Description

【発明の詳細な説明】[Detailed description of the invention]

本発明は、新規代用血液、さらに詳しくは、ヘ
モグロビン−ポリアルキレングリコール結合体を
酸素運搬物質として含有する代用血液において、
前記結合体のヘモグロビンとポリアルキレングリ
コールとがアミド結合により結合していることを
特徴とする代用血液に関する。 代用血液の酸素運搬物質として、ヘモグロビン
−ポリアルキレングリコール結合体が有用である
ことは知られている(特開昭56−12308号公報参
照。)。 本発明者は、上記物質に関し、さらに酸素供給
能の点を改良すべく鋭意研究した結果、結合体の
ヘモグロビンとポリアルキレングリコールとがア
ミド結合しているものが酸素供給能の点で一段と
優れ、またこのうち、特にポリアルキレングリコ
ールの両末端にヘモグロビンが結合している結合
体が膠質浸透圧の観点および原料費を半減できる
という工業的利点よりさらに優れており、酸素運
搬物質として実用上有効であることを見出し、こ
の発見に基づいて、本発明を完成するに至つた。 結合体に使用されるポリアルキレングリコール
としては、例えばポリエチレングリコール、ポリ
プロピレングリコール、エチレンオキサイド−プ
ロピレンオキサイド共重合体が挙げられる。 ヘモグロビンとしては、ヒト、ウシ、ブタ、ヒ
ツジ、ウマ、イヌ、サル、ウサギ、ニワトリ等の
ヘモグロビンを有する動物由来のものであればよ
い。なお、本件明細書中では、ヘモグロビンには
修飾ヘモグロビン、例えばピリドキサール誘導体
(ピリドキサール−5′−リン酸エステル、ピリド
キサール−5′−硫酸エステル、2−ノル−2−ホ
ルミルピリドキサール−5′−リン酸エステル等)、
糖リン酸誘導体(グルコース−6−リン酸エステ
ル、アデノシンモノリン酸エステル等)も含まれ
る。 前記ポリアルキレングリコールの分子量は300
〜20000程度、酸素供給能および粘度の点で好ま
しくは750〜10000程度である。 ポリアルキレングリコールをヘモグロビンとア
ミド結合により結合させるには、例えばポリアル
キレングリコールにカルボキシル基を付与せし
め、これとヘモグロビンのアミノ基との間で縮合
反応せしめアミド結合を形成せしめるとよい。 ポリアルキレングリコールにカルボキシル基を
付与するには、例えば公知方法、例えば米国特許
4179337号明細書や米国特許3941710号明細書に記
載の方法、アルカンジカルボン酸等ポリカルボン
酸のカルボキシル基の少なくとも1個とポリアル
キレングリコールの末端水酸基とを反応させてポ
リカルボン酸のエステルとする方法を採用すれば
よい。 このようなカルボキシル基を付与したアルキレ
ングリコールとヘモグロビンとの反応に際して、
例えばN−ヒドロキシコハク酸イミド、N−ヒド
ロキシフタル酸イミド、P−ニトロフエノール、
ペンタクロロフエノール等の通常のペプチド合成
におけるカルボン酸活性化剤により活性エステル
とし、これとヘモグロビンとを反応させアミド交
換することもできるし、塩化チオニル等酸ハロゲ
ン化剤を作用せしめてカルボンキシ基を有したポ
リアルキレングリコールの酸ハロゲン化物を調製
し、これとヘモグロビンを反応せしめることもで
きる。 なお、前記ポリカルボン酸の例としてはマロン
酸、コハク酸、グルタル酸、アジピン酸、リンゴ
酸およびクエン酸があるが、本酸素運搬物質の酸
素供給能はこれらポリカルボン酸の種類によつて
なんら変わるものではない。 ヘモグロビン−ポリアルキレングリコール結合
体のなかには、ヘモグロビン1分子にポリアルキ
レングリコール1分子がアミド結合しているも
の、ヘモグロビン1分子に2分子以上のポリアル
キレングリコールがアミド結合しているもの、ポ
リアルキレングリコールの両末端にヘモグロビン
が結合しているもの、その他種々ヘモグロビンと
ポリアルキレングリコールの共重合体が含まれ
る。このなかで、特にポリアルキレングリコール
1分子の両末端にヘモグロビン1分子ずつ結合し
ている結合体、もしくはこれを主成分とする共重
合体が、膠質浸透圧と酸素共給能をヒト血液に近
づけるという意味で好ましいことが見出されてい
る。 ヘモグロビン−ポリアルキレングリコール結合
体において、ポリアルキレングリコール末端水酸
基を有するものについては、保護されていてもよ
く、例えば炭素数1乃至16のアルコールによるエ
ーテル化、炭素数2乃至18の脂肪酸によるエステ
ル化、あるいは炭素数1乃至18のアミンによるイ
ミノ化による保護方法が採用される。 アミド結合によりポリアルキレングリコールが
結合したヘモグロビンにおいて1サブユニツトあ
たりのポリアルキレングリコールの結合数および
1サブユニツトあたりの分子量は、鰺坂らの方法
(K.Ajisaka and Y.Iwashita,Biochem.
