発明の背景
発明の分野
本発明は、一般に試料中の過酸化物活性物質の
分析測定、特にこのような測定に有用であつて、
試料中に存在するかもしれないアスコルビン酸か
らの有害な作用に対して抵抗性の組成物、試験手
段、試験具及び試験方法に関する。
背景技術
生物学的試料、例えば尿、糞便懸濁液及び胃腸
内容物中の過酸化物活性物質の存在を検出するた
め、現在、多数の分析方法を利用することができ
る。例えば、潜血試験によつて測定される被分析
物であるヘモグロビン及びその誘導体は、パーオ
キシダーゼ酵素と同様な挙動を示すので、過酸化
物活性物質の代表であり、これら自体はプソイド
パーオキシダーゼとも言われる。過酸化物活性物
質は過酸化物又はヒドロパーオキシドと変色のよ
うな検出可能の応答を生じる指示薬、例えばベン
ジジン、o−トリジン、3,3′,5,5′−テトラ
メチルベンジジン、2,7−ジアミノフルオレン
等との間のレドツクス反応を触媒する点で酵素に
似ている。従つて、試料中の潜血の存在を測定す
るほとんどの方法は、このプソイドパーオキシダ
ーゼ活性に依存する。
着色指示薬の過酸化物酸化の酵素様触媒作用に
依存する、過酸化物活性物質を測定する多数の分
析方法が開発された。第一、これらは湿式化学又
は溶解操作及びいわゆる「浸漬−読み取り
(dipand−read)」型試薬保持ストリツプ具を含
む。前者の代表的例は、ヘンリイ(R.M.Henry)
ら著クリニカル・ケミストリイ・プリンシプル
ス・アンド・テクニークス(Clinical Chemistry
Principles and Techniques)、第二版1124〜
1125(メリーランド州ハーガースタウン:Harper
and Row、1974)に示されている。この操作の
例は氷酢酸(緩衝剤)、ジフエニルアミン(指示
薬)及び過酸化水素を使用するものである。この
ような湿式化学法は分析に有用であることが証明
されているが、これらの方法は多くの欠点を有
し、そのうち2点を例示すると、試薬が安定性に
欠け、感度が不適当であるという欠点がある。
過酸化物活性物質を測定する別の方法で、現在
多くの臨床分析者によつて好ましいと言われてい
る方法は、いわゆる「浸漬−読み取り」試薬スト
リツプ具を利用している。このような「浸漬−読
み取り」具の代表的なものは、マイルス・ラボラ
トリーズ・インコーポレーテツド(Miles
Laboratories,Inc.)のエームス・デイビイジヨ
ン(Ames Division)から商標へマステイツクス
(HEMASTIX)の下に市販されている。この試
験具は、多孔性紙マトリツクスに、有機ヒドロパ
ーオキシドと指示薬との緩衝混合物を含浸させた
ものをプラスチツクストリツプ又はハンドルに固
定して成る。このマトリツクスをヘモグロビン、
ミオグロビン、赤血球又は他の過酸化物活性物
質、即ち、プソイドパーオキシダーゼを含む液体
中に浸漬すると、マトリツクスに青色が現れ、そ
の強度は試料中の物質の濃度に比例する。マトリ
ツクス中に現れた色を標準カラーチヤートと比較
することによつて、分析者は試料中に存在する被
分析物の量を半定量的に測定することができる。
第一に、このような試薬ストリツプの、湿式化
学法に対比した利点は、1)ストリツプ型は使用
しやすく、試薬の製造も、付随する装置も必要と
しないし、2)ストリツプ中で試薬は一層安定に
なり、精度、感度及び経済性が改善される。
過酸化物活性種に関する特定の分析を前記の方
法のどちらで行つても、両方に固有の問題が存在
する。即ち、試料中に一般に還元剤及び特にアス
コルビン酸又はアスコルビン酸イオンの存在によ
つて妨害が起こる(以下、アスコルベート妨害と
言う)。例えば、尿分析の場合、高投与量のビタ
ミンC(アスコルビン酸)を含む最近の食餌人気
は、一定の尿成分、例えば潜血を分析する際に重
大なアスコルベート妨害の問題を起こす。
早くも1938年に、アスコルビン酸塩のような還
元剤の有害な作用が認識された。コーン(R.
