Description
本発明はエシエリヒア属に属するトリプトフア
ンリプレツサー対応遺伝子(TrpR保持菌株の培
養方法に関し、更に詳しくは、菌体中にトリプト
フアンシンターゼの含有量が高く且つ高収量の菌
体を取得することのできるエシエリヒア属に属す
るTrp保持菌株の培養方法に関する。
トリプトフアンシンターゼはインドールとL又
はDL−セリンとからL−トリプトフアンを製造
する場合の酵素触媒として有用な物質である。
L−トリプトフアンは飼料や飲料等への添加物
としての用途或いは各種の工業的用途が考えられ
るが、現在までのところ工業的に安価な製造法が
開発されておらず、これらの用途は制限されてい
るのが実情である。したがつてL−トリプトフア
ンの工業的製造法の確立が望まれている。
ところで、L−トリプトフアンの製造法として
これ迄いくつかの方法が提案されている。例え
ば、(1)糖質を用いブレビバクテリウム・フラバム
を直接発酵して培養中に産生されるL−トリプト
フアンを精製採取する方法(特開昭57−174096号
公報);(2)インドールもしくはアントラニル酸と
糖質を含有する培養液中でバチルス・ズブチリス
を培養し、培養液中にL−トリプトフアンを生成
蓄積させ、その培養液からL−トリプトフアンを
精製採取する方法(特公昭53−35152号公報);(3)
インドールとセリン或いはインドールとピルビン
酸及びアンモニウムイオンから、微生物が生成す
る酵素、トリプトフアンシンターゼ又はトリプト
フアナーゼを用いて、酵素反応によりL−トリプ
トフアンを製造する方法(特開昭47−39693号公
報、特公昭49−46917号公報)等が提案されてい
る。
本発明者らはインドールとセリンとから酵素法
によるL−トリプトフアンの製造に注目し研究を
行なつてきた。この酵素法によるL−トリプトフ
アンの製造のキーポイントはその触媒作用を担う
トリプトフアンシンターゼを如何に安価に大量に
生産するかにかかつている。このトリプトフアン
シンターゼは微生物によつて生産されるものであ
るから、トリプトフアンシンターゼを安価に大量
に生産するためには、微生物菌体中のトリプトフ
アンシンターゼの含有量を高めると共に培養時に
いける該菌体が高収量で得られる培養法を開発す
ることが必須である。
微生物菌体中の特定の酵素の含有量を増大させ
るための一つの有力方法として、遺伝子組み換え
技術を利用した遺伝子増幅効果が考えられる。し
かしながら、トリプトフアンプロモーター及びオ
ペレーター領域を利用する場合には、上記のよう
な遺伝子増幅効果を有している菌株においても、
主染色体上にあるトリプトフアン抑制機構である
トリプトフアンリプレツサー対応遺伝子の作用に
よりトリプトフアンプロモーター活性が強く阻害
され、その結果菌株中のトリプトフアンシンター
ゼの含有量は一定のレベルに抑制されてしまい、
菌体中のトリプトフアンシンターゼを著量蓄積さ
せることは困難である。
また、この抑制機構であるトリプトフアンリプ
レツサーをコードする遺伝子TrpRが欠失した菌
株を用いることも考えられるが、かかるTrpR欠
失菌株は、トリプトフアン生合成に関与する遺伝
子をもつプラスミドで形質転換すると、該菌株中
ではプラスミドが極めて不安定でプラスミド全体
の脱落もしくは部分的欠失などが起こることが報
告されており〔Journal of General
Microbiology,118、253〜261(1980)参照〕工
業的には問題がある。
一方、トリプトフアンリプレツサーの機能を抑
制する方法として、培地にトリプトフアン誘導体
であるインドールアクリル酸を10μg/ml以下の
濃度で添加し、トリプトフアンリプレツサーを不
活化することによつて、菌株中のトリプトフアン
シンターゼ含有量を高めることができることが報
