本発明は、木材腐朽性担子菌の分離方法に関す
るものである。
更に詳細には、本発明は、カビその他の雑菌の
生育を抑え、木材腐朽性担子菌を効率よく分離す
る方法に関するものである。
本発明によれば、腐朽材より木材腐朽性担子菌
を効率よく分離することができ、しかもリグニン
の分解にすぐれた木材腐朽性担子菌を分離するこ
とができるので、木材腐朽処理や半材糖化工業の
技術分野及び木材腐朽性担子菌の微生物学的基礎
研究等の重要な役割を果すものである。
(従来の技術)
従来、腐朽材より木材腐朽性担子菌を分離する
培地としてはジヤガイモ・ブドウ糖寒天培地や麦
芽エキス寒天培地を用いるのが一般的である。

(発明が解決しようとする問題点)
しかしながら、ジヤガイモ・ブドウ糖寒天培地
や麦芽エキス寒天培地はカビ、酵母などの生育に
も適した培地であり、木材腐朽性担子菌の選択分
離用培地としては不適当である。
即ち、木材腐朽性担子菌の選択分離にジヤガイ
モ・ブドウ糖寒天培地や麦芽エキス寒天培地を使
用すると、カビや酵母など木材腐朽性担子菌以外
の雑菌が多量発生して目的とする木材腐朽性担子
菌の生育が抑制され、容易に選択分離できないと
いうことになつてしまう。
また、ジヤガイモ・ブドウ糖寒天培地や麦芽エ
キス寒天培地を用いた場合、生育した多くのコロ
ニーのなかで、これが木材腐朽性担子菌であると
一見して識別することができず、きわめて不便な
ものであつた。
(問題点を解決するための手段)
本発明は、これら従来法が有する欠点を一挙に
解決するためになされたものである。
本発明は、木粉を唯一の栄養源として含有する
寒天培地を用いて木材腐朽性担子菌を分離する方
法である。
本発明では木粉を唯一の栄養源として含有する
寒天培地を用いたために、カビ、酵母等の雑菌の
生育を抑制し、より多くの担子菌を生育させるこ
とに成功したものである。
また、本発明では木粉が寒天培地中に分散して
いるため、木材腐朽性担子菌が生育したとき、コ
ロニーの周辺を薄白色、薄黄色又は薄暗褐色に変
化させ、ハローを形成するので、一見して木材腐
朽性担子菌として識別できるものである。
一般に木材腐朽性担子菌は、リグニン分解能の
強い菌とセルロース分解能の強い菌とに大別され
るが、リグニン分解能の強い菌は木粉中のリグニ
ンを分解してセルロースを残すため、作用した部
分は漂白されたようになり、薄白色及び薄黄色を
呈するようになる。また、セルロース分解能の強
い菌は木粉中のセルロースを分解してリグニンを
残すため、作用した部分はリグニンの色調にな
り、薄暗褐色を呈するようになる。
本発明に用いる木粉はいかなる木の粉末でもよ
い。ブナ、ナラ、クヌギ、シデ、シイ、クリ、マ
ツ、スギなどの木から選択して、粉末化すればよ
い。
木の粉末化に際しては、木材やチツプをよく乾
燥し、細断して、粉砕すればよい。粉砕したもの
は篩別し、20メツシユを通過したもの、好ましく
は40メツシユ〜100メツシユの大きさのものを分
取し、本発明に用いる木粉とするのがよい。
木粉は0.2%以上の適宜の量で培地に添加され
るが、1〜10%程度、好ましくは2〜4%程度で
十分である。
寒天は1〜2%程度用いられ、寒天の溶解液に
木粉を添加して培地を調製し、これをシヤーレに
分注し、木材腐朽性担子菌の選択分離に供するも
のである。
本発明においては、腐朽材その他担子菌含有物
を木粉寒天培地に接種して、25〜35℃で培養すれ
ば、多くのコロニーが発生し、そのコロニーのな
かで、薄白色及び薄黄色のハローを生じたコロニ
ーを採取すれば、リグリン分解能のすぐれた担子
菌を分離することができるものである。
(発明の効果)
本発明は、上記したような独特な新規な構成を
採用することにより、きわめて簡単に木材腐朽性
担子菌を分離できるというすぐれた効果を有する
が、要約すれば次の通りである。
(1) 寒天培地で木粉を唯一の栄養源としたため
に、カビ、酵母などの雑菌の生育を抑制し、多
くの担子菌を生育させることができる。
(2) 寒天培地で木粉を含有しているので、リグニ
ン分解能の強い担子菌は、木粉のリグニンを分
解し、セルロースを残して、薄白色及び薄黄色
のハローを形成するので、リグニン分解能の強
い担子菌を容易に分離することができる。
(3) 寒天培地で木粉を含有しているので、セルロ
ース分解能の強い担子菌は、木粉のセルロース
を分解し、リグニンを残して、薄暗褐色のハロ
ーを形成するので、セルロース分解能の強い担
子菌を容易に分離することができる。
次に本発明の実施例を示す。
実施例 1
木粉(40〜100メツシユ)2.0%、寒天1.6%を
含有する培地を120℃、15分間加熱殺菌した後、
無菌状態でシヤーレ(直径90mm)に20mlずつ分注
する。
上記、木粉寒天培地に火炎処理した腐朽材を接
種し、28℃で1〜2週間培養した後、単菌分離操
作を行なつた。
なお、比較のため、ジヤガイモエキス0.4%、
ブドウ糖2.0%、寒天1.6%を含有するジヤガイ
モ・ブドウ糖寒天培地(極東製薬工業)でも同様
の実験を行なつた。
それぞれの培地での分離結果を表1に示す。