Kohn)及びワトラス(R.M.Watrous)著、ジヤ
ーナル・オブ・バイオロジカル・ケミストリイ
(Journal of Biological Chemistry)、124:163
〜168(1938)。潜血(プソイドパーオキシダーゼ)
分析を実施する場合に、同時にアスコルベート分
析も実施して潜血測定の精度を評価すべきである
という1979年の提案によつて証明されるように、
診断分析のこの分野で同じ問題になお悩まされて
いる。ニールセン(L.Nielsen)、ヨルゲンセン
(P.J.Jorgensen)及びハンセン(A.C.Hansen)
著、Ugeskrift for Laeger、141、791〜793
(1979)。
試験系、例えばグルコース感受性試薬を含む系
に対するアスコルベート妨害を除去する試みは多
数、文献に報告されている。グルコース感受性分
析に関して、アスコルベートが試薬に到達する前
にアスコルベートを濾去する方法から、その場で
アスコルベートを分解する酵素を利用する方法ま
で種々のアプローチがある。
Dahlqvistのカナダ特許第844564号明細書は、
尿又は他の媒体中でグルコースを測定する測定具
を開示しており、このものは通常のグルコース応
答性試薬で含浸した多孔性部分の他に、尿試料を
受理する付加的部分を含む。試料受理部分は、測
定具におけるただ一つの機能が尿試料中に存在す
るアスコルベートを吸着することであるイオン交
換物資を含む。
ダニンガー(Danninger)らに付与された米国
特許第4,168,205号明細書は、試験試薬の処方
に酵素であるアスコルベートオキシダーゼを配合
することを示唆している。この場合、試料中に存
在するアスコルベートは、アスコルベートオキシ
ダーゼによつて酵素酸化されて、所望の分析に悪
影響を及ぼさない化合物であるデヒドロアスコル
ベートを生じる。
アスコルベート妨害を排除する別のアプローチ
は、富士臓器製薬株式会社の特開昭58−55757号
公報に記載されている。該公報には、種々のリガ
ンド、例えばエチレンジアミン四酢酸及びジエチ
レントリアミン五酢酸の金属キレートを使用して
試料を前処理し、次にコレステロール、グルコー
ス又は他の成分、例えば尿酸を分析することが開
示されている。
クー(ku)の米国特許第3,411,887号明細
書には、アスコルベート「トラツピング系」を使
用することによつて、酵素酸化性物資、例えばグ
ルコースオキシダーゼに依存する試薬系を用いて
アスコルベート妨害を排除することが記載されて
いる。「トラツピング系」は、イオン化されたと
きに、レドツクス指示薬色素とアスコルベートと
の間で酸化−還元電位を降下するイオン化可能の
重金属化合物に関する。適当な金属は、例えばコ
バルト、鉄、水銀及びニツケルである。
マガース(Magers)らに付与され、正式に譲
渡されている米国特許第4,288,541号明細書に
は、グルコース/グルコースオキシダーゼ分析系
にアスコルベート抵抗性を付与するため水銀イオ
ン錯体、例えばサルコシン酸水銀を使用すること
を記載している。
前記文献の他に、グルコース試験に伴うアスコ
ルベートの問題に関する注意は、下記の文献に記
載されている:

前記のクーの特許のアプローチと同様に、他の
文献はコバルトを使用してアスコルベートを錯体
とし、酸化している。例えば、ブラガグノロ
(G.Bragagnolo)著、Ann.Chim.Applicata 31、
350〜368(1941)は、アスコルビン酸の溶液を金
属コバルトの存在で空気で酸化することを報告し
た。また、ジヤーナル・オブ・ザ・ケミカル・ソ
サイエテイ・オブ・ジヤパン(Journal of the
Chemical Society of Japan)、63、820〜826
(1942)に岩崎トモキチによつてCo(NH3)6CL3に
関する同様の活性が報告された。
前記の文献はグルコース測定用の分析系を広範
に扱つているが、パーオキシダーゼ及びプソイド
パーオキシダーゼのような過酸化物活性物質、例
えば潜血(ヘモグロビン)の測定に関連するアス
コルベート妨害の問題を解決するための示唆は全
くなされていない。前記のクーの特許に開示され
ているにもかかわらず、前記文献は、Co()の
ような金属イオンが実際にプソイドパーオキシダ
ーゼでもあることを示している。例えば、酢酸コ
バルト()は、クメンヒドロパーオキシドを触
媒により分解するために工業的に使用される。
〔ザ・メルク・インデツクス(The Merck
Index)、第9版311(1976)。〕ロー(K.Lohs.)、
Monatsber.Deut.Akad.Wiss.Berlin、8、657〜
659(1966)(Chem.Abstracts、67、120383z、
1967参照)によつて過酸化物を触媒により分解す
る一連のCo()錯体が報告されている。従つ
て、過酸化物活性物質を測定する典型的分析用調
整物、即ち有機ヒドロパーオキシド及び指示薬を
含む分析用調整物にこのような金属錯体を使用す
ると、ヒドロパーオキシドとの有害な反応を起こ
して、「偽陽性」の結果を生じるか、又は問題の
過酸化物活性物質、例えば潜血に対して反応しな
いようにし、従つてこのような測定に使用できな
くなると、当業者が考えるであろうことは明らか
である。実際、潜血試験に水銀錯体、例えばサル
コシン酸水銀を使用する努力は失敗した。
正式に譲渡されているブルクハルト
(Burkhardt)らの米国特許第4,310,626号明
細書は、前記問題に関して、過酸化物活性物質の
測定用組成物とのアスコルベート妨害を軽減する
ためアンモニウムCo()錯体を使用することを
記載している。この特許は有機ヒドロパーオキシ
ド及び適当な指示薬、例えば3,3′,5,5′−テ
トラメチルベンジジンをアンモニウムCo()錯
体、例えば特にCo(NH3)6CL3と一緒に含む組成
物を開示している。しかしながら、これらの錯体
は潜血試験に産業的に有利である程、充分なアス
コルベート抵抗性を与えなかつた。
過酸化物活性物質の分析測定におけるアスコル
ベート妨害に関する他のアプローチとして、例え
ば西ドイツ特許第2907628号明細書がある。この
ドイツ特許は、溶液中での尿分析に関し、尿試料
を1種以上の酸化剤で前処理してアスコルベート
を除去し、次に適当な分析試薬と接触させる。開
示されている酸化剤は、沃素酸ナトリウム、過沃
素酸ナトリウム、次亜塩素酸カルシウム、三沃化
カリウム、次亜塩素酸ナトリウム、クロロアミン
及びブロモサクシンイミドである。
要するに、過酸化物活性物質の測定においてア
スコルビン酸が起こす妨害問題を排除するための
種々のアプローチは、種々のCo()アンモニウ
ム錯体の使用、酸化剤による試料の前処理及び試
薬組成物へのアルカリ金属沃素酸塩の直接添加の
ような技術を含んでいた。
西洋わさび又はポテトから得られるパーオキシ
ダーゼのような天然パーオキシダーゼの作用のメ
カニズムを解明するため、別のパーオキシダーゼ
系としてプソイドパーオキシダーゼ、例えばヘモ
グロビンがしばしば研究される。アスコルビン酸
はパーオキシダーゼに対する代表的基質として長
い間知られており、プソイドパーオキシダーゼと
して作用する金属キレートの存在でのアスコルビ
ン酸酸化は公知の現象である。1967年及び1968年
に、カーン(M.Khan)及びマーテル(A.