告されている〔Pro.Nat.Acad.Sci.USA,
Vol.71、No.9、pp3455〜3459(1974)参照〕が、
培地としては最小栄養培地、すなわち合成培地を
使う必要があると指示されているため、単位培養
液当りの菌株の収量は低く、1菌体中のトリプト
フアンシンターゼの含有量が高くても、単位培養
液当りのトリプトフアンシンターゼの生産量は低
く、工業的に安価に酵素を得る方法としては適さ
ない。
また、真該細胞由来のペプチドをコードする遺
伝子、ベクター及びプロモーターからなる組換え
DNAを含有する該ペプチド生産能をもつ微生物
を、インドールアクリル酸を添加した培地では培
養することにより、該ペプチドの生産性を高める
方法もごく最近提案された(特開昭58−141796号
公報)が、この方法も有機窒素源の低い合成培地
を使用しており、菌体生産性は低く、工業的安価
な酵素源とはなりえない。
本発明者らは、上記のインドールアクリル酸を
添加する培養法において、合成培地の代りに、ポ
リペプトン、トリプトン、ペプトンなどのペプト
ン類、肉エキス、酵母エキス、麦芽エキス、コー
ンステイプリカー、カザミノ酸などの天然成分を
含む天然培地の使用可能性を検討すべく、大腸菌
培養用の天然培地として代表的なL培地(トリプ
トン10重量部、酵母エキス5重量部、グルコース
1重量部、NaCl5重量部、純水1000重量部、PH
7.2)を用い、上記文献の指示に従いインドール
アクリル酸を10μg/mlの濃度で添加し、大腸菌
〔YK2001株、(FERM P−7139)〕の培養を試み
たが、上記文献に記載されたような菌体中のトリ
プトフアンシンターゼ含有量の増大効果は認めら
れなかつた。
そこで、本発明者らはこの天然培地におけるイ
ンドールアクリル酸の添加効果につき、インドー
ルアクリル酸の添加量及び添加時期の両面から鋭
意検討を重ねた結果、インドールアクリル酸を上
記文献に記載されているよりもはるかに多量の少
くとも25μg/ml以上の最終濃度で添加すると、
菌体中のトリプトフアンシンターゼの含有量が増
大するばかりでなく、菌体の収量も向上するこ
と、しかも、この効果はインドールアクリル酸を
培養の初期に添加する場合のみならず、菌体が充
分に増殖するまで、すなわち対数増殖期の末期ま
での任意の時点まで添加し培養を継続する場合に
も得られることを見い出し、本発明を完成した。
かくして、本発明によれば、エシエリヒア属に
属するトリプトフアンリプレツサー対応遺伝子
(TrpR)保持菌株を、有機窒素源を窒素原子重
量に換算して少なくとも0.1重量%含有する栄養
培地で培養するに際して、遅くても該菌株の対数
増殖期の末期までに該培地にインドールアクリル
酸又はその塩を少なくとも25μg/mlの最終濃度
で添加して培養を行ない菌体中のトリプトフアン
シンダーゼの含有量が増大した上記菌株を取得す
ることを特徴とするエシエリヒア属に属する
TrpR保持菌株の培養方法が提供される。
本発明の方法においては、栄養培地として、合
成培地(最小栄養培地)と異なり、有機窒素源を
多量に含有する、いわゆる天然培地を使用する。
すなわち、本発明では有機窒素源を窒素原子重量
に換算して少なくとも0.1重量%、好ましくは0.2
〜1.0重量%、さらに好ましくは0.25〜0.6重量%
含有する天然培地を使用する。かかる天然培地に
おける有機窒素源としては、従来からエシエリヒ
ア属に属する微生物の培養のために使用されてい
る天然培地において窒素源として配合されている
ものが同様に用いられ、例えば、ポリペプトン、
トリプトン、ペプトンなどのペプトン類、肉エキ
ス、酵母エキス、麦芽エキス、コーンステイープ
リカー、カザミノ酸等の有機窒素化合物(アミノ
酸、ペプチド、ビタミン、未知生育促進因子な
ど)をそれぞれ単独で又は2種もしくはそれ以上
