この発明は、種々の酵素活性を有する固定化さ
れた微生物の原核細胞、及び真核細胞、並びに植
物細胞に基礎を置く生物変換用細胞触媒に関す
る。 この発明の細胞生物触媒は、工業的生物変換に
使用することができ、これにより薬品工業、食品
工業及び農業において重要な化合物の非連続的又
は連続的型式による生産が可能になる。 一般に、細胞を固定化する公知方法には、物理
的方法、物理化学的方法及び化学的方法があり、
細胞と担体との結合は細胞をゲル中に包埋するこ
とにより機械的に、フアン・デル・ワールス

ン、無機材料、例えばガラス、チタン、ジルコニ
ウム及びバナジウム化合物が担体として使用され

細胞を相互に固定化するための一般的方法がチエ
コスロバキア国発明者証第209265号に記載されて
いる。この明細書に引用した方法により、所望の
酵素活性を有する種々の微生物の共有結合により
相互に架橋せしめた細胞の非常に安定な生成物
を、良好な沈降性を有する一体に形成した粒子単
位として得ることができる。原理的には、この方
法は、未処理細胞を二官能価架橋剤、特にグルタ
ルアルデヒドと化学的に反応せしめることにより
個々の細胞の細胞内成分を共有結合により架橋
し、そして、これらの細胞の自然的又は人為的自
己消化に対する耐性を生じさせることから成る。
次に、架橋細胞を物理的処理、物理化学的処理又
は化学的処理を単独で又は組合わせて施すことに
より該細胞に透過性を付与し、グルタルアルデヒ
ドと反応しない細胞表面構造の一部分を構成する
高分子物質、特に脂質を放出することにより架橋
細胞の拡散障壁を喪失せしめる。最終段階とし

遊離架橋剤、特にグルタルアルデヒドの存在下で
架橋し、そして透過性を付与した細胞を相互に共
有結合せしめる。 上に引用した固定化細胞製造方法によりすでに
細胞生物触媒を十分に経済的な形で製造すること
が可能となつた。しかしながら、低分子の攻撃的
架橋剤の分子、例えばグルタルアルデヒドが非常
に低濃度であつても容易に細胞内に入るために、
細胞中に存在する幾つかの酵素は急速、且つ不可
逆的に不活性化し、このために幾つかの感受性酵
素の活性が大きく低下し、上に引用した方法は普
遍的に使用できるものではない。上記のような不
活性化は、おそらく、共有結合架橋の生成と、そ
れに続く酵素の活性部位の不可逆的な変形によつ
て生ずるものと予想される。このことは、特にラ
セマーゼ活性及びリアーゼ活性を有する細胞にお
いて実験的に示されている。 上記の欠点は、酵素活性を有する未処理細胞も
しくは場合によつては前もつて透過性付与処理を
施した細胞、もしくはこれらの細胞の断片及びこ
れらの細胞の細胞内顆粒;及び/又は未処理細胞
と細胞断片、細胞内顆粒及び細胞溶出成分との混
合物;及び/又は個々の細胞について架橋処理及
び透過性付与処理を施した細胞を、2個以上の第
一及び/又は第二アミノ基を有する水溶性化合
物、例えばポリエチレンイミン、ヘキサメチレン
ジアミン、リジン及びポリリジンと二官能価アル
デヒド、例えばグルタルアルデヒドと反応せしめ
ることにより生成する水溶性反応性重合体により
化学的に固定化し、場合によつてはさらに、該固
定化物を透過性付与処理しそして/又は機械的に
硬化せしめることにより得られるこの発明の細胞
触媒を使用することにより回避することができ
る。 この触媒には、細胞成分として、原核生物の細
胞又は真核生物の細胞、すなわち、グラム陽性細
菌、グラム陰性細菌及びグラム・ラビール細菌、
抗酸菌、放線菌、酵母、並びに糸状菌のごとき他
の顕微鏡的生物、並びに高等な、すなわち寄生生
物、並びに植物細胞、あるいは又、さらにこれら
の細胞の断片及び細胞内顆粒であつて、酵素活
性、すなわちヒドロラーゼ、イソメラーゼ、リア
ーゼ、脱炭酸酵素、酸化−還元酵素及び他の活性
を有する酵素及び/又は生化学的経路、特に分解
代謝、解糖系、ホルモンの変換、及びアルカロイ
ド、ソランアルカロイド(solane alkaloids)、
シトステロール(phtosterols)及び特に麦角ア
ルカロイドの生産を触媒する酵素系の全部又は一
部分の酵素の活性を有するものを含有せしめるこ
とができる。 上記の触媒の製造方法を提供することがこの発
明の目的であり、この方法は、細胞性材料を、2
