Description
〔産業上の利用分野〕
本発明はアミノ酸アミドの新規製造法に関す
る。
カルシトニン(Calcitonin)、性腺刺激ホルモ
ン放出ホルモン(LH−RH)、メラニン細胞刺激
ホルモン(α−MSH)、セクレチン(Secretin)、
P物質(SubstanceP)、甲状腺刺激ホルモン放出
ホルモン(TRH)、血管作用性小腸ペプチド
(VIP)、バソプレツシン(Vasopressin)のよう
に、天然の生理活性ペプチドの中にはカルボキシ
ル基末端がアミド化されたペプチドが数多く存在
する(フアルマシアレビユーNo.3、生理活性ペプ
チド、日本薬学会、1980参照)。アミノ酸アミド
はこれらの天然のペプチドおよびその関連化合物
を合成する出発原料として極めて重要な化合物で
ある。
また、N−保護アミノ酸アミドは単環β−ラク
タムの出発材料としても有用な物質である。例え
ば、N−ベンジルオキシカルボニル−L−スレオ
ニンアミドはアズスレオナムの出発原料である
(特開昭56−125362号参照)。
〔従来の技術〕
従来アミノ酸アミドまたは、N−保護アミノ酸
アミドの製造法に関しては例えば、
1 アミノ酸の低級アルキルエステルを加アミン
分解によりアミノ酸アミドを得、しかるのち必
要に応じて遊離のアミノ基を保護する方法
2 N−保護アミノ酸とアミンをジシクロヘキシ
ルカルボジイミド、エチルジメチルアミノプロ
ピルカルボジイミドなどのカルボジイミドを用
いて直接縮合させ、次いで必要に応じて保護基
を除去する方法
3 N−保護アミノ酸とアルキル炭酸の混合酸無
水物をアミンと反応させ、次いで必要に応じて
保護基を除去する方法
などが知られている。
〔発明が解決しようとする問題点〕
しかし、これらの方法は、いずれも実験室的規
模での合成には適しても大量生産には適さない。
すなわち、1)の方法を実施するには大過剰のア
ンモニアガスとアンモニアガスの漏洩を防ぐため
加圧反応釜が必要であり、価格面と安全面で問題
がある。2)の方法では高価で、かつ皮膚に付着
すると激しいカブレを引き起すカルボジイミドが
必要であり、また、3)の方法では、混合酸無水
物が不安定なため−20℃以下という反応条件が必
要であり、価格面と設備面で問題が残る。
〔問題点を解決するための手段〕
そこで、本発明者等はN−保護アミノ酸アミド
類の大量生産に適した方法を開発すべく鋭意検討
した結果、ハロゲン化リンおよびオキシハロゲン
化リンから選ばれた少なくとも1種のリン化合物
と一般式()
(式中XおよびYは同一もしくは異なつて水素
原子、炭素数1個ないし7個の低級アルキル基、
アリル基またはアラルキル基を示す。)
で表わされるアミンを反応させ、次いでN−保護
アミノ酸(側鎖に官能基ある場合、その官能基は
保護されていてもよい)を反応させ、必要に応じ
て保護基を除去すると一般式()
(式中A−COはアミノ基が保護されていても
よいアミノ酸残基を示す。)
で表わされるアミノ酸アミドが得られることを見
い出した。
本発明をさらに詳細に説明すると、本発明の原
料として用いられるハロゲン化リンとしては例え
ば三ハロゲン化リンや五ハロゲン化リンなどがあ
げられ、具体的には例えば三塩化リン、三臭化リ
ン、五塩化リン、五臭化リンなどがあげられる。
オキシハロゲン化リンとしては例えばオキシ塩化
リン、オキシ臭化リンなどがあげられる。
また使用されるアミン類は一般式()で表わ
されるアミンであり、X,Yは同一または異つて
水素原子または炭素数1〜7のアルキル基、アリ
ル基、アラルキル基を示し、分岐可能なものは分
岐していても良く環状であつても差支えない。
X,Yの具体的な例としてはメチル基、エチル
基、プロピル基、ブチル基、ペンチル基、ヘキシ
ル基、ヘプチル基等の炭素数1ないし7の低級ア
ルキル基、フエニル基およびベンジル基などがあ
げられる。
一般式()のアミンの具体的な代表例として
は、アンモニア、メチルアミン、エチルアミン、
プロピルアミン、ブチルアミン、ペンチルアミ
ン、ヘキシルアミン、ヘプチルアミン、ジメチル
アミン、ジエチルアミン、ジプロピルアミン、メ
チルベンジルアミン、エチルベンジルアミン、ア
ニリン、ベンジルアミンなどがあげられる。
本発明で使用されるN−保護アミノ酸における
アミノ酸としてはモノアミノモノカルボン酸に限
定されるものではなく、側鎖に官能基を有するア