混合して用いることができる。本発明の方法に使
用する培地には、窒素源として上記の有機窒素源
に加えて、無機窒素源、例えば、(NH4)2SO2、
NH4Cl、(NH4)2PO4、(NH4)NO3等を含ませ
ることも可能である。
また、本発明の方法に使用する培地は、栄養源
としては、上記の窒素源の他に、炭素源、ミネラ
ル分、ビタミン等を含有することができる。炭素
源としては、例えば、グルコール、ギリセロー
ル、フラクトース、シユクロース、廃糖蜜などの
種々の炭水化物を利用できる。
以上に述べた如き栄養分を含む培地は最終的に
PHを一般に5〜9の範囲、好ましくは6〜8の範
囲に調製した後、菌体の培養に供することができ
る。
本発明の方法に従い培養することのできる微生
物は、微生物菌体内にトリプトフアンシンターゼ
を産出する能力のある菌株であり、殊に、トリプ
トフアンシンターゼ生産能をもつエシエリヒア属
に属するトリプトフアンリプレツサー対応遺伝子
(本明細書においてこの遺伝子を“TrpR”と略
記する)を保持した菌株である。上記トリプトフ
アンリプレツサー対応遺伝子(TrpR)は、該菌
株の主染色体にコードされていてもよく或いはプ
ラスミド中に存在していてもよく、本発明はその
いずれのタイプの菌株に対しても適用することが
できる。
しかして、本発明の方法においては、トリプト
フアンシンターゼ生産能をもつエシエリヒア属に
属するTrpR保持菌株はいずれも使用可能である
が、特に、トリプトフアンプロモーター及びオペ
レーター領域とトリプトフアンシンターゼ対応遺
伝子をもつプラスミド、より好適にはトリプトフ
アンオペレンを有するプラスミドで形質転換する
ことにより、トリプトフアンシンターゼの生産能
が増幅された大腸菌(Escherichia coli)菌株が
有利に使用される。そのようなプラスミドを含有
する大腸菌菌株としては、例えば、本発明者らが
先に創製し提案したトリプトフアンシンターゼの
生合成を司る遺伝子を含む画分と、F因子プラス
ミドの増殖制御分画系を司る遺伝子を含む画分と
を有する新規なプラスミドで形質転換された大腸
菌菌株を有効に使用することができる。
しかして、この新規なプラスミドについてさら
に詳しく説明する。
上記のプラスミドを構成する「トリプトフアン
シンターゼの生合成を司る遺伝子を含む画分」
(以下「T画分」と略称することがある)とは、
インドールとL−又はDL−セリンからL−トリ
プトフアンの生合成を司る遺伝子を含む画分を意
味し、しかして、T画分にはトリプトフアンオペ
ロン又はトリプトフアンオペロンを含むもつと大
きな遺伝子画分、或いは、トリプトフアンオペロ
ン中のトリプトフアンシンターゼの生合成を司る
遺伝子であるTrpAとTrpBにプロモータ及びオ
ペレーターを結合した画分、又はTrpAとTrpB
にTrpC、TrpD及びTrpEの少なくとも1種とプ
ロモーター及びオペレーターを結合した画分(例
えば、TrpA、TrpB、TrpC及びTrpEにプロモ
ーター及びオペレーターが結合した画分)等が包
含される。これらのT画分としては実用的には大
腸菌由来のものが好適に使用され、中でも、トリ
プトフアンオペロンそれ自体及びフアージφ80pt
の遺伝子を制限酵素BamHIにより切断して得ら
れる約22.6kbの分子量を有するトリプトフアンオ
ペロンを含む画分が有利に使用される。
他方、F因子プラスミドは例えば「蛋白質 核
酸 酵素」第27巻第1号(1982)の98頁の図1の
遺伝子地図及びEco RIによる物理的地図に示さ
れる如き構造をもつ、分子量が94.5kb(62×106
dolton)の既知のプラスミドであり、大腸菌など
の腸内細菌中に通常細胞染色体当り1〜2個のコ
ピー数で存在し、従つて染色体が細胞当り2個あ
るとすればF因子プラスミドしては2〜4個しか
