Description
本発明は新規なヘテロポリサツカライド、水性
栄養培地中での細菌培養によるその製造方法、お
よびそのヘテロポリサツカライドを産性する微生
物に関する。本発明はまた、このヘテロポリサツ
カライドを含有する組成物に関する。
細菌の特定の菌株または菌種を用い、微生物生
合成によつてある種のヘテロポリサツカライドが
得られることはよく知られている。たとえば特公
昭42−7600号には、「271」として知られるバチル
ス・ポリミキサ(Bacillus polymyxa)の菌株
を、グルコースまたはラクトースを含有する培地
中で培養し、グルコース、マンノース、ガラクト
ース、およびグルクロン酸、約8:7:3:5の
割合で構成されるヘテロポリサツカライドの生産
が報告されている。このヘテロポリサツカライド
1%溶液の25℃における粘度は6000cPと記録さ
れている。比旋光能[α]28 D=+92゜、分子量は約
10000であるという。
またケルコ・コーポレーシヨン(Kelco
Corporation)の1973年3月1日付商品パンフレ
ツトには、キサンタンガム(xanthangum)とし
て知られた製品は微生物キサントモナス・カンペ
ストリス(Xanthomonas campestris)の発酵に
よつて生成したゴムで、3種のモノサツカライ
ド、すなわちマンノース、グルコースおよびグル
クロン酸を含有すると記載されている。キサンタ
ンガムの水溶液は擬可塑性(pseudoplastic)を
有し、1%溶液の粘度は剪断速度1 sec-1で
7000cPのオーダーであり、分子量は少なくとも
2000000のオーダーと考えられている。
この種のポリサツカライドの一部は、とくにた
とえば食品、化粧品、薬品などの水系における増
粘剤または懸濁液に用いられてきた。しかしなが
ら、一般に、微生物産生ポリサツカライドはこの
種の製品に用いる場合、増粘剤または懸濁液とし
ての機能の面で限界のあることがわかつていた。
たとえばこの種のポリサツカライドを含む薬品
は、たとえば狭い孔を通して充填または排出させ
る時のように、剪断力がかかると不安定である。
このような限界は製品中の微生物産生ポリサツカ
ライド濃度を増加させることで一部解決できる
が、他の点で製品の性質を変えるし、また原料価
格を上昇させることになる。したがつて、改良さ
れた増粘剤または懸濁液として価値があり、上述
のような欠点のない新規なポリサツカライドを発
見するための研究が続けられていた。
本発明はバチルス・ポリミキサに属する細菌種
のある菌株で栄養培地を発酵させると、以下バイ
オポリマーPS87と呼ぶ新規なヘテロポリサツカ
ライドが得られることを発見し完成されたもので
ある。このヘテロポリサツカライドはたとえば食
品、化粧品、薬品の増粘剤または懸濁液として有
用であり、また予期に反して優れた擬可塑性を有
する。
すなわち、本発明はグルコース、ガラクトー
ス、マンノース、グルクロン酸から構成され、そ
の1重量%溶液は擬可塑性を有し、20℃における
硬度は少なくとも150ポアズであり、20℃におけ
る降伏価は少なくとも30ダイン/cm2であるポリサ
ツカライド、すなわちバイオポリマーPS87を提
供するものである。
バイオポリマーPS87は化学分析の結果、少な
くともグルコース、ガラクトース、マンノースお
よびグルクロン酸の結合モノサツカライド残基か
ら構成されることが明らかにされている。このモ
ノサツカライド残基の分析は、ヘテロポリサツカ
ライドの2M−トリクロロ酢酸1%溶液を封管中
121℃において1時間加水分解したのちに実施し
た。かくして得られたモノサツカライド含有加水
分解物を水酸化アンモニウム中、20℃において水
素化ホウ素ナトリウムで1時間還元した。生成し
たアルジトールをピリジン中無水酢酸でアセチル
化した。このアセチル化生成物をシアノ−シリコ
ンOV225カラムを用い、190℃の等温クロマトグ
ラフイーに付し、標準物質と比較した。
グルクロン酸はヘテロポリサツカライド加水分
解物を塩酸で処理してラクトンに導き、グルコノ
ラクトントリメチルシリルエーテル誘導体を生成
させて同定した。
グルクロン酸は改良カルバゾール法を用いて定
量した。
バイオポリマーPS87は少量のフコースを含有
することもある。
この分析の結果から、バイオポリマーPS87中
に同定される構成モノサツカライド残基それぞれ
の重量%を計算することができる。
数回の分析から得られた結果は、バイオポリマ
ーPS87中に存在する各モノサツカライドの重量
%は次の範囲内にあることを示している。
グルコース 40〜45重量%
ガラクトース 10〜20重量%
マンノース 25〜30重量%
グルクロン酸 6〜13重量%
フコース 0〜1.5重量%
各百分率範囲の限界は、発酵の際に使用した培
地、培養条件たとえば温度、PH、酸素分圧など、
細菌発酵によるバイオポリマーPS87の異なるサ
ンプルの分析結果における変動を示している。
たとえば、以下に記述するような特定の発酵条
件で得られたバイオポリマーPS87の構成モノサ
ツカライド重量%は
グルコース 41重量%
ガラクトース 12重量%
マンノース 28重量%
グルクロン酸 9重量%
フコース 0.8重量%
であり、このバイオポリマーサンプルは以下の比
率のモノサツカライドで構成されていることにな
る。

上述したヘテロポリサツカライドの分析方法は
上述のモノサツカライドに関する分析結果を得る
のに現実に使用した方法であるが、バイオポリマ
ーPS87の組成の決定には他のモノサツカライド
分析法も使用できることは当然である。
バイオポリマーPS87は、比旋光能[α]25 D=+
60を示し、Chromatix KMX−6を用い低角度
レザー光散乱で測定したところ分子量は34±4×
104である。分子量の測定は25℃、濃度0.01重量
%から0.1重量%までの濃度範囲において、バイ
オポリマーPS87のサンプルを0.1M−KClに平衡
化させて実施した。
バイオポリマーPS87の水溶液は予期しない高
い硬度と予期しない高い降伏価を示す点で特徴的
である。
溶液の硬度はその溶液の剪断速度1 sec-1に
おける見かけの粘度と定義される。粘度は剪断応
力と剪断速度の比である。
バイオポリマーPS87の粘度は5cmのコーンと
プレートを付したWeissenberg Rheognimeterを
用い20℃で測定した。この装置はSangamo
Weston Ltd.,North Lersted,Bognor Regis,
Sussex,UKの製品である。バイオポリマー
PS87の0.5%および1.0%溶液の剪断速度
1.117sec-1における粘度はそれぞれ86および182
ポアズであつた。低剪断速度0.353sec-1では0.5%
および1%溶液でそれぞれ176および440ポアズの
値が得られた。これらのデータはこれらの溶液が
剪断減粘性すなわち擬可塑性を有することを示し
ている。この結果を補間すると、1%溶液の硬度
は約205ポアズ、0.5%溶液の値は約93ポアズとな
る。
したがつて一般的に、バイオポリマーPS87の
1%溶液は20℃において硬度少なくとも150ポア
ズ、好ましくは少なくとも200ポアズを示すとい
える。
溶液の降伏価は、溶液が流動し始めるまで溶液
に加えなくてはならない剪断応力である。バイオ
ポリマーPS87の溶液(1%w/w)は降伏価を
有しており、その降伏価の大きさは、理論的に
は、溶液に徐々に応力、例えば回転粘度計中の捩
じれた針金のトルクによつて発生する応力、を与
えることによつて測定することができる。流れが
生じた瞬間に適用されていた応力を測定すればよ
い。しかしながら、実際は、降伏価より大きな応
力を与え、ついで応力を除き、粘度計を平衡位置
まで戻らせることにより、もつと簡単に正確な降
伏価を測定できる。溶液に降伏価があれば、平衡
位置は応力のない系でのはじめの位置にはこな
い。捩れた針金に残余トルクが溶液の降伏価と平
衡に達するからである。すなわち、残余トルクを
測定すれば溶液の降伏価が得られる。この応力弛
緩法を用い、5cmのコーンとプレートを付した上
述のWeissenberg Rheogoniometerによつてバイ
オポリマーPS87の1%溶液の降伏価を測定した。
温度20℃、75時間後(平衡に達したとき)の値は
54ダイン/cm2であつた。
したがつて一般的に、バイオポリマーPS87の
1%溶液は20℃において、降伏価少なくとも30ダ
イン/cm2、好ましくは少なくとも50ダイン/cm2を
示すといえる。この濃度および温度条件下に30ダ
イン/cm2というような高い降伏価を示す擬可塑性
ポリサツカライドはほかに知られていない。実
際、多くのポリサツカライドは降伏応力を示さな
い。
バイオポリマーPS87の高い硬度と降伏価は、
またこのヘテロポリサツカライドの、たとえば砂
のような固体粒子の懸濁能を検討しても明らかで
ある。この懸濁能を明らかにするため、ヘテロポ
リサツカライドの0.5%w/v蒸留水溶液を調製
した。44〜69メツシユの酢洗浄シリカ5gを内径
45mmの容器中でヘテロポリサツカライド溶液100
ml中に懸濁した。温度25℃±2℃で懸濁させ、シ
リカの沈降程度を観察した。
7日後にもシリカの76重量%が溶液の上部80容
量%内に残つていた。同様に試験した他のポリサ
ツカライドで2日以上懸濁させるものはなかつ
た。
本発明はまた、バイオポリマーPS87産生微生
物株に関する。
バイオポリマーPS87の製造に用いて好ましい
微生物はバチルス・ポリミキサの特定の菌株また
は変異株である。
この微生物は単純な糖密と鉱酸塩を加えた寒天
培地上に海水を張つて、30℃でインキユベートす
ることにより単離された。ムコイドまたは粘稠な
外観を呈して生育したコロニーを同じ培地の新た
なプレート上に画線培養し、この微生物の純粋単
離を行つた。
純粋培養は液体糖密含有培地を用い、振盪フラ
スコ中30℃で行い、満足できる生育パターンと、
それに伴つてポリサツカライドの産性を示す粘度
の増加を確認し得た。
バチルス・ポリミキサの純粋培養では様々な不
透明度のコロニーが得られる。透明および不透明
コロニーは継代培養によつて分離し、この差は胞
子形成の程度に関係することが明らかにされた。
不透明なコロニーほど多くの胞子を形成してい
る。4種のコロニーを取り、別個に生化学的試験
に付した。結果は以下に記載したように、すべて
同一であつた。
栄養寒天上ではコロニーは小さく、平らで、白
い。馬鈴薯−イースト−グルコース寒天上ではも
つと大きく、黒く、ゴム状である。
この微生物は中程度の大きさの、真直ぐで端の
丸い、運動性グラム陽性桿菌であつて、胞子を形
成する。胞子は、未端近くで形成され、大きく、
卵型で、厚い壁をもち、細胞の膨脹を引き起こ
す。
生育試験および生化学試験の結果を以下にまと
める。