Martell)は、1.8〜3.45のPH範囲にわたつて数種
の鉄キレート及び銅キレートの存在でのアスコル
ビン酸の酸化の動的研究について報告した〔カー
ン及びマーテル著、J.Am.Chem.Soc.、89、4176
(1967);J.Am.Chem.Soc.、89、7104(1967);J.
Am.Chem.Soc.、90、3386(1968)。〕これらの研
究においては、多数の動的及び熱力学的パラメー
タが研究された。その結果は、アスコルビン酸を
酸化する能力による種々のキレートの順位であつ
た。これらの著者の研究した4種のアミノポリカ
ルボン酸のうち、Fe+3のN−(2−ヒドロキシエ
チル)エチレンジアミン三酢酸(HEDTA)キレ
ートが、最も迅速な酸化剤であることが判明し
た。マーテルの研究及びグリンステツド
(Grinstead)による更に早期の研究は、「モデ
ル」パーオキシダーゼ系を構成するためこのアス
コルビン酸酸化活性を考えている〔グリンステツ
ド、J.Am.Chem.Soc.、82、3464(1960)〕。グリ
ンステツドによる前記のような研究から、構造が
酵素パーオキシダーゼの活性部位に見られる鉄含
有ヘムの構造と類似している鉄キレートを用いて
パーオキシダーゼメカニズムを研究する試みがな
された。事実、その著者はその論文に反復して
「モデルパーオキシダーゼ系」という文言を使用
している。しかしながら、このキレート及び反応
性において類似の他の物質によつて「モデル」パ
ーオキシダーゼ活性が示されるので、このような
物質を有機ヒドロパーオキシド/指示薬系、例え
ばパーオキシダーゼ又は他の過酸化物活性物質の
測定用の分析試薬組成物及び測定具に現在使用さ
れている代表的系に組み込みうることは、確かに
予期されないであろう。更に、パーオキシダーゼ
活性に関連して本発明の譲り受け人によつて行わ
れた研究から、指示薬、例えば3,3′,5,5′−
テトラメチルベンジジン(TMB)又はo−トリ
ジン(過酸化物活性物質の存在を検出する分析系
に典型的に使用される指示薬)を用いてこのよう
な活性はアスコルビン酸とより約200倍早く反応
を起こすことを予期しうることが判明した。従つ
て、このような過酸化物活性金属キレートである
「モデル」パーオキシダーゼがアスコルビン酸を
容易に酸化する場合、(カーン、マーテル及びグ
リンステツドによりなされた仮定)、TMBのよ
うな指示薬とのパーオキシダーゼ反応が、200倍
程早くないとしても(西洋わさびで行われた後者
の研究で示唆された)、少なくともアスコルビン
酸と同じ速度で進行する。明らかに、極めて反応
性の被分析物をその被分析物の存在で変色するよ
うに構成された真の試薬調製物中に配合すると、
「偽陽性」の結果が得られるであろうと予測され
る。
発明の概要
前記の教示及び示唆にもかかわらず、ある種の
過酸化物活性金属キレート及び特にポリカルボキ
シアルキルアミンの金属キレートを本発明方法に
使用する場合には、過酸化物活性物質を測定する
試薬系を含む組成物において予期された「偽陽
性」結果を生じないばかりでなく、実際にこのよ
うな系において被分析物を測定すべき系の信頼
性、安定性及び感度の意味で意外に有利である。
更に、本発明により金属キレートを使用すると、
特に試料中に存在するアスコルビン酸イオンから
の妨害によつて起こされる不正確さを克服した点
で特に有利であることが判明した。
従つて、本発明はこの発見に基づくもので、前
記のように、一般に、アスコルベート妨害に対し
て抵抗性である過酸化物活性物質の分析測定法及
び特に本発明により有機ヒドロパーオキシド及び
レドツクス指示薬、例えばo−トリジン又は3,
3′,5,5′−テトラメチルベンジジン並びに過酸
化物活性金属キレートを含む組成物を利用する検
出法に関する。このような検出において、過酸化
物活性被分析物は、酵素パーオキシダーゼに類似
しているので、指示薬と有機ヒドロパーオキシド
との反応を触媒するか、又はその反応に関与す
る。この反応は色又は他の検出可能の応答を生
じ、その強度は被分析物の濃度の指標である。ア
スコルビン酸イオンは、存在する場合、重大な妨
害の問題を起こす。組成物中に過酸化物活性金属
キレートが存在しても、過酸化物活性被分析物の
分析測定を妨害することも予測されるであろう。
それにもかかわらず、試料中の過酸化物活性物質
の存在を検出するため、試料中のアスコルビン酸
の妨害作用に対して抵抗性の新規組成物、試験手
段(及び試験具)を有効に調整できることが判明
した。この組成物、試験手段(及び試験具)は、
「モデルパーオキシダーゼ」としても公知のポリ
カルボキシアルキルアミン誘導体の金属キレート
を含む。
従つて、本発明の組成物は、有機ヒドロパーオ
キシド、過酸化物活性物質及び過酸化物の存在で
検出可能の応答、例えば変色を生じうる指示薬並
びに、更に一般式:
〔式中R1は水素又は2〜3個の炭素原子を有
する直鎖若しくは分枝鎖のアルキルアルコール或
いはアルキルカルボン酸基を表し、R2,R3,Rx
及びRyは同一又は異なり、2〜3個の炭素原子
を有する直鎖若しくは分枝鎖のアルキルアルコー
ル或いはアルキルカルボン酸基を表し、R1、R2,
R3,Rx又はRyのうち少なくとも2個は前記のア
ルキルカルボン酸基であり、Rp及びRqは同一又
は異なり、1〜3個の炭素原子を有する直鎖又は
分枝鎖アルキレン基又は6〜9個の炭素原子を有
する2価の1,2−脂環式基を表し、nは0〜1
