Description

【発明の詳細な説明】 本発明は、牛および羊のタイレリア症
(theileriosis)の治療および予防のための化合物
および調製物およびそれらの化合物の製造方法に
関する。 タイレリア症は、Theileria属の原虫によりお
こる病気である。牛では、T. Parva(parva)、
T.Parval(lawrencei)、T. Parva(Bovis)、T.
Sergenti、T. annulataおよびT. mutansが主に中
央および東アフリカおよび中東部での実質的な損
害な原因となつている。羊では、T. hirciおよび
T.ouisの原因で中東部に遍在する。感染ダニがこ
れらの原虫を哺乳動物宿主に伝播する。淋巴球細
胞が感染されると正常の機能と逆にすみやかに分
裂し始める。死亡は破壊する淋巴球より毒性物質
の放出および寄生虫自体の有害効果でおこる。淋
巴球により放出されたあと、Theileria寄生虫は
赤血球についでダニに感染し、ダニは動物にたか
る。 野外的に、タイレリア症の効果的治療法は知ら
れていない。抗タイレリア活性のある化合物は知
られているが宿主に毒性がある。それらは、タイ
レリア感染症の治療に用いられない。牛および羊
の治療にこれまでに用いられたただひとつの薬は
オキシテトラサイクリン(Terramycin)であ
る。これは、アナプラズマ感染症および細菌感染
にふつうに用いられている。1968年Academic
Press刊、Weinman D.およびRistic M.編、S.F.
Barnett著Infectious Blood Diseases of Man
and Animals,Vol.によると、この化合物は、
感染の成立前に投与しても、非常に限られた効果
しかないが、ひとたび感染が成立すると無効であ
る。しばしば、アナプラズマ感染症が誤診され
て、タイレリア症と診断され、その結果オキシテ
トラサイクリンが投与され、しばしば大量に投与
されほとんど毒性レベルに達する。タイレリア症
より見かけ上恢復したようにみえるのも、実際は
アナプラズマ症および細菌感染より恢復しただけ
である。 英国特許No.1553424は、タイレリア症の予防お
よび治療に有効な2−ヒドロキシ−3−シクロヘ
キシルアルキルおよび2−ヒドロキシ−3−シク
ロヘキシル−1,4−ナフトキノン誘導体を記載
している。その後、後者、つまり2−ヒドロキシ
−3−シクロヘキシル−1,4−ナフトキノンは
もつとも有効な化合物であり、特にT. parvaの感
染にもつとも有効であることが分つた。 ヨーロツパ特許願No.78101426は、場合により
C1-4アルキル基、特にメチル基のような置換基を
有しているC3-12シクロアルキル環を2−位置置
換基とする3−ヒドロキシ−1,4−ナフトキノ
ンのことに言及している。しかし、置換基を有す
る具体的化合物は記載されておらず、シクロアル
キル環のどの位置に置換さるべきかも記載されて
いない。 下記する式()の化合物は、我々の特許願
UKNo.8131206に、ある種の2−置換−3−ヒド
ロキシ−1,4−ナフトキノン合成の中間体とし
て記載された。今や、下記の()の化合物は、
イン ビボーおよびイン ヒドローの條件で種々
のタイレリア種に対し高くそして有利な活性を示
すこと、そして、英国特許No.1553424の化合物と
対照的に、T. annulata感染に対して特に有効で
あることが発見された。その結果、新規治療用化
合物の製造およびそれらの生物でおこる病気に対
し感染動物を治療することが可能となる。 本発明のひとつの特徴として、式 (ただし式中、Rは1から10炭素原子数のアルキ
ル基とする)を有する化合物、それらの互変異性
体、またはそれらの薬剤として許容される塩を提
供する。 互変異性の結果、3−ヒドロキシ基はその酸性
の水素をとなりのオキソ基に与えるが、式()
がより安定な状態を表わすと信ぜられる。式中の
ヒドロキシ基は適当な塩基と塩を形成しうるの
で、化合物の薬剤として許容されうる塩は、アル