上述の結果およびBergyの“Manual of
Determinative Bacteriology”(8版)の記載か
ら、この微生物はバチルス・ポリミキサの菌株で
あつた。
この微生物の株はthe National Collection of
Industrial Bacteria,Torry Research Station,
PO Box 31,135Abbey Road、Aherdeen,
Scotlandに、1978年8目2日寄託された。寄託
番号はNCIB 11429である。
このバチルス・ポリミキサの特定の菌株を用い
るほか、バイオポリマーPS87の生産は、バイオ
ポリマーPS87の合成に適合された遺伝的改変細
菌を用いても可能である。
すなわち、本発明はまた、遺伝的形質転換又は
接合、或いはバイオポリマーPS87合成に関与す
るDNAを各々保有するプラスミド又は組換えプ
ラスミド又はフアージ又は組換えフアージ又は他
のベクターによる改変、によつて得られるバイオ
ポリマーPS87合成能を有する微生物に関する。
改変微生物の母体となり得る微生物としては、
大腸菌(E.coli)、シユードモナス
(Pseudomonas)種、クレブシエラ(Klebsiella)
種、及びバチルス(Bacillus)種を挙げることが
できる。
本発明はまた、バイオポリマーPS87合成機構
の遺伝子を保有する遺伝情報を母体となる微生物
に導入することによる遺伝的改変微生物の作製方
法に関する。
かかる方法の一つは、
a) バイオポリマーPS87合成に関与する遺伝
物質を欠く変異体を母体となる微生物から作る
工程、
b) バイオポリマーPS87合成に関与するDNA
と共有結合したプラスミドDNAから成る組換
えプラスミドを作製する工程、
c) 上記の組換えプラスミドをバイオポリマー
PS87合成能欠損微生物に導入する工程、
d) バイオポリマーPS87合成機構に適した生
育条件で得られる微生物を培養する工程、及び
e) バイオポリマーPS87合成機構に関して生
育した微生物のクローンを選択する工程、から
成る。
もう一つの方法は、
a) バイオポリマーPS87合成に関与する遺伝
物質を欠く変異体を母体となる微生物から作る
工程、
b) 母体となる微生物に対するフアージDNA
又はテンペレートフアージを固定する工程、
c) バイオポリマーPS87合成に関与するDNA
断片をフアージ又はフアージDNAに導入して、
組換えフアージを作製する工程、
d) バイオポリマーPS87合成能欠損微生物を
組換えフアージで溶原化する工程、
e) バイオポリマーPS87合成機能に適した生
育条件で得られる微生物を培養する工程、及び
f) バイオポリマーPS87合成機構に関して生
育した微生物のクローンを選択する工程、から
成る。
これらの改変を行うための適切な一般的方法は
英国特許第1521032号に記載されている。
プラスミドは、それが母体微生物内に移入し得
るものである限り、如何なる微生物から得たもの
でもよい。適当なプラスミド源としては、大腸
菌、シユードモナス種、クレブシエラ種、及びバ
チルス種が含まれ、特にバチルス・ポリミキサが
挙げられる。
組換えプラスミドは、母体微生物に存在するも
のも含めた多くのプラスミド源から直接作製する
ことができる。
フアージ法において、フアージを直接用いる場
合、フアージは母体微生物をその宿主とするもの
でなければならない。フアージDNAを用いる場
合は、本来のフアージもしくは母体微生物を宿主
としないフアージから抽出できる。
プラスミド又は組換えプラスミドを得る適切な
方法、及びフアージ又は組換えフアージの適切な
使用方法、及びフアージDNAの抽出方法は英国
特許第2003926号に記載されている。
本発明はまた、(i)バイオポリマーPS87合成に
関与する遺伝物質を有する微生物株を深部好気的
条件下に炭素源、窒素源、鉄源、マグネシウム源
及びリン源を含有する水性培地中で培養し、バイ
オポリマーPS87の実質的産性が起こるまで培地
のPHの値を4.5乃至7.5に保持する工程、及び(ii)培
地からヘテロポリサツカライドを単離する工程、
から成るバイオポリマーPS87の製造方法を供す
る。
遺伝物質は、例えば染色体DNA、プラスミド
DNA、組換えプラスミドDNA、フアージDNA、
又は組換えフアージDNAである。
本発明の好ましい態様においては、バイオポリ
マーPS87の製造プロセスはバチルス種の細菌、
好ましくはバチルス・ポリミキサを微生物として
使用する。かかる方法の一つは、(i)PH4.5乃至7.5
の水性培中で深部好気的条件下にバチルス・ポリ
ミキサNCIB11429をバイオポリマーPS87の実質
的産性が起こるまで培養する工程、(ii)培地を8以
上のPHで50℃以上に加熱する工程、(iii)次いで凝析
した菌体と菌体残渣を培地から分離して視覚上清
透なバイオポリマーPS87の溶液を得る工程、か
ら成る。
このプロセスはバツチ法でもまた連続法でも、
また他の適当な方法でも実施できる。
この本発明の方法の最初の工程を実施する好ま
しい方法によれば、適当な水性栄養培地にバチル
ス・ポリミキサNCIB11429を接種し、この菌体
を培養するため25乃至40℃、好ましくは30乃至35
℃の温度で、45乃至60時間インキユベートする。
水性栄養培地には通常、好ましくは少なくとも
1種のモノサツカライドまたはジサツカライドか
らなる炭素源を約1重量%から5重量%まで、好
ましくは約2重量%から3重量%までの濃度で含
有させる。適当な炭素源はたとえば、グルコー
ス、シユクロース、マルトース、フルクトース、
マンノース、デンプン、デンプン水解物またはコ
ーンシロツプである。炭素源としてはグルコース
を用いるのが好ましい。モノサツカライドまたは
ジサツカライドの粗製品たとえば糖密、乳漿もし
くは乳漿限外濾過液も使用できる。
培地には通常、天然有機物のような窒素源を含
有させる。たとえば大豆タンパク、大豆汁の酵素
分解液、醸造可溶成分、コーンステイープ液、イ
ースト抽出液またはカゼイン水解物が使用でき
る。水性培地中に有機窒素源を使用する場合、培
地に対して約0.05重量%から0.5重量%加え、窒
素重量%が約0.01から0.1までとなるようにする。
また、培地中に無機窒素源、たとえばアンモニ
ア、硝酸アンモニア、塩化アンモニア、硫酸アン
モニアまたは酢酸アンモニアを加えることもでき
る。無機窒素源の場合には0.5%までとする。ま
た、無機および有機窒素源の混合物を用いること
も可能である。培地には通常他の元素たとえばカ
リウム、ナトリウム、マグネシウム、リン、およ
び微量元素たとえばマンガン、鉄、亜鉛、カルシ
ウムおよびコバルトを添加する。これらは正常な
生育およびポリサツカライドの産生に必要であ
る。
培地にはまた、痕跡量、約0.5から約5μg/
のビオチンを加えることが好ましい。これはビオ
チン自体を加えてもよいし、イースト抽出液また
は糖密のようなビオチン源を加えてもよい。
細菌の適切な生育、バイオポリマーPS87の生
成には水性培地のPHが重要である。バイオポリマ
ーPS87の産生に至適なPHは4.5から7.5までであ
る。PHの調節は一般に培地に対し約0.1重量%か
ら1重量%までの濃度のリン酸二カリウムのよう
な緩衝剤の使用で達成できる。以下に挙げるよう
な任意のリン酸のナトリウム塩またはカリウム塩
すなわちKH2PO4,K2HPO4,K3PO4,
NaH2PO4,Na2HPO4またはNa3PO4が緩衝剤と
して使用できる。
必要があればPHは、適当な塩基源たとえばアル
カリ金属水酸化物たとえば水酸化カリウムまたは
ナトリウムと連動させたPHメーターを用いて調節
することもできる。細菌の生育が進むに従い、酸
生成でPHが低下するので、少量の水酸化カリウム
または水酸化ナトリウム溶液を自動的に加えてPH
を所望の範囲内に保つこともできる。
しかしながら、一般に、バツチ法の場合、本発
明の方法において培地のPHを調節するためにアル
カリを添加する必要はない。通常PHは10から20時
間後に約5に低下し、ついで約6から7に上昇
し、以後のインキユベーシヨン期間を通じて一般
にこの値に維持されることが観察されている。連
続発酵法の場合には、発酵培地のPHは必要に応じ
て上述のいずれかの方法で調節する必要がある。
急速かつ能率的にヘテロポリサツカライドを得
るためには、バチルス・ポリミキサNC11429の
生育のために十分な酸素を供給する必要がある。
培地中への酸素の供給が少なすぎると、細菌によ
るヘテロポリサツカライドの産性は低下するよう
である。撹拌および通気条件は、少なくとも0.05
g//時の移動速度で気相から液相に酸素が移
るようにする必要がある。
本発明の第二・第三の工程において、培地中の
菌体および菌体残渣から産生ヘテロポリサツカラ
イドを効果的に分離するためには、培地を50℃以
上の温度に加熱する前もしくは後で、培地をアル
カリと接触させて培地のPHを7以上に上昇させる
のが好ましい。こうすることによつて菌体及び菌
体残渣が凝析し、培地から容易に分離できるよう
になつて、ヘテロポリサツカライドの溶解した上
清が残る。
凝析した菌体及び菌体残渣の培地からの分離
は、カルシウム塩、例えば塩化カルシウム又は硝
酸カルシウム、の存在下で培地を加熱することに
よつてさらに容易になる。かかる目的に用いるカ
ルシウム塩の量は、培地の0.2重量%以下、好ま
しくは0.05乃至0.1重量%である。
培地上澄液から細胞および細胞残屑を分離する
ために、他のもつと慣用的な方法、たとえば上述
のような前処理を行わない濾過や遠心分離を用い
ても、うまく行かず、細胞および細胞残屑の除去
が不完全で、ヘテロポリサツカライドの水溶液は
常に濁つている。
培地からヘテロポリサツカライドを分離する好
ましい方法は、まず培地を少なくとも50℃、好ま
しくは60℃から70℃までの温度に加熱し、ついで
培地のPHを7以上、通常は少なくとも8、好まし
くは8から11までの値に調整する。これはアルカ
リたとえば水酸化ナトリウムを添加して行うのが
便利であるが、もちろん他のアルカリも使用でき
る。
最善の結果を得るためには、培地を再び加熱す
る。この場合は、少なくとも75℃、好ましくは80
℃から95℃までの温度とし、塩化カルシウムを培
地に対して0.08重量%存在させ、菌体と菌体残渣
が完全に凝析するまで加熱する。凝析した菌体と
菌体残渣は、たとえば自然沈降、遠心分離または
濾過によつて容易に分離でき、バイオポリマー
PS87が溶解した粘稠、澄明な上澄液が残る。
バイオポリマーPS87はこの粘稠、澄明な培地
上澄液を、ヘテロポリサツカライドを実質的に溶
解せず、また反応しない水混和性有機液体で処理
することにより回収できる。この操作でヘテロポ
リサツカライドは溶液から沈殿する。使用する有
機液体の量は、培地上澄液1容量部に対して一般
に1から2容量部でよい。使用できる適当な有機
液体の例としてはアセトン、C1〜C5アルカノー
ルたとえばメタノール、エタノール、イソプロパ
ノール、2−ブタノールおよびt−ブタノールが
ある。イソプロパノールおよびメタノールがとく
に好ましい。ヘテロポリサツカライドは、たとえ
ば濾過によつて有機液体から分離し、乾燥して、
最終的に回収できる。
バイオポリマーの製造例
実施例 1
本実施例はバチルス・ポリミキサNCIB 11429
によるシユクロース/イースト抽出液のバツチ法
での発酵によるバイオポリマーPS87の製造を例
示する。
培地の組成は次のとおりである。

