【発明の詳細な説明】[Detailed description of the invention]
本発明はL−アスパラギン酸β−脱炭酸酵素活
性を有する微生物のアラニンラセマーゼ活性を除
去する方法に関する。
L−アスパラギン酸β−脱炭酸酵素はL−アス
パラギン酸のみに使用してL−アラニンと炭酸ガ
スとを生成する反応を触媒する酵素である。
従来、上記L−アスパラギン酸β−脱炭酸酵素
を利用するL−アラニンの製造法としては、L−
アスパラギン酸β−脱炭酸酵素活性を有する微生
物を基質であるL−アスパラギン酸またはその塩
含有培地に培養する方法や該微生物の生産した酵
素を基質に作用させる方法等多数知られている。
しかしながら、一般に微生物の細胞壁の構成成分
としてD−アラニンが必須であるため、これらの
微生物は同時にアラニンラセマーゼ活性を有する
ことが知られており、L−アラニンの生成と共に
D−アラニンも副生してくる。従つてL−アスパ
ラギン酸β−脱炭酸酵素活性を有する微生物を用
いて光学純度の高いL−アラニンを効率よく製造
するためには該微生物のアラニンラセマーゼ活性
発現を阻止する必要がある。
しかしながら現在知られている酵素を高温で処
理する方法(酵素工学ハンドブツク、P.20,A・
ワイズマン編1977年、講談社発行)ではアラニン
ラセマーゼ活性は抑えられるがL−アスパラギン
酸β−脱炭酸酵素活性も共に抑えられるためL−
アラニンの生成量が低下するという難点があつ
た。
しかるに本発明者等は鋭意研究を重ねた結果、
微生物の有するL−アスパラギン酸β−脱炭酸酵
素活性には全く影響を与えることなく微生物のも
つアラニンラセマーゼ活性のみを選択的に失活さ
せる方法を見出した。
即ち、本発明によれば、L−アスパラギン酸β
−脱炭酸酵素活性を有する微生物又はその固定化
微生物を酸処理することにより当該微生物又は固
定化微生物のアラニンラセマーゼ活性を除去する
ことができる。
本発明方法において用いられるL−アスパラギ
ン酸β−脱炭酸酵素活性を有する微生物としては
上記活性を有する微生物であればいずれも用いる
ことが出来、かかる微生物としては、例えばシユ
ードモナス・ダクネーIAM1152、アセトバクタ
ー・ランセンスOUT8300、アクロモバクター・
ペスチフアーIAM1446、同ATCC23584、アルカ
リゲネス・フエーカリスATCC25094号等が好適
にあげられる。
これらの微生物は遊離菌体のまま用いることが
でき、該菌体をそれ自体公知の方法で固定化した
微生物であつてもよく、例えばポリアクリルアミ
ドゲル、含硫多糖類ゲル(カラギーナン、フアー
セレラン等、コラーゲンゲル、アルギン酸ゲル、
ポリビニルアルコールゲル、寒天ゲルで固定化し
た微生物をいずれも本発明の目的に用いることが
できる。上記の内ポリアクリルアミドゲルによる
場合は例えば特公昭53−1831号記載の通り微生物
菌体の水けん濁液にアクリルアミドモノマー、架
橋剤(例えばN,N′−メチレンビスアクリルア
ミド)、重合促進剤〔例えばβ−(ジメチルアミ
ノ)プロピオニトリル〕、重合開始剤(例えば過
硫酸カリウム)を加え、ゲル化させることにより
固定化微生物が得られる。また、カラギーナン、
フアーセレラン等の含硫多糖類による場合は例え
ば特開昭53−6483号に記載の通り微生物菌体をカ
ラギーナン水溶液にけん濁しこのけん濁液にゲル
化剤(例えば第4周期以上のアルカリ金属、アル
カリ土類金属、アンモニウムイオン等)を接触さ
せるか冷却してゲル化させることにより固定化微
生物が得られる。含硫多糖類としては分子内の硫
酸基含量10w/w%以上、とりわけ硫酸基含量12
〜62w/w%のものを用いるのが好ましい。その
他、コラーゲンゲル、アルギン酸ゲル、ポリビニ
ルアルコールゲル、寒天ゲル等による場合も、そ
れ自体公知の方法例えば特開昭51−144780号、特
開昭49−30582号、特開昭49−80285号、特開昭51
−133484号記載の方法に従つて当該微生物を処理
することにより容易に相当する固定化微生物を得
ることができる。これらの固定化微生物は例えば
立方体状(1辺約3mm程度)、球状(直径約3mm
