従つて、この発明の目的は、グラム陰性菌およ

エム−4−カルボン酸化合物およびその塩を提供

原料化合物()は次に示す方法によつて製造
することができる。

次に、この発明の目的化合物および後述の原料
化合物を表わす一般式中における種々の定義につ
いて、より詳細に説明する。
「低級」とは、特に断りのない限り、炭素数1
ないし6の基を意味する。
R1の「低級シクロアルキル基」の好ましい例
としては、シクロプロピル、シクロブチル、シク
ロペンチル、シクロヘキシルが、さらに好ましい
例としてはシクロヘキシル基が挙げられる。
R2およびR2 aのカルボキシ基のエステルとして
は、メチルエステル、エチルエステル、プロピル
エステル、イソプロピルエステル、ブチルエステ
ル、イソブチルエステル、t−ブチルエステル、

ル、サリチルエステル等の置換もしくは非置換ア
リールエステル、トリメチルシリルエステル、ト
リエチルシリルエステル、ジメチルメトキシシリ
ルエステル、ジメチルエトキシシリルエステル、
ジエチルメトキシシリルエステル、トリメトキシ
シリルエステル、トリエトキシシリルエステル等
のトリアルキルシリル化合物、ジアルキルアルコ
キシシリル化合物もしくはトリアルコキシシリル
化合物の如きシリル化合物とのエステルなどが挙
げられる。
R3およびR3 aの「置換もしくは非置換低級アル
カンアミド基」における「置換もしくは非置換低
級アルカノイル」部分としては、例えばホルミ
ル、アセチル、クロロアセチル、トリフルオロア
セチル等が例示される。
Zの「低級アルキル基」としては、メチル、エ
チル、プロピル、イソプロピル、ブチル、イソブ
チル、t−ブチル、ペンチル、ネオペンチル、ヘ
キシル等が挙げられる。
7−置換−3−セフエム−4−カルボン酸化合
物()を製造する方法A、BおよびCの反応
は、後述の実施例もしくは常法によつて行なわれ
る。
原料化合物()を製造する方法について、以
下に説明する。
方法1:エーテル化
化合物()および(a)は、化合物()
または()にエーテル化剤を作用させることに
よりそれぞれ得られる。
エーテル化剤の好ましい例としては、ブロモシ
クロヘキシルの如きハロゲン化シクロアルカンな
どが挙げられる。
この反応は通常、水、アセトン、エタノール、
ジエチルエーテル、ジメチルホルムアミド、その
他この反応に悪影響を与えない溶媒中で、冷却下
ないし加熱下に、好ましくは以下の方法3におけ
る塩基性加水分解の際に使用されるような無機も
しくは有機の塩基の存在下に行われる。
方法2:チアゾール環の形成
化合物()および(a)は化合物()ま
たは()にチオウレア化合物()を作用させ
て、また化合物(b)は化合物()にチオ尿
素を作用させてそれぞれ製造することができる。
この反応は、通常水、メタノール、エタノール
等のアルコール、ベンゼン、ジメチルホルムアミ
ド、テトラヒドロフラン、その他この反応に悪影
響を与えない溶媒中で、室温ないし加熱下に行わ
れる。
方法3:アミノ保護基の脱離
化合物(b)および(d)は化合物(
a)または(c)を、R3 aの置換もしくは非置
換低級アルカンアミド基またはトリチルアミノ基
中の保護基の脱離反応に付すことにより、それぞ
れ製造することができる。
この脱離反応は、加水分解、還元等の常法に従
つて行われる。これらの方法は、脱離すべき保護
基の種類に応じて適宜選択される。
加水分解は酸を用いる方法(酸性加水分解)、
塩基を用いる方法(塩基性加水分解)などを含
む。
この酸性加水分解において使用される酸として
は、蟻酸、トリフルオロ酢酸、ベンゼンスルホン
酸、p−トルエンスルホン酸、塩酸等の無機もし
くは有機酸、カチオン系イオン交換樹脂などが例
示される。これらのうち、好ましい酸としては、
反応生成物から中和あるいは減圧留去等の常法に
より、容易に分離できるもの、例えば蟻酸、トリ
フルオロ酢酸、塩酸等が例示される。
この反応に適した酸は、原料化合物および反応
生成物の化学的性質ならびに脱離すべき保護基の
種類を考慮して適宜選択するのがよい。この酸性
加水分解は、水の他通常の有機溶媒あるいはこれ
らと水との混合溶媒の存在下もしくは非存在下に
行い得る。
なお、酸としてトリフルオロ酢酸を用いる場合
には、アニソールを反応系に添加することによ
り、反応が促進される。
塩基を用いる加水分解は、アシル基のような保
護基、殊にトリフルオロアセチルのようなハロア
ルカノイル基の脱離に適用される。
ここで使用される塩基としては、水酸化ナトリ
ウム、水酸化カリウム等の水酸化アルカリ金属、
水酸化マグネシウム、水酸化カルシウム等の水酸
化アルカリ土類金属、炭酸ナトリウム、炭酸カリ
ウム等の炭酸アルカリ金属、炭酸マグネシウム、
炭酸カルシウム等の炭酸アルカリ土類金属、炭酸
水素ナトリウム、炭酸水素カリウム等の炭酸水素
アルカリ金属、燐酸マグネシウム、燐酸カルシウ
ム等の燐酸アルカリ土類金属、燐酸水素2ナトリ
ウム、燐酸水素2カリウム等の燐酸水素アルカリ
金属のような無機塩基、酢酸ナトリウム、酢酸カ
リウム等の酢酸アルカリ金属、トリメチルアミ
ン、トリエチルアミン等のトリアルキルアミン、
ピコリン、、N−メチルピロリジン、N−メチル
モルホリン、1,5−ジアザビシクロ[4.3.0]−
5−ノネン、1,4−ジアザビシクロ[2.2.2]
オクテン、1,5−ジアザビシクロ[5.4.0]−7
−ウンデセンのような有機塩基、アニオン系イオ
ン交換樹脂などが例示される。これらの塩基を用
いる加水分解は、通常、水または常用の有機溶媒
あるいはそれらの混合溶媒中で行われる。
還元による脱離は、トリチルなどの保護基の脱
離に適用され得る。還元方法としては、水素化硼
素ナトリウムの如き水素化硼素アルカリ金属を用
いる還元、通常の触媒を用いる接触還元などが例
示される。
反応温度は特に限定されず、原料化合物および
反応生成物の化学的性質、保護基の種類、ならび
に適用方法などを考慮して選択されるが、通常冷
却下、室温または加温下の如き緩和な条件下に行
うのが好ましい。
方法4:カルボキシ基の形成
化合物(c)および(d)は、それぞれ化
合物(a)または(b)の低級アルキル基で
エステル化されたカルボキシ基を遊離のカルボキ
シ基に変換することにより製造できる。
この方法は、加水分解等の常法が適用される。
加水分解方法としては酸もしくは塩基等を用い
る通常の方法が挙げられる。
酸および塩基の好ましい例としては、前記の方
法3において例示したようなものが挙げられ、酸
性もしくは塩基性の加水分解反応は前記の方法3
の場合と同様に行われる。
方法5:オキシム化
化合物(c)は、化合物()にヒドロキシ
ルアミン誘導体()またはその塩を作用させる
ことによつても製造できる。
ヒドロキシルアミン誘導体()は、前記のよ
うな低級シクロアルキル基をR1として有する化
合物であり、その塩としては塩酸塩、臭化水素酸
塩、硫酸塩等が例示される。
この反応は、通常、水、アルコール、テトラヒ
ドロフラン、アセトニトリル、ジメチルスルホキ
サイド、ピリジン、その他この反応に悪影響を与
えない溶媒、あるいはこれらの混合溶媒中で行わ
れ、反応温度は特に限定されない。
この反応において、ヒドロキシルアミン誘導体
の塩を使用する場合には、通常の塩基の存在下に
反応を行うのが好ましい。
本発明の7−置換−3−セフエム−4−カルボ
ン酸化合物の代表的なものについての抗菌活性試
験結果を以下に示す。
1 試験管内抗菌活性試験
(1) 試験方法:
通常の寒天平板希釈法により抗菌活性を測
定した。
各被検菌株をトリプテイケース−ソイ・ブ
ロス中で一夜培養した培養液の100倍希釈液
の1白金耳を、試験化合物を段階毎の濃度で
含むハート・インフユージヨン・アガー
(HI−agar)に接種し、37℃で20時間培養し
た。最低発育阻止濃度(MIC)を測定し、
μg/mlで表示する。
(2) 試験化合物:

