Abstract

PURPOSE:To produce a polypeptide having human tumor necrosis activity by integrating a specific DNA coding a human tumor necrosis factor(TNF) into a replicative expression vector and transforming a host microorganism with the vector. CONSTITUTION:The structure of the human TNF of formula has been determined by the following findings. (1) A human polypeptide gene and a rabbit TNF gene can be cloned by using a rabbit cDNA as a probe. (2) A DNA coding human polypeptide can be skillfully separated and its structure can be determined by comparing the homology of the base sequence with that of rabbit TNF gene, human polypeptide gene and rabbit cDNA. (3) The structure of pure DNA coding human polypeptide can be also skillfully determined and the DNA can be obtained in pure state. (4) A human polypeptide produced by using a DNA coding a human polypeptide has cytotoxic activity against L-cell, etc.

ポリペプチドであるので、1’ N Fが該TNFの出
来ではない動物に投与されるとTNFは抗原として働き
、抗体を産生せしめるからである。この理由から、1’
 N I?を人体に投与しようとするならば、ヒト出来
のT N Fの使用が非常に好ましい、しかしながら、

975年)参照〕によって解析する。 ウサギ1’NFとヒト’I’ N Fの遺伝子を含む制
限酵素分解された断片を、プラスミドベクター中に導入
し、サブクローンした後、塩基配列を解析する。 5、ウサギ”l’NFのcDNAとウサギTNF遺伝子
の塩基配列を比較して、ウサギTNF遺伝子のエクソン
(ウサギTNFのアミノ酸配列をコードする塩基配列)
と、イントロン(ウサギTNFのアミノ酸配列をコード
しない塩基配列)を決°定する。 6、そして、ウサギTNF遺伝子とヒトTNFの遺伝子
を比較して、ヒト”r N F遺伝子のエクソンとイン
トロンを決定する。 7、ウサギTNF遺伝子のイントロンを削除しエクソン
を結合することによって得られた塩基配列より決められ
るウサギTNFのアミノ酸配列は、ウサギ1”NFのc
DNへの塩基配列より決められるアミノ酸配列と一致す
ることが確認される。 8、次に、ヒトTNF遺伝子のイントロンを削除し、エ
クソンを結合することによって得られた塩基配列よりヒ
トTNFのアミノ酸配列が決められる。ヒト]’NFの
アミノ酸配列は、ウサギT N F。 のアミノ酸配列と部分的に一致することが#l認される
。 このようにして得られたヒト’r N Fをコードする
DNAから以下のようにして、ヒト’r N Fを製造
することができる。 1、ヒトI”NFをコードするDNAをin vttr
。 で修飾し、適当な発現ベクターに導入し、そのDNAを
含む組換DNAを作製する0組換DNAは適当な宿主に
導入し、これを培養液中で増殖せしめて所望のヒトTN
Fを発現せしめる。 2、このようにして得られるヒト’r N Fは成熟形
でセリンかも始まる155個のアミノ酸残基かもなる。 それがプレシーケンスにシグナルペプチドを有している
場合、シグナルペプチドは非常に疎水性の性質を有する
。 以上は、ヒトTNF遺伝子およびヒトTNFをコードす
るDNAの取得方法および該DNAを用いるヒトTNF
の製造方法を示したものである。 しかし上記の記載は本発明を限定するものではなく、本
発明の木質的な特徴及び基本概念を変えることなく当業
者によって明らかな変換を行なうことができる。 各アミノ酸に対応するコドン(遺伝暗号)の使用頻度が
異る等の理由により、アミノ酸配列を変えることなく、
ヒトTNFをコードするDNAの塩基配列の一部または
全部を、有機化学的に合成された人工のDNAに置換え
ることも可能である。 おそらく、ヒトTNFは、プレペプチド又はプレプロペ
プチドとして未成熟形で細胞内に産生され、プロセシン
グ段階でプロセスされて中間体を経て、成熟TNF’を
形成するものと考えられる。 ヒトI’ N Fの未成熟形はヒト’「NF遺伝子の塩
基配列から演鐸される。未成熟又は中間体のTNFをコ
ードするDNAからなるTNF  DNAは、また天然
又は人工合成のDNAと組換えることができる。 これらの方法の応用形態の1つは、メチオニンコドン(
ATG)を成熟あるいは未成熟あるいは中間体TNF遺
伝子の5′端に導入し、そして3′端に’rAΔもしく
は’r A GもしくはTGAの終止コドンと呼ばれる
トリプレットを、少なくとも1個導入することである。 メチオニンコドンが存在することにより、適当なプロモ
ータによって合成される+mRNAから成熟あるいは未
成熟あるいは中間体TNFが産生される。この様にTN
Fの末端に付加されたメチオニン残基は、宿主によって
は自然に除去される。終止コドンを挿入する目的は’I
’ N I=  D NAから転写されたmRNΔから
の翻訳を適当な位置〔式(1)のポリペプチドのC末端
]で止めることである。 また別の形態としては、゛′シグナル配列”と呼ばれる
疎水性に富んだ配列を付加することにより、宿主細胞の
外またはダラム陰性細菌においては。 ゛′ペリプラズム″と呼ばれる部分へ、分泌させること
も可能である。 また、開始コドンを組込んであるベクターの場合は、ベ
クターから由来するペプチドとTNFとの融合ペプチド
を形成するが、この場合は化学的または酵素的に切断す
るか、もしくはTNFの主たる活性に変化がなければ、
そのまま用いることができる。 ヒトi’ N F遺伝子を、正しく転写し、それによっ
て得られる+oflNAからの翻訳が正しく行われるよ
うな配列において、プロモーター等の5′領域の遺伝子
配列に接続し、得られたTNF  DNA−プロモータ
ー配列を細菌または高等生物細胞中で複製可能なベクタ
ーと接続した組換遺伝子を得、この組換遺伝子によって
宿主として細菌または高等生物細胞を形質転換し、この
形質転換体を増殖せしめ、i’ N F遺伝子を発現せ
しめることにより!’ N Fを得ることができる。 宿主として大腸菌[エシェリヒア・コリ(Escl+e
−ricl+ia coli) ] (以下゛′大腸菌