個以上の第一及び/又は第二アミノ基を含有する
水溶性化合物、例えばポリエチレンイミンと二官
能価アルデヒド、例えばグルタルアルデヒドと
を、0℃〜50℃にて、水中で、PH4.0〜12.0の範
囲において反応せしめることによつて得られる水
溶性、且つ化学的に活性な重合体と混合し、そし
て該細胞成分と上記の重合体とを、水性環境中−
30℃〜50℃において、PH5.0〜9.0の範囲におい
て、そして場合によつては凝集剤の存在下で反応
せしめ、これにより形成された細胞固定化物又は
凝集体を反応混合物から分離し、水又は緩衝液で
洗浄し、そして場合によつては透過性付与処理及
び/又は機械的硬化処理を行うことにより細胞成
分を固定化することができるという事実に基礎を
置いている。 水溶性重合体の調製及び細胞性材料の前凝集の
ためには、分子量100000〜800000の範囲内の水溶
性ポリエチレンイミンが有用である。 細胞性材料と重合体との反応は、0〜45℃にお
いて、1〜50000gの相対遠心力下又は0〜
50000KPaの過圧下において、反応混合物を静
置し、ゆつくり混合し又は間欠的に混合しながら
行うのが好ましい。 固定化前の未処理細胞又は固定化後の細胞凝集
体に対する透過性付与処理は、水性環境下、0〜
70℃、PH4.0〜9.0において、界面活性剤及び/又
は水混和性もしくは非混和性有機溶剤、例えばア
セトン、エーテル、イソプロパノール、石油エー
テル、酢酸ブチル、クロロホルム、ジオキサン、
ジメチルホルムアミド又はジメチルスルホキシド
の存在下で、そして場合によつては強酸及び/又
は塩基の塩を加えることにより一時に又は逐次に
イオン強度を変化せしめながら細胞懸濁液を混合
することにより行う。 固定化した細胞粒子は、有機溶剤、好ましくは
アセトンにより、又は場合によつては有機溶剤中
もしくは有機溶剤と水との混合物中接着剤溶液に
より−20℃〜25℃にて処理して部分脱水すること
によりさらに硬化せしめることができ、こうして
得た固定化物はさらに取し、そして乾燥環境又
は湿潤環境中で部分乾燥し、場合によつてはさら
に後成形することができる。 この発明の利点は、化学的に反応性の水溶性重
合体(以下RSPと略す)により、高い比活性、
良好な沈降性を有する固定化細胞触媒を、高い固
定化収率をもつて製造することができ、又、低分
子架橋剤を使用する方法と異なり、この発明に従
つて調製したRSPは細胞内に透過できない程度
に高分子であるので細胞表面と活性化RSPの反
応性アルデヒド基との間でのみ共有結合(固定
化)が生ずるので、チエコスロバキア発明者証第
197101号、第203607号及び第209265号の方法を適
用することができない高感受性酵素に適用するこ
とができる点にある。 化学的に活性な水溶性重合体の形成機構、例え
ばポリエチレンイミンと二官能価架橋剤すなわち
グルタルアルデヒドとの機構は、グルタルアルデ
ヒドの末端のアルデヒド基とポリエチレンイミン
の第一又は第二のアミノ基との間の着色シツフ塩
基(C=N結合)を優先的に生成せしめるポリエ
チレンイミン部分の内部架橋反応であつて、分枝
した、及びおそらくポリエチレンイミン鎖の末端
にも含まれている幾つかの第一又は第二アミノ基
の同時的化学的活性化と共にポリエチレンイミン
部分の分子量が増加する反応であると考えること
ができる。 こうして生成した架橋された、化学的に活性な
重合体はなお水性溶液に可溶性であり、この
RSP溶液は透明で黄褐色〜赤褐色の液体である。 一般に、この発明の方法は2つの技術的操作か

(RSP)を調製する。重合速度は、例えば、着色
シツフ塩基の濃度を反映する可視スペクトルの範
囲(例えば550nm)における吸光の変化により
追跡することができ、この濃度は第一に両反応成
分、すなわち架橋剤及び共反応化合物の比率に依
存し、そして、反応時間と温度に依存する。反応

することができる。 特に、出発材料として未処理細胞を使用する場
合、そして又、固定化した粒子を処理して粒子の
機械的安定性及び耐性を付与する場合、特にアセ
トンもしくは他の有機溶剤により、又は好ましく
は部分乾燥後粒子表面に水不溶性被膜の保護層を
形成する有機溶剤中市販接着剤のごとき溶液によ