カリ金属たとえばナトリウムまたはカリウムの塩
および有機塩基たとえばエタノールアミン、ジエ
タノールアミンおよびN−メチルグルカミンとの
塩がある。 また理解しうるように、式()の化合物は、
シスまたはトランス異性体として存在でき、シク
ロヘキシル環は、ナフトキノン−メチレン残基お
よび4−t−アルキル基で、シスまたはトランス
置換されうる。本発明は、シスおよびトランス異
性体の両者およびそれらの任意の割合の混合物を
対象とする。 留意すべきこととして、非置換1,4−ナフト
キノン環で2および3位は同じであるので、シク
ロヘキシル置換基またはヒドロキシ基を2位置に
おきうる。本明細書では便宜上、化合物を一般的
にさす場合、置換基シクロヘキシル置換基を2位
置とすることにする。 式()の化合物で、Rは直鎖状のC1-4アルキ
ル基とするのは適当である。有利な基はメチル基
またはエチル基である。 式()の化合物は、類似の構造を有する化合
物製造のための既知の方法で製造しうる。 たとえば、式()の化合物を、Rが定義のよ
うである式()の化合物より、それを3−ヒド
ロキシ−1,4−ナフトキノンに変える、標準的
文献記載の方法で製造しうる。式()の化合物
は、Fries転位により式(A)の化合物より製
造しうる。 別様には、式()の化合物を、式()の相
当する3−ハロゲノ、つまり3−クロル−または
3−ブロム類縁体(Xはハロゲン、Rは上記定義
の通りとする)を、アルカリ水解たとえば適当な
媒体中でアルカリ金属水酸化物で、3−ヒドロキ
シ置換化合物に変えうる。たとえば、水性メタノ
ール媒体中水酸化カリウムを用いるのが便利であ
ることが分つた。出発する3−ハロゲノ誘導体は
置換または非置換2−シクロヘキシルメチルフエ
ノールに出発するFierser,L.示唆の方法(J. Am.
Chem.Soc.,1948,3165)により、相当して置換
されている4a,5,7,7a−テトラヒドロ−2
−シクロヘキシルメチル−1,4−ナフトキノン
より、酸化そしてハロゲン化で得られる。 式()は、さらに別の方法として、式() (ただし、Rは、上記定義の通りとする)の化
合物の3位にヒドロキシル基を導入して製造しう
る。 それで、たとえば、式()の化合物をハロゲ
ン化(つまり塩素化または臭素化)して、上記式
()の化合物とし、これはついで、上記の式
()の化合物となしうる。 式()の化合物はエポキシ化して式() の化合物とし、これを、たとえば希塩基または希
酸水溶液で水解して式()の化合物となしう
る。 式()の化合物は、また、酸化剤(たとえば
過塩素酸)の存在でアシル化剤(たとえば無水酢
酸)と反応させてThieleアセチル化に処して式
(a)の化合物となしうる。式(a)の化合
物は水解で式(b)の化合物に変え、これら
は、たとえばOrganic Reaction19巻、222頁に記
載と類似の方法で酸化して式()の化合物に変
えうる。適当な酸化剤には、たとえば、塩化第2
鉄−鉱酸またはクロム酸がある。 上記の方法で最終生成物の立体化学は出発物質
のそれで決まる。つまり立体化学的に純粋な化合
物を反応に用いると純粋なシスまたはトランス異
性体を生ずる。 さらに有利な別法として、式()の化合物
を、 (ただし式中、R1はハロゲン、たとえば塩素
または臭素原子、またはヒドロキシ基、アセトキ
シ基またはアルコキシ基とする)の化合物と、置
換シクロヘキシルメチル基を遊離基として与えう
る形の、前記定義のようなR結合を含有するシク
ロヘキシルメチル基供与化合物とを反応させ、そ
して、望むならば、生ずる2−シクロヘキシルメ
チル基置換縮合物中の、ヒドロキシ基以外のR1
基を水解して3−ヒドロキシル基にすることによ
つても製造しうる。 有利な供与化合物は、酸化的脱炭酸を受けう
る、対応して置換されたシクロヘキシル酢酸であ
る。たとえば、この目的で、銀イオンのような触
媒を用いた過硫酸がある(Jacobson N.等、
Annalen,1972,763,135およびActa Chem.