ミノ酸であつても何ら差し障りない。アミノ酸の
具体的な代表例としては、
グリシン、アラニン、β−アラニン、オルニチ
ン、アスパラギン酸、アスパラギン、グルタミン
酸、グルタミン、ピログルタミン酸、ヒスチジ
ン、ヒドロキシリジン、ヒドロキシプロリン、イ
ソロイシン、ロイシン、リジン、フエニルアラニ
ン、プロリン、セリン、スレオニン、トリプトフ
アン、チロシン、バリン、フエニルグリシン、ヒ
ドロキシフエニルグリシン、γ−アミノ酪酸、メ
チオニン、アルギニン、システインなどがあげら
れる。
本発明のN−保護アミノ酸の保護基としてはペ
プチド化学で用いられるアミノ基の保護基を用い
ることができるが、中でも好ましい保護基として
はベンジルオキシカルボニル基、フエニル基上に
低級アルコキシ基、低級アルキル基、ハロゲン原
子、ニトロ基を有するベンジルオキシカルボニル
基(例えばp−メトキシベンジルオキシカルボニ
ル基、p−クロロベンジルオキシカルボニル基、
p−ブロモベンジルオキシカルボニル基、p−ニ
トロベンジルオキシカルボニル基)、tert−アル
コキシカルボニル基(例えばtert−ブチルオキシ
カルボニル基、tert−ペントキシカルボニル基)、
ホルミル基およびフタリル基が一般的である。
側鎖に官能基を有するアミノ酸を使用する場
合、この側鎖官能基は保護されていてもよい。こ
こで使用される保護基は側鎖官能基の種類に応
じ、ペプチド化学で通常用いられる保護基を用い
ることができる。側鎖官能基の保護されたアミノ
酸の代表例としては、δ−ベンジルオキシカルボ
ニル又はt−ブトキシカルボニル−オルニチン、
アスパラギン酸β−ベンジル又はt−ブチルエス
テル、グルタミン酸γ−ベンジル又はt−ブチル
エステル、Nim−ベンジルヒスチジン、Nim−ト
シルヒスチジン、O−ベンジル又はt−ブチルヒ
ドロキシリジン、O−ベンジル又はt−ブチルヒ
ドロキシプロリン、ε−ベンジルオキシカルボニ
ル又はt−ブトキシカルボニルリジン、O−ベン
ジル又はt−ブチルセリン、O−ベンジル又はt
−ブチルスレオニン、O−ベンジル又はt−ブチ
ルチロシン、O−ベンジル又はt−ブチルヒドロ
キシフエニルグリシン、メチオニンスルホキシ
ド、NG−トシル又はニトロ又はベンジルオキシ
カルボニルアルギニン、s−ベンジル又はメトキ
シベンジル又はアセトアミドメチルシステインな
どがあげられる。
なお、不整炭素を有するアミノ酸の立体配位は
L,D,DLのいずれでもよい。
本発明を実施するには2種類の方法を採用する
ことができる。すなわち、その一つは先述の溶媒
中にハロゲン化リンまたはオキシハロゲン化リン
(以下ハロゲン化リン類と省略する。)を溶解さ
せ、氷冷下アミン類を加えて反応させ、しかるの
ち反応生成物を単離し、溶媒中でN−保護アミノ
酸と反応させる方法であり、他の一つは生成物を
単離することなく溶液のまま次のN−保護アミノ
酸との反応に供する方法である。しかしながら、
反応生成物の安定性および操作性の両面から考え
て後者の方法が望ましい。
本発明に用いる溶媒としては反応に関与しない
溶媒であれば特に制限はなく、具体的な例として
はジクロロメタン、クロロホルム、四塩化炭素の
ようなハロゲン化炭化水素類、ジオキサン、テト
ラヒドロフランのような環状エーテル類、ベンゼ
ン、トルエン、キシレン、のような芳香族炭化水
素類、酢酸メチルエステル、酢酸エチルエステ
ル、酢酸n−プロピルエステルのようなエステル
類などがあげられる。
本発明の反応試薬であるハロゲン化リン類の量
はN−保護アミノ酸に対して0.5モル当量以上で
あれば特に制限はないが1.0ないし1.5モル当量が
好ましい。またアミンの量はハロゲン化リン類に
対し3〜10モル当量好ましくは5〜8モル当量で
ある。
本発明に用いる反応温度は特に制限はないが通
常室温から溶媒の沸点の間で行うことができるが
沸点が60〜100℃の溶媒を用い、沸点ないし沸点
に近い温度で行うことが望ましく、反応は通常2
〜8時間で完結する。
かくして生成したN−保護アミノ酸アミドはそ
のもの自身反応溶媒に溶解する場合は水を加え目
的化合物を有機溶媒層に抽出し、必要とあらば5
%炭酸水素ナトリウム水溶液、飽和食塩水で順次
洗浄を行い溶媒を減圧除去した後、水または適当
な溶媒を加え結晶化させることによつて簡単に単
離することができる。