り粒子の部分脱水を行う場合には、この発明の固
定化細胞の製造工程中に、固定化前の細胞の透過
性付与処理又は固定化後の細胞凝集体の透過性付
与処理を含める。 この発明の固定化細胞の製造のために、一般
に、目的とする酵素活性(一種類の酵素又は複数
の酵素の)が原形質又は細胞内領域(細胞の細胞
質中)のいかなる場所に存在するかにかかわら
ず、種々の種又は菌株の原核微生物細胞及び真核
微生物細胞を使用することができ、さらに植物細
胞をも使用することができる。一般に、酵素の性
質及び酵素により触媒される生化学的反応のタイ
プは限定要因ではない。原核細胞又は真核細胞の
所定のタイプに従つて特定の化学的技法、例えば
凍結法、静置法、撹拌法等、そして特に固定化の
前及び/又は後における細胞への透過性付与技法

高度に寄生的な糸状菌、並びに植物細胞を使用す
ることができる。遺伝的改良、選択、集積法及び
遺伝的処理により得られる、固定化物が高い比活
性を有するようにした変異生物の細胞を使用する
のが有利である。 この発明の方法により得られる結合細胞(粒
子)の大きさを、目盛を付した接眼レンズを使用
して顕微鏡(光学顕微鏡)的に測定した。幾つか
の標品を走査電子顕微鏡で観察した。沈降性は次
の方法により試験した。すなわち、2.5gの湿潤

例 7 β−ガラクトシダーゼ酵素を含有するイー・コ
リーの新鮮な未処理細胞ペーストを、例1に記載
した24時間反応法により調製したRSP溶液100ml
に懸濁した。但し、両反応成分の出発濃度は、セ
デイプールCL−930を5重量/容量%、グルタル
アルデヒドを2.0容量%とした。隻鮮な未処理細
胞の比活性は、0.42μmolグルコース+ガラクト
ース/分/g湿重量(37℃、PH7.0、基質として
ラクトース溶液を使用)であつた。 均一な細胞懸濁液を室温に2時間静置すること
により反応せしめた。この後、細胞凝集体のスポ
ンジ状沈澱物を前記の方法により吸引過し、得
られた湿ケーキ状の生成物を真空布袋に移し、
袋の縁を折り曲げて金属製クリツプで止め、この
袋を2枚の金属板の間に水平に配置し、室温にて
2時間、1.25×104KPa(油圧)の過圧をかけ
た。この後、円板状になつた固形物を細片に破砕
し、これを食品ミキサー中の100mlの水に移し、
ミキサーのスイツチを断続しながら低速回転によ
り小粒子に破砕した。破砕及び均一化の後、生成
物を500mlの水で3回デカントし、十分に吸引
過し、重量を測定し(16.2g)、そして0.2%の界
面活性剤(スロバフオール910)及び1%のジメ
チルスルホキシドを含有する1MNaCl溶液100ml
中に移し、この懸濁液を40℃で平衡化し、そして
3時間連続混合した。透過性付与処理の後、粒子
を1000mlの飲用水で3回デカントし、前記の方法
により十分に吸引過し、PH7.0の0.1M燐酸緩衝
液に移し、そして一夜膨潤せしめた。 固定化細胞を過し、そして洗浄した後、非常
に急速に沈降する小塊状の不規則な暗色不定形の
総重量13.2gの湿潤物を得た。このものの比活性
は0.12μmolグルコース+ガラクトース/分/g
湿重量であり、その平均粒子サイズは0.5〜2.6mm
であつた。 例 8 例7の場合と同様のβ−ガラクトシダーゼ活性
を有するイー・コリーの未処理細胞30gを、0.5
%の界面活性剤(スロバフオール910)を含有す
る0.5MNaCl溶液100mlに加え、この懸濁液を強
力な混合によつて十分に均一化し、そして500ml
の丸底フラスコに移し、このフラスコをアルミニ
ウム箔で覆い、ロータリーシエーカー(260回/
分)に装着した。未処理細胞の懸濁液を30℃にて
20分間連続的に混合することにより細胞に透過性
を付与した。次に、この細胞を遠心分離し
(20000g/10分間)、上澄液を廃棄し、ペースト
状細胞沈澱物の重量を測定した。 上記のように処理した湿細胞26gを、例2にお
いて記載したRSP溶液100ml中に懸濁し、懸濁液
を均一化し、そして例2に記載した方法に従つて
反応混合物を凍結することにより細胞を固定化
し、固定化した細胞を例2に記載した方法により
処理し、こうして得た湿粒子17gをあらかじめ−
20℃に冷却した50mlの純アセトンに懸濁した。ア
