【発明の詳細な説明】[Detailed description of the invention]

〔〕 発明の背景 本発明は、L―システイン(以下L―Cysと略
称する。)を新規酵素であるグルタチオン・スル
フイドリル・オキシダーゼ(以下GSOと略称す
る。)を用いて定量する方法に関するものである。 従来、L―Cysの定量法としては酸性ニンヒド
リン法または5,5′―ジチオビス―(2―ニトロ
安息香酸)、o―フタルアルデヒドなどのSH(チ
オール)基と反応する試薬を用いる化学的方法な
どが知られているが、特異性、簡便性、感度ある
いは夾雑物質の影響などの点で必ずしも満足すべ
き方法は確立されていなかつた。 本発明者は、先にグルタチオン(以下GSHと
略称する。)に対して特異性の高い新規なスルフ
イドリル・オキシダーゼ、すなわちGSOが微生
物によつて生産されることを発見し、本酵素の酵
素学的性質を明らかにした(特願昭56―16993号
参照)。 本発明者は、さらに本酵素をL―Cysの定量に
応用すべく研究を重ねた結果、GSHなどの他の
SH化合物が共存する試料であつても試料中のL
―CysをGSOのPHによる特異性の変化を利用する
ことによつて特異的にかつ高感度に、しかも簡便
に定量できることを知見し、本発明を完成するに
至つた。 〔〕 発明の概要 要 旨 本発明は、試料中のL―Cysを定量するに際
し、酸素存在下、GSOを試料に作用させ、反応
にともなう酸素消費量または反応物生成量を測定
することを特徴とするL―Cysの定量法を提供す
るものである。特にGSHを含む試料中のL―Cys
を定量するに際し、GSOによる酸素反応を、PH
7.0〜8.0およびPH4.5〜5.5において行い、PH7.0〜
8.0における反応液中の酸素消費量Aまたは反応
物生成量A′と、PH4.5〜5.5における反応液中の酸
素消費量Bまたは反応物生成量B′とを測定し、
それらの差(A−BまたはA′−B′)からL―Cys
含量を求める方法を提供するものである。 効 果 本発明において使用するGSOはPH4.5〜5.5にお
いてはGSHに対する特異性が高く、至適酵素量
を用いれば試料中に存在するSH化合物のうち、
GSHだけをほぼ特異的に酸化するが、PH7.0〜8.0
においては充分量の酵素が存在する場合、GSH
とともにL―Cysをも完全に酸化することができ
る。したがつて生体試料などのように、存在する
SH化合物のほとんどがGSHとL―Cysによつて
占められている試料の場合は、PH7.0〜8.0におけ
る酸素消費量Aまたは反応物生成量A′からPH4.5
〜5.5における酸素消費量Bまたは反応物生成量
B′を差し引くことによつて試料中のL―Cysの量
だけを簡単かつ正確に定量することができる。な
お、試料中にL―Cys以外のSH化合物、特に
GSHが実質的に存在しない場合には、PH7.0〜8.0
における測定だけでL―Cysを定量できることは
言うまでもない。 〔〕 発明の具体的説明 1) グルタチオン・スルフイドリル・オキシダ
ーゼの酵素化学的および理化学的性質 本発明におけるGSOの精製酵素標品の酵素

る。 (1) 作用 GSOは公知のラツト精嚢分泌物からのス
ルフイドリル・オキシターゼ(バイオケミス

てSH化合物を酸化的に2分子縮合して対応
するジスルフイド化合物を生成するが、特に
GSHに強い親和性と高い基質特異性を示し、
かつ蛋白質中のSH基に対しては微弱な活性
しか示さないという特徴を有する新規なスル
フイドリル・オキシターゼである。 GSOは、GSHを基質とした場合、下記反
