Description

【発明の詳細な説明】[Detailed description of the invention]

本発明は新規抗生物質AC−1978およびその製
造法に関する。 本発明者らは新規な抗生物質の探索を目的とし
て種々の土壌から菌株を分離し、その産生する代
識産物について研究を続けた結果、新たに土壌よ
り分離したAC−1978菌株の培養物中にグラム陽
性菌に対して抗菌活性を有する物質が産生される
ことを見い出した。そこで、該培養物から該抗菌
物質を分離し、精製し、該物質の理化学的性質を
調べた結果、後記の通りの性質を有することが判
つた。このような理化学的性質を有する物質はペ
プチド系物質と考えられるが、他に見当らないこ
とから、該物質を抗生物質AC−1978と命名した。
本発明はかゝる知見に基いて完成されたものであ
る。 本発明は後記の理化学的性質を有する抗生物質
AC−1978またはその塩およびストレプトマイセ
ス属に属する抗生物質AC−1978生産菌を培地に
培養し、得られた培養物から抗生物質AC−1978
を採取することを特徴とする抗生物質AC−1978
またはその塩の製造法である。 本抗生物質AC−1978生産菌はストレプトマイ
セス属に属するが、例えば本発明者らが分離した
AC−1978菌株は本発明に最も有効に使用される
菌株の一例であつて、本菌株の菌学的性質を示す
と次の通りである。 (a) 形態的性状 AC−1978菌株の栄養菌系は各種寒天培地で
よく発達し、通常は隔壁を有しない。気菌系は
スターチ・無機塩寒天培地、栄養寒天培地など
では豊富に着生し、ビロード状を呈する。顕微
鏡下の観察では、胞子柄は螺旋状を呈し、胞子
は楕円形で、10個以上連なり、表面は平滑であ
る。胞子の大きさは0.9×1.1μmである。菌核、
胞子のうおよび遊走子は見出されない。 (b) 次の各培地における生育状態 E.B.ShirlingおよびD.Gottliebの方法〔Int.J.

ない、特記しない限り各培地上で27℃、14日間

マイセス・スピーシーズAC−1978と命名した。 本菌株は工業技術院微生物工業研究所に寄託
番号微工研菌寄第7452号(FERM−PNo..
7452)として寄託されている。 以上、抗生物質AC−1978生産菌について説明
したが、放線菌の一般的性状として菌学上の性質
は極めて変異し易く、一定したものではなく、自
然的にあるいは通常行われる紫外線照射、X線照
射あるいは変異誘導剤などを用いる人工的変異手
段により変異することは周知の事実であり、この
ような人工的変異株は勿論、自然変異株も含め、
ストレプトマイセス属に属し、抗生物質AC−
1978を生産する能力を有する菌株はすべて本発明
に使用することができる。 本発明においては、先ずストプトマイセス属に
属する抗生物質AC−1978生産菌が適当な培地に
培養される。本菌の培養においては通常放線菌を
培養する方法が一般に用いられる。培地として
は、微生物が同化し得る炭素源、消化し得る窒素
源、さらには必要に応じ無機塩などを含有させた
栄養培地が使用される。同化し得る炭素源として
は、グルコース、フラクトース、マルトース、キ
シロース、マンニツト、ガラクトース、リボー
ス、グリセリン、糖密、澱粉、デキストリン、コ
ーン・スチープ・リカーなどの炭水化物が単独ま
たは組合せて用いられる。またグルコン酸、ピル
ビン酸、乳酸、酢酸などの有機酸、メタノール、
エタノールなどのアルコールやn−パラフインな
どの非芳香族系炭化水素、あるいは植物性または
動物性の油脂なども使用可能である。消化し得る
窒素源としては、ペプトン、肉エキス、酵母エキ
ス、乾燥酵母、大豆粉、大豆蛋白分解物、カゼイ
ン、アミノ酸、尿素、NZ−アミン、コーン・ス
チープ・リカー、フイツシユ・ミール、蛹加水分
解物などの有機窒素源、硝酸塩、アンモニウム塩
などの無機窒素化合物が単独または組合せて用い
られる。その他に、必要に応じナトリウム塩、カ
リウム塩、カルシウム塩、マグネシウム塩、リン
酸塩などの無機塩類が添加される。さらに、培地
には、必要に応じ、本菌株の生育や抗生物質AC
−1978の生産を促進する微量重金属、微量栄養
素、発育促進物質、前駆物質を適当に添加しても
よい。 培養は通常振とうまたは通気撹拌培養などの好
気的条件下で行うのがよい。工業的には深部通気
撹拌培養が好ましい。培地のPHはやゝ酸性ないし
中性附近で培養を行うのが好ましい。培養温度は
20〜40℃でも行い得るが、通常は24〜30℃、好ま
しくは27℃附近に保つのがよい。培養時間は、液
体培養の場合、通常1〜8日培養を行えばよい
が、好ましくは本抗生物質の培養物中の蓄積量が
最大に達した時に培養を終了すればよい。これら
の培地組成、培地の液性、培養温度、撹拌速度、
通気量などの培養条件は使用する菌株の種類や外
部の条件などに応じて好ましい結果が得られるよ
う適宜調節、選択されることは言うまでもない。
液体培養において発泡があるときは、シリコン
油、植物油、界面活性剤などの消泡剤を適宜使用
される。 このようにして得られた培養物中に蓄積された
本抗生物質AC−1978は菌体内および培養液中
に含有されるので、遠心分離して培養液と菌体
とに分離し、各々から本抗生物質AC−1978を採
取するのが有利である。 培養液から本抗生成物質AC−1978を採取す
るには、培養液を酢酸エチルなどの非親水性有
機溶媒で抽出するか、あるいは培養液を活性
炭、アルミナ、多孔性合成高分子樹脂、イオン交
換樹脂などに吸着させ、酢酸エチルなどの溶出溶
媒で溶出し、得られた抽出液または溶出液を減圧
濃縮するかまたはヘキサンなどの有機溶媒を加え
て沈澱させればよい。得られた粗物質はさらに脂
溶性物質の精製に通常用いられる公知の方法、例
えばシリカゲル、アルミナなどの担体を用いるカ
ラムクロマトグラフイーにより精製することがで
きる。 菌体から本抗生物質AC−1978を採取するには、
菌体を含水アセトンなどの含水親水性有機溶媒で
抽出し、得られた抽出液を減圧濃縮し、その濃縮
物を酢酸エチルで抽出し、この酢酸エチル抽出液
は前記の培養液から得た酢酸エチル抽出液と合
わせて分離精製するか、あるいは前記と同じ方法
で分離精製することができる。 次に、本抗生物質AC−1978の理化学的性質お
よび生物学的性質について述べる。 理化学的性質 元素分析炭素、水素、窒素および酸素から

