【発明の詳細な説明】[Detailed description of the invention]

〔産業上の利用分野〕 本発明は外用剤に関し、更に詳細には特定のジ
アシルグリセリンを含有してなる皮膚親和性が高
く(皮膚へのなじみ性が良く)、かつ皮膚保湿持
続効果に優れた皮膚化粧品、毛髪化粧品、外用医
薬品等の外用剤に関する。 〔従来の技術及びその問題点〕 正常な皮膚の角質層には通常10〜30%の水分が
含まれており、弾力性と柔軟性が維持されている
が、これが環境条件等の原因で10%以下になる
と、いわゆるドライスキンと呼ばれる状態にな
る。例えば冬期乾燥時のひび、あかぎれ等の症状
である。 また、加齢とともに皮脂の分泌が減少するため
皮膚が乾燥しやすくなり、掻痒を伴い落屑が生じ
る老人性乾皮症のような症状が現われてくる。 これらを解決するための方法として、従来保湿
剤を用いて皮膚水和効果を高める方法と、油剤を
用いて経皮水分損失(T.E.W.L)を抑制する方法
があつた。 前者の例としては、グリセリン、プロピレング
リコール、ピロリドンカルボン酸ナトリウム等の
保湿剤が用いられてきたが、これら保湿剤の作用
は、いずれも皮膚に適用した場合、その効果は一
時的で持続性がなく、外部環境条件の影響を受け
やすいという欠点を有していた。 一方、後者の例としては、ワセリン軟膏が良く
知られているが、効果を高めるためには塗布量を
多くしなければならず、そのことにより使用中油
性感、べたつきが強く、肌へなじみにくいといつ
た不快な感触になり、また逆に塗布量が少ないと
閉塞数が弱く持続性がないという欠点を有してい
た。 〔問題点を解決するための手段〕 本発明者らはこうした実情に鑑み、皮膚親和性
が高く、かつ皮膚保湿持続効果に優れた外用剤を
得るべく鋭意研究した結果、特定の分岐飽和脂肪
酸とミリスチン酸(テトラデカン酸)とのアシル
残基を有するジアシルグリセリンを配合すること
により、肌へのなじみ性が良く、皮膚保湿持続効
果に優れていることを見出し、本発明を完成し
た。 即ち本発明は、次の一般式()で表されるジ
アシルグリセリンを含有することを特徴とする外
用剤を提供するものである。 〔式中、R1,R2,R3のうち1つは () 次式で表されるメチル分岐イソステアリン
酸残基、 (式中、m及びnはそれぞれ4〜10の整数で、
m+n=14であり、m=n=7を中心とする分
布を有する。) () 5,7,7−トリメチル−2−(1,3,
3−トリメチルブチル)−オクタン酸残基、 () 2−ヘプチルウンデカン酸残基よりなる群
から選ばれた分岐飽和脂肪酸残基を、1つはテ
トラデカン酸残基を、残余は水素を示す。〕 本発明に用いる一般式()で示されるジアシ
ルグリセリンは1分子中に前記の如き炭素数18の
分岐飽和脂肪酸残基(イソステアリン酸残基)と
テトラデカン酸残基を有するものであり、例えば
以下の方法で合成することができる。 (1) グリセリンの脂肪酸による直接エステル化。 炭素数18の分岐飽和脂肪酸及びテトラデカン
酸(ミリスチン酸)とグリセリンから通常のア
シルグリセリンを製造する方法によつて製造さ
れる。例えば、上記の分岐飽和脂肪酸及びミリ
スチン酸とグリセリンを等モル配合し、200〜
230℃に加熱し、生成する水を減圧下で除くこ
とによつて、これらのアシルグリセリン混合物
が生成する。この生成物を蒸留もしくはカラム
クロマト処理することによつてジアシルグリセ
リンフラクシヨンを得ることができる。ただ
し、このジアシルグリセリンはランダム混合物
であるため、つまり、分岐飽和脂肪酸残基のみ
から成るジアシルグリセリン、分岐飽和脂肪酸
残基とミリスチン酸残基から成るジアシルグリ
セリン及びミリスチン酸残基のみから成るジア
シルグリセリンの混合物であるため、これらの
融点差を利用して、更にヘキサン等の溶剤を用
いた再結晶化法により目的とする本発明のジア
シルグリセリンを得る。 ここで本発明のジアシルグリセリンの製造に
用いられる炭素数18の分岐飽和脂肪酸は、石油
化学工業原料又は油脂化学工業原料より容易に
得られる。 即ち、5,7,7−トリメチル−2−(1,
3,3−トリメチルブチル)−オクタン酸は、
イソブチレン2量体のオキソ反応により炭素数
9の分岐アルデヒドとし、次いでこのアルデヒ
ドのアルドール縮合により炭素数18の分岐不飽
和アルデヒドとし、次いで水素添加、酸化する
ことにより製造することができ、これは例えば
日産化学(株)より市販されている。2−ヘプチル
ウンデカン酸はノニルアルコールをゲルベ反応
