Description
〔発明の技術分野〕
本発明は内毒素の吸着剤に関する。
〔従来技術とその問題点〕
内毒素は哺乳動物の血中に入ると発熱性と毒性
を示す物質である。これは、大腸菌、サルモネラ
菌、緑膿菌で代表されるグラム陰性菌の細胞壁外
膜から由来するリポ多糖体またはリポ多糖体・タ
ンパク質複合体であつて、高圧蒸気減菌でも分解
できない程、熱に安定な物質である。
したがつて、消化管は経ないで生体内に直接投
与する注射用の水・生理的食塩水、ブドウ糖やビ
タミンなどの栄養成分を含む輪液、血液透析用の
透析液、アルブミン等のタンパク質を含む輪液な
どには、内毒素が混入してはならない。内毒素の
吸着剤としてイオン交換樹脂が知られているが、
その吸着容量は小さく、実用化されていない。
〔発明の目的〕
内毒素に対する吸着容量の大きな吸着剤を見い
出すこと。
〔発明の構成〕
本発明は側鎖として、下記一般式(1)で示される
官能基を導入した芳香核を有するビニル系重合体
からなる内毒素吸着剤
(上式中、R1は炭素数にして4個以上、10個以
下のメチレン鎖長を有するアルキル基を示し、
R2およびR3はメチル基またはエチル基を示す。
またR4およびR5は水素原子または低級アルキル
基を示す。Xは塩素イオンで代表される通常の陰
イオンを示す。)を提供するものである。
〔効果の要約〕
本発明の重合体およびその成型品は、イオン交
換樹脂の10倍以上の内毒素の除去のほか、内毒素
血症治療に用いることができる。
〔構成および効果の説明〕
本発明でいう芳香核を有するビニル系重合体と
はスチレン、α−メチルスチレン、ビニルトルエ
ンなどで代表される芳香核を有するビニル系モノ
マの単独重合体もしくはこれらを主成分とする共
重合体を意味し、これらの重合体は架橋されてい
ればさらに好ましい。また該重合体は結晶性ポリ
プロピレン、ポリエチレンなどで代表されるポリ
α−オレフインで補強されていれば、機械的性質
が向上するので、さらに好ましい。例えば、ジビ
ニルベンゼンあるいはメチレンビスアクリルアミ
ド等で代表されるポリビニル化合物との共重合体
のほか、上記モノビニル化合物重合体成形品をホ
ルムアルデヒド、クロルスルホン酸等で架橋処理
したもの等があげられる。架橋重合体は流動性が
なく、成形が困難なので、該重合体成形品が繊
維、膜等の場合は成形後架橋処理する方法が好ま
しく採用される。
上記一般式(1)中、アルキル基R1のメチレン鎖
長は短すぎても長すぎても吸着能力が小さくな
る。R2およびR3のアルキル基はメチル基が最良
である。R4およびR5のアルキル基は水素原子で
ある場合が最も製造しやすい。
本発明の重合体を用いた成形品の表面積はあま
り小さすぎると、固定化密度が低くなるが、あま
り大きすぎても、本発明成形品を充填したカラム
の通液性が悪くなるので該成形品の表面積は0.01
以上50m2/g以下、より好ましくは、0.05以上10
m2/g以下がよい。
本発明重合体の上記一般式(1)で示される官能基
の量には特に限定はないが、少なすぎると本発明
重合体およびその形成品と溶液との親和性が悪く
なり、内毒素吸着能が低下するので、該重合体1
gあたり、0.5ミリモル以上、より好ましくは1.0
ミリモル以上存在するのがよい。
以下に実施例を示す。
実施例 1
ポリプロピレン(三井“ノーブレン”J3HG)
50分を島成分とし、ポリスチレン(スタイロン”
666)46分、ポリプロピレン(住友“ノーブレン”
WF−727−F)4分の混合物を海成分とする海
島型複合繊維(島数16、単系繊度2.6デニール、
引張強度2.9g/d、伸度50%フイラメント数42)
100gを、N−メチロール−α−クロルアセトア
ミド120g、ニトロベンゼン800g、およびパラホ
ルムアルデヒド1.7gからなる混合溶液中に浸し、
20℃で1時間反応させた。繊維を反応液から取り
出し、0℃の氷水10中に投じて、反応停止させ
たのち、水で洗浄し、次に、繊維に付着している
ニトロベンゼンをメタノールで抽出除去した。こ
の繊維(繊維A)を50℃で真空乾燥して、クロル
アセトアミドメチル化繊維71g(繊維A)を得