Scand.1973,27,3211)。なるべくはこれらの條
件で過硫酸を用いる時に、R1がヒドロキシ基以
外の基で置換された1,4−ナフトキノンを用い
る。便宜には、過硫酸アンモニウムを酸化剤とし
て使用でき、触媒は硝酸銀とする。主要カツプリ
ング反応に続いて必要ならば水解し、ヒドロキシ
基とする。水解にはアルカリ條件がふつう有利で
ある。 U.S.特許2553647に示唆されているように、そ
れ自体過酸化物基を有するシクロヘキシルメチル
供与化合物の例は、適当に置換されたシクロヘキ
シルメチルアカノイルパーオキサイドである。 供与化合物よりの自発的放出によるシクロヘキ
シル遊離基の供与は、たとえば、トリシクロヘキ
シルメチル−ボランを用いて達成されうる。この
ような試剤は、シクロアルケンとボランジメチル
スルフアイドを反応させて調製しうる。テトラヒ
ドロフランのような溶媒中で反応さすのが便宜で
ある。上記の反応スキームのいくつかに必要な2
−置換−1,4−ナフトキノンは、文献記載の既
知の合成方法で製造しうる。たとえば、Fieser,
L.,J, Am. Chem. Soc.,1948,3165または類似
の方法がある。 さらに、シクロヘキシル提供化合物として、相
当する置換シクロヘキセニルカルボン酸を使用す
ることもできる。その場合、専門家に明らかなよ
うに、式()の化合物を提供するのに、アルケ
ンの還元という追加の段階が縮合のあとに必要で
ある。 上記方法がシスおよびトランス異性体の混合物
を与えることは当然である。 式()のシスまたはトランス異性体の単一化
合物の必要の時には、異性化のおこらないような
上記反応において、異性体として純粋な出発化合
物を用いる。特に、上記の最後の方法において立
体特異的な純カルボン酸を用いる。異性体の混合
物が生ずるなら、物理的手段で混合物を分けう
る。このような方法は、この方面の技術で知られ
ており、たとえば分別結晶およびクロマトグラフ
分離がある。 本発明のさらに別の特徴は、式()の化合物
を提供することである。 (ただし式中、Rは上記定義のようであり、
R1は上記定義のようかまたはベンジルオキシ基
とする) これらの化合物は、上記合成における中間体と
して有用である。イン ビボーでヒドロキシ基に
容易に水解される基をR1として有する化合物は、
処方物中のプロドラツグ(prodrag)としてまた
はその場で加水分解させて最終生成物を与えるよ
うな治療法として用いうる。アセチル基またはベ
ンゾイル基はそのような水解を受け、それで活性
化合物の長時間作用するプリカーサーに用い得
た。 式()の化合物はTheileria属の原虫に対す
る極度の活性を有し、牛および羊のタイレリア症
の治療そして(または)予防に用いうる。特に、
これらの化合物はT. annulataの感染に有効であ
る。 タイレリア感染の治療または予防に用いるのに
必要な式()の化合物の量は、活性化合物の種
類だけでなく、感染の性質および投与ルートで変
動する。 本明細書で使用の“有効量、投与量または単位
投与量”とは、インビボーで牛または羊に投与す
る時に、Theileria微生物に十分に有効な、あら
かじめ定めた化合物の投与量とする。予防または
治療に役立ちまたは達成するためには、ある調製
物は、1頭が必要とする投与量の何倍かを含有し
てよい。 400Kgの体重の牛での最初の投与量は0.2から10
g、便宜的には0.2から2.5g、そして小牛または
羊では50mgから1.0gまたはなるべくは約0.1から
0.5グラムの活性化合物量とするが、さらに投与
量をふやしてもよい。 抗タイレリア症剤には、式()の化合物は原
料のまま投与しうるが、活性成分を治療用の調製
物として用いるのが有利である。 式()の化合物を含有する治療用調製物は、
封じた容器、または助剤のような成分を用いた成
型錠剤、または組成物の形を取りうる。調製物は
単位投与形態、または無菌の、封入された処方物
の形とするのが有利である。式()の化合物を
含有する調製物は、適当な担体とあわせて、注射
用(皮下またはなるべくは筋肉)、静注または経
口投与しうる。無菌の注射用の処方物は、なるべ
くは有機担体で形成しうる。担体は、静菌剤、抗
酸化剤、調製物を血液と等張とするための緩衝溶
質、濃厚化剤、懸濁剤または他の薬剤として許容