応式のごとく、GSH2molにつき、1molの酸
素を要求し、1molの酸化型グルタチオンと
1molの過酸化水素とを生成する。 また、L―Cysを基質としたときも、GSH
の場合と同様にL―Cysの2molにつき、
1molの酸素を要求し、1molのシスチンと
1molの過酸化水素とを生成する。 2RSH+O2→RSSR+H2O2 (式中、RSHはGSHまたはL―Cysを、

ゼA(RNase A;分子量13700)のSH基に
GSOを作用させたところ、GSOを充分量
(5U/ml)添加した場合においてさえ、
RNase A1分子当りのSH基(8個)の減少
速度は4時間に1.5個程度であり、自然酸化
によるSH基の減少速度(1.0個/4時間)を
若干上まわる程度であつた。これに対して、
公知のラツト精嚢分泌物のスルフイドリル・
オキシダーゼは、0.5U/ml添加した場合、
4時間に7.5個/mol以上の減少を示し(前
出Biochemistry 19,2639(1980)のFig7参
照)、RNase Aに対し顕著な活性を示す酵
素である。 すなわち、本発明のGSOは酸性および中
性側でGSHに強い親和性を示し、特異的に
作用する新規なフラボプロテイン・スルフイ
ドリル・オキシダーゼである。 (3) 力価の測定 GSOの力価の測定は、酸素電極法で行つ
た。すなわち、10mMのGSHを含む0.1Mり
ん酸カリウム緩衝液(PH7.4)1mlを酸素電
極セルに入れ、10μの酸素液を添加して酸
素消費速度を測定した。30℃で1分間に
1μmolの酸素を消費する酵素量を1単位
(Unit;本明細書において、Uと略称する。)
とした。 (4) 至適PH 至適PHはPH6.5〜7.7付近、特にPH7.1〜7.8
付近である。 なお、至適PHの測定は、0.15M酢酸ナトリ
ウム―塩酸緩衝液、0.15モル酢酸ナトリウム
―酢酸緩衝液、0.15Mりん酸カリウム緩衝
液、0.15Mトリス―塩酸緩衝液および0.15M
グリシン―塩化ナトリウム―水酸化ナトリウ
ム緩衝液の各緩衝液を使用し、PH2.4〜10.8
の範囲で行つた。 (5) PH安定性および熱安定性 PH安定性は、前記各緩衝液中、PH3.0〜
10.8において、45℃、15分間保持した後、酵
素活性を測定したところ、PH5〜9の範囲で
安定であつた。 また、熱安定性は、0.1Mりん酸カリウム
緩衝液(PH7.4)中、40〜80℃において15分
間保持し、酵素活性を測定した。結果は、55
℃までは完全に活性を維持していたが、80℃
では完全に失活した。 (6) 作用適温の範囲 GSOの酵素活性の測定に使用する酸素電
極法においては、温度によつて反応液の溶存
酸素量が異るので正確な範囲は特定できない
が、過酸化水素の生成を公知の方法で測定し
たところ、およそ30〜50℃の範囲が好適であ
つた。 (7) 阻害、活性化および安定化 シアン化カリウム、塩化カルシウム、塩化
カリウム、塩化ニツケル、塩化アルミニウ
ム、塩化ストロンチウム、塩化バリウム、塩
化第二鉄、硫酸マグネシウム、硫酸亜鉛、硫
酸マンガン硫酸銅、硫酸リチウム、ヨウ化カ
リウムなど種々の金属塩およびエチレンジア
ミン四酢酸(EDTA)などの阻害剤1mMを
含む0.1Mりん酸カリウム緩衝液(PH7.4)中
で酵素反応を行い、各物質による阻害の有無
を検討した。その結果、GSOの活性は硫酸
亜鉛、すなわち亜鉛イオンによつて完全に阻
害され、硫酸マンガンによつて20%程度阻害
されたが、他の阻害剤にはほとんど阻害され