目的化合物が反応溶媒に溶解しない場合は、い
つたん溶媒を減圧除去した後、抽出溶媒を加え上
記と同様の操作により単離できる。また目的化合
物が水に不溶な場合は溶媒を減圧除去後、水を加
え結晶化させることにより単離できる。単離され
たN−保護アミノ酸アミドは必要に応じてN−保
護基を公知の方法で除去してアミノ酸アミドとす
ることもできる。
保護基の除去は保護基の種類または溶媒等の種
類に応じて、溶媒の沸点以下の温度例えば、−50
℃〜150℃好ましくは−40℃〜120℃の範囲で行う
ことができ、溶媒としては無機、有機の種々の溶
媒を使用することができるが、無機溶媒としては
水、液体アンモニア、液体フツ化水素等が、有機
溶媒としてはC1〜C4のアルコール、酢酸、ジメ
チルホルムアミド、ジオキサン等の水性溶媒、酢
酸低級アルキルエステルなどをあげることがで
き、これらは場合により混合溶媒として使用する
ことができる。
保護基の除去方法としては保護基の種類によ
り、還元(例えば接触還元またはアルカリ金属と
アンモニアによる還元)、加水分解、酸分解、ヒ
ドラジン分解等の方法があげられる。好ましい保

ンモニアガス1.02g(60.0mmol)を5〜20℃の
温度範囲で導入する。導入後、N−(tert−ブチ
ルオキシカルボニル)−L−バリン2.16g
(10.0mmol)を加え、加熱還流下7時間反応させ
る。反応後反応液を減圧濃縮してクロロホルムを
留去し、残査に酢酸エチル20mlと水20mlを加え、
生成物を酢酸エチル層に抽出する。抽出液を5%
炭酸水素ナトリウム水溶液10mlと飽和食塩水10ml
で順次洗浄し、得られた酢酸エチル層を無水硫酸
ナトリウムで乾燥させ溶媒を減圧にて留去すると
N−(tert−ブチルオキシカルボニル)−L−バリ
ンアミド1.60g(収率74.3%)が得られる。
融点 156〜157℃

このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 2
クロロホルム30ml中に三塩化リン1.37g
(10.0mmol)を加え5℃に冷却し、この溶液にア
ンモニアガス0.85g(50.0mmol)を温度20℃以
下で導入する。導入後N−ベンジルオキシカルボ
ニル−L−プロリン2.49g(10.0mmol)を加え、
加熱還流下14時間反応させる。反応後反応液を減
圧濃縮してクロロホルムを留去し、残査に水20ml
を加え溶液とし、1N水酸化ナトリウム水溶液に
てPHを8.0に調整する。次に酢酸エチル30mlで2
回抽出し、酢酸エチル層を飽和食塩水15mlで洗浄
後無水硫酸ナトリウムで乾燥する。溶媒を減圧濃
縮して留去すると油状物が得られる。n−ヘキサ
ン20mlより結晶化させ取、乾燥することにより
N−ベンジルオキシカルボニル−L−プロリンア
ミド2.40g(収率97.6%)が得られる。
融点 87−89℃
〔α〕20 D −32.1゜(C=2.0,エタノール)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 3
テトラヒドロフラン40ml中に三塩化リン1.37g
(10.0mmol)を加え5℃に冷却しこの溶液にアン
モニアガス1.02g(60.0mmol)を温度20℃以下
で導入する。導入後N−(p−メトキシベンジル
オキシカルボニル)−L−アラニン2.51g
(10.0mmol)を加え加熱還流下25時間反応させ
る。溶媒を減圧濃縮して留去後実施例1と同様に
処理することによりN−(p−メトキシベンジル
オキシカルボニル)−L−アラニンアミド2.07g
(収率82.1%)が得られる。
融点 136−137℃
〔α〕20 D +19.5゜(C=1.0,ジメチルホルムア
ミド)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 4
クロロホルム30ml中に三塩化リン1.10g
(7.9mmol)を加え、5℃に冷却し、この溶液に
アンモニアガス0.67g(39.5mmol)を温度20℃
以下で導入する。導入後N−ベンジルオキシカル
ボニル−L−フエニルアラニン2.36g
(7.9mmol)を加え、加熱還流下6時間反応させ
る。反応後反応液を減圧濃縮して溶媒を留去後、

られる。さらに50%メタノール水溶液60mlより再
結晶することによりN−ベンジルオキシカルボニ