ター及びそのUV5変異、ペニシリナーゼ([3LA)
、l−リブトファン合成酵素(TRI))のプロモータ
ー、λファージのPLプロモーターあるいはトリプトフ
ァン合成酵素と、ラクトース分解酵素の融合プロモータ
ーであるLacプロモーター等が用いられる。 枯草菌[バチルス・サブチリス(3a(illus 5
ubtilis) ]  (以下゛′枯草菌パと記載す
る)を宿主とする場合には、BD170株(ATCC3

685頁〕に従ってSDSボリアクリルアミド電気泳動
(SDS r’AGE)で測定すると、約17000を
示す、尚、その際にマーカーとしては、ホスホリラーゼ
b(分子量94K)、ウシ血清アルブミン(分子ff1
67K)、オボアルブミン(分子量43K)、カルボニ
ックアンヒドラーゼ(分子量30K)、大豆トリプシン
インヒビター(分子量20、IK)とα−ラクトアルブ
ミン(分子ji14.4K)を用いる。 本発明のDNAから得られる新規生理活性ポリペプチド
は生体の正常組織は傷つけず、腫瘍の壊死を引きおこす
。本発明の活性ポリペプチドは公知の方法によって調剤
して悪性腫瘍細胞増殖のような細胞増殖を阻害するのに
有効な医薬組成物を調製することができる6本発明の活
性ポリペプチドは薬剤として使用可能な担体と混合する
こともできる0本発明の活性ポリペプチドの有効量を適
当な量の71体と混ぜて、患者に効果的に投与するのに
適した医薬組成物を調製することができる。 本発明のDNAから得られる生理活性ポリペプチドは、
抗腫瘍治療または抗ウイルス治療の必要な患者に注射剤
、点眼剤、点鼻剤、吸入剤、外用剤、経1コ投与用剤、
直腸投与用剤、膣内投与用剤などにして投与することが
できる。 本発明のポリペプチドの成人1日当りの治療量は、一般
に50単位〜too、 000.000単位であり、さ
らに好ましくは局所適用においては50〜500.00
0単位、静脈注射、筋肉注射などの全身注射においては
、1,000〜1,000,000単位、経口投与にお
いては、10.000〜too、 000.000単位
であり、用法あるいは症状に応じて適宜増減することが