程度)、繊維状(径約1mm程度)、膜状あるいは板
状等の適当な形状に成型すればつづいての処理操
作を効率よく実施できるので好ましい。
上記の如き微生物又は固定化微生物の酸処理は
当該微生物又は固定化微生物を酸溶液中に浸漬す
るか又は当該微生物又は固定化微生物を含有する
液中に酸を加えることにより実施できる。酸とし
ては例えば酢酸、乳酸、酒石酸等の有機酸、塩
酸、硫酸等の無機酸をいずれも用いることがで
き、とりわけ酢酸を用いるのが好ましい。これら
酸による処理はPH3〜6、とりわけ4〜5.5の条
件下に実施するのが好ましく、又酸は水溶液それ
自体だけでなく酸緩衝液をも用いることができ
る。
微生物又は固定化微生物含有液中に酸を加えて
実施する場合には前記酸を上記PH範囲となるよう
に酸の添加量を調整することにより行なう。更に
微生物又は固定化微生物の酸処理は約0〜60℃、
とりわけ約20〜50℃で実施するのが好ましい。接
触時間は微生物が遊離菌体の場合約3分〜24時
間、とりわけ約5分〜10時間程度とするのが好ま
しく、固定化微生物の場合には約10分〜5日間、
とりわけ約1〜48時間程度とするのが好ましい。
かくして得られたアラニンラセマーゼ活性の除
去された微生物または固定化微生物にL−アスパ
ラギン酸またはその塩を作用させるとL−アラニ
ンが得られ、またDL−アスパラギン酸またはそ
の塩を作用させるとL−アラニンとD−アスパラ
ギン酸が得られる。ここでL−アスパラギン酸ま
たはDL−アスパラギン酸の塩としては、例えば
それらのナトリウム塩、カリウム塩、カルシウム
塩、アンモニウム塩等が好ましい。酵素反応は、
0〜50℃の広い温度範囲で実施することができる
が、微生物の酵素の安定性を考慮して30〜40℃で
実施するのが好ましい。尚、上記酵素反応に際し
ては基質溶液にピリドキサールリン酸、ケト酸
(例えばピルビン酸、a−ケトグルタール酸等)
等を適宜添加することにより該酵素反応を一層促
進させることが出来、また更にコバルトイオン、
ニツケルイオン等の2価金属イオンを添加すると
微生物の酵素活性の安定性を高めることが出来
る。
上記において、遊離の微生物を用いる場合の反
応はバツチ法で実施するのが好ましい。又、固定
化微生物を用いる場合の反応は使用する固定化微
生物が水に不溶性であるため、バツチ法によるの
みならずカラム法によつて連続的に実施すること
が出来る。例えばカラム法による場合、該固定化
微生物をカラムに充填し、このカラムにL−アス
パラギン酸、DL−アスパラギン酸またはそれら
の塩を有する溶液を適当な速度で導通すればL−
アラニンのみ、あるいはL−アラニンとD−アス
パラギン酸とを含む流出液が得られる。なおこの
場合、酵素反応により生成する炭酸ガスのカラム
内への滞留を避けるため基質溶液はカラム下部よ
り上部に向けて流すのが好ましい。またバツチ法
による場合、該固定化微生物を上記基質溶液にけ
ん濁し、かく拌する如き方法によりL−アラニン
のみあるいはL−アラニンとD−アスパラギン酸
とを含む反応液が得られる。この場合には反応終
了液から固定化微生物をろ過あるいは遠心分離す
る如き方法により取得すれば再びこれを反覆使用
することができる。これらの方法において、基質
としてDL−アスパラギン酸またはその塩を用い
る場合L−アラニンとD−アスパラギン酸とを含
む溶液が得られるが、これらの両者は例えば直接
晶析法、イオン交換樹脂処理等の公知の単離精製
操作を適宜組合せることにより容易に分離採取す
ることができる。上記反応を実施するにあたつて
反応進行率は固定化微生物の量、温度、反応時
間、基質の流速その他により影響される。例えば
カラム法による場合は使用する固定化微生物の量
に従い基質溶液の導通速度を、またバツチ法によ
る場合はその反応時間を適当に調整することによ
り反応進行率を100%にまで高める至適条件を見
出すことも容易である。
以上の如く、本発明方法は(1)L−アスパラギン
酸β−脱炭酸酵素活性を有する微生物を酸処理す
るという極めて簡単な操作で該微生物のアラニン
ラセマーゼ活性を完全に除去できること;(2)該方
法は遊離の微生物にも固定化微生物にもともに適
