類、好ましくはエタノールを添
加して充分に混合撹拌し、必要ならば不溶性物質
を過、遠心分離、デカンテーシヨンなどの操作
によつて除去して抗酸化成分含有アルコール溶液
を得ることができる。 本発明方法は例えば次の如くして実施すること
ができる。上記ローズマリー及び/又はセージの
抗酸化成分含有アルコール溶液に例えば約1〜約
30重量%のアルカリ水溶液例えば、水酸化ナトリ
ウム、水酸化カリウム、炭酸水素ナトリウム、炭
酸水素カリウム、水酸化バリウム及びリン酸二ナ
トリウム或いはこれらの複数種の混合物の水溶液
を加え、必要ならば更に水を添加してアルコール
濃度約80重量%以下好ましくは約70%重量以下、
約20重量%以上で、且つ溶液のPHが約7以上とな
るように調整し、抗酸化成分のアルカリ性含水ア
ルコール溶液とする。含水アルコール濃度が80重
量%以上の場合は着色成分及び香気成分を充分に
除去することが困難であり、また20%以下の場合
は抗酸化成分の収率が減少して好ましくない。上
記アルカリ水溶液の添加条件は適宜選択できる
が、例えば、上記ローズマリー及び/又はセージ
の抗酸化成分含有アルコール溶液に室温乃至約50
℃の温度でアルカリ水溶液を添加して、約10分乃
至約3時間撹拌することによつて行うことができ
る。かかる処理によつて、緑色の色素成分を析出
させ、該色素成分を過、遠心分離、あるいはデ
カンテーシヨンなどの固液分離手段により分離す
ることにより、脱色された抗酸化成分含有アルカ
リ性アルコール溶液を得ることができ、また分離
した該色素を回収することができる。 次いで、上記アルカリ性含水アルコール溶液を
多孔性スチレン系樹脂吸着剤(以下これを“多孔
性重合樹脂”又は“多孔性樹脂”と略称すること
がある)で処理することにより、香気成分の除去
された処理液を得ることができる。かかる多孔性
スチレン系樹脂吸着剤としては、スチレンを主た
る原料として重合して得られた、多孔性樹脂であ
り例えば、スチレンとジビニルベンゼンとを重合
させて、比表面積や細孔容積を巨大化させた多孔
性の合成樹脂や多孔性の架橋ポリスチレンなどを
例示できる。上記多孔性重合樹脂処理の方法は、
例えば、抗酸化成分アルカリ性含水アルコール溶
液と多孔性重合樹脂を混合撹拌して行うことがで
きるが、好ましくは、例えば多孔性重合樹脂を充
填したカラムの上部より、抗酸化成分含有アルカ
リ性含水アルコール溶液を連続的にカラム内を通
過させ、該カラム下部より処理液を採取する方法
が良く、かかる方法は該樹脂の若生利用にも便利
である。多孔性重合樹脂の使用量は、抗酸化成分
含有アルカリ性含水アルコール溶液の香気の強さ
によつて適宜に選択することができるが、通常は
該溶液量の約0.001〜約5倍容量が好ましく例示
でき、該樹脂処理することによつて脱臭された処
理液を得ることができる。かくして得られた処理
液に酸類を添加して系の液性を酸性とし、抗酸化
成分を析出させる。かかる酸の種類としては、例
えば塩酸、硫酸、リン酸などの無機酸、或いは酢
酸、クエン酸、リンゴ酸、酒石酸、乳酸などの有
機酸、或いはこれらの混合物を例示できる。酸の
添加量は適宜に選択できるが、抗酸化成分含有ア
ルコール溶液に添加されたアルカリ量を中和する
に要する量で加えることが良く、例えばPH約5以
下となる様に添加することができる。 かかる操作によつて析出された固形物を、
過、遠心分離、あるいはデカンテーシヨン等の適
宜の固液分離手段により分離し、無色、無味、無
臭の抗酸化成分を得ることができ、更には該抗酸
化成分を熱水で洗浄して、脱水乾燥して粉末状と
することもできる。 また、本発明方法においては上記の如き抗酸化
成分含有アルコール溶液を、アルカリ性含水アル
コール溶液となす前に、該抗酸化成分含有アルコ
ール溶液を、固体吸着剤処理及び/又は陽イオン
交換樹脂処理することができる。かかる処理によ