Qの50%を殺す本発明の生理活性物質の量を意味する
。上述の量は次のようにして測定する。培養容器として
9G穴の組織培養用マイクロプレート(フロー・ラボト
リー社、米国)を用い、L−M細胞を1 v/v%のウ
シ胎児血清を含むイーグルのミニマム・エツセンシャル
培地(その組成は、たとえば、「組織培養」中外Qjl
之助池編集、朝食書店、1967年に記載されている)
中で培養する。順次培地で希釈した該生理活性ポリペプ
チド試料0.1mQとlO@個/ m Qの濃度のL−
M細胞の培地懸濁液0.1mAをプレートの各式の中で
混合し、マイクロプレートを5%の炭酸ガスを含む空気
中、37℃で48時間培養する。培養終了後、20%グ
ルタルアルデヒド水溶液20μCを加え細胞を固定する
。固定後、マイクロプレートを蒸溜水で洗浄、乾燥して
、0.05%メチレンブルー溶液をO,1mQ加え、生
き残った細胞を染色する。余分なメチレンブルーを洗い
流し乾燥した後、残ったメチレンブルーを0.36N塩
酸溶液で抽出し、その665nmにおける吸光度をタイ
ターチック・マルチスキャン(フロー・ラボラトリ−社
、米国)で測定する。この吸光度は、生き残った細胞数
に比例する。上記の本発明の生理活性ポリペプチドのl
×10°個のL−M細胞の50%を殺す量は希釈率と吸
光度をグラフ上にプロットすることによって求めること
ができる。 本発明のDNAから得られる生理活性ポリペプチドは、
非経口的に適用することができる。非経口投与用の調剤
にあたっては、アルブミン、ゼラチン、グロブリン、プ
ロタミン、プロタミン塩などの安定化剤、ショ糖、グリ
セリン、メチルセルロース、カルボキシメチルセルロー
スなどの増粘剤、各種無機塩のpl+調整剤などを添加
剤として加えることができる。また、本発明の生理活性
ポリペプチドは、錠剤の形で投与することもできる。 錠剤の調製の際に用いられる添加物の例としては、上述
の安定化剤に加えて、デンプン、乳塘なとの賦形剤を挙
げることができる。 具体的には、動物実験の結果、マウスの腫瘍も大半か−
、二回の注射投与で完治する。たとえば、マウスの皮膚
に人工の腫瘍(メスA細胞)を移植、直径6〜7ミリに
なった段階で本発明の新規生理活性ポリペプチドをわず
か0.6マイクログラム注射すると、−週間後にかさぶ
た状になり、二週間後には再び毛が生え始めて完治、そ
の後、妊娠、出産する。 以下に参考例および実施例によって本発明を詳細に記す
が、本発明はこれに限定されるものではない。 本発明の実施にあたり、組換DNAの作製、組換体の微
生物への導入は、特に断らない限り下記の実験書(1)

地g!濁液0.l mαを、96穴の組織培養用マイク
ロプレート(フロー・ラボラトリ社、米国)に加える。 培地はlv/v%のウシ胎児血清及び最終濃度5μg、
/mQのアクチノマイシンDを含むイーグルのミニマム
・エツセンシャル培地(その組成は、たとえば、「組織
培養」中耳1iff之助他編集、朝食書店、1967年
に記載されている)を用いる。マイクロプレートを5%
の炭酸ガスを含む空気中、37℃で21時間培養する。 培養終了後、20%グルタルアルデヒド水溶液20 I
t Qを加え、細胞を固定する。固定後、マイクロプレ
ートを洗浄、乾燥して、0.05%メチレンブルー溶液

加し、同緩征i液にてゲル濾過を行なう、活性画分は限
外濾過により濃縮し3.5×10@単位を回収する。比
活性は18XIO@単位/mgである・ 工程3(抗T N l?抗体) 血γ11iより得た’[’ N Fを工程2の如く部分
精製しフロイントの完全アジュバントをf、1で混合し
12週令のtli BAL[l/cマウスの背部皮下に
注射する。2週後、及び4週後にこの操作を繰返し、更
に1週後に全採血し、その血清を取得する。 この血清をL細・胞障害活性を測定する培地中に終濃度
500倍希釈となるように添加し、ウサギ血清から得た
TNFのし細胞障害活性を参考例1に示す方法で測定す
ると、L細胞障害活性は認められない、ここに得られる
マウス血清は、ウサギ血清’r N Fに対する抗体(