なかつた。 (8) 紫外線吸収スペクトル λnax272nm、365nm、443nm E280nm 1%14.78 (9) 補酵素 GSOを熱処理またはトリクロロ酢酸
(TCA)処理し、遠沈して得られた上清は、

示した。 (11) 等電点 アンフオライン(スウエーデン国、LKB
社製)を用いた等電点電気泳動法により測定
したところ、pIは4.21であつた。 (12) 分子量 GSOの分子量は、セフアデツクスG―200
(フアルマシア・フアインケミカルズ社製)
によるゲル濾過法では94000±5000と測定さ
れた。また、SDS―ポリアクリルアミドゲル
電気泳動法により、GSOは分子量47000±
3000のサブユニツト2個から構成される二量
体酵素であることが示された。 (13) 結晶構造および元素分析 GSOは結晶化されていないので測定して
いない。 (14) 精製方法 GSOの精製は塩析法、等電点沈澱法、有
機溶媒による沈澱法、けいそう土、活性炭な
どによる吸着法、各種クロマトグラフ法等を
適宜に組合せて行うことができる。精製方法
の具体例は参考例に示すとおりである。 2) GSOの製造 本発明において使用するGSOは、前記の性
質を有するものであればその起源、由来は問わ
ず、その製法によつても限定されない。製造法
の一例として微生物によるGSOの製造法を参
考例として、具体的に示す。 参考例 300ml容三角フラスコにフスマ8g、水5mlお
よびもみがら1gを入れ、120℃、30分間加圧滅
菌して調製した種用フスマ培地にペニシリウム・
アクレアタム IFO 5729を植菌し、28℃、7日
間培養して種菌を調製した。 5容三角フラスコ20本にそれぞれフスマ200
gおよび水140mlを入れ、120℃、30分間加圧滅菌
した後、前記の種菌を無菌的に接種し、28℃、7
日間培養した。得られた培養物を30の水に1時
間浸漬した後、濾過し、さらにけいそう土を通過
させて粗酵素液約27を得た。この粗酵素液に硫
酸アンモニウムを65%まで加え、生成した不溶物
を遠沈除去した。上清液にさらに硫酸アンモニウ
ムを添加して95%飽和濃度とし、生成した沈澱を
遠沈採取して0.02Mりん酸緩衝液(PH7.4)1
に溶解し、同一緩衝液で一夜透析した。透析中に
生成した沈澱を遠沈除去し、上清液を同一緩衝液
で平衡化したDEAE(ジエチルアミノエチル)―
セルロースカラム(5×70cm)に通し、吸着した
酵素を食塩0.2Mを含む同一緩衝液を用いて溶出
した。溶出された活性区分を集め、透析濃縮後、
セフアデツクスG―100(フアルマシア・フアイン
ケミカルズ社製)カラム(3.5×100cm)を用いて
ゲル濾過を行い、活性区分を集めて濃縮後、
0.02Mりん酸カリウム緩衝液(PH7.4)で透析し
た。この透析内液を遠沈し、上清液を精密濾過し
た後、凍結乾燥してGSOの精製標品(収率41%、
比活性540U/mg蛋白)40mgを得た。 使用微生物 本発明のGSOの製造に使用される微生物は、
アスペルギルス(Aspergillus;以下、A.と略
称する。)属、ペニシリウム(Penicillium;以
下、P.と略称する。)属、フザリウム
(Fusarium;以下、F.と略称する。)属、トリ
コデルマ(Trichoderma;以下、T.と略称す
る。)属、パエシロミセス(Paecilomyces;
pae.と略称する。)属またはグリオクラデイウ
ム(Gliocladium;以下、G.と略称する。)属
に属し、GSO生産能を有する微生物である。
これらの属に属するGSO生産菌の具体例とし
てはアスペルギルス・アワモリ(A.awamori)
IAM 2299、アスペルギルス・バタタエ(A.