用できること;(3)又、一旦除去されたアラニンラ
セマーゼ活性は長期間酵素反応を実施しても再び
出現することがないこと;(4)更にアラニンラセマ
ーゼ活性の除去された微生物を公知の方法で固定
化してもその得られる効果には何ら変りがない
等、種々のすぐれた特徴及び効果を有するもので
ある。又、本発明方法によりアラニンラセマーゼ
活性の除去された微生物あるいは固定化微生物を
用いてL−アスパラギン酸(又はDL−アスパラ
ギン酸)からL−アラニン(又はL−アラニンと
D−アスパラギン酸)を製造すれば生成物中に
DL−アラニンが含まれていないので再結晶等後
処理が不用であり、L−アスパラニンの工業的に
極めてすぐれた製造方法となるものである。
以下、実験例、実施例により本発明をさらに詳
細に説明する。
実験例 1
下記により調製した微生物を用いてL−アスパ
ラギン酸からL−アラニンを生成させ、その際副
生するD−アラニン生成物を比較した。
(1) 微生物の調製
(本発明方法による微生物の調製)
グルタミン酸ナトリウム3.2%、ミースト0.5
%、第1リン酸カリウム0.05%、硫酸マグネシウ
ム0.01%を含む培地(PH7.3)120mlを500ml容坂
口フラスコに入れ、これにシユードモナス・ダク
ネーIAM1152を植菌した。30℃で24時間培養し
たのち酢酸2.6mlを加えPHを4.75に調整し30℃で
1時間放置した。
(対照微生物の調製)
酢酸処理をしない以外は上記と同様にして微生
物を調製した。
(2) 実験方法
上記で得られた微生物各1gを、10-4Mピリドキ
サールリン酸と、0.05%ポリオキシエチレンオク
チルフエニルエーテルを含む1ML−アスパラギ
ン酸アンモニウム水溶液(アンモニアでPH6.0に
調整)70mlを入れた200ml容三角フラスコに加え
37℃にて振とう反応させ反応中の全アラニン量と
D−アラニン量を経時的に測定した。
尚、全アラニン量の測定はロイコノストツク・
チトロボラムATCC8081を用いるバイオアツセイ
により行ない、D−アラニン量の測定は豚腎より
調製したD−アミノ酸オキシダーゼを作用させ生
成したピルビン酸をヒドラゾンとして測定する方
法で行なつた。
(3) 結果
結果は後記図面に示す通りであり本発明方法に
より調製した微生物はD−アラニンを全く生成し
ないのに対し対照の微生物は22時間目におけるD
−アラニン生成量が約20%に達することが認めら
れた。
実験例 2
下記により調製した固定化微生物を用い連続酵
素反応を実施し反応終了液中のD−アラニン生成
量を比較した。
(1) 固定化微生物の調製
(本発明方法による固定化微生物の調製)
実験例1(1)と同様の培地を500ml容坂口フラス
コ10本に120ml宛分注しこれにシユードモナス・
ダクネーIAM1152を植菌した。30℃で24時間振
とう培養したのちピリドキサールリン酸300mgを
添加しこれに酢酸26mlを加えPHを4.75に調整し30
℃で1時間放置した。これに3N水酸化ナトリウ
ム86mlを加えPH6.0に調整したのち遠心分離する
ことによりシユードモナス・ダクネー菌体20g
(湿重量)を集めた。
別にゲニユーゲルWG(商品名、コペンハーゲ
ンペクチンフアクトリー社製のカラギーナン、以
下同)4.03gを50℃の温水85mlに溶解しカラギー
ナン水溶液を調製した。この溶液に上記で得たシ
ユードモナス・ダクネーの菌体20gを生理的食塩
水20mlにけん濁したものを45℃にて添加混合し
た。4℃にて30分間放置したのち生成したゲルを
一辺3mmの立方体に成型することにより固定化シ
ユードモナス・ダクネーを得る。
(本発明方法の固定化微生物の調製)
上記と同様の培地、菌体を用いて上記と同様に
30℃で24時間振とう培養したのち遠心分離してシ
ユードモナス・ダクネー菌体20g(湿重量)を集
めた。
別にゲニユーゲルWG4.03gを50℃の温水85ml
に溶解しカラギーナン水溶液を調製する。この溶
液に上記で得たシユードモナス・ダクネーの菌体
20gを生理的食塩水20mlにけん濁したものを45℃
にて添加混合した。4℃にて30分間放置したのち
生成したゲルを一辺3mmの立方体に成型した。こ