の数値を有する整数、mは0〜2の数値を有する
整数であり、MはFe+3である〕を有するポリカ
ルボキシアルキルアミン誘導体である金属キレー
トを含む。
好ましい化合物は、mが0であり、nが0又は
1であり、Rpがエチレン基である化合物である。
アルキルカルボン酸基が−CH2COOHであるポ
リカルボキシアルキルアミン誘導体の金属キレー
トは特に好ましい。
好ましい金属キレートはN−(2−ヒドロキシ
エチル)エチレンジアミン三酢酸の鉄キレート
(Fe−HEDTA)である。
本発明の好ましい実施態様において、組成物を
支持体マトリツクス、吸水性紙に混入して試験手
段を形成し、この試験手段を不活性支持体に固定
して試験具を形成することができる。更に、試験
手段(及び試験具)の製造方法及び試験方法も本
発明によつて提供される。
本発明により金属キレートを包含させることに
より、アスコルベート妨害に対する優れた抵抗性
ばかりでなく、良好な貯蔵性及び高温安定性及び
「偽陽性」結果の欠如を実験により確認できるよ
うに、意外に有利な安定性を有する組成物、試験
手段(及び試験具)が提供される。
発明の詳細な説明
本発明の開発中の初期の湿式化学実験は、N−
(2−ヒドロキシエチル)エチレンジアミン三酢
酸の鉄キレート(以下、Fe−HEDTAと記すが、
これは便宜のためだけに使用するもので、金属イ
オンとポリカルボキシアルキルアミン誘導体との
間に共有結合の存在を意味するものではない)、
アスコルビン酸及び緩衝剤を使用し、このキレー
トの存在でのアスコルビン酸酸化の迅速性を確認
したもので、その結果はカーン及びマーテルの前
記報告から予期されるであろう。従つて、パーオ
キシダーゼ含有組成物において、このような組成
物によるアスコルビン酸酸化と比較することによ
つて、TMBがいかに早く酸化されるかが分かつ
たので、有機ヒドロパーオキシド、酸化可能の指
示薬、例えばTMB及び更に前記のような金属キ
レートを含む組成物は極めて不安定であり、急速
に「偽陽性」結果を生じることが予想された。
しかし、更に実験したところ、このような金属
キレートを含み、過酸化物活性物質の検出に適当
であり、更に乾燥固体状態の型に適合し、製造中
及び貯蔵中に良好な試薬安定性を示し、既知のヘ
モグロビン−陰性尿と接触したときに「偽陽性」
の結果を生じない組成物を調整しうることが判明
した。こうして達成された本発明は、前記のよう
に、金属キレート、例えばFe−HEDTAは「モ
デルパーオキシダーゼ」として使用でき、従つて
過酸化物活性被分析物の濃度を測定するため組成
物に使用するには不適当であろうと示唆している
前記文献の示唆と矛盾する。
尿のような生物学的液体中の潜血(OB)、即
ちヘモグロビンを検出するための、本発明の新規
組成物から製造することができた試験手段(及び
試験具)は、尿中の異常に高いアスコルビン酸濃
度に対して抵抗性であることが判明した。前記の
ように、アスコルビン酸による阻止は、特に尿試
料の約25%が10mg/dLより高いアスコルビン酸
濃度を示すことを予測できることを考慮すると、
重大な問題である。ある型のアスコルベート妨害
遅延剤を含まない常用のOB検出具は、通常、5
mg/dL程度の低いアスコルビン酸濃度によつて
阻止される(即ちヘモグロビンの存在に対してあ
まり鋭敏でなくされる)ことが判明した。しか
し、本発明により製造される試験具は、このよう
な従来の試験具に比して、アスコルベート妨害抵
抗性の点で極めて有利であり、アスコルビン酸を
比較的高い濃度で、例えば50mg/dL程度で含む
液体中の過酸化物活性物質を検出することができ
る。
従つて、本発明は、尿のような生物学的液体中
の過酸化物活性物質の検出用の組成物、試験手段
(及び試験具)を提供する。本発明の組成物、試
験手段(及び試験具)によつて、ヘモグロビンの
他に、例えば、パーオキシダーゼ、ミオグロビ
ン、赤血球及び他のプソイドパーオキシダーゼを
含めて他の過酸化物活性物質を検出することがで
きる。本発明は、ポリカルボキシアルキルアミン
の誘導体であつて、生物学的液体について行われ
る分析に対するアスコルビン酸の有害な作用を減
少又は排除する目的で「モデルパーオキシダー
ゼ」と認識される金属キレートを使用する。この
観点で、金属キレートはこのような液体中に存在
しうるアスコルビン酸イオンの酸化を促進する作
用をする。
本発明の組成物、試験手段(及び試験具)は、
アスコルベート妨害に対して実質的に感受性でな
く、0.03mg/dL程度或いはそれ以下の痕跡のヘモ
グロビンの存在に対して肉眼で又は機器で検出し
うる応答、例えば色の応答を生じることが判明し
た。
本発明の組成物に使用する有機ヒドロパーオキ
シドは多数の周知有機ヒドロパーオキシドから選
択することができる。しかしながら、検出可能の
応答、例えば色の変化又は試験組成物によつて吸
収若しくは反射される光の量の変化を生じる指示
薬の存在で過酸化物活性物質と反応できなければ
ならない。ヒドロパーオキシドのうち特に適当な
ものはクメンヒドロパーオキシド、t−ブチルヒ
ドロパーオキシド、ジイソプロピルベンゼンヒド
ロパーオキシド、2,5−ジメチルヘキサン−
2,5−ジヒドロパーオキシド、パラメンタンヒ
ドロパーオキシド及び使用する指示薬を酸化する
のに適当な他の周知のヒドロパーオキシド、又は
これらの化合物の混合物である。前記化合物のう
ち、クメンヒドロパーオキシドが最も好ましい。