batatae)IAM 2098、アスペルギルス・イヌ
イ(A.inuii)IAM 2255、アスペルギルス・オ
クラセウス(A.ochraceus)IFO 4345、アス

はこれらの菌株に限定されるものではない。ま
た、これらGSO生産菌を通常の微生物突然変
異誘導法、たとえば紫外線、X線、γ線照射な
どの物理的処理、ニトロソグアニジンなどの薬
剤による化学的処理などの処理法によつて変異
させて得られたGSO高生産性突然変異株のい
ずれをも好適に使用できる。 GSOの調製 GSOの調製形態および精製度合は、本酵素
による酸化反応と、それに続く示標物質の検出
系の種類に応じて適宜に選択される。すなわち
本酵素による酸化反応ならびに示標物質の検出
系における諸反応および/または検出に対して
妨害的に作用する物質を含まないかぎり、
GSOの酵素剤中に夾雑物質が存在しても問題
にならない。また、採用される定量システムの
実施態様にしたがつて、可溶性酵素として、あ
るいは適宜な担体に吸着もしくは不溶化したも
のとして使用することができる。 3) L―Cysの定量 本発明におけるL―Cysの定量法は、試料中
のL―CysのGSOによる酸化反応、特にGSH
共存試料を対象とする場合にはPH4.5〜5.5およ
びPH7.0〜8.0の二点における特異的酸化反応
と、これにともなう酸素の消費量の測定もしく
は反応物の生成量の測定を行う示標物質の検出
系とからなる。 GSOによる酵素反応 L―Cysの定量におけるGSOの酵素反応条件
は次のとおりである。すなわち、反応PHは、
GSO安定PH範囲内であり、かつL―Cysに作用
するPH範囲であればよいが、特にGSH共存試
料中のL―Cysを測定する際にはPH4.5〜5.5お
よびPH7.0〜8.0のPH範囲でなければならず、両
PH範囲で反応を行うことによつてはじめて
GSOのPHによる基質特異性の変化を利用する
ことができる。上記の場合、参考例に示すペニ
シリウム属糸状菌のGSOは、特にPH5.0付近お
よびPH7.4付近で反応を行うことによりPHによ
る基質特異性の変化を好適に利用できる。な
お、GSHの共存する試料を対象とする場合、
基質に対する酵素の使用量は、PH7.0〜8.0にお
ける反応では、L―CysおよびGSHが完全に反
応する程度の過剰量であり、PH4.5〜5.5におけ
る反応では、GSHのみに作用する程度の量が
好ましい。また、酵素反応のPHを維持する目的
で、酵素反応媒質として各種の緩衝液を使用す
ることが好ましい。緩衝液としては、前記のPH
範囲を維持でき、かつ本酵素反応を阻害しない
ものであればよく、特に限定されない。たとえ
ば通常用いられるりん酸緩衝液、トリス―塩酸
緩衝液、酢酸緩衝液、くえん酸緩衝液などの緩
衝液が好適である。 反応温度は、GSOの安定温度範囲である限
り特に限定されない。 示標物質の検出 本発明における示標物質は、酸素および酵素
反応における反応物(反応生成物)である。
GSOの反応における反応物とは、過酸化水素
および基質のSH化合物に対応するジスルフイ
ドニ量体であるが、本発明の示標物質としては
過酸化水素が好適である。 L―Cysの定量は、酵素反応にともなう示標
物質の量の変化を測定することによつて行なわ
れる。具体的には、試料中のGSHの有無にか
かわらず適用できる方法として、PH7.0〜8.0に
おける酵素反応にともなう酸素の消費量Aまた
は反応物の一つ、たとえば過酸化水素の生成量
A′と、PH4.5〜5.5における酵素反応にともなう
酸素の消費量Bまたは前記と同じ反応物の一つ
の生成量B′を検出測定し、両測定値の差(A
−BまたはA′−B′)を求めることによりL―
Cysを定量する方法が挙げられる。もちろん、
明らかにGSHを実質的に含まない試料を対象
とする場合にはL―Cysが充分に反応するPH、
たとえばPH7.0〜8.0において酵素反応を行い、
酸素消費量または反応物生成量を測定すること
によりL―Cysを簡単に定量することができ
る。 示標物質の検出系の選択は任意であり、これ
らの物質の測定に通常採用される方法を適用で
きる。また、示標物質の検出のための具体的方
法にも特に限定されない。 以下、それぞれの示標物質の検出測定法につ
いてその代表的な方法を例示するが、これらの
示標物質の検出系は公知方法に限定されず、今
