本発明は、酵素の溶出が少なくかつ高い活性を
有する固定化酵素、特に膜状固定化酵素の製造法
に関する。 酵素は特定の物質と反応するという選択性によ
つて、医薬品、食品工業、分析化学等に利用され
ている。しかし酵素は一般に微生物から複雑な手
段によつて抽出精製されるため高価であり、また
酵素を工業的に利用する場合には、非連続方式に
よらざるを得ないので、その利用が制限される。
これに関し酵素を水不溶性担体上に保持させるい
わゆる固定化のための研究が1950年頃に始まり、
1960年代には工業的実用化が可能となり、現在で
は固定化酵素が広く利用されている。 酵素の固定化の方法は種々開発されており、そ
れらは包括法、吸着法、架橋化法及び共有結合法
に大別される。このうち包括法とは、高分子格子
内の空間に酵素を包括するか、マイクロカプセル
内に酵素を封入する方法である。すなわち酵素分
子は、高分子が形成する三次元網目構造内に、あ
るいは高分子膜が形成する殻内に、物理的に固定
化される。この場合の高分子物質としては、合成
高分子例えばポリアクリルアミド、エチレン―酢
酸ビニル共重合体、ポリビニルアルコール等、天
然高分子例えばコラーゲン、セルロース、アルギ
ン酸及びその塩、寒天、カラギーナン等が用いら
れる。包括法は操作が比較的簡単で、酵素は溶出
し難い利点があるが、後記の共有結合法に比べる
と若干の不利がある。更に包括法において留意す
べき点は、基質と酵素の高分子格子内における拡
散速度が、全体の反応を律速することが多く、特
に高分子量の基質や酵素が関与する場合には適用
が困難である。また重合性単量体と酵素の混合液
を適当な方法で重合する場合には、酵素を失活さ
せないような条件を設定すべきこと、また天然高
分子内に酵素を包括する場合にも、同様に酵素活
性を低減させないように低温で操作する等の配慮
が必要である。 吸着法とは、不溶性担体に酵素を吸着固定化す
る方法である。不溶性担体としてはDEAEセルロ
ース、セルロースエステル、ポリメタクリレート
等の有機高分子、ガラス、活性炭、アルミナ、珪
藻土等の無機物質が挙げられる。この方法は操作
が簡単でかつ担体表面で酵素反応が進行するため
に、前記の包括性と異なり基質や生成物の拡散の
問題がないだけでなく、吸脱着が可逆的であるた
めに、担体の繰り返し使用が可能になるという利
点があるが、他方において酵素が溶出し易いとい
う欠点を有している。 架橋化法とは、低分子量の多官能性物質を酵素
溶液又は酵素と不溶性担体の混合液に加え、酵素
分子間又は酵素と不溶性担体の間に共有結合によ
る架橋構造を形成することにより、酵素を不溶化
する方法である。多官能性物質としてはグルタル
アルデヒドが最も一般的である。また不溶性担体
としては、アルキルアミノ化多孔姓ガラス、ポリ
エチレンイミンを被覆したガラス、ゼラチン、コ
ラーゲン等が挙げられる。この方法は比較的操作
が簡単であるが、多量の酵素を要して効率が低
く、また多官能性物質による酵素の失活がしばし
ば問題になる。 共有結合法とは、不溶性担体が有する活性基と
酵素分子が有する活性基とを、直接あるいは何ら
かの化学物質を介して化学結合させることによ
り、酵素を不溶化するものであり、化学結合様式
によりペプチド結合法、ジアゾ法、アルキル化
法、シツフ塩形成法、Ugi反応法等が挙げられ
る。不溶性担体としてはデキストラン、セルロー
ス、コラーゲン等の天然有機高分子、ナイロン、
ポリスチレン、ポリアクリルアミド、ポリビニル
アルコール等の合成有機高分子、シリカ、チタニ
ア、ジルコニア等の無機物質等が挙げられる。こ
の方法により固定化された酵素は一般に高活性を
示し、元の酵素よりも高い活性を呈することもあ
る。また酵素の溶出が極めて少ないという利点を
有する。しかし固定化に使用する物質による酵素
の活性を低下するおそれがあり、更には操作が複
雑であることが欠点である。 固定化酵素の形態としては粒状、繊維状、膜状
等が挙げられる。このうち膜状固定化酵素は酵素
反応容器内の圧力損失がほとんどないという利点
があり、酵素電極等の酵素反応を利用したセンサ
ーなどに用いられている。膜状固定化酵素は合成
有機高分子膜を基材として吸着法、架橋法、共有
結合法の各種の方法で製造されてきた。しかしこ
れらの方法で製造される固定化酵素は前記の欠点
を有しており、まだ十分な性能を有しているとは
いえない。 本発明者らは、包括法の場合に酵素の活性が高
いという特色に着目し、包括法の欠点を改善すべ
く研究を重ねた結果、酸性多糖類が包括法の基材
として極めて優れていること、また4級アンモニ
ウム塩と反応させると不溶化すること、更には該
反応物が高分子物質の透過を阻止するが低分子物
質は自由に透過しうるという極めて特異な性質を
有することを見出して本発明を完成した。 本発明は、酵素を含有する酸性多糖類溶液を、
塩化ベンザルコニウム、セチルトリメチルアンモ
ニウムブロマイド、セチルピリジニウムクロライ
ド、メチレンブルーから選ばれた1種または2種
