レン化合物の製造法に関する。
【0004】前記一般式(III)で表されるジフルオ
ロメチレン化合物は、ジフルオロメチレン基を含む医農
薬の合成原料としての利用が期待できる(たとえばBi

【従来技術】前記一般式(III)で示されるジフルオ
ロメチレン化合物を合成する方法としては、(1)ケト
ンやアルデヒドを四フッ化イオウとフッ化水素(たとえ
ばOrg. React., 21, 20
(1974))あるいは三フッ化ジエチルアミノイオウ
(DAST)(たとえばJ. Org. Chem
., 40, 574 (1975))で処理す
る方法、(2)ジチオアセタールをN−ハロアミドのよ
うなハロニウムイオン発生剤と無水フッ化水素−ピリジ
ン錯体の組み合わせで処理する方法(たとえばJ.
Org. Chem., 51, 3508
(1986))がある。
【0006】(1)の方法で用いる四フッ化イオウやフ
ッ化水素は毒性の高い低沸点化合物(四フッ化イオウの
沸点は−40℃、フッ化水素の沸点は19.5℃)であ
り取扱いが困難である。また、しばしば高温下、特殊な
材質(たとえばハステロイCなど)でできたオートクレ
ーブ中で反応させる必要がある。さらに四フッ化イオウ
は反応性が高いため、たとえばアルコールのような官能
基をもつ基質のジフルオロメチレン化はできない。DA
STを用いる方法もしばしば高温、長時間を必要とする
。また、水酸基もフッ素化を受けてしまうので保護する
必要がある。(2)の方法においては、反応系が強い酸
性になるため、たとえばエポキシドのような酸に弱い官
能基を持つ基質に用いるとジフルオロメチレン化合物は
得られずに複雑な反応混合物が得られるだけである(参
考例1参照)。

【発明が解決しようとする課題】本発明者らはジフルオ
ロメチレン化号物を合成するにあたり、(1)安価で、

は50%フッ酸、フッ化カリウム及びフッ化四級アンモ
ニウムから容易に合成できる(Bull. Soc.

))。本発明に用いる二水素三フッ化四級アンモニウム
の使用量は1当量ないし大過剰量の範囲であるが、経済
性を考慮して1−4当量が好ましい。

ロモコハク酸イミド(NBS)、1,3−ジブロモ−5
,5−ジメチルヒダントイン(DBH)、N−ブロモア

イミド(NIS)などを例示することができる。本発明
に用いるハロニウムイオン発生剤の使用量は1当量ない
し大過剰量の範囲であるが、経済性を考慮して1−3当
量が好ましい。

ヒドあるいはケトンをチオールで処理することにより容
易に得られる。また、R2が水素原子のジチオアセター
ルを塩基で処理し次いで求電子剤(ハロゲン化アルキル
、カルボニル化合物、エポキシドなど)で処理すること
により、容易に得ることができる。
【0022】反応に際し、溶媒を使用してもよい。溶媒
としては反応に関与しないものならば何でも使用でき、
ヘキサン、ベンゼン、トルエン、アセトニトリル、ジエ
チルエーテル、テトラヒドロフラン、1,2−ジメトキ
シエタン、ジクロロメタン、1,2−ジクロロエタン、
四塩化炭素などを例示することができる。また、適宜こ
れらを混合物として使用してもよい。
【0023】反応は基質にもよるが、−100℃から1
00℃の温度範囲で実施できる。しかし、反応時間を短
縮し、かつ選択性を高めるためには−78℃から20℃
の間で行うのが好ましい。
【0024】本方法によれば、従来法が適用できない基
質、たとえば水酸基や酸に弱い官能基を持つ基質のジフ
ルオロメチレン化も、これらの官能基を冒すことなく=
C(SR3)(SR4)構造が選択的に=CF2構造に
変換され、収率よく、高い官能基選択性をもって目的物
が得られる利点がある(実施例18及び参考例1参照)
。