セトン処理を3分間で終了し、処理物を手早くフ
リツトガラス上で吸引過し、水で洗浄し、そし
てPH7.0の0.1M燐酸緩衝液中で、8℃にて一夜膨
潤せしめた。 こうして、0.38μmolグルコース+ガラストー
ス/分/g湿重量のβ−ガラクトシダーゼ比活性
を有する総重量12.6gの固定化湿細胞を得た。こ
のものの平均粒子サイズは85〜400μmであり、
不規則な縁を有する小板状をしており、そして5
分間の静置の後定量的に沈降した。 例 9 38アゾカゼイン単位/g湿重量(37℃、PH7.5、
基質としてアゾカゼイン使用)を有する指数増殖
期の終了前に集菌したバシルス・メガテリウム
(Bacillus megaterium)の新鮮な未処理湿細胞
2.5Kgを、10000mlの脱塩水に懸濁し、この懸濁液
を激しい混合により均一化し、そして5℃に冷却
した。この懸濁液を細孔機械的破砕機中で250気
圧にて3回破砕した。それぞれの破砕処理の間に
連続的に冷却し、破砕混合物の温度が35℃を超え
ないようにした。 全量1000mlの破砕細胞懸濁液のPHを、連続混合
しながら20%NaOH溶液により7.5に調整し、破
砕懸濁液中の最終濃度が0.25容量%となるように
凝集剤(セデイプールKA)を加えた。この懸濁
液を十分に混合し、約1時間室温に静置し、この
間に細胞片を完全に凝集せしめた。 この後、例1に記載したようにして24時間反応
により調製したRSP溶液2000mlを、1000mlの懸
濁液に加えた。但し、例1と異なり、RSPの調
製における両反応成物の出発濃度は、セデイプー
ルKAを4重量/容量%、グルタルアルデヒドを
2容量%とした。あらかじめ形成した凝集粒子が
破壊されるのを回避しながら、両成分をステンレ
ス製撹拌棒によりゆつくりと混合した。2時間静
置した後、粘度の高い部分的に反応した懸濁液を
ゆつくり混合し、そして約0.3づつを10個のポ
リエチレン袋に移した。さらに、例4の場合と同
様にして処理した。但し、固定化粒子は、激しく
撹拌しながら、あらかじめ−20℃に冷却したアセ
トンにより3分間処理した。そして、この処理物
を手早く吸引過し、水で数回デカントし、そし
てPH7.5の0.1M燐酸緩衝液1000mlに移し、そして
8℃にて一夜膨潤せしめた。 こうして得た懸濁液を篩上で洗浄しながらゆつ
くりと処理することにより1.28Kgの湿固定化粒子
を得た。この固定化物の比活性は3.62アゾカゼイ
ン単位/g湿重量であり、平均粒径サイズは0.4
〜2.0mmであり、そして2分間の静置により粒子
が定量的に沈降した。 例 10 蛋白質分解性を有するバシルス・メガテリウム
の新鮮な未処理湿細胞50gを、0.05%NaCl水溶
液100mlに懸濁し、このPHを例9において記載し
たように20%NaOHにより7.0に調製した。懸濁
液を均一化した後PHを再度7.0に調整し、凍結乾
燥した卵リゾチームを最終濃度が100μg/mlに
なるように添加し、30分後、NaOHにより懸濁
液のPHを注意深く8.6に調整し、そして、この懸
濁液を、該懸濁液の粘度が急激に変化し、そして
細胞壁が酵素的に分解されるまで数分間50℃に加
熱した。 例9に記載した反応成分を含有するRSP溶液
100mlを、あらかじめ凝集せしめることなく、懸
濁液に加えた。この懸濁液を激しく混合し、さら
に例8(反応混合物の凍結)と同様に固定化し、
粒子をアセトンで処理した。 2.4アザカゼイン単位/g湿重量の活性を有す
る固定化湿粒子30gを得た。平均粒子サイズは
0.2〜1.0mmであり、3分間の静置の後定量的に沈
降した。 例 11 0.017μmolグルコース+ガラクトース/分/mg
湿重量のβ−ガラクトシダーゼ(37℃、PH6.0、
基質としてラクトースを使用)を含有する酵母ク
ルイベロミセス・フラギリス(Kluyveromyces
fragilis)の未処理湿細胞ペースト30gを、0.1%
の界面活性剤(スロバフオール910)及び1%の
ジメチルスルホキシドを含有するPH7.0の0.1燐酸
緩衝液100ml中に懸濁し、そしてこの懸濁液を40
℃に加熱し、30分間連続撹拌した。この後、透過
性を付与した細胞を遠心分離し(5000g、10分