されうる添加物を含有しうる。 注射用の処方物は、単位投与用の容器、たとえ
ばアンプル、使いずての注射装置に加え、また多
回投与形態たとえばびんに入れて、そこから適当
量を取り出して用いうる。 経口投与のための処方物は、担体として、錠
剤、ペースト、顆粒または粉末形成のための固型
物を含有しうるし、または、懸濁または溶解のた
めの液体を含有しうる。それらには、希釈剤、結
合剤、分散剤、表面活性剤、潤滑剤、被覆材料、
着色剤、溶媒、濃厚化剤、懸濁剤または薬剤とし
て許容されうる他の添加物がある。これらの調製
物は、単位投与または多数回投与形態または食品
への添加物となしうる。式()の化合物はまた
サルトーリツク(salt−lick)として、動物が野
外で予防的処理を受けうるようにする。式()
の化合物は、また、ジメチルスルホキサイドのよ
うな適当な担体中に10重量%までの活性化合物を
含有する、潅注用の処方物にもなしうる。 本発明のさらに別の特徴として、タイレリア症
に対し哺乳動物を保護しそして治療するためのパ
ツケージ組成物を提供する。これは、上記定義の
ような調製物を、病気を保護しそして治療するた
めの使用法を示すものをあわせて含有する。 治療用には、式()の化合物またはその塩
を、動物の体温が上昇しそしてスキツオントが現
われた時、またはそれに遅れた病状の時に時には
比較的大量に与えうる。それから数日間たとえば
5日間毎日同様に投与する。この治療期間にわた
つての全投与量はなるべくは0.5から50mg/Kg、
より有利には0.5から10mg/Kgそしてもつとも有
利には1から5mg/Kgの投与量とする。別の治療
法として、毎日1回または2回投与し、全体で10
回投与に及びうる。 予防的処理のためには、たとえば動物が感染を
受けたことが疑われる時には、式()の化合物
を、第1日に1から5mg/Kg、ついで、同様の投
与を毎週1度くり返し、最高5回投与に及びう
る。感染の危険度に応じて、0.2から5mg/Kgを
毎周当たり投与しうる。予防期間は4日から20日
または120日におよびうる。別様には、化合物を、
放出性の遅い移植体に加えて、比較的に不溶の担
体とあわせてペレツトとしたものを皮膚の下にガ
ンで注入しうる。ペレツトが溶解するにつれて、
活性成分はたとえば4月に及んでゆつくり放出さ
れ、上記の投与量によるような、低レベルの予防
が維持される。 さらに別の特徴として、予防的または治療的に
牛または羊のタイレリア症を処置する方法が提供
される。この方法では、式()の化合物または
それの薬剤として許容される塩の有効量を、病原
性のTheileria種の感染を受けたか感染を受ける
可能性のある動物に、予防的または治療的に投与
する。特に、治療方法は、前記定義のような化合
物を含む、調製物または組成物を用いて達成され
る。 さらに、前記定義のような治療の別の特徴とし
て、1種または1種より多くの、有効な抗原とし
て作用する、生きたTheileria種または株を含有
するワクチンを投与する。この目的のワクチンを
製造するには、Theileria種たとえば、T.parva
(parva)およびT.parva(lawrencei)(Radley,
D.E.等(1975)、Veterinary Parasitology,51−
60)で感染された摩砕だにまたは他の感染材料
(たとえばheileria感染哺乳動物培養細胞の懸濁
液または培養だに細胞またはTheileriaで感染さ
れた唾液組織またはそれらの生成物または滲出液
より製造しうる。便宜的には、前記の活性化合物
の投与量は、宿主動物の0.5から5mg/Kgとし、
ワクチン投与日から、ワクチン投与後7日または
14日後まで投与する。つまり最初の処置は、抗原
をすぐに放出させる組合わせ生成物の投与から成
立ち、あとから活性化合物を7日または7日より
長く投与する。最初の投与につづいて、0.5から
5mg/Kgの毎週投与を10週続けうる。 例 A 4−t−アルキルシクロヘキサンカルボン酸の
製造 あとからの実施例で使用のカルボン酸はつぎの
ように製造した。 4−t−ブチルシクロヘキサン−1−カルボ
ン酸、4−t−ブチルシクロヘキシル酢酸、お
よびそれらの純シスおよびトランス異性体は、
市販品として入手するかまたは文献(J.Amer.