合物(Hopkins and Williams,Dow Chemical
製)を発酵初期に起こる発泡の抑制に使用した。
著しく粘稠で発泡性の48時間振盪フラスコ培養
物(PH5.3)400mlを発酵容器に接種した。発酵初
期は回転翼度100rpm、通気速度15/分とした。
温度は30℃に維持した。溶解酵素分圧(DOT)
は急速に2%以下の最小値に低下し、低値を保つ
た。一方、培地のPHをはじめの7.4から4.8に低下
した。PHはこの最低値から以後短時間にわたつて
徐々に上昇するが、培地の粘度はほとんど上昇し
ない。発酵培地のPHを6に上げ、この値に維持
し、回転翼の速度は200rpmに上昇させた。培地
は発酵が進むにつれて粘度を増し、最終粘度は剪
断速度1sec-1(20℃)において250ポアズとなつ
た。ヘテロポリサツカライド濃度は9g/であ
つた(供給した糖に対し収率18%に相当)。
ヘテロポリサツカライド含有培地にイソプロパ
ノール(IPA)を濃度30%(V/V)まで徐々に
加えて希釈し、室温で1時間20000gで遠心分離
して菌体を除去した。上澄液をさらにIPAと混合
し、濃度を70%に上げた。これでヘテロポリサツ
カライドの沈殿は完結した。IPAと水の混合物を
捨て、ヘテロポリサツカライドをオーブン中で乾
燥した。この生成物は容易に水に溶解し、均一粘
稠な溶液を与えた。
実施例 2
本実施例では、発酵培地にイースト抽出液0.4
%(w/v)を含有させ、シユクロースの代りに
グルコース2.0%(W/V)を用いたこと以外、
培地は実施例1の場合と同じである。3日間振盪
フラスコ中で培養したバチルス・ポリミキサ
NCIB11429の培養液1を培地(22)に接種
した。PHは初期の6.9から徐々に低下し、発酵20
時間後には6.0となつた。5%NaOHを加えてPH
がこの値より低下するのを防止した。発酵培地中
のポリサツカライド濃度を一定間隔で採取したサ
ンプルについて測定したところ、最大濃度(0.4
%、収率20%に相当)は発酵開始約45時間後に得
られた。この時点で、供給グルコースの消費およ
び溶解酸素濃度が飽和点付近まで増大したことか
ら判断して、発酵は完結したものと思われた。培
地の最終PHは6.6であつた。
実施例 3
本実施例は連続発酵によるバイオポリマー
PS87の製造を例示する。使用した培地は、
K2HPO4の濃度を0.15%に下げ、さらに0.1%
Na2HPO4を加えたほかは実施例2の場合と同じ
であつた。3の撹拌タンク発酵装置を用い、発
酵容量は1.5に維持した。発泡は消泡剤を時々
添加して予防した。調節可能な蝪動ポンプで発酵
装置への流入速度を調節し、液体の水位を調節す
るためにせきを設けた。
ヘテロポリサツカライドの濃度は、0.03から
0.10の範囲の一連の希釈率で(すなわち滞留時間
それぞれ33時間および10時間)採取したサンプル
について測定した。ポリサツカライドの濃度は希
釈率(D)の低下に応じて増加し、D=0.1での
約0.4%からD=0.03%での約0.8%までの値を示
した。
実施例 4
乳漿および乳漿パーメエートをベースとした培
地を用い実施例3記載の方法と同じ方法でバイオ
ポリマーPS87合成能製造のための連続発酵を実
施した。発酵は培地Aで10日間、さらに培地Bで
20日間行つた。この発酵時間後でも、ポリサツカ
ライドの質および量の劣化は認められなかつた。

Although the pH gradually increases from this minimum value over a short period of time, the viscosity of the medium hardly increases. The pH of the fermentation medium was raised to 6 and maintained at this value, and the rotor speed was increased to 200 rpm. The medium increased in viscosity as fermentation progressed, with a final viscosity of 250 poise at a shear rate of 1 sec -1 (20°C). The heteropolysaccharide concentration was 9 g/(corresponding to a yield of 18% based on the sugar fed). Isopropanol (IPA) was gradually added to the heteropolysaccharide-containing medium to dilute it to a concentration of 30% (V/V), and the cells were removed by centrifugation at 20,000 g for 1 hour at room temperature. The supernatant was further mixed with IPA to increase the concentration to 70%. This completed the precipitation of the heteropolysaccharide. The IPA and water mixture was discarded and the heteropolysaccharide was dried in an oven. This product easily dissolved in water to give a homogeneous viscous solution. Example 2 In this example, 0.4 g of yeast extract was added to the fermentation medium.

