酸ジエステル誘導体の2位にヒドロキシアルキル
基を有する化合物を合成する際には、保護された
ヒドロキシアルキル基を有する該ジヒドロピリジ
ン誘導体を経ることが必須であるか、または別途
合成された2−ホルミル−1,4−ジヒドロピリ
ジン誘導体のホルミル基を還元することによりヒ
ドロキシメチル基へと導く製造法が開示されてい
るのみである。即ち、従来の製造法においては2
−ヒドロキシアルキル−1,4−ジヒドロピリジ
ンを製造するには多段階の反応が必要とされ、か
つその製造収率は充分とはいえない。従つて原料
物質より単段階の反応で目的とする2−ヒドロキ
シアルキル−1,4−ジヒドロピリジン誘導体を
工業的に収率よく合成する方法が求められてい
る。 課題を解決するための手段 本発明者らは鋭意研究の結果、前記一般式
()で示される新規エナミン化合物および一般
式()で示されるアセチレン化合物を原料物質
として使用し、該化合物のヒドロキシル基を保護
することなく、2−ヒドロキシアルキル−1,4
−ジヒドロピリジン−3,5−ジカルボン酸ジエ
ステル誘導体()が収率よく合成できることを
発見し、本発明を完成するに至つた。 以下、本明細書中に記載された各種用語につい
て説明する。 低級アルキル基とは、炭素数1乃至6個からな
る直鎖状または分岐状のアルキル基を示し、例え
ばメチル、エチル、プロピル、イソプロピル、ブ
チル、イソブチル、第二級ブチル、第三級ブチ
ル、ペンチルおよびヘキシルなどが挙げられる。 低級ハロアルキル基とは、前記の低級アルキル
基にハロゲン原子が置換したハロアルキル基を示
し、例えばβ−クロロエチル、β−ブロモエチ
ル、β−クロロプロピル、γ−クロロプロピル、
ω−クロロブチルおよびトリフルオロメチルなど
が挙げられる。 低級アルケニル基とは、炭素数2乃至4個から
なるアルケニル基を示し、例えばビニル、アリ
ル、3−ブテニルおよびイソプロペニルなどが挙
げられる。 低級アルキニル基とは、炭素数2乃至4個から
なるアルキニル基を示し、例えばプロパルキルお
よび2−ブチニルなどが挙げられる。 アラルキル基としては、例えばベンジル、α−

は結果に悪影響を及ぼすことなく広範囲に変える
ことができる。 アンモニアまたはその塩としては、アンモニア
または酢酸、ギ酸、酒石酸、安息香酸もしくはフ
エニル酢酸などの有機酸のアンモニウム塩または
炭酸、重炭酸もしくはホウ酸などの無機酸のアン
モニウム塩などが使用される。 反応温度は通常冷却下、室温または加温乃至加
熱下で実施される。 反応溶媒としては水、不活性有機溶媒または水
と不活性有機溶媒との混合溶媒を用いることがで
きる。不活性有機溶媒としては、メタノール、エ
タノール、プロパノール、イソプロパノールもし

えん酸などの有機酸の付加塩とすることができ
る。 本発明に使用する一般式()で表わされるエ
ナミン化合物は文献未載の新規物質であり、次に
示す方法により製造される。 方法(イ)および(ロ)で使用する原料物質()は(A)
および(B)の反応式で示すように、保護オキシアル
キルβ−ケトエステル()または保護オキシア
ルキルプロピオール酸エステル()をアンモニ
アまたはその塩と反応させ、次いで所望により脱
保護処理を行うことによつて製造できる。 本原料化合物()は本発明者らに係る、特開
昭59−231049号公報に詳しく説明されている。 本発明により供される一般式()で示される
化合物は血管拡張作用および血圧降下作用を有
し、特に冠血管拡張作用が強くかつ毒性が極めて
弱いので高血圧症、心機能不全、狭心症、心筋梗
塞および脳血管障害などの循環器系疾病の治療に
有用であると期待される医薬品である。 薬 理 本発明の化合物()について薬効薬理試験お
