Description
本発明は、新規抗生物質AM―5344―A1およ
びその製造方法に関するものである。
本発明者らは、放線菌の生産する抗生物質の探
索の過程で、土壌分離の一放線菌AM―5344―
M81株が抗生物質を生産することを見い出し、本
菌株の菌学的性質を調べ、また、この抗生物質を
単離した後、その理化学的および生物学的性質を
調べべることによつて、本発明を完成した。
本発明によつて得られる抗生物質M―5344―
A1は、次に示すような理化学的性質を有してい
る。
(1) 元素分析値{炭素、水素、窒素、酸素から成
り、イオウ、ハロゲンを含まない。本物質の元
素分析値は下に示すとおりである。
C64.51%、H4.19%、N2.46%
(2) 分子量:マススペクトル(EIマススペクト
ルおよびFDマススペクトル)分析において、
質量数529に大きなピークが観察された。
(3) 融点:明確な融点は示さない。240℃付近か
ら茶色に変化し、275℃で黒褐色に変色する。
(4) 比旋光度:
〔α〕23 D=−92゜(C0.05、クロロホルム)
(5) 紫外線吸収スペクトル:
クロロホルム中での吸収極大(E1%
1cm)は、
303nm(516)、376nm(219)、385nm(214)を示
す。

(7) 溶剤に対する溶解性:
クロロホルムに溶け、アセトン、酢酸エチ
ル、ベンゼン等の有機溶媒および低級アルコー
ルに難溶で、n―ヘキサン、エチルエーテル、
水に不溶である。
(8) 呈色反応:
塩化第二鉄反応に陽性であり、ドラーゲンド
ルフ反応、ニンヒドリン反応には陰性である。
(9) 塩基性、中性、酸性の区分:
中性物質である。
(10) 物質の色:
黄色
(11) Rf値:
シリカゲル薄層クロマトグラフイー(メルク
社製TLCアルミシート、シリカゲル60F254、厚
さ0.2mm)を通常法にて行なつた時のRf値は次

抗生物質AM―5344―A1の生物学的性質につ
いて述べると次のごとくである。
(1) 抗菌性

(2) 抗トリコモナス活性
液体培地希釈法による最小阻止濃度(MIC)
は、次に示すとおりである。なお、液体培地に
はTrichosel broth(BBL社製)を用いた。

以上のように、本抗生物質はグラム陽性菌、嫌
気性菌およびトリコモナスに対して抗菌、抗原虫
活性を示すことから、抗生物質AM―5344―A1
は抗菌剤としての用途が考えられ、特に嫌気性菌
感染症の予防および治療に有効と思われる。また
本物質は、その抗菌スペクトルから動物薬として
の使用も十分期待される。
上記の理化学的性質および生物学的性質を既知
抗生物質の性質と比較した結果、抗生物質AM―
5344―A1は新規物質であることがわかつた。す
なわち、中性、脂溶性で紫外線吸収スペクトルが
303nm、376nm、385nm付近に吸収極大を有し、
かつグラム陽性菌に対して有効な類似の抗生物質
として、たとえばアルボフアンジン
〔Albofungin,Antibiotiki4,10―13(1959)〕、
クロラルボフアンジン〔Chloralbofungin,
Dokl.Akad,Nauk,SSSR207,1347―1350
(1972)〕があげれる。しかし、アルボフアンジン
は、元素分析値よりの窒素含量(5.3%)および
比旋光度(−670゜)が、抗生物質AM―5344―A1
の元素分析値からの窒素含量(2.46%)と比較し
て大きく異なることから明らかに異なる。クロラ
ルボフアアジンは、元素分析値より塩素を含み、
抗生物質M―5344―A1は塩素を含まないことか
ら明らかに異なる。以上のとおり、抗生物質AM
―5344―A1は既知抗生物質と明らかに区別され
るので新規である。
本発明の抗生物質AM―5344―A1を生産する
ために使用される微生物の実用的な例は本発明者
らによつて千葉県千葉市幸町の土壌から新たに分
離されたストレプトミセス・エスピー・AM―
5344―M81があげられる。
(1) 形態学的性質
栄養菌糸は、天然培地、合成培地の両方でよ
く発達し、通常は隔壁を有しない。気菌糸は、
グリセロース・アスパラギン寒天、チロシン寒
天等で中程度に着生するが、その他の培地では
着性が貧弱かあるいは着生しない。気菌糸の色
調は黄色ないし灰色で、粉状またはベルベツト
状を呈する。
顕微鏡下の観察では胞子柄は直線状ないしは
ループ状を呈し、グリセロール・アスパラギン
寒天、チロシン寒天では胞子の着生が見られ
ず、スターチ無機塩寒天で、10〜50個の胞子の
連鎖が観察される。胞子の表面は平滑である。
菌核、胞子嚢および遊走子は見出されない。
(2) 各種培地上での性状
イー・ビー・シヤーリングら(lnt.J.Syst.
Bacteriol、16巻,313頁,1966)の方法にした
がい、それに公知の培地および実験方法を併せ
て使用した。その結果を第2表に示す。色調は
標準色として、カラー・ハーモニー・マニユア
ル第4版(コンテナー・コーポレーシヨン・オ
ブ・アメリカ・シカゴ,1958年)を用いて決定
し、色票名とともに括弧内にそのコードを併せ
て記した。以下は特記しない限り、27℃、2週
間目の各培地における観察である。