反応して、有機ヒドロパーオキシド及び過酸化
物活性物質の存在で検出可能の応答を生じること
ができる限り、本発明の組成物に使用するため多
くの指示薬が適当である。指示薬は、例えば、い
わゆる「ベンジジン−型」化合物;ベンジジン;
o−トリジン;3,3′,5,5′−テトラ(低級ア
ルキル)ベンジジン;2,7−ジアミノフルオレ
ン;及びこれらの混合物又は他の種々の指示薬の
混合物である。本明細書に使用する用語「低級ア
ルキル」は、メチル基、エチル基、n−プロピル
基及びイソプロピル基、及び種々のブチル異性
体、ペンチル異性体及びヘキシル異性体を含めて
炭素原子数1〜6個のアルキル基である。指示薬
である3,3′,5,5′−テトラメチルベンジジン
(TMB)が特に好ましい。
使用するため選択された特定の金属キレートの
適格性は、アスコルビン酸の酸化を促進する能力
によつてばかりでなく、組成物の他の成分との相
溶性によつても決定される。従つて、このような
適当なキレートは、下記の一般式で表されるポリ
カルボキシアルキルアミン誘導体の金属キレート
を包含することが分かつた:
従つて、本発明に使用するため適当であること
の分かつた金属キレートは、例えばN−(2−ヒ
ドロキシエチル)エチレンジアミン三酢酸鉄キレ
ート(Fe−HEDTA)、エチレンジアミン四酢酸
鉄キレート(Fe−EDTA)、シクロヘキシレンジ
アミン四酢酸鉄キレート(Fe−CDTA)、ニトリ
ロ三酢酸鉄キレート(Fe−NTA)、イミノ二酢
酸鉄キレート(Fe−IMDA)、エチレンジアミン
二酢酸二プロピオン酸鉄キレート(Fe−EDDP、
α型及びβ型)、及びヒドロキシエチルイミノ二
酢酸鉄キレート(Fe−HIMDA)並びにこれら
の混合物である。鉄キレートの形が一般に好まし
く、最も好ましいのは化合物はFe−HEDTA及
びFe−EDTAであり、最も望ましいのはFe−
HEDTAである。しかしながら、本明細書を読ん
だ当業者には明らかなとおり、特に上に掲げたキ
レートの他に多数の適当なキレートが本発明の範
囲内にある。
従つて、Fe−HEDTA及びFe−EDTA、特に、
Fe−HEDTAは本発明の組成物、試験手段(及
び試験具)において優れた結果を生じ、潜在出血
試験において最も満足にアスコルビン酸妨害抵抗
性を生じ(ヘモグロビンを検出しうる)、使用前
の良好な試薬安定性を示し、「偽陽性」の結果を
生じないことが実験により分かつた。更に、記載
した他の金属キレート並びに多数の他のキレート
も満足に作用するであろう。しかし、このような
他の化合物を使用する場合に過酸化物活性物質を
検出できる速度に著しい変動があることを予想す
ることができる。それというのは、アスコルビン
酸の酸化速度が変動するからである。従つて、本
発明に使用するため適当な金属キレートは、前記
の化合物類内のポリカルボキシアルキルアミン誘
導体であるものから選択することができ、このよ
うな化合物はすべて、アスコルビン酸塩を満足に
酸化することができ、本発明の範囲内に置くこと
ができるものと理解すべきである。しかしなが
ら、この観点で多数のものが緩徐に作用し、従つ
て産業的用途ではあまり実用的でなく、好ましく
ない。
本発明に使用するのに適当な金属キレートは、
アルドリツチ・ケミカル社(Aldrich Chemical
Co.)、シグマ・ケミカル・カンパニイ(Sigma
Chemical Company)又は同様の供給者から市
販されているポリカルボキシアルキルアミン誘導
体を使用して常用の実験操作によつて製造するこ
とができる。例えば、金属キレートであるFe−
HEDTAは、市販のHEDTA及びFeCl3・6H2O
の等モル量を水溶液中で混合して鉄:キレートの
1:1(モル:モル)Fe−HEDTA溶液を製造す
ることによつて製造することができる。金属:キ
レートの他の溶解濃度比は、単に、混合する溶液
のそれぞれの濃度を変動することによつて容易に
調整することができる。アスコルビン酸妨害を克
服する点で最も良好な結果は、キレートにおける
金属イオン:ポリカルボキシアルキルアミン誘導
体の濃度が約1:1(モル:モル)の関係にある
場合に得られることが判明した。
本発明の種々の実施態様において所定の金属キ
レートの好ましい濃度範囲は広範に変動する。例
えば、Fe−HEDTAの場合に、有機ヒドロパー
オキシド及びテトラ(低級アルキル)ベンジジン
指示薬を含む組成物に使用する場合、好ましい濃
度範囲は約0.5ミリモル(mM)〜約50mMであ
る。この範囲は、尿試料中約50mg/dLのアスコ
ルベート濃度まで抵抗するのに最適であることが
測定された。更に、実験では、適当な鉄キレート
を比較的低濃度で使用すると、比較的迅速にヘモ
グロビンを検出することができるが、同じキレー
ト又は他のキレートを高い濃度で使用してもあま
り有効でない。これらの明らかに変則的な結果に
ついては以下に詳述するが、これらの結果は、本
発明の組成物におけるキレート濃度と機能又は適
格性との間に一般的相関関係はないことを証明す
る。
本発明の組成物において満足に作用する適当な
金属キレートの大部分は、構造的にアルキルアミ
ン基又はアミン中心基及びカルボン酸基−CH2
COOHを有する。しかしながら、このような特
徴を有しないが、前記の一般的化合物範囲に属す
る他のキレートは、一般に、アスコルベート妨害
を克服するのに有効であり、満足な感度及び安定
性を示し、従つて申し分なく使用される。
使用する際に、本発明の一般的概念に基づく組