間)、上澄液を廃棄し、そして沈澱した細胞ペー
ストの重量を測定した。 透過性を付与した湿細胞25gを、例8の方法に
従つて固定化した。但し、固定化後の粒子をアセ
トン中カナゴン1%溶液で処理した。処理及び膨
潤における他の条件は例8に記載した通りとし
た。 0.02μmolグルコース+ガラクトース/分/mg
湿重量の比活性を有する固定化湿細胞15gを得
た。粒子サイズの分布は0.2〜1.5mmの範囲であ
り、そして粒子は2分間の静置の後定量的に沈降
した。凝集体は線状であり、その沈澱物は比較的
大であつた。 例 12 比活性5.9μmolL−リジン/時/mg湿重量(37
℃、PH7.5、L−α−アミノ−ε−カプロラクタ
ム水溶液)のL−α−アミノ−ε−アミノカプロ
ラクタムヒドロラーゼ活性を有する酵母クリプト
コツカス・ラウレンテイー(Cryptococcus
laurentii)の新鮮な未処理細胞25gを、例2の方
法に従つて調製したRSP溶液100mlに懸濁した。
1時間ゆつくり撹拌した後、高粘性線状懸濁液
を、例2の凍結技法を用いて固定化し、固定化後
の凝集体を例2の方法に従つてアセトン中接着剤
溶液により処理した。吸引過して得られた凝集
体を、8℃においてPH7.0の燐酸緩衝液中で一夜
膨潤せしめた。 3.2μmolL−リジン/時/mg湿重量の比活性を
有する固定化湿細胞15gを得た。平均粒子サイズ
は0.125〜1.5mmの範囲であり、この定量的沈降は
2分間の静置の後に生じた。この固定化物は、肉
眼的には不定形の線状構造を有し、顕微鏡的には
不規則な縁を有する不規則な構造を有していた。 例 13 7.1単位/g湿重量(1単位は30℃、PH5.5にお
いて5分間でL−アスパラギン酸から100μの
CO2を発生する酵素量である)の比活性のL−ア
スパラギン酸脱炭酸酵素を含有するミコバクテリ
ウム・エスピー(Mycobacterium sp.)の新鮮
な未処理細胞の湿ペースト30gを、例2の場合と
同じRSP溶液100mlに懸濁し、均一な懸濁液を生
成せしめた。そして例2と同様にして反応混合物
を凍結し、デカントし、洗浄し、そして吸引過
し、そして19.6gの固定化湿細胞を得た。 上記の量の粒子を、0.5%の界面活性剤(スロ
バフオール910)を含有するPH6.5の1MNaCl溶液
100mlに懸濁した。この懸濁液にクロロホルム
(5容量%)を加え、そして混合しながらこの懸
濁液の温度を35℃にし、この温度において1時間
激しく混合した。これを1000mlの水で希釈し、
0.5の水で3回デカントし、透過性を付与した
粒子を前記のように吸引過し、あらかじめ−25
℃に冷却した50mlのアセトンに懸濁し、そして室
温にて3分間激しく混合した。粒子を再度吸引
過し、フリツトガラス上で十分に水洗し、PH6.0
の0.05M酢酸緩衝液50mlに懸濁し、そして8℃に
て一夜膨潤せしめた。 4.0単位/g湿重量の比活性を有する固定化湿
細胞14.2gを得た。粒子サイズ分布は75〜800μm
の範囲であつた。この粒子は、肉眼的には湿潤表
面の少ない不定形線状フレーク形を有していた。
固定化細胞はほとんど自然沈降しなかつた。すな
わち、粒子は液の上層に残留した。しかし過性
及び吸引過性は良好であつた。 例 14 比活性16.4単位/g湿サンプル(1単位は、30
℃、PH5.5においてL−リジンから100μのCO2
を発生せしめる酵素量)のL−リジン脱炭酸酵素
を含有するバクテリウム・キヤダベリス
(Bacterium cadaveris)の新鮮な未処理湿細胞
30gを、例2の場合と同様のRSP溶液100mlに懸
濁した。さらに例2の場合と同様にして凍結、デ
カント、洗浄、及び吸引過を含む処理を行つ
た。24gの固定化湿細胞を得た。 上記の量は吸引過粒子を、0.1%の界面活性
剤(スロバフオール909)を含有するPH6.5の
1MNaCl溶液100mlに懸濁し、この懸濁液を37℃
にて1時間連続混合した。固定化し、そして透過
性付与処理を行つた菌体を、例13に記載した方法
によりさらに処理した。 11.8単位/g湿重量の比活性を有する固定化湿
細胞19.1gを得た。この固定化物は、平均粒子サ
イズが180〜600μmであり、粒子は顕微鏡的には