後さらに開発されるべき種々の方法をも採用可
能と考えられる。 酸素の消費量を測定する方法としては、ワー
ルブルグの検圧法、酸素電極法が知られている
(生化学実験講座5 酵素研究法(上) 第35
〜52頁、東京化学同人、1975年8月20日発行)。
さらに、酸素電極の先端部にGSOを固定化し
た酵素電極法も採用できる。 過酸化水素を検出測定する方法には、大別し
て分光学的測定法、けい光測定法、電気化学的
測定法の三種の方法がある。 過酸化水素の分光学的測定法としては、過酸

なお指示薬としては、前記のほかに定量条件に
おいて定量的に酸化されて色調変化を示しうる
ものである限り、いずれも使用可能である。他
の過酸化水素の分光学的測定法には、メタノー
ルとカタラーゼの存在下で過酸化水素からホル
ムアルデヒドを生成させ、これを酸化剤(たと
えばカリウムシアノフエラート、塩化第二鉄な
ど)の存在下でヒドラゾン(たとえば3―メチ
ル―2―(スルホニル)―ベンゾチアゾロンヒ
ドラゾンなど)と反応させる方法がある。さら
に、カタラーゼ―アセチルアセトン法、アルデ
ヒドデヒドロゲナーゼを用いるアルコールの酸
化による方法、銅イオン―ヒスタミン系でイン
ジゴカルミンを酸化脱色し、その色度の減量か
ら定量する方法などが知られている。 過酸化水素の電気化学的測定法としては、白
金電極を用いるポーラログラフ法などが知られ
ている。また、カタラーゼを共用した酵素電極
を用いて酸素生成にともなう酸素レベルの上昇
幅および速度を測定し、過酸化水素を定量する
方法もある。このほかヨウ素アニオンをモリブ
デン酸塩の触媒存在下で過酸化水素によりヨウ
素に酸化し、その生成速度を電位差計もしくは
電流計により検出して定量する方法などもあ
る。 過酸化水素のけい光法による定量法として、
ホモバニリン酸を過酸化水素とパーオキシダー
ゼにより2,2′―ジヒドロキシ―3,3′―ジメ
トキシ―ジフエニル―5,5′―ジアセチル酸の
けい光体に変換して測定する方法が知られてい
る。ホモバニリン酸のほかに過酸化水素とパー
オキシダーゼによりけい光物質に変換されるも
のとしてはp―ハイドロキシフエニル酢酸、ジ
アセチル―2′,7′―ジクロロフルオレセインな
どが挙げられる。また6―メトキシ―7―ハイ
ドロキシ―1,2―ベンゾピロン、3,5―ジ
アセチル―1,4―ジハイドロルチジンなどの
けい光物質を過酸化水素―パーオキシダーゼに
より酸化して非けい光物質に変換し、そのけい
光の減少を測定する方法がある。 妨害物質の影響の除去 前記のような示標物質の検出系において、定
量目的物中の示標物質以外の物質が検出を妨害
する場合がある。このような場合には妨害物質
の影響を除去する必要がある。 たとえば、PH7.0〜8.0における酵素反応で
は、定量目的物中のGSHおよびL―Cys以外の
SH化合物あるいはPH4.5〜5.5における酵素反
応ではGSH以外のSH化合物は、GSOを作用さ
せた後もそのまま残存するが、SH化合物が存
在すると前記の発色試薬による発色が阻害さ
れ、L―Cys測定を妨害する。このような場合
SH基封鎖剤によつてSH化合物を封鎖する必要
がある。SH基封鎖剤としては、N―エチルマ
レイミド(以下、NEMと略称する。)が好ま
しい。 以下、本発明方法の実施の一例を示して具体
的な説明を加える。しかしながら、これらは単
なる例示であつて、本発明方法を何ら限定する

L―システインを定量するに際し、酸素存在下、
グルタチオン・スルフイドル・オキシダーゼを試
料に作用させ、反応にともなう酸素消費量または
反応物生成量を測定し、該測定値から試料中のL
―システイン含量を求めることを特徴とするL―
システインの定量法。 2 測定する反応物生成量として過酸化水素生成
量を用いる特許請求の範囲第1項記載のL―シス
テインの定量法。 3 グルタチオンを含有する試料中のL―システ
インを定量するに際し、酸素存在下、グルタチオ
ン・スルフイドリル・オキシダーゼによる酵素反
応を、PH7.0〜8.0およびPH4.5〜5.5において行い、
PH7.0〜8.0における反応液中の酸素消費量Aまた
は反応物生成量A′と、PH4.5〜5.5における反応液
中の酸素消費量Bまたは反応物生成量B′とを測
定し、それらの差(A−BまたはA′−B′)から
試料中のL―システイン含量を求めることを特徴