水を添加する。エンドトキシンを添加した培養上清にL
細胞障害活性が出現し7時間で最高値に達する。 この活性は抗T N F抗体で消去されるが、正常マウ
ス血清ではtl’l去されない。 一方、エンドトキシンを添加しない細胞培養上清にはL

上清に、分子量的17500の物質の生成が認められる

Sポリアクリルアミドゲル電気泳動に付した後、2.5
%NP40  (カルビオケム社、米国)で1時間、水
中で2時間様とう後、各泳動レーンを切断分離し、泳動
方向に直角に2IIIl中にスライスする。各スライス
断片をL細胞と共に培養することにより、し細胞障害活
性を調べる。エンドトキシン存在下に培養上清を展開し
たレーンの分子量的17500の位置にし細胞障害活性
が認められ、その池の位置には細胞障害活性は認められ
ない。 工程3(mflNAの取得) 工程lと同様な細胞培養においてエンドトキシンを添加
後2時間培養したのち、遠心分離にて細胞(ウサギ肺洗
浄細胞)を集める。細胞質RNΔおよびその中からのm
RNΔの抽出は下記の如くチャーブウィン(Cbirg
win)らの条件[チャーブウィン、ジェイ、エム、(
Chirgwin、 J、 M、)ら、バイオケミスト

94頁(1979年)〕に従って行なう。細胞3XIO
1個に対して4mfiの4Mグアニジンチオシアネート
溶液を加え、ホモジナイザー(AM−7、日本精機製作
所)にて破砕する。残査を遠心除去後、2.4gの塩化
セシウムを溶解し、あらかじめ2゜5mQの5.7M塩
化セシウム、0. I M EDTA溶液(pH7、5
)を入れであるポリアロマ−チューブへ静かに!■層す
る。 ベックマン3W410−ター(ベックマン社、米国)を
用いて20℃3oooorpmにて12時間、超遠心分
離を行った後、上清を除き、ペレットをlomMhリス
−塩酸縁tfli?IJL(5mM EDTA、 l 
w/V%SDSを含有する)1m+1にて溶解するにの
溶液を1mQ、のクロロホルムと1−ブタノールの混液
(容積比4・l)で抽出し、水層に0.05容積の2M
酢酸ナトリウムと、2.5容積のエタノールを加え、−
20℃で2時間以上放置してRNAを沈澱させる。遠心
にて沈澱を集め、乾燥させたのち滅菌水500μαに溶
解して、細胞質RNA溶液を得る。 上記細胞質RNArB液を68℃、2分間加熱後急冷し
、500μQの2倍濃度のl領1トリスEDTA緩tf
li液pH7、4(1mM EDTA、 0,1w/v
% SDS及び0.5M塩化リチウムを含む)を加え、
200mgのオリゴ(dT)−セルロース(ピーアール
エル(BRL)社、米国)カラムに展開、1倍濃度の上
記バッファー10mAで洗浄、溶出縁1#液(lO州ト
リス−塩酸pa17 、4.1 mM EDTA、0,
1w/v%sDsを含む)2mQで溶出する。溶出液に
0゜05容積の酢酸ナトリウム溶液、2.5容積のエタ
ノールを加えて一20℃に冷却して沈澱せしめる。沈澱
を遠心にて集め、再度同様にオリゴ(dT)セルロース
カラムに吸着する両分を集める。 紫外吸収スペクI・ル分析により、85ILgのmRN
Aを回収する。 工程4<mRNAのサイズ分画) 工程3と同様にしてlj)られる+5RNA880μg
を250μ0の水に溶解し、5−25%直線シヨ糖密度
勾配置 0mAに重層する。ショ糖密度勾配は、5およ
び25%のショ糖を各々含むトリス緩衝液(25mMト