つて更に精製度の高い、すぐれた抗酸化性成分を
得ることができる。固体吸着剤としては、例えば
活性炭、硅藻土、活性アルミナ、活性白土、酸性
白土、ベントナイト、微結晶セルロース、上記多
孔性重合樹脂、あるいはこれらの複数種の混合物
もしくは併用を例示することができ、好ましくは
活性炭及び/又は多孔性重合樹脂を例示できる。
更に、最も好ましくは多孔性重合樹脂を例示する
ことができる。また陽イオン交換樹脂としては、
例えばスチレンとジビニルベンゼンとを重合させ
て製造されたゲル型或いはポーラス型の三次元高
分子基体にスルフオン酸基を導入した強酸性陽イ
オン交換樹脂、該高分子基体にカルボキシル基を
導入した弱酸性陽イオン交換樹脂などを例示で
き、殊に強酸性陽イオン交換樹脂を好ましく例示
できる。かかる固体吸着剤処理、又はイオン交換
樹脂処理は、例えば次の如くして行うことができ
る。 上記ローズマリー及び/又はセージの抗酸化成
分含有アルコール溶液に固体吸着剤を添加して室
温乃至該アルコール溶液の沸点温度で約10分乃至
約30時間静置、或いは撹拌する。固体吸着剤の使
用量は、該アルコール溶液中の水不溶性色素成分
含有量によつて適宜選択することができるが、通
常は該色素成分含有量と少なくとも等量の該吸着
剤を使用して吸着剤処理を行なうのが好ましい。 次いで上記処理物を過、デカンテーシヨンあ
るいは遠心分離などの操作によつて固体吸着剤を
分離、除去して処理液を得ることができる。 また固体吸着剤として多孔性重合樹脂を用いる
場合は、該樹脂をカラムに充填し、前記多孔性樹
脂処理と同様の操作により、カラム上部より、抗
酸化成分含有アルコール溶液を通過させ、該カラ
ム下部より色素成分の除去された処理液を得るこ
とができる。 更にまた多孔性重合樹脂を用いて吸着剤処理を
行なつた場合は、該処理後の該樹脂をアセトンで
抽出処理することによつて該樹脂に吸着している
天然緑色色素を回収することができる。 かゝる緑色色素の回収は、上記の如きカラムを
用いての多孔性重合樹脂処理後の該樹脂について
行なうのが好ましく、かゝる場合は、該樹脂の充
填されたカラム上部より、連続的にアセトンを該
カラム内を通過させて該樹脂に吸着している色素
成分を溶出させ、該カラム下部から流出する溶出
液が殆んど無色になるまでアセトンによる抽出処
理を行ない、次いで得られたたアセトン溶液を常
圧乃至減圧下で蒸留し溶媒を溶去させることによ
り、殆んど無臭の天然緑色色素を回収することが
できる。 また抗酸化成分含有アルコール溶液の陽イオン
交換樹脂処理は、例えば、カラムを用いて多孔性
重合樹脂処理する場合と同様にして行なうことが
できる。更に上記抗酸化成分含有アルコール溶液
を陽イオン交換樹脂処理して得られた処理液を多
孔性重合樹脂処理しても良いし、上記の如き固体
吸着剤処理によつて脱色された抗酸化成分含有ア
ルコール溶液を陽イオン交換樹脂を用いて処理し
ても良い。 陽イオン交換樹脂の使用量は、上記アルコール
溶液中の香気成分含有量、或いは該溶液の香気の
強さによつて適宜選択することができるが、通常
は、該容液量の約0.001〜約5倍容量が好ましく、
該樹脂処理することによつて脱臭された処理液を
得ることができる。 かかる固体吸着剤処理及び/又は陽イオン交換
樹脂処理によつて得られた脱色及び/又は脱臭さ
れたローズマリー及び/又はセージの抗酸化成分
含有アルコール溶液は、前記した方法と同様に、
アルコール濃度約80重量%以下のアルカリ性含水
アルコール溶液となし、該溶液を多孔性重合樹脂
で処理して得られた処理液を酸性にして形成され
る固形分を採取することによつて無色、無味、無
臭の天然源抗酸化成分を得ることができる。 本発明によつて得られた抗酸化性物質は、例え
ば、ラード、ヘツド、チキンオイル、タロー、乳
脂、魚油、肝油、などの動物性油脂、サフラワー
油、大豆油、コーン油、菜種油、ごま油、やし
