Description
本発明は、禿げた部分のウブ毛を通常の硬毛に
生育せしめる発毛、養毛剤に係るものである。
禿げは、硬毛が生え変るたびに細く小さくなり
ウブ毛化した状態であつて、毛包の機能が全く消
失したものではないが、その原因については従来
から種々の説明がなされている。
その代表的なものをあげると、皮脂腺からの皮
脂の分泌が多くなり頭皮の垢などと混じつて皮脂
漏となり、毛孔に詰つて毛根の栄養障碍,循環障
碍を来たすためであるとする説(皮脂漏説)、男
性ホルモン(テストステロン)が過剰に生産さ
れ、それまでの男女の両性ホルモンのバランスに
よつて支配されていた頭頂部にアンバランスが生
じるためであるとする説(ホルモン説)、その他
遺伝、栄養と関連させた説などがある。しかし、
いずれも決定的な原因とみることはできなかつ
た。
それは、前記の諸説が毛の発生に関する従来の
誤まつた皮膚科学上の定説、つまり、皮膚に毛球
が形成されるとその内部に毛母細胞が形成され、
この細胞から分化が始まり漸次角化して毛となる
という定説に支配されていたためである。
このような実情から、従来用いられている発毛
又は養毛剤は、毛球部の血液循環を良好にするこ
とを主眼としていた。例えば、ヒノキオール、メ
ントール、トウガラシチンキなどの水溶液を主体
とし、これを頭皮に塗布して頭皮の血液循環を良
好に保ち、2次的に毛球部の血液循環の改善をは
かり、毛の養毛又は再生を待するものであるが、
充分な効果をあげ得ないのが現状である。
本発明は、前述した毛の発生に関する従来の定
説とは異なり、本発明者が長年にわたつて研究
し、その結果、見出された新しい毛の発生理論に
基づいて完成されたものであり、この発生理論は
本発明を理解する上で不可欠のものであるから、
以下にその大要を説明する。
第1図は本理論による毛周期の説明図である。
すなわち、毛包とこれに付随している皮脂腺1と
が1つのセツトになり、成長期には、毛は毛球部
4の毛母細胞5から形成されてくる。しかし、永
久的に生え続けるのではなく、移行期を経て休止
期になり、抜毛し、次の再生が始まつてくる。こ
れが毛周期である。
従来の定説では、この休止期の毛包の下端部よ
り再生が始まるとしていたが、本発明による新し
い毛の発生説によれば、皮脂腺1の排出管の出口
の部分である峡部毛鞘2から毛芽が形成され(毛
芽期)、その毛包3下部の上皮性の胚芽細胞層よ
り毛が形成され、毛を作りながら下降し(毛杭
期)、その後に毛球が形成される。しかし、この
毛球部4に毛母細胞5が形成されるまではなお下
降し(毛球性毛杭期)、その毛母細胞5が充分に
機能を発揮するようになると下降が止り、皮表の
方にだけ毛が出てくる(完成毛)のである。この
ように、毛は毛球部4の毛母細胞5のみから形成
されるのではなく、表皮と同様に毛包の上皮性細
胞からも形成されること、また毛の形成には前記
皮脂腺1が重要な基盤になつていることは、毛の
発生に関する新しい学説(皮脂腺説又は稲葉説)
として内外に承認されているのである。
他方、毛の成長に関与する男性ホルモンである
テストステロンは睾丸8で形成され、従来は第2
図の鎖線aで示すように直接に毛球部4の毛乳頭
7から毛母細胞5に作用するとみられていた。
本発明者はこれに対し、このテストステロンは
毛球部4だけでなく直接皮脂腺1に血行によつて
運れるのであり(実線b)、皮脂腺1内に存在す
る還元酵素5αリダクターゼによつて還元され、
強力な性ホルモンである5αデイハイドロテスト
ステロン(以下「5αDHT」という)に転換さ
れ、次に、この5αDHTが毛包の周囲の血管によ
つて血乳頭7、毛母細胞5に運ばれc、毛母細胞
5の細胞分裂を抑制するという全く新しい説を見
出すに至つた。
したがつて、栄養,ホルモンなどの影響により
皮脂腺が肥大してくると、5αリダクターゼの量
が多くなり、テストステロンが強力な性ホルモン
5αDHTに転換され、毛包は漸次縮少して毛がウ
ブ毛化し禿げの状態になるのである。このよう
に、テストステロンそれ自体あるいは少量の
5αDHTは毛の生成を促進するものであるが、こ
れが多量に転換された5αDHTはその作用が余り
にも強過ぎるため、あたかも植物に過剰に肥料を
与えるとその成育が阻害されるのを同様に、毛母
細胞に対して抑制的に作用するのである。
本発明は以上に述べた禿げの形成過程から、前
記皮脂腺内に存在している5αリダクターゼの酵
素作用を抑制すれば、テストステロンが強力な性
ホルモン5αDHTに転換されることが少ない。し
たがつて、毛母細胞に対する抑制作用が減弱又は
消失してくることに着目し、皮脂腺内に存在して
いる5αリダクターゼの作用を抑制又は消失させ
る物質を毛の発毛、養毛剤中に含ましめたことを
要旨としている。
現在まで、5αリダクターゼの活性を抑制する
目的に競合抵抗剤例えば黄体ホルモン、その他の
男性ホルモンを用いる試みがあつたが、副作用が
多く、発生剤としては適当でない。
本発明者は5αリダクターゼが還元性酵素であ
ることから、この活性を減弱又は消失させる一つ
の方法として、強い酸化作用を有し、かつ人体に
無害で副作用を生ずることがない物質を用いるこ
とである。
このような物質としては、例えば過ホウ酸ナト
リウム,ブロム酸ナトリウム等がある。なお、前
記の2条件を満す限り、これ以外の酸化物質を用
いることもできる。
これら酸化剤は5αリダクターゼの作用を抑制
又は消失させるのに十分で、かつ人体に影響を与
えないような組成,濃度とし、これを必要に応じ
他成分とともに、1日に数回、禿げた頭皮に塗
り、軽くマツサージし、これを長期間に亘つて連
続的に使用する。
これにより、各種の酸化作用物質が頭皮の毛孔
から皮脂腺に浸透し、皮脂腺に存在する還元型補
酵素(NADH,NADPH(TPNH))並びに5αリ
ダクターゼの活性を減弱又は消失させる。このた
め睾丸から運ばれたテストステロンは5aDHTに
転換されることが少なく、テストステロンあるい
は少量の5αDHTのみの作用が毛母細胞に働くこ
ととなる。この5αリダクターゼは皮脂腺に多量
に、毛球部には少量のみ存在するとの報告があ
る。したがつて、睾丸より経路aによつて、直接
毛母にテストステロンが運ばれ少量の5αDHTが
形成され、毛の発育を促進するので、本発明の効
果が損なわれることはない。
このテストステロンあるいは少量の5αDHT
は、あごひげ、胸毛どの男性の思春期以後の発生
と同様に頭髪にも好結果をもたらし、前述した毛
の再生過程図に示すように毛包が発育形成され、
毛母細胞に活溌な細胞分裂が生起して硬毛に成生
してくるのである。
(実施例)
ブロム酸ナトリウムおよび過ホウ酸ナトリウム
の2種につき、臨床テストを下記の要領でおこな
つた。なお、この臨床試験は米国において、各テ
ストにつき30〜50才の男性のみ5名ずつを対象と
しておこない、1日約5時間おきに3回使用し
た。
その結果を表に示すが、表中の効果は使用者に
よつてそれぞれ個人差はあつたが、全体を総合し
て判定を下したものである。