明瞭な小板状であり、肉眼的には不定形褐色粒子
であり、3分間の静置の後定量的に沈降した。 例 15 148単位/g湿重量(30℃、PH4.75、基質とし
てシユークロースを使用)の比活性のインベルタ
ーゼを含有する酵母サツカロミセス・セレビシエ
ー(Saccharomyces cerevisiae)の新鮮は未処
理湿細胞20gを、例2の場合と同じRSP溶液100
mlに懸濁し均一な懸濁液を得た。さらに、例2に
記載した方法により、凍結及びその他のすべての
操作を含む固定化処理を行つた。細胞を例8と同
様にアセトンで処理した。 90単位/g湿重量の比活性を有する固定化湿細
胞12.1gを得た。粒子サイズの平均分布は0.32〜
2.0mmであり、粒子は、顕微鏡的には縁に亀裂を
有する不規則な形状の不明瞭な構造を有し、肉眼
的には淡褐色〜ベージユ色の不定形粒子であり、
2分間の静置の後定量的に沈降した。 例 16 酵母サツカロミセス・コレアヌス
(Saccharomyces coreanus)を液体培養した後
の培地1を遠心分離〔ジツネツキ(Janetzki)
S−70、3000g、10分間〕し、上澄液は廃棄し、
そして新鮮な未処理湿菌体28gを得た。 こうして得た量のペースト状の新鮮な酵母菌体
を、例2に記載した方法により調製したRSP溶
液100mlに懸濁し、この懸濁液を激しく混合して
十分に均一化し、そして反応混合物を小型往復式
実験室振とう機に装着し、温度にて1時間振とう
(50回/分)した。そして混合を停止し、懸濁液
を上記の温度にて3時間静置した。この後、生成
した凝集体の高粘性スポンジ状マツシユを500ml
のポリエチレン製キユベツトに移し、そして上記
のようにして遠心分離し、上澄液を廃棄し、沈澱
物は脱イオン水に懸濁し、そしてゆつくり混合し
て250mlの水で3回デカントした。凝集体のデカ
ント及び定量的沈澱の後、固定化物を真空過
し、約100mlの水で再度洗浄し、そして重量を測
定した(14g)。これをあらかじめ−25℃に冷却
したアセトン25ml中に懸濁して強く混合し、フリ
ツトガラス上で手早く吸引過しそして約1000ml
の脱イオン水で洗浄し、再度十分に吸引過し、
そして重量を測定した(9g)。得られた固定化
湿菌体を上記の遠心分離上澄液(酵母を培養した
後の培地)に懸濁し、そして室温にて数時間膨潤
せしめた。 固定化物を再度十分に真空過し、湿菌体100
mgをワールブルグ容器に入れ、遠心分離した透明
な発酵液(PH4.6)を2ml加え、12.5%シユーク
ロース0.5mlをワールブルグ容器の側室に入れ、
第1の対象容器には未処理固定化菌体(100mgの
未処理湿ペースト)を入れ、そして2mlの遠心分
離した発酵液を加えた。第2の対象容器には上澄
液のみ(菌体を加えない)を入れ、そしてシユー
クロースを側室に入れ、そしてマノメーターを取
り付け、振とう機のスイツチを入れ、30℃にて15
分間インキユベートした。マノメーターの脱気を
行い、シユークローズ溶液を側室から注入し、そ
して発生するCO2の測定を開始した。 第1の対象(未処理未固定化菌体)においては
3064μ/時のCO2が発生し、第2の対象(菌体
を含まない上澄液のみ)ではCO2の発生がなく、
第3の測定(第1の対象の場合と同濃度の固定化
菌体を使用)においては165μ/時のCO2が発生
した。この量は、最初の(固定化してない)菌体
の全代謝活性の5.4%に相当する。 酵母固定化物は、顕微鏡的には不規則な形状の
明瞭な粒子として観察され、肉眼的にはベージユ
色の不定形粒子として観察された。1分間の静置
により定量的に沈降し粒子サイズの分布は300〜
900μmであつた。 例 17 比活性100μmolL−アスパラギン酸/分/g湿
重量(37℃、PH5.0、基質としてL−アスパラギ
ンを使用)のL−アスパラギンアミノヒドロラー
ゼ酵素を含有するエルウイニア・アロイデア
(Erwinia aroidea)の新鮮な未処理湿菌体ペー
ストを、例7に記載した方法によりRSP溶液100
ml中に懸濁した。この懸濁液を激しく混合して均
一化し、例16の場合と同様に30分間ゆるやかに撹
拌し、そして1時間室温において。この後、粘調
