〔技術分野〕 本発明は、新規なアミラーゼG4とβ−アミラ
ーゼを組み合せ、澱粉から収量よくマルトースを
製造する方法に関するものである。 〔従来技術〕 β−アミラーゼはアミロース及びアミロペクチ
ンの非還元性末端からマルトースを生成するが、
β−アミラーゼはアミロペクチン中の分岐結合
(α−1,6−グルコシド結合)を加水分解する
ことができないため、アミロペクチンやこれを含
む澱粉からのマルトースの収量は50〜60%であ
る。そして、これ以上に収量よくマルトースを得
るためには、α−1,6−グルコシド結合を加水
分解する、プルラナーゼ、イソアミラーゼなどの
α−1,6−グルコシダーゼをβ−アミラーゼと
併用して使用する必要がある。 〔目的〕 本発明は、澱粉からマルトースを70%以上の高
い収量で収得する新規なマルトースの製造方法を
提供するものである。 本発明者は数年来、澱粉からマルトース以上の
各種オリゴ糖の製法を確立することを目的とし
て、広く自然界より、新規なアミラーゼ産菌の検
索を行つたきた結果、アミロース、アミロペクチ
ン、澱粉などのα−グルカンを特異的にマルトテ
トラオース(G4)に加水分解する新規なアミラ
ーゼがバシルス・サーキユランス(Bacillus
circulans)と同定した細菌により生産されるこ
とを認めた。 澱粉からマルテトラオースを生成するアミラー
ゼとして、シユードモナス・スツツツエリ
(Pseudomonas stuzeri)の生産するアミラーゼ
が、アミロースやアミロペクチンの非還元性末端
からマルトテトラオース単位で加水分解すること
が知られている。しかし、この酵素はアミロペク
チンやグリコーゲンからは高分子量のリミツトデ

以上の通り、アミラーゼG4はα−1,6−グ
ルコシド結合を分解することはできないが、アミ
ロースがアミロペクチンをマルトテトラオース単
位で規則的に分解し、且つアミロペクチンの分岐
内鎖を切断する能力をもつているため、リミツト
デキストリンを殆んど残すことなく、アミロペク
チンや澱粉を分解することができる。このため、
本酵素はβ−アミラーゼのようなマルトース生成
酵素を併用して用いるとき、アミロペクチンやこ
れを含む澱粉からも70〜80%の高い収量でマルト
ースを得ることができる。本発明は、この知見に
基づいてなされたものである。 〔構成〕 すなわち、本発明は、澱粉、又はその派生物で
ある液化澱粉に、バシルス属アミラーゼG4とマ
ルトース生成酵素を作用させることを特徴とする
マルトースの製造方法に関するものである。以下
に、本発明の内容を更に具体的に説明する。 本発明において使用するアミラーゼG4は、土
壌中より分離し、バシルス・サーキユランス
(Bacillus circulans)と同定された細菌の生産す
る菌体外酵素であり、下記の酵素的性質を有す
る。 (1) 作用;アミロース、アミロペクチン、グリコ
ーゲンなどのα−グルカンを、マルトテトラオ
ースを主成分をする分解物に分解する。本酵素
はエンド型の分解様式を持つα−アミラーゼの
一種であり、液化澱粉に作用させるとき、約65
〜75%の収量でマルテトラオースが得られる。 (2) 作用温度範囲及び最適温度;1%可溶性澱
粉、0.05Mリン酸緩衝液の下で作用させたと
き、約75℃まで作用し、最適作用温度は約50℃
である〔第1図a〕。 (3) 作用PH範囲及び最適PH;約4〜約12の広い範
囲に作用する。最適作用PHは6〜8.5である
(0.05M酢酸またはリン酸緩衝液、1%可溶性
澱粉下で作用、第1図b)。 (4) 熱安定性;0.1Mトリス緩衝液(PH7.0)の下
で加熱した場合、50℃、10分間の加熱で約40%
失活し、55℃、10分間の加熱で約85%失活し
た。〔第1図c〕。 (5) PH安定性;0.1M緩衝液の下で、室温(25℃)
で3時間放置後、残存活性を測定した。その結
果、PH6〜9の範囲で安定であつた。〔第1図