担われていない。このプラスミドは細胞分裂時に
ちようど倍化してそれぞれの娘細胞に正確に伝達
されるような機構を備えている(このように、コ
ピー数を低レベルに保ちつつ、正確に宿主の増殖
とペースを合わせて増やす仕組みをstringentな
増殖の制御と呼んでいる)。F因子プラスミドに
おけるこのようなstringentな増殖の制御機能が、
mini−Fと呼ばれる分子量が9.1kbの自律増殖で
きる断片に担われていることも既に究明されてお
り、このmini−F因子プラスミドより制限酵素
EcoRIにより切り出し可能であることも知られて
いる。
上記新規なプラスミドはこのmini−Fに担わ
れている増殖制御分配系を利用するものであり、
しかして「F因子プラスミドの増殖制御分配系を
司る遺伝子を含む画分」(以下F画分と略称する
ことがある)とは、上述したようなF因子プラス
ミドを娘細胞に正確に伝達する機構を備えた遺伝
子画分を意味し、そのようなF画分の代表例とし
ては約9.1kbの分子量を有するmini−F画分が挙
げられる。
前記新規なプラスミドは、以上に述べたT画分
とF画分に加えて、さらに他の遺伝子画分、例え
ば各種細菌の薬剤耐性を司る、例えばカナマイシ
ン耐性等の遺伝子画分、例えばカナマイシン耐性
を司る遺伝子画分等を含有していてもよい。
そのようなプラスミドの代表例として、トリプ
トフアンオペロン画分及びF因子プラスミドの
mini−F画分を含有し、分子量が約31.7kbであ
り且つ下記の制限酵素に対して下記の感受性を示
す、すなわち
(a) EcoRIに対する認識部位が5ケ所であり、
(b) BamHIに対する認識部位が2ケ所であり、
(c) Hindに対する認識部位が3ケ所であるこ
とを特徴とするプラスミドpMTY−1が挙げ
られる。
このプラスミドpMTY−1は上記のとおり約
31.7kbの分子量を有するが、この分子量はアガロ
ース・ゲル電気泳動法により測定したものであ
る。
また、プラスミドpMTY−1の各種制限酵素
に対する感受性は上記(a)〜(c)に示すとおりであ
り、より詳細に各制限酵素による分解断片の分子
量(kb)を示せば次のとおりである。
(a) EcoRI:9.1,7.0,6.4,6.1,3.1
(b) BamHI:22.6,9.1
(c) Hind:18.0,10.8,2.9
以上に述べた如き特性をもつプラスミド
pMTY−1は例えば次のようにして製造するこ
とができる。
まず、トリプトフアンオペロン画分の調製は、
例えば、染色体遺伝子中にトリプトフアンオペロ
ンをもつ大腸菌、例えば、Escherichia coliK−
12(IFO3301,ATCC10798,ATCC e23562)な
どに、フアージ、例えばフアージφ80
(ATCC11456a−B1)などを感染させ、溶源化及
び誘発現象を利用して、フアージDNA中にトリ
プトフアンオペロンを取り込んだフアージを大量
に調製し〔R.M.Denney、C.Yanofsky;J.
Bacteriol.,118、505(1974)参照〕、それから常
法〔E.F.Fritsch,Sambrook,“Molecular
cloning”(1982)p.164〜165、Cold Spin
Harbor Laboratory参照〕に従つてフアージ
DNAを抽出し、制限酵素、例えばBamHI,
EcoRI等を用いてトリプトフアンオペロンを含む
遺伝子画分を切り出すことにより行なうことがで
きる。
他方、miniF画分の調製は、例えば、F因子プ
ラスミドを保持する微生物、例えば、大腸菌(E.
coli)K−12株(ATCC15153,ATCC e23589,
ATCC e23590)等からそれ自体公知の方法で、
例えばP.Guerry、D.L.LeBlanc、S.Falkow;J.