なスポンジ状物を500mlのポリエチレン製キユベ
ツトに注入し、5000×gで30分間遠心分離した。
上澄液を廃棄し、例7に記載したようにして沈澱
物に過圧をかけた。但し、沈澱粒を約5分間油
圧手段により圧搾し(RSP液部分を搾除する)、
脆い固形物を破砕し、特殊な鋼製の1mm孔径のイ
ンサートを有する肉用ホモジナイザーに入れ、こ
のインサートを通して棒状顆粒形に押出し、この
顆粒を25℃にて約3時間空気乾燥し、水で3回デ
カントし、脱イオン水中で一夜膨潤せしめた。お
よび1〜3mmの長さの棒状で不規則な粒子サイズ
を有する比活性64μmolL−アスパラギン酸/g湿
重量の固定化菌体の総重量8gの顆粒状固形物を
得た。このものは数10秒間で定量的に沈降した。 例 18 比活性0.21μmolフラクトース/分/g湿重量
(60℃、PH6.85、基質としてグルコースを使用)
のグルコースイソメラーゼ酵素を含有するストレ
プトマイセス・パエオクロモゲネス
(Streptomyces phaeochromogenes)の新鮮な
未処理湿菌体ペースト50gを、PH7.0の0.1MNaCl
溶液200mlに懸濁し、この懸濁液を70℃にて10分
間連続撹拌しながら加熱した。次に、この懸濁液
を冷却し、そして遠心分離した(5000×g、15
分)。上澄液は廃棄し、沈澱物は、例7の方法に
従つて調製したRSP溶液150mlに懸濁した。さら
に、遠心分離、短時間の圧搾、破砕、及び乾燥を
含む例17に記載した方法による固定化操作を行つ
た。但し、後成形した固定化菌体の顆粒は、2%
の接着剤(カナゴン)を含有するあらかじめ冷却
したアセトン50mlで−28℃にて3分間処理した。
そして粒子を手早く吸引過し、約1000mlの
0.05M燐酸緩衝液で洗浄し、再度吸引過し、そ
して100mlの同じ緩衝液に懸濁し、そして8℃に
て一夜貯蔵した。 総量20gの固定化菌体の顆粒状湿固形物を得
た。比活性は0.1μmolフラクトース/g湿重量で
あり、粒子は小棒状であり、粒子サイズは不規則
で0.5〜3.0mmの範囲であつた。固定化物は数10秒
の間に定量的に沈降した。 例 19 比活性0.1μmolH2O2/分/mg湿重量のグルコー
スオキシダーゼ酵素とカタラーゼを含有するアス
ペルギルス・ニガー(Aspergillus niger)の未
処理湿菌体ペースト45gを、例7の方法により調
製したRRP溶液150mlに加えた。さらに、例7に
記載した方法により固定化処理を行つた。但し、
後成形した顆粒は、室温にて24時間、40%湿度の
空気(H2SO4)で乾燥した。この後、菌体を脱
イオン水に移し、そして8℃にて24時間膨潤せし
めた。 0.045μmolH2O2/ml湿菌体の比活性を有する総
計29gの固定化湿菌体を得た。粒子サイズは不規
則で0.5〜3.0mmの範囲であつた。粒子の形は棒状
であつた。この固定化物は数10秒間に定量的に沈
降した。 例 20 シユークロースによる高張圧環境下でソランア
ルカロイド及びシトステロールを合成する植物ソ
ロニウム・アビクラレ(Solanum aviclare)(単
細胞培養)の新鮮な洗浄湿細胞20gを、例2の方
法に従つて調製したRSP溶液100mlに懸濁し、激
しく混合して得た均一懸濁液をアルミニウム製皿
(直径15cm)に移し、そしてこれを−19℃のフリ
ーザー中に4時間置いた。凍結した反応混合物を
フリーザーから取り出し、そして0.75MのKClを
含有するPH5.7の0.05M燐酸緩衝液約50mlで覆つ
た。2時間室温においた後、植物細胞の凝集体を
1000mlの上記の緩衝液に移し、常に粒子を定量的
に沈降せしめながらこの緩衝液で数回デカントし
た。次に固定化細胞を十分に真空過し、フリツ
トガラス上で緩衝液により洗浄し、再度吸引過
し、そして重量を測定した(18g)。 次に、粒子を、殺菌した8重量/容量%のシユ
ークロース水溶液100mlに移し、そしてこの懸濁
液を20℃にて24時間間隔で3回電磁撹拌機で撹拌
した。凝集体を分離した後、ソランアルカロイド
及びシトステロールの細胞外生産を、ジルク
(Jirku)等(Biotechnol.Letters、3、No.8、
447、1981年)により記載された方法により、