回収する。各分画はエタノール沈澱し、遠心後滅菌水に
溶解する。 工程5(mRNAの翻訳実験) アフリカッメガエル卵母細胞によるmRNAの翻訳は、
実験8(例えば寺岡宏、五木幹男、田中兼太部、蛋白質
、核酸、酵素、臨時増刊、遺伝子操作、602頁198
1年)に依る。アフリカッメガエルは、浜松生物教材よ
り得る。工程4で得た分画mRNAをlμg/μCにな
るように滅菌水に溶解し、卵母細胞1個あたり、50n
fiずつを微量注入し、l m g / m Qのウシ
血清アルブミンを含有するパース(Barth)溶液(
7,5mMhリスー塩酸pl+ 7 、6.88吐塩化
ナトリウム、1mM塩化カリ、0.33taM硝酸カル
シウム、0.41mM塩化カルシウム、0.82mM硫
酸マグネシウム、2.411口M重炭酸ナトリウム、1
80/mQペニシリンG、■8μg、/mQストレプト
マイシンを含有する)中で24時間培養する。培養液の
まま卵母細胞をガラス棒でつぶし、遠心後、上清のし細
胞障害活性を測定する。沈降定数163 (」近におい
て、L細胞障害活性が最高値を与える。またこの活性は
参考例2工程3で得た抗TNF抗体により消去されるが
正常マウス血γNでは消去されない。 工程6 (形質転換体の取得) 工程4で得た分画nRNA  5μgを用い、実験′#
1(1) 96頁以降に従って二mInDNAを調製す
る。逆転写rIP素はライフ倶イエンス社(米国)のも
のを使用する。二重鎖DNAを3゜5%ゲル濃度のポリ
アクリルアミドゲル電気泳動にて分画し、長さ約100
0〜2000bpの画分330口gを得る。この両分7
ngを用い同上の実験群に従い、ターミナルデオキシヌ
クレオチジルトランスフェラゼ[ピーアールエル(B 
RL)社、米国]を用いてデオキシCAJ1をつなぎ、

ール後の混合物を用いて大腸α1K−12株(II[3

IO’単位の1’ N Fを使用し、蛋白として約18
0μg回収すこのうち150μ区を1%重炭酸アンモニ
ウム75μCに溶解、 TPCK トリプシン(ワーシ
ントン・バイオケミカル社、米国)3μgを添加し、3
7℃、4時間インキュベートする0反応液をコスモシル
5C8(牛丼化学)を担体とする高速液体クロマトグラ
フィーにより分画し、トリプシン消化断片を得る。 上記の如く高純度に精製したTNFおよびそのトリプシ
ン消化断片は、次に、セファデックスG25のカラムで

2種類の混合物として合成する。 各々を常法に従って脱保護し、セファデックスG−50
(ファルマシア社、スウェーチン)を用いるカラムグロ
マトグラフイー、7M尿素を含む20%ポリアクリルア
ミドゲル電気泳動及び、DE52(ワットマン社、米国
)カラムグロマトグラフイーにより精製し、O,ll1
1Mトリス−EDTA緩1重i液に対し透析する。 各々の精製オリゴDNAを常法によりT4ポリヌクレオ
チドキナーゼ(ベセスダリサーチラボラトリーズ社、米
国)およびγ−12−アデノシントリリン酸を用いて放
射性ラベルし、DE52カラム(ワットマン社、米国)

放射能が導入される。 各グループの混合物の形で得られたオリゴDNAプロー

℃60分間処理したのち、1.1重量%アガロースゲル
電気泳動により分画する0分画後のmRNAを、f!気
気泳動トランスファープロティング装置(バイオラッド
社、米国)を用いて、メーカーのマニュアルに従い、移
動せしめる9次いで、この膜上のmRNAと、5xSS
C:及び150μg/ m Qの変性サケ精子DNAを
含む5×デンハルト溶液で65℃、2時間処理したのち
、放射性標識したオリゴDNAをI X 10’cpm
/mQ、  5 X5SC溶液を含む5Xデンハルト溶
液で50℃、2時間処理する。次いでこの膜を6xss
cで室温、40℃、50℃、60℃で順次洗浄し、X線
フィルムXAR−5(イーストマン・コダック社、米国
)に対しn光せしめる。この結果、mRNAと最も強く
ハイブリダイズするオリゴDNAはMJであって、MJ
混合物中に+aRNAと完全に相補的な配列を有するオ