ル−L−フエニルアラニンアミド1.35g(収率
87.5%)が得られる。

クロロホルム30ml中に三塩化リン1.10g
(7.9mmol)を加え5℃に冷却し、この溶液にア
ンモニアガス0.80g(47.4mmol)を温度20℃以
下で導入する。導入後、N−ベンジルオキシカル
ボニルスレオニン2.00g(7.9mmol)を加え、加
熱還流下10時間反応させる。反応後反応液を減圧
濃縮して溶媒を留去したのち、実施例1と同様に
処理して水10mlを加え、結晶化させ析出した結晶
を取、乾燥することによりN−ベンジルオキシ
カルボニル−L−スレオニンアミド0.99g(収率
50%)が得られる。
融点 82−84℃

このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 7
クロロホルム200ml中にオキシ塩化リン12.10g
(79.0mmol)を加え5℃に冷却し、この溶液にア
ンモニアガス8.00g(0.47mol)を温度20℃以下
で導入する。導入後、N−ベンジルオキシカルボ
ニル−L−スレオニン20.0g(79.0mmol)を加
え、加熱還流下6時間反応を行う。反応後反応液
を減圧濃縮して溶媒を留去し水100mlを加え、加
熱溶解させる。この溶液を冷却し、析出した結晶
を取、乾燥させることによりN−ベンジルオキ
シカルボニル−L−スレオニンアミド16.90g
(収率85.0%)が得られる。
実施例 8
実施例7のクロロホルムに変えてテトラヒドロ
フランを使用し、以下実施例7と同様に処理する
ことによりN−ベンジルオキシカルボニル−L−
スレオニンアミド16.30g(収率82.0%)が得ら
れる。
実施例 9
実施例7のクロロホルムに変えて酢酸エチルを
反応溶媒として使用し、以下実施例7と同様に処
理することによりN−ベンジルオキシカルボニル
−L−スレオニンアミド16.50g(収率83.0%)
が得られる。
実施例 10
クロロホルム50ml中にオキシ塩化リン3.10g
(20.0mmol)を加え5℃に冷却し、この溶液にア
ンモニアガス2.10g(60.0mmol)を温度20℃以
下で導入する。導入後室温で30分間放置し析出し
ている結晶を取し、洗浄、乾燥すると塩化アン
モニウムを含む反応生成物5.20gが得られる。
得られた反応生成物5.20gとN−ベンジルオキ
シカルボニル−L−スレオニン5.0g
(19.8mmol)をベンゼン50ml中75〜85℃の温度で
3時間反応させる。反応後反応液を減圧濃縮して
溶媒を留去後、水30mlを加え加熱溶解させ溶液と
する。冷却し析出した結晶を取、洗浄、乾燥す
ることによりN−ベンジルオキシカルボニル−L
−スレオニンアミド4.40g(収率88.0%)が得ら
れる。
実施例 11
クロロホルム30ml中に三塩化リン1.37g
(10.0mmol)を溶解させ、冷却下ジエチルアミン
3.66g(50.0mmol)を滴下し室温で30分間反応
させる。この液にN−ベンジルオキシカルボニル
グリシン2.10g(10.0mmol)を加え加熱還流下
7時間反応させる。反応後反応液を減圧濃縮して
溶媒を留去し、以下実施例1と同様に処理するこ
とにより油状のN−ベンジルオキシカルボニルグ
リシンジエチルアミド1.58g(収率60.0%)が得
られる。
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 12
酢酸エチル50ml中にオキシ塩化リン1.53g
(10.0mmol)を溶解させ、冷却下ベンジルアミン
6.43g(60.0mmol)を滴下し室温で30分間反応
させる。この液にN−ベンジルオキシカルボニル
−L−アラニン2.23g(10.0mmol)を加え加熱
還流下4時間反応させる。反応後、水30mlを加え
生成物を溶媒に抽出する。抽出液を無水硫酸ナト
リウムで乾燥後、溶媒を減圧濃縮して留去すると
粗生成物が得られる。酢酸エチルとシクロヘキサ
ンより再結晶するとN−ベンジルオキシカルボニ
ル−L−アラニンベンジルアミド2.90g(収率
92.0%)が得られる。
融点 141−142℃
〔α〕20 D +8.7゜(C=2.0,ジメチルホルムアミ
ド)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 13