成物、試験手段(及び試験具)の特定の実施態様
の性能は多数の異なるフアクターに左右される。
多量のアスコルビン酸(ビタミンC)を摂取する
習慣のある人からの代表的尿試料は、しばしばア
スコルベートを25〜100mg/dL又はそれ以上含
む。研究のための参照アスコルベート濃度を約50
mg/dLに選択した。本発明のアスコルベート抵
抗性組成物、試験手段(及び試験具)及び試験方
法の好ましい実施態様は、このような試料中の過
酸化物活性物質を約50mg/dLの参照濃度でばか
りでなく、他の種々のアスコルベート濃度で検出
することができることが判明した。ほとんどの場
合、アスコルベート濃度がキレート濃度よりはる
かに高いと、応答時間の遅延が起こる。下記のよ
うに、「ラグタイム」、即ち、観察可能の応答が起
こるまでの時間は、50mg/dLのアスコルベート
の存在で尿中のヘモグロビンを検出する能力につ
いて試験した、キレート濃度の変動する本発明の
好ましい実施態様に関して、実験により1/3分よ
り短時間から約1/2時間に及ぶことが判明した。
好ましい実施態様においては、本発明の組成物
を使用して、過酸化物活性物質の測定用の試験手
段(及び試験具)を製造する。このような好まし
い実施態様では、組成物を適当な支持体マトリツ
クスに混入させて試験手段を形成することができ
る。支持体マトリツクスは多くの形を取ることが
でき、例えば米国特許第3846247号明細書に開示
されているもの(フエルト、多孔性セラミツクス
トリツプ及び織られた又はマツト状に編まれたガ
ラス繊維)であつてよい。米国特許第3552928号
明細書に記載されているマトリツクス(木材ステ
イツク、布、スポンジ材料及び粘土質物質)も適
当である。英国特許第1369139号明細書には、支
持体マトリツクスとして合成樹脂フリース及びガ
ラス繊維フエルトを使用することが示唆されてい
る。また、英国特許第1349623号明細書は、下層
紙マトリツクスに対する被覆として細いフイラメ
ントの光透過性網の使用を提案している。フラン
ス特許第2170623号明細書には、ポリアミド繊維
が開示されている。このような開示があるにもか
かわらず、支持体マトリツクスとして従来使用さ
れ、本発明に使用するのに特に好ましく、適当で
ある材料は濾紙等のような吸水性材料である。し
かしながら、支持体マトリツクスは前記のような
種々の物理的形及び他の形であつてよく、このよ
うな形がすべて本発明に使用するのに適当であ
り、使用するものである。
本発明の試験手段を製造する際には、組成物の
成分を種々の方法で支持体マトリツクスに混入す
ることができる。例えば、成分を水又は他の適当
な溶剤、好ましくは有機溶剤、例えばエタノー
ル、アセトン若しくはジメチルホルムアミド
(DMF)並びにこれらの溶剤の混合物及び他の溶
剤の混合物中に溶解又は懸濁することができる。
次に、この溶液又は懸濁液を使用して吸水性濾紙
を含浸させることができ、例えば、試薬を適当な
マトリツクス上に印刷するインクのようにして含
浸することができる。また、支持体マトリツクス
を組成物中に浸漬するか、又は例えばドクターブ
レードを用いて組成物で被覆することができる。
組成物の成分を支持体マトリツクスに混入する
現在好ましい方法は、吸水性濾紙を成分の2種以
上の溶液又は懸濁液で含浸することである。含浸
は、濾紙片をこのような溶液又は懸濁液中に2回
以上浸漬し、各浸漬後に浸漬した紙を乾燥器中で
乾燥することによつて達成される。こうして形成
した試験手段を両面接着テープ片の片側に積層
し、積層物をストリツプに切断し、各ストリツプ
をプラスチツク裏張り材料(例えばポリスチレ
ン)の長いシートに固定し、これを次に短い寸法
に平行に切断して一方の端部に含浸された紙を有
し、他端がハンドルとして役立つ長方形試験具を
形成する。こうして形成した試験具は、2回乾
燥、含浸した試験手段片を便利なハンドルを構成
する長いプラスチツク支持体の平坦な片面に一方
の端部で固定して成る。
本発明の試験手段を作る好ましい方法は、例え
ばFe−HEDTAを有機ヒドロパーオキシドと一
緒に、指示薬の添加前に、水性の第1回の浸漬で
濾紙中に導入することである。即ち、まず、濾紙
を1種以上の適当な溶剤及び/又は緩衝剤、例え
ばトリエタノールアミンボレート及びトリス(ヒ
ドロキシメチル)アミノメタン−マロネート(以
下トリス−マロネートと記す)と一緒にFe−
HEDTA及びヒドロパーオキシドの水溶液を含浸
させ、乾燥し、適当な溶剤、例えばエタノール中
の指示薬の第二浸漬溶液で再び含浸し、第2回の
乾燥を行う。このような、紙中にまず金属キレー
トを含浸させ、その後に他の活性試薬を含浸させ
る「2回浸漬」法は、優れたアスコルベート抵抗
性及び貯蔵安定性を示す試験具を生じることが判
明した。
本発明の試験手段を調整する特に好ましい方法
は、金属キレート及び指示薬以外の試薬を濾紙中
に、前記のような試薬の第一溶液中に浸漬し、そ
の後紙を乾燥することによつて導入し、その後、
乾燥した紙を適当な溶剤中の指示薬及び増粘剤、
例えばポリビニルピロリドンの溶液中に浸漬する
ことによつて指示薬を添加し、次いで2回目の乾
燥を行うことである。
前記の試験組成物試薬及び他の成分の他に、他
の成分、例えば種々の増粘剤、湿潤剤、緩衝剤、
乳化剤及び周知の助剤を本発明の組成物、試験手
段(及び試験具)に添加することもできる。例え
ば、増粘剤として、ポリビニルピロリドンの他に