調節可能な蠕動ポンプによつて発酵装置中への培
地の流入速度を調整し、培地の水位を調節するた
めにせきを用いた。
流入速度は45ml/分に調整した。培地の希釈率
は次式で表される。

保持した。
ヘテロポリサツカライドの濃度を、この範囲内
の一連の希釈率で採取した発酵培地サンプルにつ
いて測定したところ、希釈率(D)の低下によつ
て上昇することが明らかになつた。すなわちD=
0.1では約0.4%、D=0.03では約0.8%であつた。
温度、通気速度、溶存酸素分圧およびPHは連続
供給開始前の2日間の発酵時に採用した値に等し
くまたほぼ等しく保持した。
連続培養は無限に続けることができる。1つの
実験では50日間操作して全く問題がなかつた。
ヘテロポリサツカライドは発酵装置から取り出
した発酵培地から以下の方法で単離した。
この培地3をパドル型撹拌器を用にてたえず
混合して60℃の温度に加熱した。ついでPHを5%
NaOHを加えて9.0に調整した。2.4gの塩化カル
シウムを加えて培地をコニカルフラスコに移し、
これを85℃のオープンに12時間置いた。この操作
の間に菌体と菌体残渣の凝析が起つた。凝析物の
殆どは底に沈殿したが、一部は気体の泡に付着し
たらしく表面に乳遊した。この細胞物質の二つの
層の間に、透明な液体の帯域が存在していた。こ
の帯域の液体を吸引して、計2の透明な液体を
得た。吸引濾過して、計2の透明な液体が得ら
れる。冷却後、イソプロパノール3をこの液体
に混合しながら添加した。ポリサツカライドの糸
をひく沈殿が得られた。これを回収し、真空乾燥
した。収量は11gであつた。
このヘテロポリサツカライドを水に1%W/V
の濃度で再溶解すると、20℃における硬度約200
ポアズ、20℃における降伏価約54ダインを示す溶
液が得られた。このヘテロポリサツカライドは比
旋光能[α]25 D=+60を示し、かつクロマテイツ
クスKMX−6(Chromatix KMX−6)を用い
た低角度レーザー光散乱法で分子量を測定したと
ころその分子量は34±4×104であつた。
バイオポリマーPS87の家庭用、工業用用途
本発明はまた、バイオポリマーPS87を含有す
る、含水製品、または水を加えて含水製品を製造
するのに適した乾燥ミツクスをも提供する。
バイオポリマーPS87はこのようなユニークな
物理的特性を有しているので、懸濁剤、乳化剤、
凝集剤、安定剤、および増粘剤として、家庭用品
並びに工業用品に幅広い用途を有する。
以下の実施例はバイオポリマーPS87の使用に
より改善される製品、方法および適用の例を示
す。
ミルク製品
本発明はまた、食品とくにミルク−ゲル化組成
物に関する。
ピロリン酸四アルカリ塩および食用カルシウム
塩を含有するミルクプデイングはよく知られてい
る(たとえば米国特許第2607692号参照)。補助成
分として、ミルクプデイングには通常、乾燥した
あらかじめゼラチン化したデンプンの添加を必要
とする。この種の組成物から生成するミルクゲル
はペースト様または糊状の堅さを持ち、好ましく
ない液体の分離、すなわち時間がたつと離漿を起
こしやすい。
本発明はピロリン酸四アルカリ金属塩、食用カ
ルシウム塩およびバイオポリマーPS87の混合物
よりなる組成物をも提供する。これらの成分は乾
燥微粉末組成物とすることが好ましく、これは冷
ミルクと撹拌してミルクゲルを生成させることが
できる。
ピロリン酸四アルカリ金属塩としては、たとえ
ば、ピロリン酸四ナトリウムまたはピロリン酸四
カリウムが使用できる。適当な食用、水溶性カル
シウム塩の例には、酢酸カルシウム、乳酸カルシ
ウム、硫酸カルシウム、クエン酸カルシウム、グ
ルコン酸カルシウム、プロピオン酸カルシウム、
サツカリンカルシウム塩および酒石酸カルシウム
がある。
ピロリン酸四アルカリ金属塩、食用カルシウム
塩、並びにバイオポリマーPS87の量は、所望す
るミルクプデイング製品の性質に応じて変化させ
ることができる。しかしながら一般に、本発明に
使用するための適当な組成物はピロリン酸四アル
カリ金属塩1.5から3.5重量部、食用カルシウム塩
1から5重量部、および0.5から4重量部のバイ
オポリマーPS87から構成される。
本発明のミルクゲルまたはミルクプデイングを
生成させるには、上述のようなゲル化組成物の均
一乾燥配合物を冷ミルクに加え、この成分をミキ
サーで混合する。混合材料を次に適当な容器中に
注ぎ、ついで冷蔵庫に入れるのが好ましい。約15
分後に、この混合物は十分食べられる程に硬化す
る。
本組成物にはゲル化のための主成分のほかに、
通常ミルクプデイングの矯味および着色に用いら
れる他の成分も添加することもできる。たとえば
通常の任意の矯味剤、ココア、バニラ、シナモン
またはフルーツもしくはナツツ、ペカン、レーズ
ン、バナナ、いちじく、やし等を添加できる。さ
らに消泡剤たとえば、ベニバナ油、ヤシ油、落花
生油または綿実油を加えることもできる。
この組成物からミルクプデイングを生成させる
に際しては、全乳でも液状スキムミルクでも使用
できる。さらにミルクの代りに、ドライミツクス
に、全乳またはミキムミルクから誘導された乾燥
粉乳を添加してもよい。この場合には水を加え、
得られた混合物を撹拌すればミルクプデイングが
生成する。
さらに本発明を詳細に説明するため、以下に実
施例を示す。

いて数分間混合し、ついで適当な容器中に注ぐ。
この混合物を次に約15分間冷蔵する。この時点
で、混合物は食べられる。生成物はきめのきわめ
て滑らかな、さくさくして容易にくずれ、きわめ
て純な舌ざわりを有し、しかも離漿に安定であ
る。
この種のミルク製品については英国特許第
1257057号に記載されている。この開示を本明細
書に導入する。
サラダドレツシング
本発明はまたサラダドレツシングに関する。サ
ラダドレツシングは通常、凍結し、解凍すること
はできない。これはエマルジヨンが破壊され、ほ
とんどすべての油が解凍後、ドレツシングから分
離し、きめが劣化するからである。本発明は凍結
−解凍安定性を有するサラダドレツシング、およ
びその製造方法に関する。
サラダドレツシングは食用植物油、酸性化成
分、卵黄含有成分、および食用デンプン、タピオ
カ、小麦粉、ライ麦粉もしくはこれらの2種もし
くは3種以上から調整した調理もしくは部分調理
粘稠ペーストからなる乳化半固体食品である。
さらにサラダドレツシングは、各種成分たとえ
ば食塩および/またはグルタミン酸一ナトリウム
塩、甘味成分たとえば砂糖、デキストロース、コ
ーンシロツプ、転化糖シロツプ、非糖化マルトー
スシロツプ、グルコースシロツプおよび蜂蜜、他
のフレーバー成分たとえばマスタード、パプリ
カ、その他のスパイスまたは任意スパイス油もし
くはスパイス抽出液で味つけし、フレーバーを添
加される。
本発明は食用油、水、乳化剤、調理デンプン、
酸性化剤、食塩およびバイオポリマーPS87から
なるサラダドレツシングを提供する。
適当な食用油としては、綿実油、大豆油、ベニ
バナ油、トウモロコシ油およびオリーブ油を挙げ
ることができる。サラダドレツシング中の油の量
は通常、ドレツシング重量に対して30から70%、
好ましくは40%から50%までである。
本発明によつて製造されるサラダ中に添加され
る酸性化剤には、酢、ライムジユースまたはレモ
ンジユースがある。使用される酸性化剤の量は最
終製品中の氷酢酸含量に換算して1重量%から
1.5重量%までである。
その他の成分は乳化剤、たとえば卵黄含有成分
または合成乳化剤たとえば商品名「ツイーン60」
(ポリオキシエチレンソルビタンモノステアレー
ト)がある。サラダドレツシング中に加える乳化
剤の量は食品総重量に対して0.25重量%から4重
量%までである。さらにサラダドレツシングに添
加する食塩は最終生成物の重量に対し1重量%か
ら3.5重量%までである。
上述の成分に加えて、サラダドレツシングには
バイオポリマーPS87を0.2重量%から0.3重量%ま
でを添加する。
本発明はまた、サラダドレツシングに対して30
重量%から70重量%までの食用油、サラダドレツ
シングに対して30重量%から70重量%までの水、
乳化物を形成するのに有効な量の乳化剤、スプー
ンで掬えるサラダドレツシングの特徴的本質を与
えるのに十分な量の調理デンプン、サラダドレツ
シングに対して氷酢酸換算量で1重量%から1.5
重量%の酸性化成分、ドレツシングに対して0.2
重量%から0.3重量%までのバイオポリマー
PS87、およびドレツシングに対して、1.0重量%
から3.5重量%までの食塩を含有するさじて掬え
るサラダドレツシングの製造方法も提供する。す
なわち、バイオポリマーPS87と半量の水を含有
する第一の混合物を生成させ、残りの水を調理テ
ンプン、乳化剤およびドレツシングに対して約
0.75%までの食塩を配合した第二の混合物を生成
させ、第一の混合物と第二の混合物を配合して両
者の混合物とし、この混合物を食用油および酸性
化成分と均一な混合物が得られるまで撹拌し、こ
の均一な混合物を均一に乳化し、乳化混合物に残
りの食塩を加えてさじで掬えるサラダドレツシン
グを製造する方法である。
次に本発明のこの態様について、以下の実施例
でさらに詳細に説明する。