この発明の目的化合物()は、通常の担体と
共にカプセル剤、錠剤、顆粒剤、トローチ剤、坐
剤等の固形製剤、軟膏剤あるいは液剤、懸濁液
剤、乳剤等の液状製剤として、経口投与または直
腸投与もしくは注射等の非経口投与などにより患
者に投与される。上記で例示したような各種製剤
には、必要に応じて賦形剤、結合剤、安定化剤、
懸濁化剤、乳化剤、溶解補助剤、矯味剤、緩衝剤
等の通常の添加剤を添加してもよい。
抗菌剤の投与量は、患者の年令、体重等、疾患
の種類、程度等あるいは薬剤の種類等に応じて適
宜定められるが、通常10mg、50mg、100mg、250
mg、500mg等を含有する製剤として投与される。
そして、一般的には1回1mg/Kg〜100mg/Kgの
範囲で、好ましくは5mg/Kg〜50mg/Kgの範囲で
投与される。
次に、この発明を実施例により説明する。
目的化合物()の製造法
実施例 1
(1) オキシ塩化燐(557mg)をN,N−ジメチル
ホルムアミド(266mg)およびテトラヒドロフ
ラン(2ml)の混液に−5℃で撹拌下に適下
し、同温度で30分撹拌する。これにテトラヒド
ロフラン(8ml)および2−(2−ホルムアミ
ドチアゾール−4−イル)−2−シクロヘキシ
ルオキシイミノ酢酸(シン異性体、0.9g)を
−5℃で加え、同温度で30分撹拌する。この溶
液を7−アミノ−3−セフエム−4−カルボン
酸の4−ニトロベンジルエステル(1.05g)、
テトラヒドロフラン(10ml)、アセトン(3ml)
および水(3ml)からなる懸濁液に−5〜5℃
で15分間を要して撹拌下に適下する。この間、
20%炭酸ナトリウム水溶液を加えて反応液の液
性をPH7〜7.5に保つ。次いでこの溶液を30分
間撹拌する。不溶物を濾去した後、濾液に塩化
ナトリウム飽和水溶液を加え、テトラヒドロフ
ランで2度抽出する。抽出液を塩化ナトリウム
飽和水溶液で洗浄し、硫酸マグネシウムで乾燥
した後、減圧濃縮する。残渣にジイソプロピル
エーテルを加え結晶化すると、7−[2−(2−
ホルムアミドチアゾール−4−イル)−2−シ
クロヘキシルオキシイミノアセトアミド]−3
−セフエム−4−カルボン酸の4−ニトロベン
ジルエステル(シン異性体、1.69g)を得る。