Biophys.Res.Commun.97,1076〜1081(1980)に
より測定した。ヘモグロビンサブユニツト1分子
当り重合物、例えばポリエーテルカルボン酸は1
〜10分子程度が結合している。 本発明において使用する、アミド結合により重
合物が結合したヘモグロビンは、例えば次の如く
して調製することができる。 (1) ポリエーテルカルボン酸と、1〜10倍モル好
ましくは2倍モルのN−ヒドロキシコハク酸イ
ミドとを、N,N−ジメチルホルムアミド中、
1〜10倍モル好ましくは2倍モルのジシクロヘ
キシルカルボジイミドの存在下3〜20時間好ま
しくは8〜14時間室温または加熱下撹拌する。
反応後生成したジシクロヘキシル尿素を濾過除
去後エーテル等を加えてポリエーテルカルボン
酸活性化エステルを得る。得られた物質をPH7
〜9、好ましくはPH7〜9、好ましくは8〜
8.5で水溶液中または緩衝溶液中1〜1/100倍好
ましくは1/5〜1/30倍程度のヘモグロビンとを
反応せしめる。 上記反応においてN−ヒドロキシコハク酸イ
ミドに代えてN−ヒドロキシフタル酸イミド、
p−ニトロフエノール、ペンタクロロフエノー
ルを用いても同様に調製することができる。 (2) ポリエーテルカルボン酸と1〜10倍モル、好
ましくは5倍モルのイミダゾールとをN,N−
ジメチルホルムアミドに溶解し、これに1〜10
倍モル、好ましくは5倍モルのジシクロヘキシ
ルカルボジイミドを加えて加熱環流下5〜20時
間、好ましくは10時間反応せしめる。反応後冷
却し生じたジシクロヘキシル尿素を濾過除去
し、濾液にエーテル等を加えてポリエーテルカ
ルボン酸アミドを得る。このポリエーテルカル
ボン酸アミドをPH7〜9、好ましくはPH8〜
8.5で水溶液中または緩衝液中1倍〜1/100倍、
好ましくは1/5〜1/30倍程度のヘモグロビンと
反応せしめる。上記反応においてイミダゾール
に代えてコハク酸イミドもしくはフタル酸イミ
ドを用いてもよい。 またカルボン酸アミドの調製法としてポリエ
ーテルカルボン酸と1〜10倍モル、好ましくは
2倍モルのカルボニルジイミダゾールとをN,
N−ジメチルホルムアミド中反応させてもよ
い。 (3) ポリエーテルカルボン酸と大過剰量の塩化チ
オニルとを60〜90℃、好ましくは75〜80℃で1
〜5時間、好ましくは1.5〜2時間反応後減圧
下塩化チオニルを留去し、得られたポリエーテ
ルカルボン酸塩化物と1/10〜1/100倍程度、好
ましくは1/30〜1/50倍程度のヘモグロビンとを
PH8.5〜9.5の水溶液中もしくは緩衝液中反応せ
しめる。 上記反応において塩化チオニルに代えてオキ
シ塩化リン、あるいは5塩化リン等の酸ハロゲ
ン化剤を用いてもよい。 (4) ポリエーテルジカルボン酸と1〜10倍モル好
ましくは3倍モルのN−ヒドロキシコハク酸イ
ミドとをN,N−ジメチルホルムアミド中、1
〜10倍モル好ましくは3倍モルのジシクロヘキ
シルカルボジイミドの存在下3〜20時間好まし
くは8〜14時間室温または加熱下撹拌する。反
応後生成したジシクロヘキシル尿素を濾過除去
後、エーテル等を加えてポリエーテルジカルボ
ン酸活性化ジエステルを得る。その後の反応は
(1)と同様にしてヘモグロビンと反応せしめる。 なお、(1)〜(4)においてヘモグロビンには前述の
如く修飾ヘモグロビンが含まれることはもちろん
である。 以下、実施例および各種試験により本発明を詳
細に説明する。 実施例 1 モノメトキシポリオキシエチレンコハク酸モノ
エステル(平均分子量5000)5g(0.001モル)、
N−ヒドロキシコハク酸イミド0.23g(0.002モ