のゲルを10mML−アスパラギン酸および2%塩
化カリウムを含む0.2M酢酸緩衝液(PH5.0)260
ml中に浸漬し37℃にて24時間放置した。その後2
%塩化カリウム水溶液で洗浄することにより固定
化シユードモナス・ダクネーを得る。
(対照微生物の調製)
上記本発明方法による微生物と同様にして調
製し、一辺3mmの立方体に成型した固定化微生物
を以後何ら処理することなく用いた。
(2) 実験方法
固定化シユードモナス・ダクネー40gをそれぞ
れ外とう管付カラム(内径1.6cm×長さ19cm)に
充填し、37℃にて10-4Mピリドキサールリン酸を
含む1ML−アスパラギン酸アンモニウム溶液
(アンモニアにてPH6.0に調整)をカラム下部より
上方に向つて12ml/hrの流速で導通した。定常状
態に達した後、流出液中の全アラニンとD−アラ
ニン濃度を経時的に測定した。
尚、全アラニン量およびD−アラニン量の測定
は実験例1と同様にして行なつた。
(3) 結果
結果は下記表に示す通りであり、本発明方法に
より調製した固定化微生物はD−アラニンを全く
生成しないのに対し、対照の固定化微生物は約5
%のD−アラニンを生成することが認められた。

脂処理により回収したL−アラニンを合せ活性炭
処理することにより、L−アラニン61.8gを結晶

に溶解しカラギーナン水溶液を調製した。この溶
液に上記で得たシユードモナス・ダクネーの菌体
20gを生理的食塩水20mlにけん濁したものを45℃
にて添加混合した。4℃にて30分間放置したのち
生成したゲルを1辺約3mmの立方体に成型するこ
とにより固定化シユードモナス・ダクネー130g
(湿重量)を得た。
(2) L−アラニンの製造
上記(1)で得た固定化シユードモナス・ダクネー
40gを外とう管付カラム(内径1.6cm×長さ19cm)
に充填し37℃にて10-4Mピリドキサールリン酸を
含む1ML−アスパラギン酸アンモニウム溶液
(アンモニアにてPH6.0に調整)500mlをカラム下
部より上方に向つて12ml/hrの流速で導通した。
流出液500mlを全量150mlになるまで濃縮しエタノ
ール150mlを加えた。析出晶をろ取し冷メタノー
ル30mlで洗浄することによりL−アラニン40.5g
を結晶として得る。

実施例 3
(1) 固定化微生物の調製
実施例2(1)で用いた培地と同様の培地を500ml
容坂口フラスコ10本に120ml宛分注しこれにシユ
ードモナス・ダクネーIAM1152を植菌した。30
℃で24時間振とう培養したのち遠心分離すること
によりシユードモナス・ダクネー菌体20g(湿重
量)を集めた。
別にゲニユーゲルWG4.03gを50℃の温水85ml
に溶解しカラギーナン水溶液を調製する。この溶
液に上記で得たシユードモナス・ダクネーの菌体
20gを生理的食塩水20mlにけん濁したものを45℃
にて添加混合した。4℃にて30分間放置したの
ち、生成したゲルを一辺3mmの立方体に成型し
た。このゲルを10mML−アスパラギン酸および
2%塩化カリウムを含む0.2M酢酸緩衝液(PH
5.0)260ml中に浸漬し37℃にて24時間放置した。
その後、2%塩化カリウム水溶液で洗浄すること
によりアラニンラセマーゼ活性を有しない固定化
シユードモナス・ダクネー130g(湿重量)を得
た。
(2) L−アラニンとD−アスパラギン酸の製造
上記(1)で得た固定化シユードモナス・ダクネー
40gを外とう管付きカラム(内径1.6cm×長さ19
cm)に充填し、37℃にて10-4Mピリドキサールリ
ン酸を含む1MDL−アスパラギン酸アンモニウム
水溶液(アンモニアにてPH6.0に調整)500mlをカ
ラム下部より上方に向つて15ml/hrの流速で導通
し、流出液500mlを約100mlとなるまで濃縮しPH
3.0に調整した。析出晶をろ取し冷水で洗浄する
ことによりD−アスパラギン酸30.4gを得る。
〔α〕25 D−24.9°(C=2,5N−HCl)
D−アスパラギン酸をろ取したろ液をアンバー
ライトR4B(OH-型)樹脂カラムに導通した。
流出液を減圧濃縮しエタノールを加えることによ
り、L−アラニン19.0gを得た。