又はその代わりに、種々の材料、例えばゼラチ
ン、アルギン、カラゲニン、カゼイン、アルブミ
ン、メチルセルロース等を使用することができ
る。湿潤剤としては、ドデシル硫酸ナトリウムを
使用するのが好ましいが、長鎖有機硫酸塩及びス
ルホン酸塩、例えばジオクチルスルホコハク酸ナ
トリウム又はドデシルベンゼンスルホン酸ナトリ
ウムを使用することもできる。緩衝剤系には、ト
リエタノールアミンボレート及びトリス−マロネ
ートの他に、酒石酸塩、燐酸塩、フタル酸塩、ク
エン酸塩、酢酸塩、コハク酸塩又は他の緩衝剤を
使用することができる。組成物を約6.0〜7.0のPH
値に緩衝するのが好ましい。乳化剤としては、ポ
リビニルアルコール、アラビアゴム、カルボキシ
ビニルポリマー等を使用することができる。指示
薬を懸濁するため有用な有機溶剤はほとんど非反
応性有機揮発性溶剤、例えばエタノール、アセト
ン、DMF、クロロホルム、エチレンジクロリド、
ベンゼン、酢酸エチル等を包含する。勿論、他の
適当な溶剤を選択することは、当業者には可能な
ことである。
試験手段(又は試験具)を使用する際には、こ
れを試験すべき液体又は試験すべき物資の液体懸
濁中に浸漬し、直ちに取り出すか、又は液体、固
体或いは半固体の試料を試験手段(又は試験具)
に施すことができる。試料中に過酸化物活性物質
が存在すると、試験組成物は色の変化又は他の検
出可能の応答を生じる。応答が色である場合に
は、これを試料中に含まれる過酸化物活性物質の
定量的量を評価するための予め検定された色標準
と比較することができる。完全な過酸化物活性物
質、例えば完全な赤血球は、その他は未着色のマ
トリツクス上に着色した斑点として現れる。溶血
された過酸化物活性物質は均一にマトリツクスを
着色することができる。現れた色又は他の応答の
量を測定するため、肉眼による比較の他、種々の
機器測定法を使用することができ、人間の目の主
観的測定を省くことによつて試験の精度を高める
ことができる。
下記の実施例は、単に本発明の概念及び利点を
説明するためのものであり、本発明の範囲を限定
するものではない。このような限定は特許請求の
範囲のみによつてなされる。
実施例
A 試験組成物
例 ……Fe−HEDTA
試料中のパーオキシダーゼ又は別の過酸化物活
性物質及び特にヘモグロビンの存在を測定しうる
本発明の組成物を製造する実験を行つた。組成物
は、アスコルベート妨害遅延剤としてN−(2−
ヒドロキシエチル)エチレンジアミン三酢酸の
1:1(モル:モル、M:Mと記す)鉄キレート
(Fe−HEDTA)を含むものである。
HEDTA0.278gを蒸留水100mlに溶解して、
10mMHEDTA溶液を作り、10mMHEDTA溶液
にFeCl3・6H2O0.270gを溶解することによつて
Fe−HEDTAキレートを製造した。5mMの濃度
のアスコルビン酸を組成物に、溶液の最終溶量中
で50マイクロモル濃度を生じるのに充分な量で添
加した。組成物の成分及びアスコルビン酸を下記
の表に示した順序及び量で混合した。最終組成物
溶液は、存在する他の成分の濃度と同様に、固体
試験手段に混入するため同様の組成物に使用する
より著しく低い濃度である100マイクロモル
(μM)濃度のFe−HEDTAを含んでいた。

こうして製造した本発明の組成物は、溶液中に
1デシリツトル当たり0.139mgの最終濃度を生じ
るように含水血液を添加したとき、青色を形成す
るのが観察され、これは、試料のアスコルベート
濃度が50μMであるにもかかわらず、組成物が存
在するヘモグロビンを検出することができること
を示す。
例 ……Fe−EDTA
Fe−HEDTAではなく、エチレンジアミン四
酢酸の鉄キレート(Fe−EDTA)の10mM溶液
を使用することを除いて、例の実験を繰り返し
た。EDTA0.292gを蒸留水100mlに溶解し、例
に記載したようにFeCl3・6H2Oを添加すること
によつて例と実質的に同様にしてFe−EDTA
を製造した。こうして製造した組成物は例と同
様に、ヘモグロビン0.139mg/dL及びアスコルベ
ート50μMの存在で青色を形成した。
例 ……Fe−CDTA
Fe−HEDTAではなく、シクロヘキシレンジ
アミン四酢酸の鉄キレート(Fe−CDTA)の
10mM溶液を使用することを除いて例1の実験を
繰り返した。CDTA0.346gを蒸留水100mlに溶解
し、例に記載したようにFeCl3・6H2Oを添加
することによつてFe−CDTA溶液を製造した。
こうして製造した組成物は例と同様に、ヘモグ
ロビン0.139mg/dL及びアスコルベート50μMの
存在で青色を形成した。
例 ……Fe−IMDA
Fe−HEDTAではなく、イミノ二酢酸の鉄キ
レート(Fe−IMDA)の10mM溶液を使用する
ことを除いて、例の実験を繰り返した。
IMDA0.133gを蒸留水100mlに溶解し、例に記
載したようにFeCl3・6H2Oを添加することによ
つてFe−IMDA溶液を製造した。こうして製造
した組成物は、例と同様に、ヘモグロビン
0.139mg/dL及びアスコルベート50μMの存在で
青色を形成した。
例 ……Fe−NTA
Fe−HEDTAではなく、ニトリロ三酢酸の鉄
キレート(Fe−NTA)の10mM溶液を使用する
ことを除いて、例の実験を繰り返した。
NTA0.191gを蒸留水100mlに溶解し、例に記
載したようにFeCl3・6H2Oを添加することによ