ル)、ジシクロヘキシルカルボジイミド0.42g
(0.002モル)を、300mlのN,N−ジメチルホル
ムアミドに溶解し、室温で一夜撹拌した。 析出したジシクロヘキシル尿素を濾過し、濾液
に600mlのエチルエーテルを加え、生成したモノ
メトキシポリエチレングリコールモノ(サクシイ
ミジルサクシネート)結晶を濾過し、エーテルで
よく洗い乾燥し、4.6gの白色結晶を得た。ピリ
ドキサールリン酸結合ヘモグロビン0.5g
(0.0077ミリモル)を100mlのホウ酸緩衝液(PH
8.5)に溶解した液に氷冷下上記活性エステル0.5
g(0.1ミリモル)を加えた。氷冷下4時間撹拌
した後分子量阻止3万の限外濾過膜により限外濾
過をくり返し、未反応の活性エステルまたはその
分解物を除去することにより修飾ヘモグロビン溶
液を得た。 このものはTSK G3000SWカラム(東洋ソー
ダ(株)製)を用いた高速液クロにおいて単一ピーク
を示した。このものを凍結乾燥して0.58gの修飾
ヘモグロビンを得た。置換度6.0、分子量95000。
但し、置換度および分子量はヘモグロビン1分子
(α2β2型)に対応している。 実施例 2 ポリオキシエチレンアジピン酸モノエステル
(平均分子量2000)2g(1ミリモル)、ペンタク
ロロフエノール0.27g(1ミリモル)、ジシクロ
ヘキシルカルボジイミド0.25g(1.2ミリモル)
を30mlのN,N−ジメチルホルムアミド中室温に
て一夜反応させた後生成したジシクロヘキシル尿
素を濾別した。濾液にエチルエーテルを加え析出
した結晶を濾過し、トルエンから再結晶し1.5g
の結晶を得た。 上記活性エステル0.22g(0.1ミリモル)を0.05
%一酸化炭素型ヘモグロビン水溶液20ml(0.016
ミリモル)に少量ずつ添加した。その際0.2Nカ
セイソーダ水溶液によりPHを8.5に保つた。 反応溶液を分子量阻止3万のメンブランにて限
外濾過をくり返すことにより0.4%修飾ヘモグロ
ビン溶液2mlを得た。こおものは高速液体クロマ
トグラフイにおいて単一ピークを示した。置換度
3.5、分子量72000。 実施例 3 ポリエチレングリコールモノマロン酸エステル
(平均分子量850)8.5g(0.01モル)、パラニトロ
フエノール2.07g(0.015モル)、ジシクロヘキシ
ルカルボジイミド2.3g(0.015モル)を300mlの
N,N−ジメチルホルムアミドに溶解し室温にて
一夜反応させた。反応終了後生成したジシクロヘ
キシル尿素を濾別し、濾液にエチルエーテル600
mlを加えて6.2gの結晶を得た。 上記活性エステルを用いて実施例1と同様の方
法により1%ピリドキサール硫酸結合ヘモグロビ
ン溶液10mlに反応させることにより0.8%修飾ヘ
モグロビン溶液10.5mlを得た。置換度6.2、分子
量70000。 実施例 4 モノメトキシポリオキシエチレンコハク酸モノ
エステル(平均分子量2000)4g(0.002モル)、
N,N−カルボジイミダゾール0.7g(0.004モ
ル)を100ml、N,N−ジメチルホルムアミド中
室温で一夜撹拌を行つた。反応液にエチルエーテ
ル200mlを加えて沈澱を濾過後エチルエーテルで
よく洗浄することによりイミダゾールのモノメチ
ルポリオキシエチレンコハク酸モノイミド2.2g
を得た。 6%ピリドキサールリン酸結合ヘモグロビン6
ml(0.0055ミリモル)を0.1Mトリス緩衝液(PH
8.0)60mlに溶解し、これに上記イミド1g(0.5
ミリモル)を加えた。氷冷下4時間反応後、分子
量阻止3万の限外濾過膜により限外濾過をくり返