Bact.,116,1064(1973)等の文献に記載の方法
でF因子プラスミドを取り出し、それから制限酵
素EcoRIを用いて分子量が約9.1kbのminiF断片
を切り出すことにより調製することができる。
このようにして調製されたminiF断片は、上記
トリプトフアンオペロンを含む遺伝子画分の切り
出しに用いたと同じ制限酵素で処理した後、上記
で調製したトリプトフアンオペロンを含む遺伝子
画分と一緒にし、リガーゼ、例えばT4フアージ
由来のリガーゼ等を作用させて結合させることに
より、目的とするプラスミドpMTY−1が得ら
れる。
なお、プラスミドpMTY−1の具体的調製法
については後記参考例1に詳細に説明されてい
る。
このようにして調製されるプラスミドは、コピ
ー数は少ないが、宿主の細胞分裂に際して娘細胞
に正確に受け継がれ、脱落することが少ないとい
う優れた特性を有する。
従つて、このプラスミドをトリプトフアンの製
造において工業的に応用するに際して、該プラス
ミドで宿主が形質転換される。この形質転換に利
用できる宿主菌としては、例えば、大腸菌、殊に
トリプトフアン要求性変異株としたものが特に好
ましいものである。
これら宿主菌に対する上記のプラスミドの導入
はそれ自体公知の方法、例えばM.Mandel,A.
Higa;J.Mol.Biol,53,159(1970)等の文献に
記載の方法で行なうことができる。
以上に述べた菌株を前述した如き天然培地で培
養するに際し、本発明の方法は、培地にインドー
ルアクリル酸又はその塩(塩としては例えば、イ
ンドールアクリル酸のNa塩、K塩、NH4塩等が
挙げられる)を少なくとも25μg/mlの最終濃度
が達成されるような量で添加することを本質的特
徴とするものである。この特定の高濃度のインド
ールアクリル酸又はその塩の添加により、トリプ
トフアンリプレツサーが不活化されてトリプトフ
アンプロモーター及びオペレーターが抑制解除状
態となり、該菌株はトリプトフアンシンターゼを
高生産性でもつ生成すると考えられる。
しかして、インドールアクリル酸又はその塩の
添加量は、培地に対する添加量が最終的に
25μg/ml以上となるような割合で添加される。
添加量の上限は特に制限はないが、培地に対する
溶解度以上に加えても無意味であり、一般には
500μg/ml以下とすることができる。好適な添加
量範囲は用いる菌株等にもよるが、通常80〜
200μg/mlであり、特に90〜150μg/mlの範囲が
好ましい。インドールアクリル酸又はその塩は上
記量を一度に添加してもよく、或いは場合によつ
ては数回に分けて添加してもよい。
また、インドールアクリル酸又はその塩の添加
時期は、菌体の充分に増殖し終るまで、すなわち
対数増殖期の末期までであり、その間であればい
つの時点で添加してもほぼ同じトリプトフアンシ
ンターゼの増収効果が達成されることが判明し
た。しかして、インドールアクリル酸又はその塩
は培養の初期(開始時)に添加してもよく、或い
は菌体の対数増殖期の終る直前に添加してもよ
く、或いはまた、上記の期間中数回に分けて添加
してもよい。いずれにしても、対数増殖期の末期
における培地に対するインドールアクリル酸又は
その塩の添加量の合計が最終的に25μg/ml培地
以上となるようにすべきである。
前記菌株の培養は、上記のようにしてインドー