ドを総量の半分の水に激しく撹拌しながら加え
る。撹拌は30分間続けて、第一の混合物を形成さ
せる。次に総量の35%の食塩を、デンプンととも
に残つた水に加える。この混合物をデンプンの水
和が完全に起こるまで加熱する。次にこの混合物
を放冷し、卵黄を加えて完全に混合し、第二の混
合物を形成させる。
第一の混合物および第二の混合物をついで完全
に混合し、油を最初のうちは徐々に、次いで急速
に加える。加え終わるまでに約2分を要し、この
間撹拌を行う。続いて撹拌しながら酢を加え、混
合物が均一になるまで撹拌する。次に混合物をコ

間がかかりすぎること、新鮮な食品に匹敵するほ
どの品質のものを得ることができないことなどで
ある。
本発明は、とくに乾燥または脱水食品につい
て、急速かつ完全な再水和または水和が可能な食
品、およびこの目的を達成する方法に関する。
本発明は乾燥バイオポリマーPS87および乾燥
食品成分を含有し、食品中のバイオポリマー

ら、余分な工程が不必要であること、優れた製品

歯 磨
本発明はまた、液体またはペースト状歯磨並び
にその製造方法に関する。さらに詳しくは、本発
明は、バイオポリマーPS87よりなる増粘および
懸濁液を、歯磨100重量部に対して好ましくは0.3
重量部から2重量部までの割合で均一に含有する
液体またはペースト状歯磨を提供する。
本発明は、バイオポリマーPS87を含有する液
体またはペースト状歯磨は、キサンタンガム
(xanthan gum)を含有する通常の液体またはペ
ースト状歯磨と比較した場合、独特な擬可塑性を
示し、歯磨は低圧で容易に流出し、圧を除けば流
動性を失うという点で性能が向上することを発見
し完成されたものである。さらに詳しく説明する
と、使用時に容器中の歯磨を歯ブラシ上に押し出
すと、軽い圧力で良好な流動性をもつて歯磨が押
し出され、押し出し口から離れると直ちに流動性
を失つて押し出された形を保持する。
すなわち、本発明は、主として(i)水25重量%か
ら35重量%、(ii)湿潤剤20重量%から35重量%、(iii)
界面活性剤0.5重量%から2重量%、(iv)バイオポ
リマーPS87、0.3重量%から2重量%、(v)固体粒
子研摩剤20重量%から35重量%を含有した固体研
摩性歯磨粒子成分の相分離に安定な揺変性ゲル歯
磨であつて、ゲル歯磨は低圧で良好な流動性をも
つて容易に押し出され、一旦押し出されると押出
圧力の除去によつてその流動性を失い、押し出さ
れたままの形状を保持する液体揺変性ゲル歯磨を
提供する。
歯磨に使用するバイオポリマーPS87の量は0.5
重量%から1重量%が好ましい。
本発明の歯磨は通常のよく知られた歯磨成分、
たとえば湿潤剤、洗浄剤もしくは界面活性剤、甘
味剤、研摩剤、着色剤、抗カリエス剤、抗菌もし
くは抗細菌剤または水を含有する。これらの歯磨
慣用成分の量は常用される範囲に調整すればよ
い。通常、液体歯磨の場合、水25〜45重量%、研
摩剤20〜35重量%、保湿剤20〜35重量%、洗浄剤
もしくは界面活性剤0.5〜2重量%、バイオポリ
マーPS87を0.3〜2重量%、その他慣用の歯磨成
分を含有する。ペースト状歯磨の場合、有用な処
方は、水25〜35重量%、研摩剤35〜50重量%、保
湿剤15〜30重量%、洗浄剤もしくは界面活性剤
0.5〜2重量%、バイオポリマーPS87を0.3〜2重
量%、およびその他の慣用歯磨成分を含有する。
増粘剤および懸濁剤として用いられるバイオポ
リマーPS87の一部分を歯磨用の公知増粘剤およ
び懸濁剤で置き換えてもよい。この種の公知増粘
剤および懸濁剤の適量はバイオポリマーPS87の
重量の約1.4倍以下、好ましくはバイオポリマー
PS87と等量もしくはそれ以下である。
慣用される公知増粘剤および懸濁剤の例として
はカルボキシメチルセルロース、そのアルカリ金
属塩、カラゲニン、アルギン酸ナトリウム、ヒド
ロキシエチルセルロース、メチルセルロース、ト
ラガカントゴム、イナゴ豆ゴム、タマリンド種子
ポリサツカライドがある。
保湿剤の例としては、グリセロール、ソルビト
ール、マルチトール、グルコース、プロピレング
リコール、ポリエチレングリコール、ピロリドン
カルボン酸ナトリウムがある。
研摩材の例としては、リン酸二カルシウム二水
和物、ピロリン酸カルシウム、無水リン酸二カル
シウム、不溶性メタリン酸ナトリウム、水和アル
ミナ、炭酸カルシウム、炭酸マグネシウム、酸化
マグネシウムおよび微粉末シリカがある。
洗浄剤または界面活性剤の例としては、ラウリ
ル硫酸ナトリウム、N−ラウロイルザルコシンナ
トリウム、α−オレフインスルホネート、2−ヒ
ドロキシアルキル硫酸ナトリウム、ラウリエーテ
ル硫酸ナトリウム、ヤシ油モノグラセライド硫酸
ナトリウム、ヤシ油モノグリセライドスルホン酸
ナトリウム、ラウロイルエタノールアミドスルホ
コハク酸モノエステルのナトリウム塩、ポリオキ
シエチレン脂肪酸エステルたとえばポリオキシエ
チレンソルビタンモノラウレート、ポリオキシエ
チレンソルビタンモノパルミテート、ポリオキシ
エチレンソルビタンモノステアレートまたは重合
度少なくとも25のポリオキシエチレンステアレー
ト、およびポリオキシエチレンポリオキシプロピ
レンブロツク共重合体がある。
フレーバーの例としてはペパーミント油、スペ
アミント油、サツサフラス油、丁子油、セージ
油、ユーカリ油、マヨナラ油、レモン油、シナモ
ン油、オレンジ油およびサリチル酸メチルナトリ
ウム塩がある。
甘味料はたとえばサツカリンナトリウムであ
る。
着色剤、抗カリエス剤および抗菌もしくは抗細
菌剤の例としては、フツ化ナトリウム、フツ化

バイオポリマーPS87含有水性ペイント
本発明はまた水性ペイント、さらに詳しくは、
擬可塑性および揺変性の利点を有するしたたり落
ちない水性ペイントに関する。
水性ペイントはモノマーの重合によつて生じた
重合樹脂乳化液である。この種のペイントは顔料
のほかに他の成分、たとえば増量剤、消泡剤、分
散剤、凍結−解凍安定剤、増粘剤および防腐剤を
含有する。
水性ペイントでは、水は通常の油性ペイントの
場合のシンナーの役をしていて、ペイントが表面
に薄いフイルムとして塗布された場合、水は蒸発
し、樹脂様またはゴム様物質が連続性フイルムを
形成し、この場合、酸化、重合または樹脂粒子の
合着によつて、フイルムは耐水性となる。
水性ペイントの組成はその使用条件に適合する
ように変化させる。結合剤またはフイルム形成剤
は、ペイント技術の分野の文献に記載されている
ように、ゴム様物質で組成は多様である。以下の
合成プラスチツク半固体、たとえば可塑化前また
は後両者のスチレン−ブタジエン重合体もしくは
ポリスチレン、ポリアクリレート乳化剤、ポリ塩
化ビニル乳化剤、ポリ酢酸ビニル乳化液、酢酸ビ
ニル共重合体乳化液たとえば酢酸ビニル−エチレ
ン共重合体、塩化ビニル−酢酸ビニル共重合体お
よびポリ酢酸ビニル−ブタジエン共重合体乳化液
は、とくによく知られていて、市販の水性ペイン
ト中に結合剤またはフイルム形成剤として使用さ
れている。この種の合成水性ペイント結合剤には
乳化剤および合成ラテツクスの両名称が使われて
いるが、用語的にはラテツクスの方が好ましい。
天然ラテツクスは水性ペイントの結合剤またはフ
イルム形成剤として文献に記載されているが、一
般には使用されていない。
水性ペイントの性質に改良の必要があることは
当業者にはよく知られている。すなわち、伸展性
の刷子のりがよく、しかもしたたり落ちる性質の
ない、良好な流動性と平滑なガラス様の仕上りを
与える平坦性、温度やPHなどの条件が変化しても
比較的均一な粘度を示す性質、良好な遮閉性と非
垂下性、良好なゲル構造と強度、製造の容易性等
が問題で、とくに水性ペイント組成物には上述の
望ましい性質のすべてまたは大部分について改良
の必要があつた。
本発明は、ラテツクスゴム様水性ペイント結合
剤、顔料、結合剤と顔料に対する水性ビヒクル、
およびペイントに擬可塑性を与えるのに十分な量
のバイオポリマーPS87を含有する水性ペイント
を提供する。
バイオポリマーPS87の適量はペイント重量に
対して0.01重量%から4重量%まで、好ましくは
0.2重量%から2重量%までである。
ペイントを増粘するのに有効な量のバイオポリ
マーPS87を含有する水性ペイントを生成させる
場合には、ペイントには樹脂様フイルム形成剤お
よび顔料含有水性乳化ラテツクス組成物を加え
る。ペイントには他の組成たとえば増量剤、消泡
剤、分散剤、凍結−解凍安定剤および防腐剤を添
加することができる。
本発明はまた、顔料と水およびバイオポリマー
PS87の顔料粉砕物を形成させ、ついでこのよう
に処理した顔料をペイント結合剤および所望によ
り他の成分を導入する、ラテツクスゴム様水性ペ
イント結合剤から構成される水性ペイントの製造
方法を提供する。
この種のバイオポリマーPS87含有水性ペイン
トの処方およびその製造方法については米国特許
第3894976号に一般的に記載されている。この開
示を参考として本発明に導入する。

