Description
Claims (16)
- 1.機能的蛋白質を表現し得る遺伝子:それと有効に会合した上記遺伝子のため の調節領域;相補ストランド上の隣接する第一および第二DNA配列によって限 定される上記調節領域内の蛋白質結合部位;上記第一DNA配列に対して同系の 第一DNA結合ドメインを有する第一蛋白質モノマー; および上記第二DNA配列に対して同系の第二結合ドメインを有する第二蛋白質 モノマー; 会合してヘテロダイマーを形成し得る上記第一および第二蛋白質モノマーを有し : それによって上記ヘテロダイマーが上記蛋白質結合部位に対して結合でき、従っ て上記遺伝子の表現を調節し:上記蛋白質結合部位が回転対称性を有さないよう に上記第一および第二DNA結合領域および上記第一および第二DNA配列が本 質的に異なっている、 ことから成る細胞。
- 2.蛋白質結合部位上に結合した上記ヘテロダイマーの存在が機能的蛋白質の生 産を阻害することによって遺伝子の調節を行なう請求の範囲1による細胞。
- 3.蛋白質結合部位上に結合した上記ヘテロダイマーの存在が機能的蛋白質の生 産を促進することによって遺伝子の調節を行なう請求の範囲1による細胞。
- 4.細胞、即ち 機能的蛋白質を表現し得る遺伝子: 上記遺伝子と有効に会合した調節領域;および相補ストランド上の隣接する異な るDNAによって限定される上記調節領域内の蛋白質結合部位を有し; それぞれ第一および第二の異なるDNA結合ドメインを各々が有する第一および 第二の異なる蛋白質モノマーを細胞中に供給し、上記モノマーが会合して、上記 蛋白質結合部位に対して結合でさそして機能的蛋白質の表現を調節できるヘテロ ダイマーを、形成することができる、ことから成る細胞、中で遺伝子表現を調節 する方法。
- 5.機械的蛋白質を表現するための遺伝子;上記遺伝子と有効に会合した調節領 域;および上記調節領域内の蛋白質結合部位を有し;結合した時、機械的蛋白質 の表現を調節する第一および第二の異なる蛋白質モノマーの会合から形成される ヘテロダイマーに対して結合するための相補ストランド上の隣接ずる異なる第一 および第二DNA配列によって蛋白質結合部位が限定される、 ことから成る組み換え遺伝子系。
- 6.機能的蛋白質を表現するための第一遺伝子;上記遺伝子と有効に会合した第 一調節領域;および上記第一調節領域内の蛋白質結合部位を有し;該蛋白質結合 部位が相補ストランド上の隣接する異なる第一および第二DNA配列によって限 定されており:第一蛋白質モノマーを表現するための第二遺伝子で上記蛋白質が 上記第一DNA配列に対して同系の第一DNA結合ドメインから成っており;そ して 第二蛋白質モノマーを表現するための第三遺伝子で、上記第二DNA配列に対し て同系の第二DNA結合ドメインから成っており;これによって上記第二および 第三遺伝子の表現が上記第一遺伝子の表現を調節する働きをしてもよい、 ことから成る原核生物もしくは真核生物の有機体のゲノム。
- 7.第二調節領域が上記第二遺伝子のために与えられており、上記第二領域が、 一個以上の調節子の存在または不在に応答して上記第一蛋白質モノマーの生産を 調節する働きをする請求の範囲6によるゲノム。
- 8.第三調節領域が、上記第三遺伝子のために与えられており、上記第三調節領 域が、請求の範囲7の調節子と同じか異っていてもよい一個以上の調節子の存在 または不在に応答して上記第二蛋白質モノマーの生産を調節する働きする請求の 範囲6または7によるゲノム。
- 9.上記調節子(類)の少なくとも一個が、請求の範囲5に従う遺伝子系によっ て生産される機能的蛋白質である請求の範囲第7または8によるゲノム。
- 10.隣接する異なる第一および第二DNA配列によって限定される蛋白質結合 部位に関して、第一および第二DNA結合ドメインに対して同系のそれぞれ異な る第一および第二DNA結合ドメインから成る異なる第一および第二蛋白質モノ マーの会合によって形成されるヘテロダイマー調節子蛋白質。
- 11.第一DNA結合ドメインがDNA配列:5′AXTXAAGXX3′ に対して同系であり、そして第二DNA結合ドメインがDNA配列:5′ACA AXXX3′ [式中、Xは、ヌクレオチド塩基A、T、CまたはGである]に対して同系であ る請求の範囲10によるヘテロダイマー。
- 12.第一DNA結合ドメインがDNA配列:5′ATTTAAGTT3′ に対して同系であり、そして第二DNA結合部位がDNA配列:5′ACAAT AA3′ に対して同系である請求の範囲11によるヘテロダイマー。
- 13.DNA配列: 5′AXTXAAGXXXXXTTGT3′〔式中、Xは、ヌクレオチド塩基A 、T、CまたはG]或いはその補体から成る蛋白質結合部位。
