Description
(産業上の利用分野)
本発明は、アルギン酸のゲル又は固定化ゲルを
強化する方法に関するものである。
更に詳細には、本発明は、カルシウムイオンで
形成されたアルギン酸のゲル又は固定化ゲルをス
トロンチウムイオンで処理し、ゲル又は固定化ゲ
ルを強化する方法に関するものである。
本発明において強化されたゲル又は固定化ゲル
は燐酸等を含む培養液によつて脱弱化させること
なく、長期間の生化学的反応等に耐え得るもの
で、生化学界に益するところ大なるものがある。
(従来の技術)
一般に、酵素、微生物、動物細胞等をアルギン
酸ナトリウム溶液に添加し、カルシウムイオンを
含む溶液中に、滴下したり、紡糸したりして、ゲ
ル化して、生化学的反応等に利用することはよく
知られている。
(発明が解決しようとする問題点)
しかし、従来のアルギン酸ナトリウムとカルシ
ウムイオンを反応させて得られたゲルが反応液中
に含まれる種々の影響、或はPHの変化によつてゲ
ル強度が著しく低下したり、リン酸イオン等のキ
レート剤の存在によりゲルが溶解してしまうとい
う問題点があつた。
そこでゲルの強度を向上させるため反応液中に
カルシウムイオンを多量に添加することも行われ
るが、多量のカルシウムイオンによつて、酵素、
微生物、動物細胞等の生化学的活性が阻害される
という問題が生じるのである。
(問題点を解決するための手段)
本発明者らは、アルギン酸ゲルを長期間の培養
等に耐える強固なものにするために鋭意研究をし
た結果、カルシウムイオンを用いてゲル化したア
ルギン酸ゲルをストロンチウムイオンで処理する
ことによつて、強固なアルギン酸ゲルを得ること
に成功したものである。
本発明は、アルギン酸ナトリウム溶液をカルシ
ウムイオンで適宜形状に成型し、次いでストロン
チウムイオンで処理し、強化することを特徴とす
る強化されたゲルの製造法である。
また、本発明は、生化学的活性体を添加したア
ルギン酸ナトリウム溶液をカルシウムイオンで適
宜形状に成型し、次いでストロンチウムイオンで
処理し、強化することを特徴とする強化された固
定化ゲルの製造法である。
本発明に使用するアルギン酸ナトリウムはカツ
藻類中に含まれるアルギン酸を化学操作により抽
出しナトリウム塩にしたもので、市販のアルギン
酸ナトリウムなどいずれも使用できる。しかしな
がら、アルギン酸はD−マンヌロン酸(M)とL
−グルロン酸(G)とから構成されていて、両者
の成分比(M/G比)が物性に大きな影響を与え
ている。
そして、Mのみからなる部分、Gのみからなる
部分、MとGが混在する部分などがあるがゲル化
剤の金属イオンと結合して強固なゲルを形成する
のは主としてGブロツクの働きによることが明ら
かとなつている。
本発明では、アルギン酸を構成するD−マンヌ
ロン酸(M)とL−グルロン酸(G)の比、すな
わちM/G比が0.1〜0.8より好ましくは0.1〜0.2
のものを選んで用いるのがよい。
この様なM/G比の低いアルギン酸ナトリウム
は、G含量の高い海藻類、若しくは多く含まれて
いる部位〔茎(幹)部〕、及びとれた季節(5〜
8月)を選択して抽出調整するか、又はG含量の
高いものをプレゼントするなどの方法によつて、
特別に調製して得ることができる。
M/G比が上記範囲にあるものはゲル強度が大
である。
本発明においては、カルシウムイオンで処理し
たアルギン酸ゲルをストロンチウムイオンで処理
することによつて、ゲルを強化することを特色と
するものである。
カルシウムイオンとしては硝酸カルシウム、酢
酸カルシウム、乳酸カルシウム、酪酸カルシウ
ム、塩化カルシウム等が用いられる。又、ストロ
ンチウムイオンとしては塩化ストロンチウム、硝
酸ストロンチウム、酢酸ストロンチウム等の水溶
性の塩が用いられ、特に塩化ストロンチウムが容
易に入手できて使いやすい。
具体的には、生化学的活性体を添加もしくは添
加しないアルギン酸ソーダの溶液をカルシウムイ
オンを含む溶液で、粒状、繊維状等に成型した
後、その成型ゲルをストロンチウムイオンを含む
溶液で処理し、燐酸等に対して強化されるもので
ある。
ナトリウムと置換したカルシウムの一部又は大
部分がストロンチウムと置き換つたり、新たにス
トロンチウムが残つたカトリウムと置換したり、
更にはこれらの組合せも起り、全体としてゲルが
燐酸等に対して強化されるものと考えられるが、
カルシウムイオンでゲル化して、更にストロンチ
ウムイオンで処理することによつて、ゲルが燐酸
等に対して強化される理由の詳細は不明である。