められなかつた。 上記の通り本抗生物質AC−1978は毒性が低く、
主としてある特定のグラム陽性菌の抗菌活性を有
するので、ヒトおよび動物の細菌感染症に対する
治療薬あるいは予防薬として有用である。 次に実施例を挙げて本発明を具体的に示すが、
これにより本発明の製造を限定するものではな
い。 実施例 500ml容三角フラスコにグルコース1.0%、デキ
ストリン1.0%、酵母エキス0.5%、NZ−アミン
(Aタイプ)0.5%、炭酸カルシウムを含む液体培
地(PH7.0)100mlを分注し、121℃で15分間蒸気
滅菌し、これらに上記組成に寒天1.2%を含む寒
天斜面培地上で27℃で培養したストプトマイセ
ス・スピシーズAC−1978(FERM−PNo.7452)
の斜面培養から1白金耳づつ接種し、回転式振と
う機を用い27℃で2日間振とう培養して種母を得
た。 100ml容ジヤー・フアーメンターにグリセリン
2.0%、コーン・スチープ・リカー1.5%、大豆粉
0.5%、炭酸カルシウム0.3%、塩化コバルト
0.0002%を含む液体培地(PH6.5)70を仕込み、
121℃で15分間蒸気滅菌した。これに上記の種母
21本分を移植し、撹拌速度250r.p.m.通気量20
/分の条件下で30℃で69時間通気撹拌培養し
た。培養液をシヤープレス型遠心機で遠心分離
(10000r.p.m.)して菌体と培養上清に分別した。 得られた菌体に75%アセトン水16を加えて撹
拌抽出し、これを別してアセトン抽出液を得
た。この抽出液を減圧下1まで濃縮した。濃縮
液に酢酸エチル500mlを加えて撹拌抽出する操作
を10回行なつた。得られた酢酸エチル抽出液を減
圧で50mlまで濃縮し、濃縮液にヘキサン1を加
え、4℃で一夜放置した。生じた沈澱物を取、
乾燥して粗製物2.7gを得た。 一方、培養上清に酢酸エチル30を加え、よく
撹拌した後、遠心分離により分液した。酢酸エチ
ル層を減圧下50mlまで濃縮し、濃縮液にヘキサン
1を加え、4℃で一夜放置した。生じた沈澱物
を取、乾燥して粗製物4.3gを得た。これを前
記の粗製物と合わせ、できるだけ少量のクロロホ
ルム−メタノール(50:1)に溶かし、これをク
ロロホルム−メタノール(50:1)で充填したシ
リカゲル(メルク社製、キーゼルゲル60,
Art7734)200gのカラムにチヤージし、クロロ
ホルム−メタノール(50:1)で溶出するカラム
クロマトグラフイーを行た。各フラクシヨンをシ
リカゲル薄層クロマトグラフイー(理化学的性質
の項参照)およびミクロコツカス・ルテウスを
用いる生物検定法により追跡して活性成分を含む
フラクシヨンを集めて減圧濃縮して粗製物質820
mgを得た。これをできるだけ少量のクロロホルム
−アセトン(10:1)に溶かし、予めクロロホル
ム−アセトン(10:1)で充填したシリカゲル
(メルク社製、キーゼルゲル60,Art7734)200g
のカラムにチヤージし、クロロホルム−アセトン
(10:1)1、クロロホルム−アセトン(5:
1)1、クロロホルム−アセトン(3:1)
400ml、アセトン500mlの順で溶出するカラムクロ
マトグラフイーを行つた。溶出液は20mlづつ分画
し、クロロホルム−アセトン(5:1)で溶出さ
れた48〜100本目のフラクシヨンを集めて減圧乾
固して抗生物質AC−1978を無色定形物として得
た。収量350mg。