Chem.Soc.1970,92,2800およびそれに記載の
文献)に従つて製造した。 4−t−ペンチルシクロヘキシル−1−カル
ボン酸(シス/トランス混合物)および純トラ
ンス−4−t−ペンチルシクロヘキシル−1−
カルボン酸はつぎのように製造した。 a シス/トランス−4−t−ペンチルシクロヘ
キシル−1−カルボン酸の製造 4−t−ペンチルシクロヘキサノン(49.2g)
をエーテル(200ml)に溶解し、シアン化ナトリ
ウム(24.46g)および水(30ml)を加えた。混
合物0℃に冷却しはげしくかきまぜながら、濃塩
酸を1時間かけて加えた。さらに3時間かくはん
し、1夜放置した。混合物は、メタ重亜硫酸ナト
リウム飽和溶液で洗い(2×200ml)そしてエー
テル層を硫酸ナトリウム乾燥した。減圧で溶媒を
留去し、淡黄色油状のシアノヒドリン54.0gを得
た。 シアノヒドリン(54.0g)を無水ピリジン(70
ml)および無水ベンゼン(70ml)に溶解し、0℃
に冷却かくはんした。ピリジン(83ml)中塩化ホ
スホリル(90ml)含有溶液を45分かけて添加し
た。そのあいだ温度は0℃に保つた。反応混合物
はさらに30分間加温還流させた。混合物は放冷
し、氷にあけ、30分かくはんし、エーテル抽出
し、水洗し、硫酸ナトリウム乾燥し、減圧で蒸発
乾こし、4−t−ペンチルシクロヘキス−1−エ
ン−1−ニトリルとした。48.4gで油状。1−4
−t−ペンチルシクロヘキス−1−エン−1−ニ
トリル(48.0g)を、水酸化カリウム(23.3g)
を水(34ml)に含有する溶液およびエタノール
(150ml)の混合物に加えた。混合物は72時間加熱
還流させ、氷冷し、水(175ml)で希釈し、濃塩
酸酸性とした。沈殿無色固型物を取し、水洗
し、乾燥した。固型物は、酢酸エチルと水酸化ナ
トリウム(2N)のあいだに分配し、塩基層を分
け濃塩酸酸性とした。生ずる無色固型物を取
し、水洗し、乾燥し、4−t−ペンチルシクロヘ
キス−1−エン−1−カルボン酸、33.65g、融
点123−125℃を得た。 4−t−ペンチルシクロヘキス−1−エン−1
−カルボン酸(33.5g)をエタノール(275ml)
に溶解し、10%パラジウム−炭(1.0g)を加え
た。混合物は10気圧で水素添加し、理論量の水素
を吸収させた。触媒を去し、無色の液を減圧
で蒸発乾こし4−t−ペンチルシクロヘキシル−
1−カルボン酸を得た。27.0g(シス/トランス
混合物)。 b 純トランス−4−t−ペンチルシクロヘキシ
ル−1−カルボン酸の製造 シス/トランス混合酸(12g、方法aで製造)
を、濃流度(60ml)の存在で16時間、蒸気浴中で
加熱した。反応混合物を冷却し、氷にあけると黒
色固体が生成した。固型物を取し、軽石油(40
−60℃)でこね、大部分の固型物を溶解した。石
油抽出物は炭末処理し、減圧で蒸発乾こしトラン
ス−1−4−t−ペンチルシクロヘキシル−1−
カルボン酸を得た。5.7g。融点92−100℃。
NMRスペクトルから95−97%純度のトランス異
性体である。 例 1 2−〔トランス−(4−t−ブチルシクロヘキ
シル)メチル〕−3−ヒドロキシ−1,4−ナ
フトキノンの製造 A 1−トランス−(4−t−ブチルシクロヘキ
シル)−酢酸の製造 4−t−ブチルシクロヘキサン−1−カルボン
酸(50g、シスおよびトランス異性体の混合物)
と、水酸化カリウム(150g)およびエチレング