以上の4級アンモニウム塩で処理することを特徴
とする固定化酵素の製造法である 本発明に用いられる酵素としては任意のものが
用いられ、例えば酸化酵素例えばカタラーゼ、グ
ルコースオキシダーゼ、ペルオキシダーゼ等、転
移酵素例えばグリシンアミノトランスフエラー
ゼ、ロイシンアミノペプチダーゼ等、加水分解酵
素例えばアスパラギナーゼ、インベルターゼ、ウ
レアーゼ、リパーゼ等、リアーゼ例えばアスパル
ターゼ、グルタミン酸デカルボキシラーゼ、フマ
ール酸ヒドラーゼ等、異性化酵素例えばグルコー
スイソメラーゼ、アラニンラセマーゼ、グルタミ
ン酸ラセマーゼ等、リガーゼ例えばグルタチオン
シンテターゼ、アスパラギンシンテターゼ等が挙
げられる。 酸性多糖類としては、カルボキシル基、硫酸基
等の酸性基を有する水溶性多糖類、例えばアルギ
ン酸ソーダ、カラギーナン、フアーセレラン、寒
天、ペクチン、アラビアガム、更にヘパリン、コ
ンドロイチン硫酸塩等のムコ多糖類、中性多糖類
に酸性基を導入した多糖類変性物等が用いられ
る。中性多糖類としてはセルロース、殿粉、グア
ガム、ローカストビーンガムなどが挙げられる。
酸性基はカルボキシメチル化又は硫酸化反応によ
り導入することができる。 本発明を実施するに際しては、酸性多糖類を常
法により水に溶解する。この際必要に応じて加温
する。固定化しようとする酵素は酸性多糖類と同
時あるいは前後のいずれの時期に添加してもよ
い。ただし酵素が加えられた状態で酸性多糖類を
加温溶解する際には酸素の活性が低下しない温度
範囲で行うことが必要である。また酸性多糖類溶
液に酸素を加える方法の場合には該溶液は加える
酵素の活性が十分に高い温度範囲で十分な流動性
を有することが必要であり、ゲル化能力が大きい
酸性多糖類溶液を用いる場合は、その濃度は低い
方がよい。 次いで酵素を含有する多糖類水溶液を4級アン
モニウム塩で不溶化処理する。本発明において使
用される4級アンモニウム塩としては、塩化ベン
ザルコニウム、セチルトリメチルアンモニウムブ
ロマイド、セチルピリジニウムクロライド、メチ
レンブルーの中から選ばれた4級アンモニウム塩
で、これらを単独または2種以上の混合物として
使用される。4級アンモニウム塩は水溶液でも固
体のままでも使用できる。 本発明においては、上記のような特定の4級ア
ンモニウム塩を使用することにより、ゲル化能力
を有しないアラビアガム等の酸性多糖類の使用も
可能となり、従来の包括法において著しく限定さ
れていた基材を、広い範囲から選択することが可
能となるとともに、低濃度の多糖類溶液の使用も
可能となり、基質の移動の阻害、酸素活性の低下
等の問題点をも解消できるものである。 処理の方法としては両者が接触するのであれば
いかなる方法でもよい。特に膜状固定化酵素を製
造する場合には、酵素を含有する多糖類溶液を溶
膜状としたのち水溶化処理すればよい。液膜状と
するにはガラス板、ステンレス板、合成樹脂板な
ど水不溶性板状物上に流延するなどの方法が用い
られる。不溶化処理は数秒で完了する。 不溶化処理することにより、酸性多糖類と4級
アンモニウム塩との不溶性反応生成物の薄い層及
び酸性多糖類溶液から成る二重構造が形成され
る。このうち外層の不溶性反応生成物は分子量分
画能を有し、内部の酵素の流出が完全に抑制され
ると共に、基質及び反応物の移動が極めて自由に
行われるので、この構造は理想的な固定化酵素の
形態である。 公知の包括法は酵素の活性を維持しうるが酵素
の流出が欠点として挙げられる。これに対し本発
明方法により得られた固定化酵素は、前記の特色
を生かしながら酵素流出を防ぐことができる。 公知の包括法では基材としての多糖類はゲル化
能力が要求されるため、基材の選択範囲が著しく
限定されていたが、本発明方法ではゲル化能力を
有しない酸性多糖類を用いることもできるので、
広い範囲から選択できる。さらに公知の包括法で
は基材がゲル化することを要件としているので、
多糖類溶液の濃度は十分に高いことが必要であ
る。一般に包括法では基質の移動速度が極めて重
要であり、基材としての多糖類の濃度を高くする
ことは、基質の移動を阻害するだけでなく、酵素
の立体構造にも制約が加えられ、酵素活性が低下
することがある。これに対し本発明方法では低濃
度の多糖類溶液を用いることができるので、従来
法の欠点を解消することができる。 本発明方法により得られる固定化酵素は、例え
ばバイオリアクターあるいはセンサーに用いら

1. A method for producing an immobilized enzyme, characterized by treatment with an ammonium salt. 2. Claim 1, characterized in that the acidic polysaccharide solution containing the enzyme is treated as a liquid film.
