Abstract

PURPOSE:To enhance oxygen permeability, transparency, mechanical strength and bio-compatibility by forming an oxygen permeable membrane containing crystalline substance and insoluble in water using a mixture of silk fibroin and silk sericin. CONSTITUTION:A membrane composed of a mixture of silk fibroin and silk sericin is immersed in a solution mixture consisting of a solvent and water to form an oxygen permeable membrane containing crystalline substance and insoluble in water. In this case, the oxygen permeability of the membrane can be controlled by changing the composition, temp. and treatment time of the solution mixture and the drying temp. of the membrane is 3 deg.C or more, pref., 5-80 deg.C. When the oxygen permeable membrane is formed by crystallizing the silk fibroin membrane, a degree of crystallization is set to 10% or more, pref., to 15-50%. The obtained oxygen permeable membrane has excellent oxygen permeability and is also well in transparency, mechanical strength and bio- compatibility.

Description

【発明の詳細な説明】 [技術分野] 本発明は絹タンパク質からなる酸素透過膜に関するもの
である. [従来技術] 近年各種高分子膜が開発され、そのガス透過性、水蒸気
透過性、溶質透過性等を利用して各種分野に応用されて
いる.それらの高分子膜のうち、酸素透過性を有するも
のは,コンタクトレンズや、人工角膜,人工皮膚等に応
用されている.従来一般に知られている酸素透過膜は、
シリコーン系高分子やポリアクリル酸系高分子からなる
ものであるが、前者の場合、薄膜の加工が困難で、また
汚染されやすい等の問題があり、また後者の場合、酸素
透過性に問題があり,未だ満足し得るものではなく、新
しい素材からなる酸素透過膜の開発が望まれている. [目 的] 本発明は、前記の如き問題を含まない新しい素材からな
る酸素透過膜を提供することを目的とする. [構 成コ 本発明によれば、家蚕あるいは野蚕由来の絹フィブロイ
ンと絹セリシンとの混合物から調製された膜からなり、
該膜は結晶質を含み、水不溶性であることを特徴とする
酸素透過膜が提供される.本発明の酸素透過膜は、絹フ
ィブロインと絹セリシンとの混合物からなる水溶液を製
膜し、絹フィブロインと絹セリシンとの混合物からなる
膜を得る.この膜は非結晶質で水溶性を示す.本発明で
は、この膜を結晶化させ、その膜中に結晶質を含有させ
、水不溶性のものとする.絹フィブロインと絹セリシン
とめ混合物からなる膜を不溶・結晶質化させるにはエタ
ノールやメタノール等の絹フィブロインと絹セリシンに
対する貧溶媒を用いて行うことができる. 原料として用いる絹フィブロインおよび絹セリシンは、
家蚕あるいは野蚕由来のものでよい.また絹フィブロイ
ン水溶液と絹セリシン水溶液は、熟蚕体内の絹糸腺より
取出した液状絹フィブロイン、絹セリシンを用いること
ができるし、精練生糸から再生した絹フィブロイン溶液
ならびに生糸の精練時に得られる絹セリシン溶液を用い
ることもできる.絹フィブロインと絹セリシンとの混合
割合の異なった膜を得るためには、絹ブイブロイン水溶
液と絹セリシン水溶液とを所定爪混合して溶液を調製し
、この溶液を平らなプラスチック板上に流延して水を蒸
発させる. 絹フィブロインと絹セリシンとの混合物からなる膜の結
晶化処理は、溶媒と水からなる混合溶液中で、絹ブイブ
ロインと絹セリシンの混合物からなる膜を浸漬処理した
後,乾燥することによって実施することができる。この
場合,混合溶液の組成、温度,処理時間を変えることに
よって、膜の酸素透過性を制御することができる。乾燥
温度は3℃以上、望ましくは5〜80゜Cである.本発
明において,絹フィブロイン膜を結晶化させて酸素透過
膜とする場合、その結晶化度は少なくとも10%、好ま
しくは15〜50%の範囲に規定するのがよい.本発明
における酸素透過膜は、酸素透過性を利用して使用され
る膜一般を意味するもので、このような酸素透過膜の具
体例としては、コンタクトレンズ、人工皮膚、人工角膜
、人工肺用膜、血液バッグ、溶存酸素取り込み膜(人工
えら)、細胞培養床材等が挙げられる. [効 果] 本発明の酸素透過膜は、すぐれた酸素透過性を示すと共
に、透明性及び機械的強度にすぐれ、生体適合性もよく
、かつ人体に対しても高い安全性を示す.本発明の酸素
透過膜の殊にすぐれた利点は、含水状態においても高い
酸素透過性と機械的強度を示すことである.このような
含水状態の膜は、生体遣合性(人体に対する親和性)に
すぐれ、その取り扱いが容易である.本発明の酸素?i
過膜においては、その含水率は、10〜70重量%、好
ましくは20〜60%に規定するのが好ましい.[実施
例] 次に本発明を実施例によりさらに詳細に説明する. 実施例1 桑葉で飼育した家蚕(日l37×支137)の熟蚕休内
より中部糸腺な取り出し、蒸留水で充分水洗した後、絹