リコール(500ml)を1夜加熱還流させた。反応
混合物は砕氷(2000g)にあけ、濃塩酸で酸性と
し、生ずる色沈殿を去した。湿固型物は酢酸エ
チルに溶解し、水層を除き、有機層を硫酸マグネ
シウムで乾燥した。減圧で溶媒を留去すると白色
固型物を与えた。これをトルエンより再結すると
1−トランス−(4−t−ブチルシクロヘキシル)
−カルボン酸(30.9g、融点172−174℃)を与え
た。 この1−トランス−(4−t−ブチルシクロヘ
キシル)−カルボン酸(35g)を無水エーテル
(250ml)に溶解し、この溶液を、水素化リチウム
アルミニウム(7.3g)を無水エーテル(200ml)
に含有する溶液に、おだやかな還流を保つように
ゆつくり添加した。添加を完了してから、混合物
はさらに1.5時間加熱還流させた。ついで反応混
合物は氷浴中で冷却し、氷冷塩酸2N,200ml)を
加えた。水層を分け、エーテル(100ml)で洗い、
エーテル抽出物を合併して希水酸化ナトリウムで
洗い、水で洗い、硫酸マグネシウムで乾燥し、溶
媒を減圧留去した。28gの無色の油を与えた。上
記の1−トランス−(4−t−ブチル−シクロヘ
キシル)−メタノール(28g)を乾燥アセトニト
リル(330ml)に溶解し、無水臭化リチウム
(28.7g)およびクロルトリメチルシラン(44.5
g)を加え、混合物は60時間加熱還流させた。反
応混合物は放冷し、減圧で溶媒を留去した。残留
物はエーテルに溶解し、水(100ml)、10%重炭酸
ナトリウム(100ml)、水(100ml)で洗い、
MgSO4乾燥し、溶媒を減圧留去した。粗生成物
をカラムクロマトグラフイン−し、油状の1−ト
ランス−(4−t−ブチルシクロヘキシル)−メタ
ンブロマイド(15.5g)を得た。 上記のブロマイド(15g)を、ヘキサメチルリ
ン酸トリアミド中シアン化ナトリウム(4.83g)
に加え常温で1夜かくはんした。粗反応混合物は
水(250ml)で希釈しエーテル抽出(2×200ml)
した。有機層を合併し、水洗し(3×100ml)、乾
燥し(MgSO4)、溶媒を減圧留去し、無色液体状
の1−トランス−(4−t−ブチルシクロヘキシ
ル)メタンニトリル(10.5g)を得た。 1−トランス−(4−t−ブチルシクロヘキシ
ル)−メタンニトリル(10g)、水酸化カリウム
(20g)、エタノール(200ml)および水(50ml)
を65時間加熱還流させた。大部分の溶媒を減圧留
去し、残留物を水(300ml)に溶解し、酢酸エチ
ル(100ml)で抽出した。水層は塩酸で酸性とし、

ナフトキノンを与えた。融点154−156℃。NMR
スペクトルは、2−〔トランス−(4−t−ブチル
シクロヘキシル)メチル〕−3−クロル−1,4
−ナフトキノンの構造を示した。また100%トラ
ンス異性体であることも示した。融点は154−156
℃。 上記のように得られたクロルキノン(6g)を
ジメトキシエタン(60ml)および水(60ml)に含
有する溶液を加熱還流しておき、水酸化カリウム
(6.0g)を水(60ml)に含有する溶液を10分間に
わたり滴加した。さらに15分還流させ、混合物は
常温まで急冷し、濃塩酸で酸性とした。得られる
明黄色固型物を集め、水洗し、乾燥し、2−〔ト
ランス−(4−t−ブチルシクロヘキシル)メチ
ル〕−3−ヒドロキシ−1,4−ナフトキノンを

ノンを水解し、2−〔シス−4−t−ブチルシク
ロヘキシル)メチル〕−3−ヒドロキシ−1,4
