Claims (48)
- (1)ヌクレオチド/非ヌクレオチドポリマーの製造に適した非ヌクレオチド試 薬であって、 (a)化学部分および非不適条件下で活性化されることにより化学部分と連結し 得るリンキングアームから選択されるリガンドを有する、非ヌクレオチドモノマ ー単位、 (b)モノマー単位に連結された第1および第2非ヌクレオチド結合基(ただし 、前記第1結合基は非ヌクレオチドモノマー単位を第1追加モノマー単位に連結 させ得、第2結合基は不活化された状態であるため実質的には結合し得ないが、 その後非不適条件下で活性化されることにより、非ヌクレオチドモノマー単位を 第2追加モノマー単位に結合させ得、第1および第2追加モノマー単位の少なく とも一方はヌクレオチドモノマー単位である)を含む試薬。
- (2)リガンドが、標識、挿入剤、金属キレート剤、薬剤、ホルモン、蛋白質、 ペプチド、ハプテン、ラジカル生成剤、核酸分解剤、蛋白質分解剤、触媒、特異 結合物質、DNA輸送修飾剤、およびヌクレオチド多量体の溶解性を変える物質 、並びに前記化学部分のいずれかに連結され得る連結機能基から選択されるもの である、請求項1記載の試薬。
- (3)ヌクレオチド/非ヌクレオチドポリマーの製造に適した非ヌクレオチド試 薬であって、 (a)化学部分および非不適条件下で脱保護されることにより化学部分と連結し 得る保護リンキングアームから選択されるリガンドを有する、非ヌクレオチドモ ノマー単位、(b)モノマー単位に連結された、非ヌクレオチドの、第1結合基 および保護第2結合基(ただし、前記第1結合基は非ヌクレオチドモノマー単位 を第1追加モノマー単位に連結させ得、第2結合基は実質的には結合し得ないが 、その後非不適条件下で脱保護されることにより、非ヌクレオチドモノマー単位 を第2追加モノマー単位に結合させ得、第1および第2追加モノマー単位の少な くとも一方はヌクレオチドモノマー単位である) を含む試薬。
- (4)リガンドが、標識、挿入剤、金属キレート剤、薬剤、ホルモン、蛋白質、 ペプチド、ハプテン、ラジカル生成剤、核酸分解剤、蛋白質分解剤、触媒、特異 結合物質、DNA輸送修飾剤およびヌクレオチド多量体の溶解性を変える物質、 並びに前述の化学部分のいずれかに連結され得る連結機能基から選択される、請 求項3記載の試薬。
- (5)第1および第2結合基が、非ヌクレオチドモノマー単位をモノマー単位の 5′ヒドロキシおよび3′ホスフェートと各々結合させ得るものである、請求項 4記載の試薬。
- (6)非ヌクレオチドモノマー単位が非環状骨格を有し、その各末端に第1およ び第2結合基が連結されている、請求項5記載の試薬。
- (7)非ヌクレオチドモノマー単位が、その各末端に第1および第2結合基が連 結されている骨格を有し、前記骨格が1〜20個の原子鎖で構成されている、請 求項5記載の試薬。
- (8)非ヌクレオチドモノマー単位が、その各末端に結合基が連結されている骨 格を有し、その骨格が1〜10個の炭素原子から成る非環状炭化水素鎖である、 請求項5記載の試薬。
- (9)非ヌクレオチドモノマー単位が、各末端に結合基が連結されている骨格を 有し、連結アームが前記骨格から延びている1〜25個の原子鎖である、請求項 5記載の試薬。

- (19) (a)官能部分および官能部分が連結され得る保護リンキングアームから選択さ れるリガンドを有する非ヌクレオチドモノマー単位、および (b)非ヌクレオチドモノマー単位に連結された第1結合基および保護第2結合 基 を有する試薬から置換ヌクレオチドを製造する方法であって、非不適条件下、 (a)まず、第1結合基を介して非ヌクレオチドモノマー単位を第1追加モノマ ー単位に結合させ、 (b)次いで第2結合基を脱保護し、 (c)次いで第2結合基を介して非ヌクレオチドモノマー単位を第2追加モノマ ー単位に結合させる ことを含む(ただし、第1および第2モノマー単位の少なくとも一方がヌクレオ チドモノマー単位であり、他方がモノマー単位および固体支持体から選ばれる) 方法。
- (20)第1および第2追加モノマー単位の両方がヌクレオチドモノマー単位で ある、請求項18または19記載の方法。

- (24)リガンドが、標識、挿入剤、金属キレート剤、並びに標識、挿入剤およ び金属キレート剤のうちの一つと連結され得る連結官能基から選択されるもので ある、請求項23記載の方法。
- (25)X2=Clまたは第2アミノ、R3=クロロフェノキシ、メトキシ、エ トキシまたはベータ−シアノエトキシ、 X4=Cl、第2アミノまたはR3=Hの場合O、R5=メトキシ、エトキシ、 モノクロロフェノキシまたはべータ−シアノエトキシ、またはX4=O−の場合 のみHであり得る、請求項24記載の方法。
- (26)試薬の第2アミノ基が、ジアルキルアミノおよび複素環N−アミン類か ら選択されるものである、請求項24記載の方法。
- (27)試薬のR1がトリフェニルメチルまたはそのアルコキシ誘導体である、 請求項23または26記載の方法。

- (33)追加単位がヌクレオチドモノマー単位である、請求項31または32記 載の組成物。
- (34)追加モノマー単位が、別の非ヌクレオチドモノマー単位に結合している 非ヌクレオチドモノマー単位である、請求項31または32記載の組成物。
- (35)ヌクレオチド/非ヌクレオチドポリマー含有組成物であって、(8)飽 和連結基を介して骨格に連結されている官能部分、および非不適条件下でリガン ドに連結され得る飽和リンキングアームから選択されるリガンドを連結している 状態で有する非ヌクレオチドモノマー単位、 (b)非ヌクレオチドモノマー単位に結合した第1ヌクレオチドモノマー単位、 および (c)非ヌクレオチドモノマー単位に結合した追加モノマー単位を含む組成物。
- (36)リガンドが、標識、挿入剤、金属キレート剤並びに標識および挿入剤の うちの一方に連結され得る飽和連結アームから選択されるものである、請求項3 5記載の組成物。
- (37)リガンド基が飽和連結アームである請求項36記載の組成物。
- (38)非ヌクレオチドモノマー単位が、その両端に結合基が連結されている骨 格を有し、リガンドが、アルキルアミノおよびアルキルチオから選ばれる連結基 を介して骨格に連結された標識を含んでいる、請求項34記載の組成物。