ルアクリル酸又はその塩を添加することを除け
ば、それ自体公知の方法で行なうことができる。
例えば、培養は、振盪培養或いは通気攪拌深部培
養などの好気的条件下に行うことができる。培養
温度は一般に20〜50℃の範囲とすることができ、
培地のPHは中性または微アルカリ性附近、通常PH
6〜8の範囲に維持することが望ましい。培養は
菌体内にトリプトフアンシンターゼが充分に蓄積
するまで行なうことができ、培養期間は通常4〜
48時間程度で充分である。
以上に述べた本発明の培養法により、菌体中に
おけるトリプトフアンシンターゼの含有量が高い
菌体を高収量で取得することができ、単位培地当
りのトリプトフアンシンターゼの生産量は、イン
ドールアクリル酸又はその塩を添加せずに培養を
行なつた場合の約2〜10倍にすることが可能であ
る。
しかして、本発明の方法よれば、トリプトフア
ンシンターゼを工業的に安価に且つ大量に提供す
ることを可能にするものであり、インドールとL
−又はDL−セリンとからのL−トリプトフアン
の工業的生産に大いに貢献するものである。
本発明の方法により得られる菌体をL−トリプ
トフアンの生産に利用するに際して、該菌体は培
養物から分離した後そのまま使用することもでき
るが、普通一般に行なわれるように超音波破砕処
理等の破砕処理を行なつた後に使用してもよく、
或いはまた、該破砕処理物をさらに水等で抽出し
た抽出物、又は該抽出物をさらに硫安等で処理し
て酵素成分を沈殿させた粗精製物の形で使用する
こともでき、さらに、該菌体又はこれら処理物は
必要により固定化して用いることもできる。
このようにして調製される菌体又はその処理物
は、インドールとL−又はDL−セリンとの反応
の触媒として使用される。該菌体又はその処理物
の存在下でのインドールとL−又はDL−セリン
との反応は、通常の酵素反応と同様に、例えば
0.1Mリン酸緩衝液(PH7.0〜9.0)あるいは水(PH
7.0〜9.0)等の溶媒中で、約20〜約50℃、好まし
くは約30〜約40℃の温度で通常約10〜約72時間行
なわれる。
インドールとL−又はLD−セリンの反応時の
使用量には特に制限はないが、一般にはそれぞれ
を0.1〜20%(wt/vol)の濃度範囲で使用するの
が適当である。また、該菌体又はその処理物の使
用量も特に制限されるものではないが、一般に1
〜10%(wt/vol)の濃度で使用することができ
る。
上記のような培養方法によつて得られた菌体又
はその処理物を用いてインドールとL−、又は
DL−セリンを反応せしめて得られる、反応液中
に生成したL−トリプトフアンの分離・精製は、
イオン交換樹脂、活性炭等による吸着、脱着処理
等の公知の方法により行うことができる。
次に実施例及び参考例により本発明をさらに説
明する。
参考例1:プラスミドpMTY−1の調製
(A) miniF画分の調製
大腸菌(E.coli)K−12菌株(ATCC15153)
を1のL培地(Bactoトリプトン10g、酵母
エキス5g、NaCl5g、グルコース1g、水1
;PH7.2)に接種し、約37℃で約4時間振盪
培養した後、菌体を集め、リゾチウム処理を行
ない且つドデシル硫酸ナトリウム(SDS)を添
加して溶菌させた。この溶菌液を32000×gで
40分間遠心分離処理し、上清を分画し、次いで
塩化セシウム−エチジウムブロマイド平衡密度