ル化、分離、顔料沈殿を起さず、良好な流動性を
示す。
油井の発掘液
本発明はまた、新規な穿孔液、および油井やガ
ス井等の発掘に際し、発掘孔内を循環させる粘性
液体としてのその使用に関する。
油井やガス井等の発掘に用いられる穿孔泥は、
一般にクレーと他のコロイド性物質を含有する水
性液体である。このコロイド性物質が穿孔土や増
量剤の輸送や懸濁に必要な液に所望の粘度とゲル
力を付与し、周囲の岩層に液体が失われるのを減
少させるのに必要な濾塊の組成に役立つ。コロイ
ド状クレー等の使用により穿孔液に粘度を与える
ことは、得られた液のポンプによる輸送が困難で
あり、またドリルビツトに適当な滑沢性を付与し
ないという逆効果を生じることが多い。したがつ
てその粘性が濾塊に必要な液体内に固体を懸濁さ
せ、しかも液体のポンプ輸送性およびドリルビツ
トへの滑沢性付与能が悪影響を受けないような穿
孔液が望まれていた。
すなわち、本発明はバイオポリマーPS87を含
有する穿孔液を提供する。本発明はバイオポリマ
ーPS87を含有する穿孔液が著しい擬可塑性を示
し、剪断力がかかつた場合には粘度が著しく低下
することを発見し完成されたものである。この性
質は静止時またはわずかな剪断力がかかつている
場合にはきわめて高粘度を示し、液中に存在する
穿孔クレーや増量剤を輸送し懸濁が可能である。
同時に、穿孔液に剪断力が負荷された場合、たと
えばポンプ輸送の場合には粘度が著しく低下し、
ポンプ輸送が容易になる。また、穿孔液がドリル
ビツトにより生じる強い剪断力を受けたときは、
また粘度が著しく低下して、ドリルビツトの滑沢
性の改善が可能である。
本発明の穿孔泥は、水性穿孔液にバイオポリマ
ーPS87を添加することによつて製造できる。バ
イオポリマーPS87は約0.05重量%から約2.5重量
%まで、またはそれ以上の濃度で添加できる。
バイオポリマーPS87を含有する溶液は他の物
質を添加しないので、そのまま穿孔泥または破壊
液に使用できる。たとえば穿孔時にクレー型の泥
板岩を含む地層があれば、これが穿孔液の密度を
増加させるのに役立つので、穿孔液は単なるヘテ
ロポリサツカライドの水溶液でよい。穿孔中に地
層から穿孔クレーが取り込まれて穿孔液の密度が
増加する。
場合により、増量剤を穿孔液に加えることもで
きる。増量剤の例としては硫酸バリウム、無定形
シリカもしくは炭酸カルシウム、硫化鉛、炭酸バ
リウム等がある。穿孔液はさらに、防腐剤とし
て、ホルムアルデヒド、パラホルムアルデヒドま
たトリクロロフエンナトリウム、ペンタクロロフ
エンナトリウム等を添加してもよい。増量剤は所
望の泥密度を与えるように濃度を決定し、防腐剤
は一般に系の液体重量に対して約0.001重量%か
ら約0.1重量%の濃度で添加される。
増量剤、防腐剤のほか、本発明の泥もしくは類
似の液体には、他の添加剤を加えることもでき
る。たとえばゲル形成物質たとえばベントナイト
およびアタプルガイトクレー、また液体損失防止
剤たとえばカルボキシメチルセルロース、デンプ
ン等を添加できる。さらに他の添加剤、粘度改変
剤たとえば鉄クロムリグノスルホネート、ナトリ
ウムリグノスルホネート、ケブラゴおよびカルシ
ウムリグノスルホネート、またカルシウム処理剤
たとえば石灰、硫酸カルシウム、塩化カルシウ
ム、乳化剤たとえばペトロリウムスルホネート、
トールオールソープ、およよびナトリウムリグノ
スルホネートおよび、乳化液体が所望の場合は原
油もしくはジーゼル油のような物質を加えてもよ
い。上述の添加剤は常に特定の穿孔泥や他の液体
にすべて含まれているものではない。この特殊添
加物の量は液体中の他の成分の量ともある程度関
連して、特定の使用目的に合致するように決定さ
れる。
バイオポリマーPS87は乳化剤の穿孔液にも使
用され、水をベースとして得られた液を激しく混
合すれば安定な油中水型乳化液が得られる。油中
水型乳化液は液体の喪失性が低く、乳化型穿孔液
に代表的な望ましい特性を持つている。
この種のバイオポリマーPS87含有穿孔液の処
理およびその製法、使用は米国特許第3979303号
に一般的に記載されている。この開示を参考とし
て本明細書に導入する。
本発明のこの態様の実施例を以下に示す。本実
施例は典型的な穿孔液である。
実施例 18
実施例5記載のバイオポリマーPS87を食塩水
に溶解し、バイオポリマーの濃度を0.3重量%に
することにより穿孔液を調製できる。
三次原油の回収
通常、原油は地下の埋蔵部から一連の連続操作
によつて回収される。新しい油井は一般に油井の
内圧が放出する結果、限られた量の原油しか採取
されない。この圧力が消失すると、機械を用い
て、さらに原油を汲み出す必要がある。この操作
では埋蔵されている原油の総量の約25%しか回収
されない。かなりの量の原油が地層の孔に捕えら
れている。さらに回収を進めるためには二次回収
が行われる。一つの回収方法では油井または一連
の油井に水をポンプで送つて洪水を起こさせて多
孔性の岩に捕えられていた原油の一部を置換し、
置換された原油を周囲の油井から集める。しかし
ながら、水による洪水では、地層に捕集された原
油の約55〜60%はそのまま残つてしまう。この現
象は、水は原油に比べて粘度が低く、抵抗の少な
い道を通る傾向があり、原油に触れるだけで、大
きな鉱穴には届かないということで説明できる。
さらに地層内の表面力が油を結合させて置換を妨
げる傾向がある。
最近になつて、置換液の粘油度や滲透性を増大
させて、原魅油の置換効率を高めるように可動性
を調整された液体を使用することにより、これら
の埋蔵部からさらに原油を回収するための多くの
方法が開発されてきた。このうち特に興味がある
のは、置換液の粘度を増大させるためポリサツカ
ライドやポリアクリルアミドによるポリマー洪水
法を用いる回収量の上昇方法である。この方法に
は、界面活性剤や界面活性補助剤を用いて岩層か
ら原油を放出させる変法もある。ある種のポリア
クリルアミドは塩水中で粘度を失うという欠点の
あることがわかつた。
本発明はバイオポリマーPS87が塩に対して強
く、使用の通常条件では沈殿を生じたり粘度を失
うことはなく、広範なPH域で剪断、熱および粘度
が安定していることを発見して完成された。バイ
オポリマーPS87は油に対して良好な置換作用を
示し、多孔性地層の要素に吸収され難く、原油回
収率の上昇に適当な粘度を示す(1%溶液が20℃
において少なくとも150ポアズの粘度を示す)。
すなわち、本発明は、対照溶液の重量に対して
0.005重量%から0.5重量%までのバイオポリマー
PS87を含有する水溶液からなる採油用可動性調
整溶液を提供する。この水溶液には食塩のような
塩を添加することもできる。
本発明はまた、バイオポリマーPS87を0.005重
量%から0.5重量%を含有する可動性調整溶液を
岩層内に注入して、石油埋蔵地下岩層から原油を
回収する方法を提供する。
石油埋蔵部に注入する大量の可動性調整溶液を
輸送することは条件によつては実用的でないか
ら、この目的にはバイオポリマーPS87を乾燥ま
たは脱水した形で提供し、必要な場所で必要なと
きに水または食塩水で再生するのが便利である。
バイオポリマーPS87の液を使用前に保存する必
要がある場合は、対照溶液の重量に対して0.02重
量%から1重量%の濃度でホルムアルデヒドを添
加すれば、一般に夾雑微生物による有害な変化を
防止することができる。
本発明のこの態様に関して、さらにデータが英
国特許第1531970号に記載されている。この開示
を参考として本明細書に導入する。
懸濁重合
本発明はまた、広い意味での、たとえば
Encyclopedia of Polymer Science and
Technology、第13巻に述べられているようにモ
ノマーおよびコモノマーに関する操作といつた意
味での、懸濁重合に関する。
懸濁重合は、はじめポリスチレンの製造に導入
された昔の粒状重合から発展したものであり、液
体モノマーの小滴が水中に分散していて、重合し
て固体球形粒子を生じる塊状重合の1種である。
懸濁重合における主要点はモノマー小滴の均一
な懸濁の生成と維持である。この小滴は粘稠な物
質から堅い顆粒状固体に徐々に変換するので、粒
子の沈殿や凝集が起こると凝塊を形成してしま
う。この操作には小滴または粒子の接触が最小限
に押えられるように懸濁を維持するための適切な
撹拌と安定化剤が要求される。
懸濁重合に用いられる操作は使用するモノマー
に無関係なくほぼ同じで、少量の懸濁剤および分
散剤を小量含有する水からなる安定化媒質を撹拌
しておき、これにモノマーを液体小滴として分散
させる。触媒(開始剤)はモノマー添加後に反応
混合物中に加えられる。