(4) 細胞壁組成
デイアミノピメリン酸はLL―型であり、ア
ラビノース、ガラクトースは認められない。
以上、本菌の菌学的性状を要約すると次のとお
りになる。細胞壁組成は、LL―デイアミノピメ
リン酸を有する。また、形態的には直線状または
ループ状の胞子柄を形成し、胞子の表面は平滑で
ある。培養上の諸性質としては、栄養菌糸は黄色
から暗褐色を呈し、PHの変化によつて変化せず、
気菌糸の色調は黄色ないし灰色を呈する。シユー
クロース・硝酸塩寒天、グルコース・アスパラギ
ン寒天等で黄色の可溶性色素を生産する。メラニ
ン色素は生産しない。
これらの結果から、本菌株はストレプトミセス
属に属する菌種であり、しかもプリムドハムとス
トレナーの分類(バージーズ・マニユアル・オ
ブ・デタミネーテイブ・バクテリオロジー第8
版、1974年,748〜829頁)によるイエローあるい
はグレイシリーズに属する菌種と考えられる。
なお、本菌株はストレプトミセス・エスピー・
AM―5344―M81(Streptomyces sp.AM―5344
―M81)として、工業技術院微生物工業技術研究
所に寄託されている。(微工研菌第6006号)。
本発明における使用菌として、ストレプトミセ
ス・エスピー・M―5344―M81株はその具体例で
あつて、この菌をたとえば紫外線、エツクス線、
放射線、化学薬剤等を用いる人工的変異手段で変
異して得られる変異株は、もちろんストレレプト
ミセスに属する微生物であつて、抗生物質AM―
5344―A1の生産能を有するものはすべて本発明
に用いることができる。
本発明において、抗生物質AM―5344―A1を
生産する微生物を培養する培地としては、炭素
源、窒素源、無機物等を含む微生物の培養に通常
用いられる培地が広く使用されうる。培地の炭素
源としては、同化可能な炭素化合物であればよ
く、たとえば、ブドウ糖、麦芽糖、乳糖、シヨ
糖、デンプン、デキストリン、グリセリン、糖蜜
などが使用される。また培地の窒素源としては、
利用可能な窒素化合物でればよく、たとえば、大
豆粉、コーンスチーブリカー、綿実粉、ペプト
ン、肉エキス、酵母エキス、乾燥酵母、カゼイン
加水分解物、アンモニウム塩、硝酸塩などが利用
される。その他に、培地の調製にあたり、リン酸
塩、マグネシウム、カリウム、カルシウム、ナト
リウム、鉄、マンガン、などの塩類が必要に応じ
て使用される。培養は通常好気的に行なわれ、通
気撹拌培養が好適である。培養温度は微生物が発
育し抗生物質AM―5344―A1を生産する範囲で
適宜変更でるが、特に好ましいのは25〜30℃であ
る。PHは6〜7が好ましい。培養時間は種々の条
件によつて異なるが、通常50〜100時間程度であ
つて、抗生物質AM―5344―A1が最高力価に達
する時間を見計つて適当な時間に培養を終了す
る。
このようにして得られたストレプトミセスに属
する抗生物質AM―5344―A1生産微生物の培養
物からの抗生物質AM―5344―A1の採取は、微
生物の培養物より抗生物質を分離精製する公知の
手段を適宜選択組合わせて行なうことができる。
培養物から抗生物質AM―5344―A1を分離精製
する手段の一例を示すと、次のとおりである。
すなわち、培養物を菌体と液に分別して、菌
体からはアセトンや酢酸エチルなどで抽出する
か、メタノールで菌体体処理した後、酢酸エチ
ル、クロロホルム等の有機溶媒で抽出する。液
からは酢酸エチル、ベンゼン等の水と分離し、抗
生物質AM―5344―A1を溶解せしめる有機溶媒
で抽出した後、脂溶性物質の精製において通常用
いられる公知の方法により、抗生物質AM―5344
―A1を回収する。たとえば、抽出液(酢酸エチ
ル層)を減圧下で濃縮し、生じた沈澱を分離し、
これをn―ヘキサンで洗浄した後、シリカゲルカ
ラムクロマトグラフイーにより、溶出溶媒にクロ
ロホルム、メタノールの混合溶媒系を用いて、抗
生物質AM―5344―A1を単離する。
なお、生産される抗生物質AM―5344―A1の
検出および定量は、シリカゲル薄層クロマトグラ
フイー(メルク社製シリカゲル60F254、厚さ0.2
mm、展開溶媒;クロロホルム:メタノール=40:
1、抗生物質AM―5344―A1のRf値0.39)および
アコレプラズマ・レイデルウイ(Acholeplasma
laidlawii)PG―8を用いた生物学的検定法によ
つた。
次に本発明の抗生物質AM―5344―A1の実施
例を示すが、この実施例は単なる一例を示すもの
であつて、本発明を限定するものではない。
実施例 1
ストレプトミセ・エスピー・AM―5344―M81
株(Streptomyces sp.AM―5344―M81;微工研
寄託受理番号第6006号)の斜面培養から一白金耳
を種培地に接種し、27℃で2日間培養後、30の
培地を含む50容ジヤーフアーメンターに1%の
割合で種培養を接種し、27℃で89時間、通気撹拌
培養を行つた。培地は種培養には、グルコース