油、綿実油、ひまわり油、落花生油、パーム油、
パーム核油、コーヒー油、米油、などの植物性油
脂、バター、チーズ、マーガリン、シヨートニン
グ、マヨネーズ、ドレツシングなどの油脂性食
品、ハム、ソーセージなどの畜肉加工食品、水産
加工食品、揚麺類、揚菓子類、珍味、焼肉たれ、
乾燥野菜類などの食品類、乳飲料、清涼飲料、酒
類などの飲料類などの如き飲食品、更には錠剤、
液状経口薬、粉末状経口薬、湿布薬などの如き保
健医薬品、石けん、洗剤、シヤンプーなどの如き
香粧品、例えば、レモン油、ライム油、グレープ
油、オレンジ油などの如き天然精油類添加剤など
広汎な分野に利用することができる。 以下実施例により本発明方法実施の数態様につ
いて更に詳しく例示する。 実施例 1 セージ粉末100gに95%エタノール500gを加え
て、約75℃で2時間撹拌した。次いで遠心分野に
よつて不溶性固形物を分離、除去して抗酸化成分
含有アルコール溶液を得た。次いでこの溶液に10
%水酸化ナトリウム水溶液液10gを室温にて徐々
に添加してアルカリ性とし、次いで水250gを加
えてアルコール濃度約65%に調整し、緑色色素を
沈澱させた。次いで、沈澱物を過して除き、脱
色された抗酸化成分含有アルカリ性含水アルコー
ル溶液701gと、粗製緑色色素1gを得た。 次いで、あらかじめ約65%の含水アルコールに
浸漬した多孔性重合樹脂(商品名ダイヤイオン
HP−20)100mlを充填したガラスカラム(直径
4cm、高さ20cm)の上部から、上記アルカリ性含
水アルコール溶液を滴下して、多孔性重合樹脂処
理液780gを得た。次いで、上記処理液を撹拌し
ながら、10%塩酸9gを徐々に添加してPH約3.5
とし室温で30分間撹拌して、不溶性の抗酸化成分
を析出させた。次いでこの不溶性固形物を紙
過し、水洗、減圧乾燥して、無色、無味、無臭の
抗酸化性成分6.1gを得た(本発明品No.1)。 実施例 2 ローズマリー粉末100gにn−ヘキサン1000g
を加えて約60℃で3時間撹拌した。次いで遠心分
離によつて不溶性固形物を分離、除去後得られた
ヘキサン溶液を常圧下約70℃で蒸留して溶媒を除
去しローズマリーオレオレジン5gを得た。次い
でこれに95%エタノール200gを加えて充分撹拌
混合し、不溶物を紙過してローズマリーオレ
オレジンのアルコール溶液195gを得た。次いで
このアルコール溶液に活性炭2gを添加して、室
温にて約30分間撹拌後紙過し、得られた活性
炭処理液をあらかじめ95%エタノールに浸漬した
強酸性陽イオン交換樹脂(商品名ダイヤイオン
SK−116)50mlを充填したガラスカラム(直径3
cm、高さ15cm)の上部から滴下してイオン交換樹
脂処理液215gを得た。 次いで、この処理液に10%水酸化ナトリウム水
溶液5gと水200gを添加しアルコール濃度約50
%のアルカリ性含水アルコール溶液とし、次いで
この溶液を不溶物を除去後実施例1と同じ多孔性
重合樹脂カラムを通して吸着剤処理液465gを得
た。この処理液に10%塩酸を約4g添加してPH約
4.0として抗酸化成分を沈澱させ、その沈澱物を
約85℃の熱水で2回洗滌し、次いで乾燥して無
色、無味、無臭の天然源抗酸化成分3.2gを得た。
(本発明品No.2) 実施例 3 ローズマリー80g及びセージ120gの混合粉末

【特許請求の範囲】 1 ローズマリー及び/又はセージの抗酸化成分
含有アルコール溶液を、アルコール濃度約80重量
%以下のアルカリ性含水アルコール溶液となし、
析出した色素を分離回収し、得られた溶液を多孔
性スチレン系樹脂吸着剤で処理し、次いで得られ
た処理液を酸性にして形成される固形分を採取す
ることを特徴とする天然源抗酸化成分の製造方
法。 2 アルカリ性含水アルコール溶液となす前に、
該抗酸化成分含有アルコール溶液を、固体吸着剤
処理及び/又は陽イオン交換樹脂処理することを