た。
上記で得た繊維A100gを100gのヨウ化カリウ
ムを含む10%含水エタノール2に浸し、50℃で
4時間加熱して、ヨードアセトアミドメチル化繊
維(繊維B)を得た。
繊維B10gをN,N−ジメチルヘキシルアミン
30gおよびジメチルホルムアミド170gからなる
溶液に浸し、80℃で5時間加熱した。さらに、こ
の繊維をクロマトカラムにつめ、1の1N−塩
酸、5の水、1の1N−カセイソーダ水、5
の水および10の1M−食塩水で、順次、洗浄
して、塩化N,N−ジメチル−N−ヘキシルアン
モニウムアセトアミドメチル化繊維(本発明試料
1)を得た。このものは、中性塩分解容量が1.83
ミリ当量/g、弱塩基性基量が0.05ミリ当量/g
で、Cl型含水度は1.9であつた。
実施例 2
実施例1で得た繊維B10gをN,N−ジメチル
オクチルアミン30gおよびジメチルホルムアミド
270gの混合溶液に浸し、80℃で5時間加熱した。
さらに、この繊維を、ソツクスレー抽出器で、10
時間メタノール抽出したのち、クロマトカラムに
つめ、1の1N−塩酸、5の水、1の1N−
カセイソーダ水、5の水および10の1N−食
塩水で洗浄して、塩化N,N−ジメタル−N−オ
クチルアンモニウムアセトアミドメチル化繊維
(本発明試料2)を得た。このものは、中性塩分
解容量が1.72ミリ当量/g、弱塩基性基量が0.05
ミリ当量/gで、Cl型含水度は1.3であつた。
実施例 3
本発明試料1、本発明試料2および比較試料1
について、以下の内毒素吸着実験を行なつた。
各試料毎に6本の試験管(外形24mm)を用意
し、その中に15mg、30mg、50mg、80mg、160mgお
よび300mgの試料を入れ、つぎに、10mlのリポ多
糖体水溶液(E.Coli 055:B5、トリクロル酢酸
抽出法、デイフコラボラトリーズ社製、0.11mg/
ml)を入れて、37℃で6時間振とうしたのち、No.
2の定性濾紙で濾過し、濾液についてリポ多糖体
の濃度をフエノール・硫酸法(試料2ml+5%フ
エノール水1ml+98%硫酸5ml;485mμ)で測
定した。母液中のリポ多糖体濃度と初期濃度
(0.11mg/ml)との差を吸着されたリポ多糖体量
とみなし、等温吸着線を求めた。各試料の吸着能
を比較するため、母液濃度が0.05mg/mlのときの
吸着能を等温吸着線から求めた結果を表1に示
す。

ル(1:1)溶液中55℃で8時間加熱して得た塩
化トリn−プロピルアンモニウムアセトアミドメ
チル化繊維であり、その中性塩分解容量は1.64ミ
リ当量/gで、弱塩基性基はなく、Cl型含水度は
1.57であつた。比較試料4は、実施例1の繊維B
をトリエチルアンモニウムの25%エタノール溶液
中55℃で5時間加熱して得た塩化トリエチルアン
モニウムアセトアミドメチル化繊維であり、その
中性塩分解容量は1.56ミリ当量/gで、弱塩基性
基量は0.09ミリ当量/gで、Cl型含水度は1.20で
あつた。
表1から本発明試料はイオン交換樹脂の10倍以
上の内毒素吸着能があることがわかる。また、比
較試料1〜4は官能基の種類や炭素数の点で本発
明試料とほとんど変わらないにもかかわらず、内
毒素に対する吸着能ははるかに小さい。このこと
から、本発明重合体の一般式(1)で示される官能基
の構造が、内毒素に対して特別な親和性を持つて
いることがわかる。
実施例 4
実施例1と同様にして下記一般式()で示さ
れる官能基を有する繊維状ポリスチレン誘導体
(本発明試料3)を調製した。
比較のために下記一般式()で示される官能
基を有する繊維状ポリスチレン誘導体(比較試料
5)を調製した。
E−coli055:B5B由来のリポ多糖体を注射用
蒸溜水に溶かし0.1mg/mlの溶液を調製した。
乾燥減菌された試験管にこの溶液を10mlずつ分
注した。
本発明試料3および比較試料5をそれぞれ20
mg、30mg、60mg、100mg、200mg用意して、各試験
管投入し、37℃で10時間振とうし、残液のリポ多
糖体濃度をフエノール・硫酸法にて定量し、等温
吸着線より平衡吸着量を算出した。

は48.4mg/g繊維であつた。