酢酸エチル50ml中にオキシ塩化リン1.53g
(10.0mmol)を溶解させ、冷却下ベンジルアミン
6.43g(60.0mmol)を滴下し室温で30分間反応
させる。この液にN−ベンジルオキシカルボニル
グリシン2.10g(10.0mmol)を加え、加熱還流
下6時間反応させる。以下実施例12と同様に処理
することにより粗生成物が得られる。このものを
アセトンと水より再結晶するとN−ベンジルオキ
シカルボニルグリシンベンジルアミド2.30g(収
率88.5%)が得られる。
融点 119−120℃
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 14
テトラヒドロフラン50ml中にオキシ塩化リン
1.53g(10.0mmol)を溶解させ、冷却下イソペ
ンチルアミン5.23g(60.0mmol)を滴下し、室
温で30分間反応させる。この液にN−ベンジルオ
キシカルボニル−L−ロイシン2.65g
(10.0mmol)を加え、加熱還流下6時間反応させ
る。反応後反応液を減圧濃縮し溶媒を留去し、残
査に酢酸エチル50mlと水50mlを加え生成物を酢酸
エチルに抽出する。抽出液を0.5N塩酸溶液30ml
と5%炭酸水素ナトリウム水溶液30mlおよび飽和
食塩水30mlで順次洗浄し、得られた酢酸エチル溶
液を無水硫酸ナトリウムで乾燥させ、溶媒を減圧
濃縮にて留去すると粗生成物が得られる。得られ
た粗生成物をイソプロピルエーテルから再結晶す
るとN−ベンジルオキシカルボニル−L−ロイシ
ンイソペンチルアミド2.80g(収率83.7%)が得
られる。
融点 93−94℃
〔α〕25 D −18.7゜(C=2.0,エタノール)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 15
クロロホルム50ml中にオキシ塩化リン1.53g
(10.0mmol)を溶解させ、冷却下エチルベンジル
アミン8.10g(60.0mmol)を滴下し室温で30分
間反応させる。この液にN−ベンジルオキシカル
ボニル−L−フエニルアラニン2.99g
(10.0mmol)を加え加熱還流下2時間反応させ
る。以下実施例14と同様に処理すると油状の粗生
成物が得られる。これをシリカゲル300gおよび
展開溶媒としてトルエン−酢酸エチルの混合溶媒
を用いるカラムクロマトグラフイーを行い、目的
物を含むフラクシヨンを集め減圧濃縮により溶媒
を留去すると油状のN−ベンジルオキシカルボニ
ル−L−フエニルアラニンエチルベンジルアミド
2.62g(収率63.0%)が得られる。
〔α〕25 D −2.8゜(C=1.0,メタノール)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 16
N−ベンジルオキシカルボニル−L−プロリン
アミド2.00g(8.1mmol)をメタノール30mlに溶
解し、パラジウム黒0.20gを加え室温、常圧で2
時間接触還元を行う。反応後触媒を別し液を
減圧濃縮して溶媒を留去すると油状の目的物が得
られる。これにエーテル20mlを加え結晶化させ、
析出した結晶を取、乾燥するとL−プロリンア
ミド0.90g(収率97.0%)が得られる。
融点 101−102℃
〔α〕25 D −86.6゜(C=1.0,メタノール)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 17
N−(p−メトキシベンジルオキシカルボニル)
−L−アラニンアミド2.00g(7.9mmol)を用い
実施例16と同様に処理すると油状の目的物が得ら
れる。これにイソプロピルエーテル10mlとn−ヘ
キサン20mlを加え結晶化させ、析出した結晶を
取し乾燥するとL−アラニンアミド0.68g(収率
98.0%)が得られる。
融点 71−72℃
〔α〕25 D +6.5゜(C=2.0,水)
このものはNMR,IRにおいて別途合成した標
準品と完全に一致した。
実施例 18
N−tert−ブチルオキシカルボニル−L−バリ
ンアミド1.50g(7.0mmol)を4N塩化水素ジオ
キサン溶液10mlに溶かし室温で1時間撹拌する。
溶媒を減圧濃縮により留去し、乾燥エーテルで洗
浄するとL−バリンアミド塩酸塩1.06g(収率
99.0%)が得られる。
融点 262−263℃