つてFe−NTA溶液を製造した。こうして製造し
た組成物は例と同様にヘモグロビン0.139mg/
dL及びアスコルベート50μMの存在で青色を形成
した。

Fe−HEDTAではなく、α−エチレンジアミ
ン二酢酸二プロピオン酸の鉄キレート(Fe−
EDDPα)の10mM溶液を使用することを除いて、
例の実験を繰り返した。EDDP〓0.320gを蒸留
水100mlに溶解し、例に記載したように
FeCl3・6H2Oを添加することによつてFe−
EDDPα溶液を製造した。こうして製造した組成
物は例と同様にヘモグロビン0.139mg/dL及び
アスコルベート50μM存在で青色を形成した。

Fe−HEDTAではなく、α−エチレンジアミ
ン二酢酸二プロピオン酸の鉄キレート(Fe−
EDDP〓)の10mM溶液を使用することを除いて、
例の実験を繰り返した。EDDP〓0.320gを蒸留
水100mlに溶解し、例に記載したように
FeCl3・6H2Oを添加することによつてFe−
EDDP〓溶液を製造した。こうして製造した組成
物は例と同様にヘモグロビン0.139mg/dL及び
アスコルベート50μMの存在で青色を形成した。

Fe−HEDTAではなく、ヒドロキシエチルイ
ミノ二酢酸の鉄キレート(Fe−HIMDA)の
10mM溶液を使用することを除いて、例の実験
を繰り返した。HIMDA0.177gを蒸留水100mlに
溶解し、例に記載したようにFeCl3・6H2Oを
添加することによつてFe−HIMDA溶液を製造
した。こうして製造した組成物は例と同様にヘ
モグロビン0.139mg/dL及びアスコルベート
50μMの存在で青色を形成した。
例 −
各例において0.2Mクエン酸ナトリウム緩衝剤
を9.5mlではなく、9.4ml使用し、各鉄キレートを
0.1mlではなく、0.2ml使用する以外は、実質的に
例〜に記載したように実験を行つて、本発明
による組成物を製造した。これは各組成物中に
200μMの濃度の鉄キレートを存在させたことに
なる。それぞれの場合に、200μMの鉄キレート
を含む組成物を前記のように試験すると、該組成
物がヘモグロビン0.139mg/dL及びアスコルベー
ト50μMの存在で青色を形成するのが観察され、
このことは試料中にアスコルベートが存在して
も、ヘモグロビンを検出しうることを示す。しか
し、例〜の100μM鉄キレート組成物を同様
に試験したときに観察された色形成と比較して、
色形成に必要な時間の間に差が認められた。それ
ぞれの場合の色形成時間(「ラグタイム」と言わ
れる)を下記の表に示す。この時間は前記のよう
に、本発明の組成物に使用する金属キレート、例
えば前記鉄錯体の1種の濃度とその組成物のアス
コルベート妨害に抵抗し、過酸化物活性物質の検
出しうる能力との間には一般的関係又は相関関係
はないことを示す。

C 試験具のアスコルベート妨害抵抗性及び安定
性
前記の例に記載したようにして製造し
た試験具の、長期貯蔵に似せたストレスの後にア
スコルベートの存在で尿中のヘモグロビンを検出
しうる能力を評価するため、更に実験を行つた。
新しく製造した直後の試験具及び高温条件で長時
間貯蔵した後の試験具について、実験を行つた。
特に、環境温度(23℃)で製造直後、並びに乾燥
器中で50℃で10日及び28日の「熱ストレス」後
に、試験具を性能について試験し、比較した。
種々のヘモグロビン濃度を含む試験尿溶液を調整
した。50mg/dLの濃度でアスコルベートを含む
2つのヘモグロビン溶液も製造した。
全血を蒸留水で、水100ml当たりヘモグロビン
15.4mgの濃度に希釈することによつてヘモグロビ
ン15.4mg/dLを含むストツク溶液を製造した。全
血のヘモグロビン含有量は従来技術により予め測
定しておいた。プールし、予めスクリーニングし
たヘモグロビン及びアスコルビン酸が陰性である
尿試料を、盲検として併置した。次に、血液溶液
をプールした尿中にピペツトで加えて、ヘモグロ
ビンを0.015、0.031、0.062及び0.139mg/dL含む
尿溶液を作ることによつて試験溶液を製造した。
ヘモグロビンを0.031及び0.062mg/dL含む尿溶液
を別の容器に分離し、アスコルビン酸を添加し
て、試験直前に溶液のアスコルベート濃度を50
mg/dLにした。
例と同様にして製造した試験具及びFe−
HEDTAを含まない以外は、その例に記載したよ
うにして製造した対照試験具を盲検及び各ヘモグ
ロビン/尿溶液で試験した。試験具を各溶液中に
瞬間的に浸漬し、取り出し、1分後、試験具にお
ける色形成を観察した。形成した色を浸漬後1分
に相対強度について、別の試験具及び標準カラー
チヤートと肉眼で比較した。色は、無色(盲検)
から0.139mg/dLヘモグロビン溶液で暗帯緑青色
に及ぶ。
この試験の結果は、下記の表に示すとおり、本
発明による試験具が、アスコルベートを含む2つ
のヘモグロビン試料で、新しく製造した後及び50
℃の高温に10日曝露した後に試験すると、アスコ
ルベートを含まない尿試料を試験するため使用し
た試験具の応答と同様に、ヘモグロビンの存在に
対する色応答を生じることを示す。Fe−

つた結果は、対照試験具の応答が試料中のアスコ
ルベートによつてほぼ完全に損なわれたことを示

素原子を有する直鎖又は分枝鎖アルキレン基又
は6〜9個の炭素原子を有する2価の1,2−
脂環式基を表し; (c) nは0〜1の数値を有する整数、mは0〜2
の数値を有する整数であり、mが0より大きい