勾配遠心分離処理を行なつた後、透析処理によ
り、F因子プラスミドを含む溶液を分取した。
この溶液にエタノール沈殿を行ない、最終的に
約20μgのF因子プラスミドを採取した。
次に、miniF画分の調製に際して、上記のF
因子プラスミドを5μgとり、制限酵素である
EcoRIを37℃、1時間作用させて該F因子プラ
スミドDNA鎖を切断し、約9.1kbのminiF断片
を含むDNA溶液を調製した。
(B) フアージφ80ptの調製
大腸菌(E.coli)K−12株(IFO3301)を100
mlのL培地(組成は前記と同じ)に接種し、37
℃で約4時間振盪した培養物0.2mlと、フアー
ジφ80(ATCC11456a−B1)水溶液(105ケ/
ml)の0.1mlとを、L培地軟寒天(L培地+寒
天沫)中に混合したのち、L培地寒天プレート

37℃で約3時間振盪培養し、対数増殖期に25%
(w/v)グルコース溶液10mlと上記で調製し
たフアージφ80pt溶液を1011ケ/mlの濃度で添
加し(moi2.0)、5時間振盪を継続後常法通り
クロロホルムの添加により、フアージφ80ptを

衝液(PH7.8)にて透析後、フエノール法によ

精製し、これに制限酵素BamHIを与え30℃で
30分間反応させ、トリプトフアンオペロンを含
む遺伝子画分を得た。
(D) トリプトフアンオペロン画分とminiF画分の
結合
前記(A)で得たminiF画分に制限酵素BamHI
を30℃にて30分間反応させ、次いで、60℃で5
分間熱処理してBamHIを失活させた後、前記
(C)で得たトリプトフアンオペロン画分を添加混
合し、ATP(アデノシントリホスフエート)及
びジチオスレイトールを加え、さらにT4フア
ージ由来のDNAリガーゼ(DNA結合作用を有
する;宝酒造(株)製「T4DNAリガーゼ」)
を添加した後、12℃で17時間反応を行なつた。
次いで60℃で5分間熱処理して、DNAリガー
ゼを失活させた後、エタノール沈殿法にて、プ
ラスミドpMTY−1を採取した。
得られたプラスミドpMTY−1の分子量を、
アガロース・ゲル電気泳動法により測定した。
使用したアガロース・ゲル電気泳動装置は、マ
リソル産業(株)製フラツトアガロースゲル電
気泳動装置KS−8422FAE型であり、電気泳動
条件は、0.6%寒天ゲルを作製し、緩衝液
(0.04M Tris、0.04M酢酸ソーダ、0.008M
EDTA、酢酸でPH8.3に調整)中に製限酵素
(EcoRI、BamHI又はHind)を用いてプラ
スミドpMTY−1を分解させたDNA断片の混
合物を入れ、80V、4〜5時間電気泳動させ
た。
泳動後、寒天ゲルを取り出し、エチジウムブ
ロマイドにて染色後、254nmUV光で照射して
DNAの移動位置を検出し、分子量既知のDNA
断片(λDNAをHindで分解して得られた
DNA:和光純薬工業(株)製「λDNA−Hind
digests」)との比較により分子量を決定し
た。
参考例2:プラスミドpMTY−1による大腸菌
K−12系変異株の形質転換
(A) 宿主菌の準備
大腸菌(E.coli)K−12株(IFO3301)から
常法に従い、NTG(N−メチル−N′−ニトロ
−N−ニトログアニジン)処理によつてトリプ
トフアン要求性変異株を調製した〔E.A.
Adelberg et al.,Biochem,Biophys,Res.