2頁の方法に従ってセルロースフィルター上に移し、そ
のDNAと、参考例3工程8で選択される放射性標識オ
リゴDNA(MJ)とを参考例3工程8と同様の条件で
ハイブリダイズせしめる(コロニー・ハイブリダイゼー
ション)0強くハイブリダイズする株49個を選び更に
フィルター上に固定して再度コロニーハイブリダイゼー
ションを実施し、標識オリゴDNΔ(プローブMJ)に
より強くハイブリダイズする9個を選ぶ。 この9個の株から、実験書(1)の6頁の迅速プラスミ
ド分離法に従って各々約5μgのプラスミドを取得する
。このプラスミドを制限酵素地図1、T!AI 、均翌
1、均世■(いずれも[3RL社、米国)を用い、メー
カーのマニュアルに従って切断し、1重量%アガロース
ゲル電気泳動で、各々の酵素による切断片の長さを比較
する。 この結果、9株すべてが、約50bpのI’vu II
とRsa Iによる断片を有し、9株のほとんどがRs
a Iによる約200bpの断片を有し、部分的に共通
の配列を有することが示唆される。第1図に制限酵素に
よる解析結果を図示する。 第1図に開示の内容を説明する。 即ち、発現が見られるのは、pB2−2とpR12の両
者についてであり、pB2−7.pR9゜pR17,p
R18及・びpR25についてはいずれも発現はみられ
ない。挿入遺伝子の発現にはβ−ラクタマーゼのプロモ
ーターが使用され、そして該発現の際に生じる蛋白は、
β−ラクタマーゼとの融合蛋白(fusion pro
teinlである。ところで、挿入遺伝子が正しく発現
されるためには、β−ラクタマーゼのプロモーターの転
写方向とが、同じでなければならない。故に、転写方向
が逆であるpR17,pR18及びpR25が発現しな
いのは当然である。また、β−ラクタマーゼのプロモー
ターの転写方向と挿入遺伝子の読みとり方向が同じでも
β−ラクタマーゼのフレームと、cDNAのコードする
蛋白のフレームが合わないと正しく発現されない。因に
pB2−7とpR9の場合は、β−ラクタマーゼのプロ
モーターの転写方向が同じであるにも拘らず発現がみら
れないことから、フレーム不一致によるものと判断され
る。尚、塩基配列からTNFのコーディング領域は、第
1図のPvuIIの上流135bp及び下流330bp
にあることがわかっている。従ってpB2−2とpR1
2の三者の場合は、β−ラクタマーゼのプロモーターの
転写方向が同じであり且つβ−ラクタマーゼのフレーム
と、cDNAのコードする蛋白のフレームが合い、更に
TNFのコーディング領域を含むことから発現がみられ
るものである。また第1図に示す内容から明らかなよう
に、クローニングのために用いたPst Iサイトを除
き、全てのクローンの制限酵素地図は一致しており、基
本的には全てのクローンは同じ蛋白をコードしている。 したがってTNFのコーディング領域を含むものであれ
ば発現構築のためにはいずれのクローンを用いてもよく
、本例ではpRt7を発現構築のために用いている。因
に、pR17のコーディング領域の塩基配列もpR12
およびpR18のそれと一致した。 また、このうち7株を10μg / m 12のテトラ
サイクリンを含む2mj2のLB培地中で吸光度が第2
表に示す値になるまで培養し、遠心にて集めた菌体を2
mβの生理食塩水で超音波により破砕し、遠心上溝のし
細胞障害活性を測定すると、第2表に示す如く、L細胞
障害活性を示す。対照実験としてプラスミドpBR32
2を含有する株を用いて同様の操作を繰返して行なう。 結果を第2表に示す。 (以下余白) 第2表 またこの活性は抗TNF抗体により消去され、正常マウ
ス血清では消去されない、従ってこの9株すべてがTN
F遺伝子を含むプラスミドを有していることが示される
。 工程10(ウサギTNFill伝子の塩基配列の決定) プラスミドp[+2−7、およびpR18を含有する大
am株を、IOμg/mQのテトラサイクリンを含有す
るM9培地〔実験#J(3)440頁)IQ中で培養し
た後、実験書(3)90頁の方法に従ってプラスミドを
単離し、各々約150μgを得る。 各々の塩基配列をマキサム−ギルバート(Maxa*−

にプラスミドの(a築を行なう、第2図に示す様にIO
μgのプラスミドpl?12をlOユニットの±1〔ピ
ーアールエル(I3RL)社〕で37℃で2時間tl’
l化し、4%のポリアクリルアミドゲル電気泳動で約6
30bp断片を単離する。約lILgの断片がゲルから
電気泳動溶出する。参考例31程8と同様の方法によっ
て第2図に示す2個デオキシオリゴヌクレオチド即ち、

