Abstract
Description
Claims (30)
- 1.存在する場合、サンプル中のヒト乳頭種ウィルス(HPV)を検出し、そし てそのHPVの型を分類するための方法であって: (a)HPVが存在する場合、コンセンサスプライマーの拡張生成物が合成され 得るような条件下でコンセンサスHPVプライマー、重合剤及びデオキシヌクレ オシド5′−トリホスフェートによりサンプルを処理し、ここで前記コンセンサ スプライマーは、一対のプライマーの1つのメンバーから合成される拡張生成物 が、その相補的鎖から分離される場合、前記対の他のメンバーの拡張生成物の合 成のための鋳型として作用することができるように、それらにハイプリダイズす るHPVDNAの一本鎖を分離するために十分に相補的な少なくとも1対のプラ イマーを含んで成るオリゴヌクレオチドの混合物であり; (b)もし存在するなら、前記鋳型からプライマー拡張生成物を分離し、これに 基づいて拡張生成物が一本鎖分子を形成するために合成され; (c)もしあるなら、段階(b)で生成された一本鎖分子を、プライマー拡張生 成物が鋳型として段階(b)で生成された一本鎖分子のそれぞれを用いて合成さ れるような条件下で段階(a)のコンセンサスプライマーにより処理し;(d) 段階(b)及び(c)を少なくとも1度くり返し;(e)増幅が生じたかどうか を決定し;そして増幅が生じた場合、 (f)前記増幅されたDNAにタイプ特異的DNAプロープをハイプリダイズし ;そして (g)ハイブリダイゼーションが生じたかどうかを決定することを含んで成る方 法。
- 2.前記HPVが生殖器HPVである請求の範囲第1項記載の方法。
- 3.前記段階(e)が、ハイブリダイゼーション条件下で段階(d)において調 製された反応混合物をコンセンサスプローブにより処理し、そしてハイブリダイ ゼーションが生じたかどうかを決定することを含んで成る請求の範囲第2項記載 の方法。
- 4.前記コンセンサスプライマーが前記生殖器HPVの転写調節領域の配列を増 幅することができる請求の範囲第2項記載の方法。
- 5.前記コンセンサスプライマーが前記生殖器HPVのL1読み取り枠配列を増 幅することができる請求の範囲第2項記載の方法。
- 6.前記コンセンサスプライマーが前記生殖器HPVのE6読み取り枠配列を増 幅することができる請求の範囲第2項記載の方法。
- 7.前記コンセンサスプライマーが前記生殖器HPVのE1読み取り枠配列を増 幅することができる請求の範囲第2項記載の方法。
- 8.前記コンセンサスプライマーが前記生殖器HPVのE7読み取り枠配列を増 幅することができる請求の範囲第2項記載の方法。
- 9.前記コンセンサスプライマーが前記生殖器HPVのE1,E6及びE7読み 取り枠配列を増幅することができる請求の範囲第2項記載の方法。
- 10.前記タイプ特異的プローブが前記生殖器HPVの転写調節領域の配列にハ イプリダイズすることができる請求の範囲第2項記載の方法。
- 11.前記タイプ特異的プローブが前記生殖器HPVのL1読み取り枠配列にハ イプリダイズすることができる請求の範囲第2項記載の方法。
- 12.前記タイプ特異的プローブが前記生殖器HPVのE6及びE7配列から成 る群から選択されたE6読み取り枠配列にハイプリダイズすることができる請求 の範囲第2項記載の方法。
- 13.前記コンセンサスプローブが前記生殖器HPVの転写調節領域の配列にハ イプリダイズすることができる請求の範囲第3項記載の方法。
- 14.前記コンセンサスプローブが前記生殖器HPVのL1読み取り枠配列にハ イブリダイズすることができる請求の範囲第3項記載の方法。
- 15.前記コンセンサスプローブが前記生殖器HPVのE6読み取り枠配列にハ イプリダイズすることができる請求の範囲第3項記載の方法。
- 16.前記コンセンサスプローブが前記生殖器HPVのE1読み取り枠配列にハ イブリダイズすることができる請求の範囲第3項記載の方法。
- 17.前記コンセンサスプローブが前記生殖器HPVのE7読み取り枠配列にハ イプリダイズすることができる請求の範囲第3項記載の方法。
- 18.前記生殖器HPVの完全なURR領域の配列が増幅される請求の範囲第4 項記載の方法。
- 19.前記プライマーがFS10,JS15,JS16,FS17及びMY01 から成る群から選択される請求の範囲第4項記載の方法。
- 20.前記プライマーがMY11及びMY09である請求の範囲第5項記載の方 法。
- 21.前記プライマーがWD64,WD65,WD66,WD67,WD68, WD69,WD70,WD71,WD72,WD73,W076,WD77,W D83,WD84,WD154,WD155,WD157,WD158,WD1 59,WD160,WD161,WO162,WD163、及びWD164から 成る群から選択される請求の範囲第6項記載の方法。
- 22.前記プライマーがTYN01,TYN02,TYN03,TYN04,T YN05,TYN06,TYN07,TYN08,TYP01,TYP02,T YP03,TYP04,TYP05、及びTYP06から成る群から選択される 請求の範囲第7項記載の方法。
- 23.前記生殖器HPVの完全なE7領域の配列が増幅される請求の範囲第9項 記載の方法。
- 24.前記プローブがMY12,MY13,MY14,MY16,MY58,M Y59,MY60,MY61,MY62,MY63,MY64,MY65,MY 69,MY70,MD74,MD126,MD127,MD128,MD150 ,MD151,MD152、及びWD153から成る群から選択される請求の範 囲第11項記載の方法。
- 25.前記プローブがWD78,WD79,WD80,WD81,WD82,W D102,WD103,WD104,WD132,WD133,WD134,W D165,UD166,WD167,WD168、及びWD169から成る群か ら選択される請求の範囲第12項記載の方法。
- 26.前記プライマーが少なくとも1つのE1プライマー及び少なくとも1つの E6プライマーを含んで成る請求の範囲第13項記載の方法。
- 27.前記コンセンサスプローブがFS10,MY18,MY19,MY39, MY55,MY56,MY57,MY66,MYXX、及びWD147から成る 群から選択されたプローブを含んで成る請求の範囲第14項記載の方法。