(Guerbet reaction)に付し、次いで酸化する
ことにより製造することができ、これは例えば
三菱化成(株)より市販されている。また、メチル
分岐イソステアリン酸は、例えばオレイン酸の
ダイマー製造時の副産物として得られるもので
〔例えばJ.Amer.Oil Chem. Soc.,51、522
(1974)〕、例えば米国エメリー社などから市販
されている。 (2) 脂肪酸の反応性誘導体とモノグリセライドと
の反応。 より有利な製造法として分岐飽和脂肪酸もし
くはミリスチン酸のいずれか一方をより反応性
の高い誘導体に導きエステル化することもでき
る。反応性の高い誘導体の一つは、脂肪酸の酸
ハライドであり、これを経る場合の好ましい具
体例においては、先ず、原料分岐飽和脂肪酸
に、この分岐飽和脂肪酸1モル当たり1〜5モ
ル、好ましくは1〜2モル塩化チオニル、臭化
チオニル、三塩化リン等のハロゲン化試薬を0
〜100℃、好ましくは20〜80℃で反応せしめて
酸ハライドに導く。片やミリスチン酸1モルと
グリセリン1モルとを200〜230℃に加熱し、生
成する水を減圧下で除くことによつてミリスト
イルグリセリンが得られる。このものを、分子
蒸留することによつて、モノミリストイルグリ
セリンを得る。そこで前述した分岐飽和脂肪酸
ハライドと、分岐飽和脂肪酸ハライド1モル当
たり0.5〜3モル、好ましくは1〜1.5モルのモ
ノミリストイルグリセリンとを、酸ハライドに
対して1〜3モル、好ましくは1〜1.5モルの
ピリジン、キノリン等の脱ハロゲン化水素剤を
用い、例えばベンゼン、トルエン、キシレン、
ヘキサン等の適当な不活性溶剤中(酸ハライド
に対して1〜10倍容量、好ましくは(3〜5倍
容量)で50〜100℃、好ましくは60〜80℃で加
熱撹拌しながら反応させる。生成するハロゲン
化水素アミン塩を除去したアシルグリセリン
を、分子蒸留もしくはカラムクロマト処理する
ことによつて目的とする分岐飽和脂肪酸残基と
ミリスチン酸残基とを含むジアシルグリセリン
を得ることができる。 (3) リパーゼを用いる方法。 更に有利な製造方法として反応性の高い誘導
体を経ることなく、リパーゼを用いて製造する
ことができる。先ず、分岐飽和脂肪酸1モルと
グリセリン1モルとを200〜230℃に加熱し、生
成する水を減圧下で除くことによつて、分岐飽
和脂肪酸残基を有するアシルグリセリンを得
て、分子蒸留を通して分岐飽和脂肪酸残基を有
するモノアシルグリセリンを得る。このモノア
シルグリセリンとミリスチン酸を等モル量配合
し、リパーゼを触媒としてエステル化反応を行
うことによつて目的とする分岐飽和脂肪酸残基
とミリスチン酸残基とを含むジアシルグリセリ
ンが得られる。エステル化の程度に応じて、後
処理として分子蒸留を行う。例えばミリスチン
酸のエステル化率が低い場合は、残余のミリス
チン酸及びモノアシルグリセリンを除去する必
要がある。また、トリアシルグリセリンの副生
も分子蒸留カラムクロマトにより除去すること
ができる。リパーゼを触媒として用いる場合、
α−位選択性を有するリパーゼあるいは部分グ
リセリドに選択的なリパーゼは最も有効であ
る。 本発明のジアシルグリセリンの外用剤における
配合量は第1表に示した通り、剤型に応じて幅広
く変動し得るものの、概ね0.1〜90重量%、好ま
しくは0.1〜50重量%の範囲である。 一方、本発明で使用される水以外の他の原料成
分としては、通常、外用剤に適用される油分、界
面活性剤、多価アルコール類を含む保湿剤、各種
薬効剤、防腐剤、香料等、ほとんどの成分が挙げ
られ、その中から目的、用途、剤型等に応じて適
宜選択され、配合される。例えば、油分について
は、ステアリン酸、ミリスチン酸、セタノール、
ステアリルアルコール、コレステロール、ワセリ
ン等に代表される固体脂とスクワラン、コレステ
リルイソステアレート、ホホバ油、オリーブ油、
ジカプリン酸ネオペンチルグリコール、流動パラ
フイン等に代表される液体脂がある。一方、多価
アルコールについては、グリセリン、1,3−ブ
チレングリコール、プロピレングリコール、ジプ
ロピレングリコール、ポリエチレングリコールな
どが挙げられる。更に界面活性剤については、ポ
リオキシエチレン硬化ヒマシ油、ソルビタン脂肪
酸エステル、ポリオキシエチレンソルビタン脂肪
酸エステル、ポリエチレングリコール脂肪酸エス
テル、アルキルグリセリルエーテル、脂肪酸塩、
アルキルリン酸塩などが挙げられる。 また、本発明の対象となる外用剤としては、軟