し8.3%の修飾ヘモグロビン水溶液3.6mlを得た。
置換度10.4、分子量86000。 実施例 5 モノメトキシポリオキシエチレンコハク酸エス
テル(平均分子量5000)5g(0.001モル)、コハ
ク酸イミド0.5g(0.005モル)、ジシクロヘキシ
ルカルボジイミド1g(0.005モル)をN,N−
ジメチルホルムアミド50mlに溶解し12時間加熱還
流した。生成したジシクロヘキシル尿素を濾過後
エチルエーテル150mlを加え生じた沈澱を濾過し
た。沈澱物をエチルエーテルでよく洗浄、乾燥し
て3.2gの結晶を得た。 上記イミドを実施例4と同様にして3.2%のピ
リドキサールリン酸結合ヘモグロビン7mlと反応
させることにより4.8%修飾ヘモグロビン溶液3.8
mlを得た。置換度11.2、分子量120000。 実施例 6 ポリエチレングリコールマロン酸エステル(平
均分子量5200)5.2g(0.001モル)を20mlの塩化
チオニルに溶解し75〜80℃に1.5時間加熱した。
減圧下塩化チオニルを留去し、生じた酸塩化物の
結晶をよく乾燥させた。1%ピリドキサールリン
酸結合一酸化炭素型ヘモグロビン20mlを200mlの
0.7Mホウ酸緩衝液(PH10.0)に溶解し、氷冷下、
上記酸塩化物5gを少量ずつ添加した。 氷冷下3時間撹拌後分子量阻止3万のメンブラ
ンにより限外濾過をくり返し、4.1%修飾ヘモグ
ロビン溶液4.6mlを得た。置換度9.3、分子量
114000。 実施例 7 局方ポリエチレングリコール(平均分子量
4000)100g(0.025モル)、無水コハク酸6.3g
(0.063モル)をN,N−ジメチルホルムアミド
100mlに溶解し、100℃で3時間撹拌した。冷却後
エチルエーテル400mlを加えて生じる沈澱を濾過
した。沈澱物をエチルエーテルでよく洗浄、乾燥
して97.5gのポリエチレングリコールコハク酸ジ
エステルの結晶を得た。 上記結晶97.5g(0.024モル)、N−ヒドロキシ
コハク酸イミド6.3g(0.054モル)、ジシクロヘ
キシルカルボジイミド11.4g(0.054モル)を100
mlのN,N−ジメチルホルムアミドに加熱して溶
解したのち30℃にて一夜撹拌した。生成したジシ
クロヘキシル尿素を濾過し、濾液に300mlのエチ
ルエーテルを加え生成したポリエチレングリコー
ルジ(サクシイミジルサクシネート)結晶を濾過
し、エーテルでよく洗い乾燥し、95gの白色結晶
を得た。 16.4%ヘモグロビン溶液10ml(0.025ミリモル)
を0.2Mホウ酸緩衝液(PH8.5)35mlに溶解し、こ
れにポリエチレングリコールジ(サクシイミジル
サクシネート)結晶4.3g(1.0ミリモル)を加え
た。4℃にて一夜撹拌後、分子量阻止10万の限外
濾過膜により限外濾過をくり返し、3.2%の修飾
ヘモグロビン水溶液51mlを得た。置換度11.2、分
子量122000。 実施例 8 15%のピリドキサールリン酸結合ヘモグロビン
水溶液14.3ml(0.1ミリモル)を270mlのリン酸緩
衝液(PH7.0)に溶解し、4℃にて実施例7にお
いて調製されたポリエチレングリコールジ(サク
シイミジルサクシネート)結晶4.43g(1.1ミリ
モル)を少量ずつ加えた。4℃にて4時間撹拌し
た後分子量阻止10万の限外濾過膜により限外濾過
をくり返すことにより、11.6%濃度の修飾ヘモグ
ロビン溶液11mlを得た。置換度6.6、分子量
93000。 試験 1 実施例1〜8の方法により調製した修飾ヘモグ
ロビンについて今井らの方法(K.Imai,H.
Morimoto,M.Kotani,H.Watari.H.Waka,