懸濁剤は通常、保護コロイド剤(水溶性高分
子)たとえば広範に使用されている保護コロイ
ド、たとえばカルボキシメチルセルロース、カル
ボキシメチルセルロースとデンプンの1:1混合
物、ヒドロキシエチルセルロース、メチルセルロ
ース、ポリアクリル酸、ポリビニルアルコール、
ゼラチン、アルギン酸塩、アラビアゴムおよびト
ラガカントゴムが使用される。
液体モノマーの懸濁重合による固体粒子ポリマ
ーの製造に際してバイオポリマーPS87を懸濁剤
として使用すると有利であることが明らかにされ
た。添加するバイオポリマーPS87の量はこの方
法に用いたモノマーの重量に対して0.1重量%か
ら5重量%である。
本発明はしたがつて、0.1〜5重量%のバイオ
ポリマーPS87存在下に、アクリル酸もしくはそ
の重合性誘導体、メタクリル酸もしくはその重合
性誘導体、ビニルアルコールのエステルもしくは
エーテル、ビニリデンクロライド、スチレンまた
はメチルスチレンのような液体モノマーを粒状ポ
リマーに水性懸濁重合させる方法を提供する。
この懸濁剤の改良は懸濁重合で重合可能なすべ
てのモノマーに適用できる。改良が適用できるモ
ノマーは
(1) アクリル酸およびメタクリル酸の重合性誘導
体、たとえば、メタクリル酸自身、アクリル酸
メチル、アクリル酸エチル、アクリル酸フエニ
ルエチル、アクリル酸メトキシエチル、メタク
リル酸メチル、メタクリル酸エチル、メタクリ
ル酸イソプロピル、メタクリル酸イソプロピ
ル、メタクリル酸フエニル、メタクリル酸フエ
ニルエチル、メタクリル酸メトキシエチル、ア
クリロニトリルおよびメタクリロニトリル、
(2) ビニルアルコールのエステルおよびエーテ
ル、たとえば塩化ビニル、酢酸ビニル、プロピ
オン酸ビニル、酪酸ビニル、ビニルメチルエー
テル、ビニルエチルエーテルおよびビニルフエ
ニルエーテル、
(3) 塩化ビニリデン、および
(4) スチレンおよびそのα−アルキル誘導体、た
とえばスチレンおよびα−メチルスチレンであ
る。
メタクリル酸メチルをモノマーとして用いた特
定の態様では、メタクリル酸メチルに対して0.1
〜1重量%のオーダーのバイオポリマーPS87を
添加した。
他のスチレンをモノマーとして用いた態様で
は、スチレンに対して0.1〜0.5重量%のオーダー
のバイオポリマーPS87を添加した。
さらに他のスチレンをモノマーとして用いた態
様では、スチレンに対して2.0〜4.0重量%のオー
ダーのバイオポリマーPS87を添加した。
バイオポリマーPS87の存在下の懸濁重合法に
用いられる操作および実例はさらに詳細に米国特
許第3852257号に記載されている。この開示を参
考として本明細書に導入する。
本発明はまた、物理的貯蔵安定性を改良した水
性、ビルド液体洗浄剤に関する。
水性、ビルド液体洗浄組成物の持つている大き
な問題のひとつに、この種の組成物が十分な物理
的貯蔵安定性を持たないことがあることはよく知
られている。特殊な方策をとらなければ、この種
の組成物中のビルダー塩、また場合により他の成
分が分離してしまう。とくにかかるビルダー塩を
高濃度に加えた場合にはこの傾向が顕著である。
したがつて、この技術分野では、組成物中にビ
ルダー塩を安定に懸濁させるための特定の安定剤
を加えるなど多くの提案がなされてきた。公知の
安定剤の例としては、合成ポリマーたとえば無水
マレイン酸とエチレンの共重合体、ビニルメチル
エーテル、ポリアクリレート等がある。通常この
種のポリマーは0.5〜2重量%の濃度で用いられ
る。この種のポリマーでは満足できる貯蔵安定性
は達成されないが、このようにして安定化された
組成物は粘度が望ましい値よりも高くなり、粘度
を下げる手段がとられている場合が多い。したが
つて安定性と粘度の間に注意深くバランスをとる
ことが要求される。
本発明は、水性、ビルド液体洗浄剤組成物にご
く少量のバイオポリマーPS87を添加すると、す
ぐれた物理的貯蔵安定性が付与され、この方法で
安定化された組成物は従来技術における他のポリ
マーを添加した場合よりも粘度が低く、許容でき
る粘度を示すことが明らかにされ、完成されたも
のである。
したがつて本発明はまた、活性洗浄剤物質、ビ
ルダーおよび、安定剤としてバイオポリマー
PS87を含有する水性、ビルド液体洗浄剤組成物
を提供する。
本発明の水性、ビルド液体洗浄剤組成物に用い
られるバイオポリマーPS87の量は総組成物重量
に対して0.05〜1重量%、好ましくは0.1〜0.5重
量%である。本発明の液体洗浄剤組成物はさらに
主成分として活性洗浄剤組成物、すなわち、重合
脂肪酸を含むC10〜C24脂肪酸のアルカリ金属また
はアルカノールアミン石鹸、あるいは陰イオン
性、非イオン性、両イオン性(両性)合成洗浄剤
またはこれらの任意の混合物を含有することもで
きる。
陰イオン界面活性剤の例としては、C9〜C20ア
ルキルベンゼンスルホン酸、C8〜C22一級もしく
は二級アルキルベンゼンスルホン酸、C8〜C24オ
レフインスルホン酸、たとえば英国特許第
1082179号に記載されているクエン酸アルカリ土
類金属塩の熱分解物のスルホン化によつて得られ
るスルホン化ポリカルボン酸、C8〜C22アルキル
スルホン酸、C8〜C24アルキルポリグリコールエ
ーテルスルホン酸(エチレンオキサイド10モルま
でを含有)の塩(ナトリウム、カリウム、アンモ
ニウムおよび置換アンモニウム塩たとえばモノ、
ジおよびトリエタノールアミン塩を含む)があ
る。その他の例はSchwartz PerryおよびBerch
著“Surface Active Agents and Detergents”
第1巻および第2巻に記載されている。
非イオン性合成洗浄剤の例には、C8〜C18アル
キルフエノール、C8〜C18一級もしくは二級脂肪
族アルコール、C8〜C18脂肪酸アミドとエチレン
オキサイド、プロピレンオキサイドおよび/また
はブチレレンオキサイドの縮合生成物がある。非
イオン性洗浄剤の例にはそのほか1個のC8〜C18
アルキル鎖および2個のC1〜3アルキル鎖を有す
る二級アミンオキサイドがある。上級の文献には
非イオン性洗浄剤の他の例も記載されている。
上記非イオン性洗浄剤に存在するエチレンオキ
サイドおよび/またはプロピレンオキサイド分子
の平均モル数は1〜30である。各種非イオン活性
剤は、アルキル化の程度が高いものと低いものと
の混合物も含めて、混合して使用できる。
両性すなわち両非イオン性洗浄剤の例にはN−
アルキルアミノ酸、スルホベタイン、脂肪酸と蛋
白水解物の縮合生成物がある。しかしながら通常
は高価なため、陰イオン性または非イオン性洗浄
剤と配合して使用される。各種の界面活性剤を混
合して使用することもできる。C10〜C20脂肪酸な
らびに重合脂肪酸の石鹸(そのナトリウム、カリ
ウムおよび置換アンモニウム塩たとえばトリエタ
ノールアミン塩)も使用でき、好ましくは陰イオ
ン性および/または非イオン性合成洗浄剤と配合
して用いられ、最終組成物の発泡性に好ましい影
響を付与する。
活性洗浄剤の量は1〜60%、好ましくは2〜40
%、とくに好ましくは5〜25%である。たとえば
陰イオン性と非イオン性の洗浄剤を混合して用い
る場合には、その重量比は10:1から1:10まで
変化させることができる。好ましくは6:1から
1:6までである。石鹸を加える場合には、その
量は1〜40重量%である。
本発明の液体組成物にはさらに60%までの適当
なビルダー、たとえばピロおよびトリポリリン
酸、ニトリロトリ酢酸、エーテルポリカルボン
酸、コハク酸、炭酸、オルトリン酸、ゼオライ
ト、カルボキシメチルオキシコハク酸等のナトリ
ウム、カリウムおよびアンモニウムもしくは置換
アンモニウム塩等を添加できる。とくに好ましい
ものはポリリン酸ビルダー塩、ニトリロトリ酢酸
塩、ゼオライトおよびそれらの混合物である。一
般には、ビルダーは最終組成物の1〜60重量%、
好ましくは5〜50重量%添加する。
本発明の洗浄組成物中の水の量は5〜70重量%
である。
本発明の液体洗浄剤組成物には他の慣用成分も
添加できる。たとえば固定懸濁剤、懸濁剤、腐食
防止剤、着色剤、香料、シリケート、光輝剤、発
汁増強剤、発汁抑制剤、殺菌剤、変色防止剤、乳
濁剤、プロテアーゼ、アミラーゼおよびセルラー
ゼのような酵素、繊維柔軟剤、漂白剤駆体の存在
もしくは不存在下における酸素放出漂白剤たとえ
ば過酸化水素、過ホウ素酸ナトリウムもしくは過
カルボン酸、ジ過フタル酸無水物、緩衝剤等であ
る。この場合、バイオポリマーPS87で効率が改
善されているので、少量の塩たとえば食塩を加え
ることが有利な場合が多い。
次に本発明を以下の例によりさらに詳細に説明
する。