アルギン酸ナトリウムの濃度は、0.5(W/V)
%〜8(W/V)%濃度が良く、より好ましくは
0.8(W/V)%〜3(W/V)%濃度である。
また、アルギン酸ナトリウム溶液に添加される
生化学的活性体としては、酵素、微生物、動物細
胞、植物細胞などがある。
酵素としては、アルコールデヒドロゲナーゼ、
D−アミノ酸オキシダーゼ、カタラーゼ等の酸化
還元酵素、トランスケトラーゼ、アデニレートキ
ナーゼ、ヘキソキナーゼ等の転移酵素、β−ガラ
クトシターゼ、ペニシリナーゼ、リパーゼ、エス
テラーゼなどの加水分解酵素、フマラーゼ、アス
パルターゼ、スレオニンアルドラーゼ、β−チロ
シターゼなどのリアーゼ酵素、グルコースイソメ
ラーゼ、アラニンイソメラーゼなどの異性化酵
素、グルタチオンシンターゼ、グルタミンシンタ
ーゼなどのリガーゼ酵素などがあげられる。
又、微生物としては細菌、酵母、カビ、放線菌
などの酵素活性を有する微生物であれば特に限定
されることはなく、また、動物細胞としては、各
種生理活性物質を生産する細胞株、抗体を生産す
るハイブリドーマなどがあり、更に植物細胞とし
ては各種生理活性物質を生産する細胞がある。
生化学的活性体は0.01〜50%、好ましくは0.1
〜20%程度アルギン酸ナトリウム溶液に添加さ
れ、混合される。ただし、動物細胞の場合は、ア
ルギン酸ナトリウム溶液1mlに対し1×104〜1
×109、好ましくは1×105〜5×108個添加され
る。
生化学的活性体を添加、もしくは添加しないア
ルギン酸ナトリウム溶液は次の1〜3の方法等で
ゲル化される。ゲル化に用いるカルシウムイオン
の濃度は0.01〜1.0モル濃度程度で、好ましくは
0.02〜0.3モル濃度程度である。
1 注射器やピペツト等のノズルからカルシウム
イオンを含有する水溶液中に滴下することによ
りビーズ状のゲルが得られる。
2 カルシウムイオンを含有する水溶液中で注射
器、ピペツト等のノズルから連続的に吐出させ
ることにより繊維状のゲルが得られる。
3 平板上にキヤストするか濾紙もしくはガーゼ
などに含浸させた後、水溶液中のカルシウムイ
オンと接触させることにより膜状のゲルを得る
ことができる。
ここにられたゲルはストロンチウムイオンを含
む溶液に浸漬したり、該溶液を噴霧したりして処
理される。ストロンチウムイオンの濃度は0.01〜
1.0モル、好ましくは0.02〜0.3モル程度である。
処理時間は1分〜1時間、PH3〜11、温度4〜50
℃程度である。
本発明のストロンチウムイオンで強化処理され
たアルギン酸ゲルは燐酸に対して強い抵抗力をも
つようになる。動物細胞株、ハイブリドーマ等を
ゲル中に含有させた場合、培養液にリン酸緩衝液
を使用するので、特に有効である。即ち、ストロ
ンチウムイオンで強化処理したものは1週間以上
の長期間リン酸緩衝液に浸漬しても崩壊しないが
ストロンチウムイオンで強化処理しないものはリ
ン酸緩衝液に浸漬して1日以内ですべて崩壊して
しまうものである。
次に本発明の実施例を示す。
実施例 1
2%アルギン酸ナトリウム溶液100mlに、市販
のマウス細胞(大日本製薬K.K製)Bリンパ球を
常法により融合したハイブリドーマ培養懸濁液を
1.0×106ケ/gゲルになるように添加、混合し、
注射器につめ、氷水中の0.1M塩化カルシウム溶
液に滴下し、60分放置し、粒状化ゲルを得た。
得られたゲルを無血清RDF培地で3日間37℃
で静置培養した。
さらに粒状化ゲルを5つに分けて、氷水中の
0.1M塩化ストロンチウム溶液に0分、5分、10
分、20分、30分それぞれ浸漬した。
得られた各強化ゲルを無血清RDF培地で3日
間37℃で静置培養し、グルコースの消費量をみ
た。
結果は表1に示される。

幼ハムスター腎細胞(大日本製薬K.K製)を4.7
×106ケ/gになるように添加、混合し、注射器
につめ、氷水中の0.1M塩化カルシウム溶液に滴
下し、60分放置し、粒状化ゲルを得た。
得られたゲルをカラムにつめ、3%グルコース
添加BME培地300mlづつ毎日とりかえながら65日
間30℃で循環培養し、グルコース消費量を測定
し、65日間後ゲルを取り出し、氷水中の0.1M塩
化ストロンチウム溶液に20分間浸漬し、後再びカ
ラムにつめ、3%グルコース添加BME培地300ml
づつを毎日とりかえながらあと10日間30℃で循環
培養し、グルコース消費量を測定した。その結果
は第1図に示されるが、この図から塩化ストロン
チウムによる処理が細胞に全く影響を及ぼさない
