- 【特許請求の範囲】 1、個々の哺乳動物の染色体を有糸分裂細胞または間期細胞の中で染色体特異的 プローブと正常部位ハイブリダイゼーションさせて、染色体特異的シグナルを生 成することによって、個々の哺乳動物の染色体を標識する方法。 2、個々のヒトの染色体を有糸分裂細胞または間期細胞の中で染色体特異的プロ ーブと正常部位ハイプリダイゼーションさせて、染色体特異的シグナルを生成す ることによって、個々のヒトの染色体を標識する方法。 3、染色体特異的である標識したDNAプローブをヒト有糸分裂細胞またはヒト 間期細胞の中でDNAに正常部位抑制ハイブリダイゼーションすることからなる 、個々のヒト標的染色体の高度に特異的なデコレーションを生成する方法。 4、DNAプローブは、全組み換えライブラリーのDNA、染色体誘導組み換え DNAライブラリーから精製したDNAインサート、および染色体から誘導した 特異的DNA断片から成る群より選択される、上記第3項記載の方法。 5、標識したDNAプローブは、少なくとも1種の蛍光色素で標識したDNAプ ローブ、特異的結合対の少なくとも1つの構成員で標識したDNAプローブ、お よび酵素で標識したDNAプローブから成る群より選択される、上記第4項記載 の方法。 6、蛍光色素は、フルオレセイン、ローダミン、テキサスレッド(Texasr ed)、ルツファーイエロー(LuciferyelIow)、フィコビリンタ ンパク質およびシアニン色素から成る群より選択され、そして特異的結合対の構 成員はビオチンである、上記第5項記載の方法。 7、染色体の正常部位ハイブリダイゼーションにより、ヒト細胞中の染色体異常 を評価する方法。 8、ヒト細胞は、分裂中期細胞、前期細胞および間期細胞から成る群より選択さ れる、上記第7項記載の方法。 9、工程: a)1)存在する核酸配列を相補的核酸配列とのハイプリダイゼーションに利用 可能に処理した、ヒト異数性細胞;および2)特異的染色体から誘導された標識 したヒトDNA、競争DNA、およびヒト以外のゲノムのDNAからなるハイブ リダイゼーション混合物を、相補的核酸配列のハイブリダイゼーションが起こる ために適当な条件下に、一緒にし、そして b)前記異数性細胞からの核酸配列にハイブリダイゼーションした特異的染色体 から誘導された標識したヒトDNAを検出する、からなる、ヒト異数性細胞中の 染色体異常を検出する方法。 10、異数性細胞はヒト腫瘍細胞である、上記第9項記載の方法。 11、ヒト腫瘍細胞は、分裂中期細胞、前期細胞および間期細胞から成る群より 選択される、上記第10項記載の方法。 12、ヒト腫瘍細胞はヒトグリオーム細胞である、上記第10項記載の方法。 13、工程: a)1)試料の中に存在する核酸配列を相補的核酸配列とのハイブリダイゼーシ ョンに利用可能に処理した、試料;および2)選択した染色体から誘導された標 識したヒトDNA、競争DNA、およびヒト以外のゲノムのDNAからなるハイ ブリダイゼーション混合物を、相補的核酸配列のハイプリダイゼーションが起こ るために適当な条件下に、一緒にし、そして b)試料の中に存在する核酸配列にハイブリダイゼーションした選択した染色体 から誘導された標識したヒトDNAを検出する、からなる、試料の中に存在する ヒトコスミドの数の変更を試料の中で検出する方法。 14、選択したヒト染色体は、染色体13、18、21、XおよびYから成る群 より選択される、上記第13項記載の方法。 15、選択したヒト染色体は染色体番号21であり、そして選択した染色体から 誘導された標識したヒトDNAは染色体誘導組み換えDNAライブラリーから精 製したDNAインサートである、上記第13項記載の方法。 16、工程: a)1)ヒト腫瘍細胞の中に存在する核酸配列を相補的核酸配列とのハイブリダ イゼーションに利用可能に処理した、ヒト腫瘍細胞;および2)選択した染色体 から誘導された標識したDNA断片、競争DNA、およびヒト以外のゲノムのD NAからなるハイブリダイゼーション混合物を、相補的核酸配列のハイブリダイ ゼーションが起こるために適当な条件下に、一緒にし、そして b)前記腫瘍細胞からの核酸配列にハイブリダイゼーションした標識したヒト染 色体特異的DNA断片を検出する、からなる、ヒト腫瘍細胞中の選択した染色体 またはその一部分のリプリゼンテイションの過剰または低下を検出する方法。 17、工程: a)次の配列:1)選択した哺乳動物の染色体、2)検出可能な標識を有する選 択した哺乳動物の染色体から誘導したDNA断片、3)競争DNA、およびキャ リヤーDNAを、相補的核酸配列のハイブリダイゼーションが起こるために適当 な条件下に、一緒にして、検出可能な標識を有するDNA断片と選択した哺乳動 物の染色体との複合体を形成し、そして b)工程(a)において形成した複合体を検出する、からなる、選択した哺乳動 物の染色体の中に存在する染色体特異的DNAを同定する方法。 18、工程(a)において一緒にした残りの物質から形成した複合体を分離する ことによって、選択した哺乳動物の染色体中の染色体特異的DNAを単離するこ とをさらに含む、上記第17項記載の方法。 19、存在するDNAが相補的核酸配列とのハイプリダイゼーションに利用可能 とするように予備処理された、哺乳動物の有糸分裂細胞または哺乳動物の間期細 胞を、2またはそれ以上の選択した検出可能に標識した、高い遺伝的複雑性をも つ、染色体特異的ヌクレオチドプローブと、CISSハイブリダイゼーション条 件下に、一緒にして、選択した染色体を標識し、そして標識した選択した染色体 を検出することからなる、哺乳動物の有糸分裂細胞または哺乳動物の間期細胞の 中で選択した染色体を同定する方法。 20、存在するDNAが相補的核酸配列とのハイブリダイゼーションに利用可能 とするように予備処理された、哺乳動物の有糸分裂細胞または哺乳動物の間期細 胞を、同定すべき異なる染色体の1つに対して相補的な少なくとも第1の検出可 能に標識した核酸プローブ2および同定すべき異なる染色体の第2のものに対し て相補的な第2の検出可能に標識した核酸プローブからなる、検出可能に標識し た染色体特異的プローブの組と、CISSハイブリダイゼーション条件下に、一 緒にして、検出可能に標識した選択した染色体を生成し、前記第1の検出可能に 標識した核酸プローブ上の検出可能な標識は前記第2の検出可能に標識した核酸 プローブ上の検出可能な標識と異なり、そして標識した選択した染色体を検出す ることからなる、哺乳動物の有糸分裂細胞または哺乳動物の間期細胞の中で少な くとも2つの異なる選択した染色体を同定する方法。 21、検出可能に標識した染色体特異的プローブは蛍光的に標識されている、上 記第20項記載の方法。