1.0%、スターチ2.0%、酵母エキス0.5%、ペプト
ン0.5%、炭酸カルシウム0.4%のものを、本培養
には、グリセリン2.0%、キナ粉2.0%、食塩0.3%
のものを、それぞれPH7.0に調整した後、121℃で
15分間滅菌したものを用いた。
菌体を含む培養物30に酢酸エチル10を加え
てよく撹拌した後、遠心分離により酢酸エチル層
を分離した。この酢酸チル抽出液を減圧下で500
mlまで濃縮し、n―ヘキサン300mlを加えて油性
物質を除き、褐色粉末3.0gを得た。これをシリカ
ゲルカラム(メルク社製キーゼルゲル60、120g)
にのせた後、クロロホルム1.5で展開し、次に
クロロホルム:メタノール(50:1)の混合溶媒
5.0で溶出して得られた活性分画(分画番号130
番〜550番、12ml/tube)を減圧下で濃縮乾固す
ることにより、褐色粉末500mgを得た。この褐色
粉末をクロロルム:メタノール(40:1)の混合
溶媒を用いた分取薄層クマトグラフイー(Rf値
0.39)により精製し、抗生物質AM―5344―A1の
橙色粉末25mgを得た。このものの性質は、前記し
た理化学的性質および生物学的性質と一致した。
実施例 2
ストレプトミセス・エスピー・AM―5344―
M81(Streptomyces sp.AM―5344―M81)の斜
面培養から一白金耳宛を種培地100mlを含む坂口
フラスコ25本に接種し、25℃2日間培養後、60
の本培養地を含む100容ジヤーフアーメンター
に5%の割合で種培養液を接種し、28℃で65時
間、通常撹拌培養を行つた。培地は種培養には、
グルコース1.0%、澱粉2.0%、酵母エキス0.5%、
ペプトン0.5%、CaCO30.4%のものを、本培養に
は、グリセリン2.0%、キナ粉2.0%、NaCl0.3%
のものを、それぞれPH7.0に調整した後、121℃で
30分間滅菌したものを用いた。
培養液54をドラムフイルターで過して得た
菌体に、メタノール30を加えてよく撹拌した
後、大型ブフナーロートで再び過し、得られた
菌体にクロロホルム22を加え、撹拌抽出した。
これを別して得られたクロロホルム層を減圧下
でアメ状になるまで濃縮し、これにn―ヘキサン
1を加え、生じた沈澱物を別し、乾燥するこ
とにより23gの粗物質を得た。
この粗物質10gをクロロホルムで充填したシリ
カゲルカラム(和光純薬工業社製ワコーゲルC―
200、400g)にのせ、クロロホルム2で展開
し、次にクロロホルム:メタノール(50:1)の
混合溶媒3で展開溶出して得られた活性分画No.
22―36,100ml/fractiontube)を減圧下で250ml
まで濃縮し、アセトニトリル20mlを加え冷却し、
得られた沈澱を別、乾燥することにより抗生物
質AM―5344―A1の橙黄色粉末2.5gを得た。この
ものの性質は、前記した理化学的性質および生物

AM―5344―M81 (Streptomyces sp.AM―5344
-M81) has been deposited at the Institute of Microbial Technology, Agency of Industrial Science and Technology. (Feikokenboku No. 6006). Streptomyces sp. strain M-5344-M81 is a specific example of the bacteria used in the present invention.

An example of a means for separating and purifying the antibiotic AM-5344-A 1 from a culture is as follows. That is, the culture is separated into bacterial cells and liquid, and the bacterial cells are extracted with acetone, ethyl acetate, etc., or the bacterial cells are treated with methanol and then extracted with an organic solvent such as ethyl acetate, chloroform, etc. The solution is separated from water such as ethyl acetate and benzene, extracted with an organic solvent that dissolves the antibiotic AM-5344-A 1 , and then extracted using a known method commonly used in the purification of fat-soluble substances. 5344