熱安定性の増加が認められた。〔第1図cの破
線〕 (7) 阻害剤;本酵素は、5×10-3MのHgCl2、
CuSO4、ZnSO4、AgNO3により、それぞれ約
100%、約95%、約50%、約30%阻害された。 (8) 精製方法;本酵素は、液体培養物の遠心上澄
液から、硫安分画、DEAE−セフアロース
(Sepharose)カラム、クロマトグラフイーと
バイオゲル(Biogel)A0.5mカラム、クロマ
トグラフイー及び同ゲルによる再クルマトグラ
フイーにより電気泳動的に均一まで精製するこ
とができる。 (9) 分子量;バイオゲル(Biogel)A0.5mを用
いたゲル濾過法により測定した分子量は約1万
であつた。 (10) 力価測定法;0.1Mリン酸緩衝液(PH7.0)に
溶解させた2%可溶性澱粉0.5mlに、適量の酵
素を加え、水で全量1mlとし、40℃で反応させ
る。この条件で、1分間に1μモルのグルコー
スに相当する還元力を生成する酵素量を1単位
とした。 以上の酵素的性質について、本発明以前に知ら
れているシユードモナス スツツツエリのマルト
テトラオース生成アミラーゼと比較すると(1)最適
作用PHは本発明の酵素はPH6〜8.5の広いPH範囲
にあるのに対し、シユードモナス スツツツエリ
の酵素はPH8付近にあること、(2)本発明の酵素を
澱粉に作用させたとき、約65〜75%の収量でマル
テトラオースが得られるに対し、シユードモナス
スツツツエリの酵素の場合は約55%であるこ
と、(3)本発明の酵素の分子量は1万前後にあるの
に対し、シユードモナス スツツツエリの酵素
は、48000と58000にある〔バイオケミストリーア
ンド バイオフイジクス アクタ
(Biochemistry and Biophysics Acta)566巻、
88頁(1979)〕など、本発明の酵素とシユードモ
ナス スツツツエリの酵素とは、澱粉からのマル
トテトラオースの収量、最適作用PH、分子量など
の性質において顕著な違いのあるものであり、新
規な酵素ということがきる。 本発明の酵素を生産するバシルス サーキユラ
ンス(Bacillus circulans)G−4の菌学的性質
は下記に示す通りであり、微工研条寄第820号と
して工業技術院微生物工業技術研究所に寄託され
ている。 (1) 形態;桿菌、巾0.5〜1×5〜6μ、運動性な
し、グラム陰性。 (2) 胞子;通常、末端近くに1個、胞子嚢のふく
らみは認められない。 (3) 肉汁;混濁、沈降する。 (4) 肉汁寒天;生育良好、淡黄〜淡褐色、周辺及
び表面凹凸あり。 (5) グルコース肉汁寒天;生育良好、淡黄〜淡褐
色。 (6) グルコース硝酸寒天;生育悪い。 (7) ポテト;生育良好、乳白色に生育。 (8) グルコース・アスパラギン寒天;生育あまり
良くない。淡黄〜黄褐色。 (9) チロリン寒天;良く生育する、わずかに褐
色。 (10) ミルク;ゆつくり凝固、ペプトン化。 (11) インドール;生成しない。 (12) カタラーゼ;生成する。 (13) アセチルメルカルビノール;生成しない。 (14) 硫化水素;生成しない。 (15) クエン酸;利用する。 (16) 硝酸塩の還元;陰性。 (17) クエン酸;利用する。 (18) 食塩肉汁;8%食塩含有培地までよく生育
し10%食塩でも少し生育する。 (19) 炭水化物の利用;D−グルコース、D−フ
ラクトース、D−マンノース、L−アラビノー
ス、D−リボース、マルトース、シユークロ
ス、澱粉などから酸を生成するが、ガスの生成
はしない。D−キシロース、L−ラムノース、
L−ソルボースの利用性は良くない。 (20) 最適生育温度;26℃前後。 (21) 最高生育温度;約60℃。 (22) 死滅温度;100℃、で30分間加熱しても死
滅しない。 以上の菌学的性質について、バージエイス マ

(The Willams and Wilkins Company)、(1957
年及び1974年)を参照し、本菌をバシルス サー
キユランス(Bacillus circulans)の一種と同定
し、バシルス サーキユランスG−4と命名し
た。 本発明によるアミラーゼG4を生産するための
培養は、窒素源として、肉エキス、ペプトン、酵
母エキス、カゼイン、コース・ステイープ・リカ
ー、大豆粕など、通常、微生物の培養に対しよく
用いられる有機窒素源が使用され、炭素源として
は、澱粉、デキストリン、マルトース、グルコー
ス、シユークロスなどが使用される。そして、こ
れに補足する栄養源として、無機窒素源、リン酸
塩、マグネシウム塩と各種金属塩を含む培地が使
用される。培養はPH5〜9、温度20〜60℃で好気
的に行われる。 アミラーゼG4は、菌体外に生産される酵素で
あるので、培養終了後、濾過または遠心分離によ
り除菌し、上澄液を回収する。必要により濃縮
し、硫安、硫酸ナトリウムによる塩析によるか、
または、アセトン、イソプロパノール、エタノー
ル、メタノールなどの有機溶媒を加えて、酵素を
沈澱物として収得し、乾燥、保存する。 本発明において、アミラーゼG4と併用される
マルトース生成酵素としては、通常、大豆、小
麦、大麦など、植物起源のβ−アミラーゼや、バ
シルス属など細菌の生産するβ−アミラーゼ〔ア
グリカルチヤ・バイオロジカル・ケミストリー

1515頁(1976)など〕が使用されるが、この他、
バシルス属、アスペルギルス属、ストレプトマイ
セス属などの微生物の生産するマルトース生成α
−アミラーゼを使用することができる。 アミラーゼG4とマルトース生成酵素と組み合
せて行う澱粉の糖化反応は、次のようにして行
う。 澱粉は、通常、あらかじめ酸または液化型α−
アミラーゼにより液化される。液化度はマルトー
スの収量に影響し、なるべく低DEであることが
望ましい。通常、DE5以下の液化澱粉が使用され
る。反応は、基質濃度、通常5〜40%において通
常PH5〜8、温度は40℃〜60℃で行われる。アミ
ラーゼG4はカルシウムイオンの存在により熱安
定化されるので、糖化に際し、通常5×10-4〜2
×10-2M程度のカルシウム塩が添加される。 アミラーゼG4とβ−アミラーゼなどマルトー
ス生成酵素は、同時に澱粉に作用されるのが望ま
しいが、あらかじめ、マルトース生成酵素で糖化
し、反応途中でアミラーゼG4を添加して反応を
継続するか、あるいは、最初、アミラーゼG4で
反応を開始し、次いでマルトース生成酵素を添加
して反応を行なう。 次に、実施例により、本発明の詳細を説明す
る。 実施例 1 ポリペプトン1%、リン酸2カリ0.3%、硫酸
マグネシウム(7水塩)0.1%、可溶性澱粉1%
からなる培地30mlを200ml容三角フラスコに入れ、

を用い、常法により液化した、DE4.2のポテト液
化澱粉1g(形分として)に、アミラーゼG4を
4単位又は/及び大麦β−アミラーゼ500単位