を含む寒天培地で培養して約200個の白色コロニーを
得る。これらの形質転換体100個からプラスミドDN
Aを調製し、Ba+all+で消化したところ、15個
が目的の約670bpのBam1l I断片を含んでい
る。さらに、挿入の方向を調べるために、上記15個の
プラスミドをpUc−8上に1ケ所しか8μ部位がない
Ec。 R1とPvu II (約670bpの断片上に認識部
位がある)を用いて消化し、6重量%ポリアクリルアミ
ドゲル電気泳動を用いて調べることにより、7個のプラ
スミドから目的の約140bpの断片が確認され、pu
c−s上のlacプロモーターから順方向であることが
判明する。 塩基配列の解析により、この7個のプラスミドは同一で
、lacプロモーター、合成りNA及びcDNA間の結
合部に所望のヌクレオチド配列を有することが確認され
る。 プラスミドpH17を用いて、大1湯菌内で1acU 
V 5をプロモーターとしてTNFを直接発現させるこ
とを目的にプラスミドの構築を行なう、第3図に示す様
に10μgのプラスミドpR17をIOユニットのM副

化し、4%のポリアクリルアミドゲル電気泳動で約63

する。 プラスミドpOP95−15は、フラーの方法〔エフ、
フラー(1”、Fuller)、ジーン(Gene)、
19巻42頁〜54頁、1982年1に従って調製する
。 pOP95−15のIμgをEcoRIで消化してフェ
ノール抽出、クロロフォルム抽出、エタノール沈澱をし
てベクターを調製する。ベクター0.5μgは、上記の
如く合成デオキシオリゴヌクレオチドと、TNFをコー
ドするDNAを結合して得られる約670bpの断片に
、T、DNAリガーゼを用いて結合される。実験w(4
)、20頁に従って、大腸菌を形質転換してI nMI

5重量%アガロースゲル電気泳動を用いて調べると4個
のプラスミドから目的の約1280bp及び2600b
pの断片が確認され、1acUV5プロモーターから順
方向にTNF遺伝子が接続されていることが判明する。 塩基配列の解析により、この4個のプラスミドは同一で
、1acUV5プロモーター、合成デオキシオリゴヌク
レオチド、及びcDNAが正しく結合されていることが
確認される。得られるプラスミドをpTNF−1acU
V5−1と命名する。 工程12(大腸菌が産生ずる’rNFの精製)工程11
で得られたプラスミドを含有する大腸菌株をアンピシリ
ンを含有するLB培地に50mΩ中37℃で!夜培養し
、5Q、の同上の培地に移して更に3時間培養する。イ
ソプロピルβ−ローチオガラクトピラノシド(シグマ社
米国)を終濃度l−になる様に添加し更に、6時間培養
を続けたのち冷却し、遠心分離により菌株を集める。工
程11におけると同様に菌体を0.04Mトリス−塩酸
緩衝液(pH7,8) 5 Q、中で超音波破砕し、菌
体蛋白溶液を得る。 この溶液は5X10“単位/Qのし細胞障害活性を示す
。 この溶液を参考例2工程2と同様に精製し1゜2×10
°単位のTNFを得る。このものの比活性は6.8X1