HPLCを用いて24時間間隔で追跡した。 24時間×3期の間、上記の固定化細胞による上
記の物質の生産が維持された。粒子を走査電子顕
微鏡で観察したところ、植物細胞相互間の結合が
認められた。 例 21 例20と同様の新鮮な未洗浄植物細胞30gを、
0.75MのKClを含有する0.05M燐酸緩衝液100mlに
懸濁した。混合しながら、均一な細胞懸濁液に凝
集剤(セデイプールKA)を加えた(0.5容量%)。
混合した後、細胞の凝集が視覚的に観察できるよ
うになるまで室温に1時間置いた。この後、懸濁
液を注意深く(生成したフレークが崩壊しないよ
うに)フリツトガラス上に移し、そして繊維
(布)を通してゆつくりと、且つ十分に真空過

この懸濁液を往復式実験室振とう機上で1時間混
合し、そして2時間静置し、次に粒子を遠心分離
し(10分間、5000g)、沈澱物を針を付けてない

かり型撹拌機)しながら行つた。使用した条件下
で酵素的転化は3時間で完結した。同じバツチの

出成分との混合物;及び/又は個々の細胞につい
て架橋処理及び透過性付与処理を施した細胞を化
学的に固定化し、場合によつてはさらに該固定化
物に透過性付与処理を施し、そして/又は該固定
化物を機械的に硬化せしめて成る生物的変換用細
胞触媒。 2 ヒドロラーゼ、イソメラーゼ、リアーゼ、脱
炭酸酵素、酸化還元酵素、もしくは他の酵素、及
び/又は生化学的代謝経路の一部を成す酵素群、
もしくはホルモンの生物変換もしくはアルカロイ

菌、放線菌、酵母、糸状菌、もしくは植物細胞、
又は前記の細胞の断片及び細胞内顆粒を含んで成
る特許請求の範囲第1項記載の細胞触媒。 3 分子量100000〜800000のポリエチレンイミン
とグルタルアルデヒドとの反応により生成した水
溶性、且つ化学的に活性な重合体により前記細胞
性材料を化学的に固定化して成る特許請求の範囲
第1項又は第2項記載の細胞触媒。 4 未処理細胞もしくは場合によつては透化性付
与処理を施した細胞、もしくはこれらの細胞の断
片及び細胞内顆粒;及び/又は未処理細胞と細胞
断片、細胞内顆粒及び細胞溶出成分との混合物;
及び/又は個々の細胞について架橋処理及び透過
性付与処理を施した細胞を、水性媒体中−30℃〜
50℃、PH5.0〜9.0の範囲において、そして場合に
よつては凝集剤の存在下で、ポリエチレンイミ
ン、ヘキサメチレンジアミン、リジン又はポリリ
ジンと二官能価アルデヒドとを水性媒体中で0℃
〜60℃、PH4.0〜12.0において反応せしめて得ら
れる水溶性且つ化学的に活性な重合体と接触せし
めることにより固定化し、反応混合物から固定化
物を分離し、水又は緩衝液で洗浄し、そして場合
によつては透過性付与処理及び/又は機械的硬化
処理を行うことを特徴とする生物的変換用細胞触
媒の製造方法。 5 分子量100000〜800000のポリエチレンイミン
を水溶性重合体の製造、及び場合によつては細胞
類の予備凝固に使用することを特徴とする特許請
求の範囲第4項記載の方法。 6 前記の細胞類と前記の重合体との反応を0℃
〜45℃において、そして1〜50000gの相対遠心
力下、又は0〜50000Kpaの過圧下で、静置し
て、もしくはゆつくり混合しながら、もしくは静
置と混合を繰り返しながら行うことを特徴とする
特許請求の範囲第4項又は第5項記載の方法。 7 固定化前の細胞又は固定化後の細胞凝集体の
透過絶付与処理を、水性環境中0℃〜70℃、PH
4.0〜9.0において、界面活性剤、及び/又は水混
和性有機溶剤及び/又は水不混和性有機溶剤の存
在下で、場合によつては強酸及び/又は塩基の塩
を加えることにより懸濁液中のイオン強度を一時
に又は逐次に変化せしめながら、細胞懸濁液を混
合することによつて行うことを特徴とする特許請
求の範囲第4項又は第6項記載の方法。 8 場合によつてはさらに、有機溶剤による部分
脱水により、又は有機溶剤もしくは有機溶剤と水
との混合物中接着剤溶液により、−25℃〜20℃の
範囲の温度において固定化細胞の機械的硬化を行
い、そしてこの固定化物を吸引過し、そして乾
燥条件下もしくは湿潤条件下で部分乾燥を行い、
又は場合によつては適当に後成形することを特徴
とする特許請求の範囲第4項又は第5項記載の方
法。