Comm.,18,788(1965)参照〕。
すなわち、上記E.coli K−12株をL培地に
て対数増殖中期まで培養し、遠心分離により集
菌して菌株をTris−マレイン酸緩衝液(PH6.0)
に懸濁そた後、100μg/ml〜200μg/mlNTGを
添加し、15〜30分間処理する。該処理物を同じ
緩衝液にて洗浄後、L培地に添加し、37℃にて
振盪培養を行ない。該培養物は常法に従い、ペ
ニシリン濃縮法およびレプリカ法を併用してト
リプトフアン要求性変異株を単離した。
(B) 形質転換
上記(A)で得た大腸菌K−12系株(トリプトフ
アン要求性変異株)を、前記参考例1で用いた
と同じL培地10mlに接種し、37℃にて振盪培養
を行ない、対数増殖中期もしくは、後期まで生
育させた後遠心分離にて集菌し、これを0℃の
氷冷下に0.1M MgCl2および0.1MCaCl2の水溶
液に懸濁させてDNA受容状態菌(コンピーテ
ント菌)を調製した。これに前記参考例1で得
たプラスミドpMTY−1を加え、0℃の氷冷
下に40分間反応させ、次いで40℃にて3分間加
温処理した後、再度0℃の氷冷下にて2時間反
応させた。
次にL培地に上記処理菌体を接種し、37℃に
て1時間30分振盪培養後、遠心分離にて集菌
し、菌体を洗浄後、最少培地(Davis培地:K2
HPO47g、KH2PO42g、MgSO4・7H2O0.1
g、(NH4)2SO41g、グルコース2g、純水
1)にて調製した平板培地に塗抹し、37℃に
て培養し、生育したコロニーを再び同培地上で
純化した後、形質転換株(Trp要求性の消失、
すなわちプラスミド上のトリプトフアンオペロ
ンにより生合成可能となり、最少培地上にて生
育可能となつた菌株)を得た。この形質転換株
は茨城県筑波群谷田部町東1丁目1番3号の工
業技術院微生物工業技術研究所に、昭和58年7
月6日付で受託番号:微工研菌寄第7139号
(FERM P−7139)にて寄託されている。
参考例3:形質転換株の安定性
前述のL培地100mlを500ml容三角フラスコに分
注し、120℃で15分間滅菌処理したものに、参考
例2で得た形質転換株を植菌し、37℃にて24時間
振盪培養を行なつた後、同様にして調製したL培
地に2ml植継し、同じく37℃にて24時間振盪培養
を行ない、この操作を繰返し実施して計5回の培
養を行なつた。
各培養終了時(24時間培養時)に集菌し、菌体
を洗浄後、前記最少培地にL−トリプトフアン20
mg/を添加して調製した平板培地に一定量塗抹
し、37℃にて1日培養した。生育コロニー200ケ
(無差別選択)をし、トリプトフアンを含まない
最少培地に植菌し、37℃にて2日間培養後、再度
同様の最少培地に植菌し、生育コロニーをカウン
トする。
この結果、トリプトフアン添加培地に生育した
コロニーはすべてトリプトフアンを含まない最少
培地にも生育すること、すなわち該プラスミドの
高度な安定性を確認した。
なお。形質転換株中のプラスミド存在の確認試
験として、形質転換株のアクリジンオレンジ処理
法により、Trp要求性株が出現し、また、前記の
SDS−リゾチーム法及び塩化セシウム−エチジウ
ムブロマイド法により、プラスミド確認を併行し
て行ない、プラスミドの分子量に変化の無いこと
を確認した。

次に、上記と同じ組成のL培地100mlを容量500
mlの振盪フラスコに入れ120℃で15分間殺菌した
培養液に上記の前培養物2mlを植菌し、同時にイ
ンドールアクリル酸を下記第1表に示す量添加し
た後、37℃で7時間振盪培養した。
培養終了後、遠心分離によつて集菌し、該菌体
を、Yanofskyらの方法〔Method in Enzy−
mology,vol.5、794(1962)参照〕に準じて、0.2
g(湿菌体)はかりとり、100mMトリス(ヒド
ロキシメチルアミノメタン)緩衝液PH7.8に懸濁
し、超音波処理により菌体を破壊した。この溶液
を12000rpmで40分間遠心分離を行ない、上清を
粗酵素液とした。反応液組成としては、インドー
ル0.4μmole、DL−セリン80μmole、塩化ナトリ
ウム180μmole、ピリドキサール5リン酸
0.03μmole、トリス(ヒドロキシメチルアミノメ
タン)100μmole、組酵素液0.1ml、純水と1N塩酸
を加えて全容1ml(PH7.8)にしたものを使用す
る。反応は37℃で20分間行ない、2N水酸化ナト
リウム反応を停止させる。その後、トルエン4ml
を加え残残インドールを抽出し、その1mlを呈色
液(p−ジメチルベンズアルデヒド3.6%(w/