ロニが得られる。 工程2(pr+2−7とpR18プラスミドDNAの調
製) 工程lで得られるプラスミドp[+2−7とpRI8の
形質転換体を下記の報文の方法に従って培養し、プラス
ミドを増幅させる6次いで得られる形質転換体を集菌し
、破砕したのち、プラスミドDNAを精製する。〔実験
11F(3)、88−96頁〕すなわち、30μg/m
Qのテトラサイクリンを含有するり、B培地に、それぞ
れの形質転換体を植菌し、激しく振とうしながら37℃
で培養する6次いで、この工程をくりかえして形質転換
体を増殖させ、更にプラスミドを増幅させる0次に、得
られる形質転換体の培養液を4℃に冷却しながら、40
00gで10分間遠心を行ない、上清を除去する。 氷冷STE [0,1M塩化ナトリウム、10+mMト
リス−塩酸緩衝液(pl+ 7 、8 )と1mM E
DTA )の100mAを用いて洗浄し、続いて101
1Mトリス−塩酸縁tljn (pH8,0)の中に2
0mg/mQ、のりゾチームを含む水溶液を用いて細胞
を煮沸溶菌する。得られる粘性液体を超遠心チューブに
移し入れ25,0OOrp+m 30分間4℃で遠心を
行なってDNA溶液を得る。 該DNA溶液の容量を測った後、該溶液1mQ当りに、
固体の塩化セシウムIgを加え、塩化セシウムが完全に
溶けるまで、ゆっくりと注意深く撹拌する。該塩化セシ
ウム水溶液のI OmQ毎に、10tmg/mQのエチ
ジウムブロマイド水溶fi0.8mQを加える。この結
果、該溶液の最終比重は1.55g/mQ、エチジウム
ブロマイドの最終濃度は約600μg/mαとなる。 該塩化セシウム水溶液を適当な遠心チューブに移し、空
隙に軽パラフィンオイルを加え、20℃で36時間、4
5.OOOrpmの遠心を続けると上層に鎖状の微生物
由来DNAとニックの入った環状プラスミドDNAと、
下層に閉環状プラスミドDNAがくる。下部のDNAの
バンドをチューブの横に注射針をさしこんで採取しガラ
スチューブに移す。エチジウムブロマイドを除去し、水
層をTΔE緩衝液に透析する。プラスミドDNA溶液を
I?Nアーゼ(RNase)処理し、等量の飽和フエノ
ルrEI液で抽出する。水層をあらかじめ0,1%SD
Sを含むTAE緩衝液(pl+ 8 、0 )で平衡化
したバイオゲル([tiogel) A −150に付
す、DNAを洗い込み、活性画分を取得する為に0.1
%SDSを含むTAE緩挿i液で溶出する6分画液をエ
タノールで沈殿させ、精製したプラスミドDNAを得る

る0次いで得られる混合液を3息のNZブロスに加え、
37℃、23時間撹拌培養する1次いで60mAのクロ

て採取する。得られるバクテリオファージをSMの28
mQに懸濁し、次いでクロロホルムを等量加える。ポル
テックスミキサーで30秒1?IJ混合した後、遠心し
て水層を集め、その全量をSMで30mQにする。これ
に26.4gの塩化セシウムを加え、静かに溶解した後
、超遠心(45000rp+*、20時間)でファージ
のバンドを採取する。10mM塩化ナトリウムと10m
M塩化マグネシウムを含む50nMトリス緩衝液(pH
8、0)に透析した後、それぞれの最終濃度が2011

3株をLI3培地中で、培養ブロスの550nmにおけ
る吸光度が0.3になるまで培養する。50mQの増殖
した大腸菌K12JM83株を集め、25m2の10m
MM0PS(pH7,0) −10mM RbC1で洗
い、25mαの0、1 M MOP 5(pH6、5)
 −50mM CaC1,−10mM RbC1中に懸
濁する。この懸濁液203μQに、10ngの上記結合
生成物を含むlOμαの水溶液を加える。この混合物を
水中にて30分間冷却し、40℃で60秒間加熱する。 その後すぐに、あらかじめ37℃にしておいたLI3ブ
ロス5m(Lに、加熱した混合物を加え、37℃で1時
間培養する。得られる培養ブロスを遠心し上清を除去す
る。遠沈した細胞にLB培地を加えて、301tg/

むL [3プレートに植菌する。インサートを含むプラ
スミドが導入された大腸菌Kl 2JM83株のコロニ
ーは白色であるが、プラスミドのみが導入された株のコ
ロニーは青色である。得られる白色コロニーは再び、3
0μg/mαのアンピシリンと40μg/mQ、のx−
gal を含むLBプレートに確認のため植菌する。 上で得られる白色コロニーより、10個のコロニー(バ
クチリアルクローン)を選び、迅速法を用いてスクリー
ニングする。 詳しくは、それぞれのコロニーを30μg/mQのアン

以外は工程2と同じ操作によって、1.89mgのp 
I−I CE  D N Aを得る。 工程10 (Ecol目分解flG−1のサブクローニ
ング) 工程6において得られる30μgのRG−1をEcoR
Iによりll!I化する。得られる各種断片の混合物よ
り、上記各種断片の混合物と0.8%の低融点アガロー
スゲルを用いる以外は工程9と実質的に同じ操作によっ
て、約3.5kbの長さを持つ断片を回収し、1.0μ
gのIEcoR1分解RG−1断片(3,5kb)を得
る。この断片とEcoRIで消化したりUCl3を、