バイオポリマーPS87は食餌療法用食品や特殊
病人用食事の増量剤として使用できる。

バイオポリマーPS87はビールのボデイー剤と

バイオポリマーPS87の工業的応用
研摩剤
バイオポリマーPS87の高剪断力下での低粘度
により、急速な研摩作用と研ぎ屑の除去が簡単に
なるほか、研摩剤の懸濁も改善される。
接着剤
バイオポリマーPS87のレオロジー特性により
接着剤の滲透が調節できる。接着剤は容易にポン
プ輸送が可能で、適用時には低粘度であるが低剪
断力条件下には高粘度を示す。乾燥時の水の放出
も早い。
農 業
バイオポリマーPS87は除草剤、殺虫剤、肥料、
殺菌剤の懸濁剤として使用できる。乳遊性は粘着
性が改善されるので、スプレー時の接触時間が延
長する。また流動性の農業用化学薬品にすぐれた
安定剤として使用することができる。
窯 業
バイオポリマーPS87はうわぐすり中の成分を
懸濁し、粘度を維持する。押出しはすべりが良く
なり、焼く前の製品の強度が改善される。
クリーナー
バイオポリマーPS87のレオロジー特性により
垂直面に付着する時間が延長される。工業用のゲ
ル型の酸およびアルカリクリーナーの処方を可能
にすると考えられる。
ゲ ル
バイオポリマーPS87ボラツクスは爆薬のゲル
化剤として作用する。バイオポリマーPS87鉄ゲ
ルは粒子板で尿素−ホルムアルデヒド接着剤の使
用を低下させる作用がある。
鉱 業
バイオポリマーPS87は沈降工程での鉱石の沈
降速度を調整することができ、分離工程では沈降
剤として作用し、発泡安定化作用がある。スラリ
ーの輸送では65%抵抗を低下させ、またとくに低
剪断力条件では懸濁性を示す。
製紙業
製紙業には高固体サイズプレスおよびロールコ
ーテイングに際しレオロジー改変剤として抗にじ
み作用を示す。湿端生成の補助、ジエトクツカー
に対する原料デンプンの懸濁、エアナイフコーテ
イングにおける脱水抑制に用いることができる。
顔 料
バイオポリマーPS87は輸送および貯蔵時に顔
料スラリーの懸濁に用いられ、凝結を抑制する働
きがある。
艶出し
バイオポリマーPS87は靴磨きにおける固形サ
スペンジヨン、銀と黄銅の艶出しにおける研摩サ
スペンジヨンおよびワツクス艶出し剤におけるエ
マルジヨン安定化を供することができる。
生 地
染・顔料の懸濁剤として、バイオポリマー
PS87はスペースプリントやKuester染色における
適用に調整でき、かつプリント使用中流動改良剤
として作用する。
壁 紙
バイオポリマーPS87はプリント使用中流動改
良剤および懸濁剤として使用することができる。
溶接棒
バイオポリマーPS87は押出し中潤滑にするこ
とができかつ生強度を供することができる。
脱臭ゲル
バイオポリマーPS87で作つた脱臭ゲルは熱的
に可塑性(フイルドホツトした時、冷却によりゲ
ル化する)である。しつかりしたゴム状ゲルが生
成し、ゆつくり脱臭剤を放出する。
フアイヤーフアイテイング
低濃度における高粘性により、バイオポリマー
PS87は木やかん木に対するフアイヤーフアイテ
イングのドロツプパターンおよび粘着性を改善す
ることができる。
紙のサイズ剤
紙工業によつてバイオポリマーPS87を使うと、
ロジン−フラムサイズ性の効果を促進し、
Mullenreadingを増大させ、そして内部耐水性を
改善することができる。
サスペンシヨン
バイオポリマーPS87溶液の高イールドストレ
ス値は各種材料の安定なサスペンシヨンを得るこ
とができる。
導火爆発物
高濃度の無機硝酸塩と匹敵する以外に、バイオ
ポリマーPS87は容易にゲル化して耐水性スラリ
ーを得ることができる。
各種写真処理
バイオポリマーPS87は写真処理溶液と非常に
混和性であり、写真処理の温度調節では熱的に可

凍結組成物
1〜6%W/Vの濃度のバイオポリマーPS87
溶液は、室温で流動しないゲルを生成する。この
ようなゲルは高電解質濃度の存在下でつくること
ができ、冷却用の凍結組成物として適当である。
例えば、3%W/VりバイオポリマーPS87およ
び22%W/VNaClを含有するゲル、遊離の電解
質溶液の放出がなく、伸流動性のロスがなく、繰
り返し凍結−解凍に堪える。
エマルジヨンの適用
例えば、ハンドクリーム、フオーム、ワツクス
ポリツシユ、アイスクリーム、マーガリンその他
非乳製スプレツド、チヨコレート、化粧品(リツ
プステイツク等)。
フオームの安定化
例えばミルクセーキ、ビール。
ゲルタイプの「cling」の適用
例えば、酸/中性クリーナー、浴槽の脱サビ、

硬度と20℃における降伏価がそれぞれ200ポアズ
以上及び50ダイン/cm2以上であることを特徴とす
るヘテロポリサツカライド。 2 40乃至45重量%のグルコース、10乃至20重量
%のガラクトース、25乃至30重量%のマンノー
ス、6乃至13重量%のグルクロン酸、及び0乃至
1.5重量%のフコースから成ることを特徴とする、
特許請求の範囲第1項記載のヘテロポリサツカラ
イド。 3 41重量%のグルコース、12重量%のガラクト
ース、28重量%のマンノース、9重量%のグルク
ロン酸、及び0.8重量%のフコースから成ること
を特徴とする、特許請求の範囲第1項又は第2項
記載のヘテロポリサツカライド。 4 グルコース、ガラクトース、マンノース、及
びグルクロン酸から主として構成され、任意には
フコースを含有するヘテロポリサツカライドにし
て、比旋光能[α]25 D=+60であり、かつ分子量
が34±4×104であつて、その1重量%溶液は擬

カライドの製造方法にして、該ヘテロポリサツカ
ライド合成に関与する遺伝物質を有するバチルス
属に属する微生物株を深部好気的条件下に、炭素
源、窒素源、鉄源、マグネシウム源及びリン源を
含有する水性培地中で培養し、ヘテロポリサツカ
ライドの実質的産生が起こるまで培地のPHの値を
4.5乃至7.5に保持し、次いで培地からヘテロポリ
サツカライドを単離することを特徴とする方法。 5 前記ヘテロポリサツカライドが実質的に蓄積
された培地を、8以上のPHにおいて、少なくとも
50℃に加熱し、ついで凝析した菌体と菌体残渣を
培地から分離して視覚上透明なヘテロポリサツカ
ライドを得ることを特徴とする特許請求の範囲第
