Description
〔産業上の利用分野〕
本発明は、インターフエロン−α(以下、IFN
−αという)の製造方法に関する。さらに詳しく
は、本発明はIFN−α産生細胞を、培地で培養し
てIFN−αを産生する方法の改良に関するもので
あり、IFN−αの産生を増強せんとするものであ
る。
〔従来技術〕
IFN−αは、一般にウイルスによつて誘発され
た白血球などの細胞から産生されるものであり、
ウイルスに起因する各種疾病に対して有用である
とされている。
ところが、IFN−αをウイルス性疾患の予防ま
たは治療に使用するためには、大量のIFN−αを
採取する必要がある。
従来、IFN−αの大量産生方法としては、培養
培地中に、カルボン酸、多価アルコールあるいは
グルココルチコイドを添加する方法などが提案さ
れている。
しかしながら、かかる方法においても満足する
に足る産生量が得られないのが実情である。
〔発明が解決しようとする問題点〕
従つて本発明の目的は、IFN−αの産生を増強
せしめることである。
〔発明の構成〕
かかる観点から、本発明者らは種々研究を重ね
てきたとこら、IFN−α産生細胞を誘発前に低級
アルコールと接触させることによつてIFN−αの
産生量が増強されることを見いだした。
本発明はかかる新知見に基づいて完成されたも
のであり、その要旨は、IFN−α産生細胞を、培
地で培養してIFN−αを産生させる方法におい
て、誘発前に当該IFN−α産生細胞を低級アルコ
ールと接触させることを特徴とするIFN−αの製
造方法に関する。
本発明にて作用されるIFN−α産生細胞は、
IFN−αを産生しうるものであれば特に制限はな
く、たとえば培養リンパ芽球様細胞などがあげら
れる。好ましいIFN−α産生細胞としては、産生
能の観点から、培養リンパ芽球様細胞があげられ
る。培養リンパ芽様球細胞としては、特にナマル
バ株が好ましい。ナマルバ株は、20種のバーキツ
トリンパ腫および白血病患者の株化リンパ球の中
から選択された株化細胞であり、現在この細胞は
IFN−αを最もよく産出するとされている
〔Inter.J.Cancer、11、327(1973)、J.Clinical
Microbiol.、1、1(1975)〕。
IFN−α産生細胞と低級アルコールとの接触
は、IFN−αの誘発前に行われ、通常、その接触
は、低級アルコールを添加した培地中で培養する
ことによつて行われ、IFN−α誘発時には当該低
級アルコールは除去されるが、誘発時に存在して
も差し支えはない。また、培地交換時に低級アル
コール含有液〔溶培としては、後述する培地の
他、PBS(−)、ハンクス液が例示される。〕と一
過性に接触せしめることによつても行われる。
培地に低級アルコールを添加して接触をおこな
う場合、その培地としてはRPMI1640培地が一般
的であるが、Eagle′sMEM培地、DEM培地、
Ham′sF−12培地、RITC56−2培地などが使用
可能であり、好ましくは牛新生児血清を0.1〜15
(v/v)%、更に好ましくは、10(v/v)%程
度添加したRPMI1640培地、Eagle′s MEM培地、
DEM培地、Ham′sF−12培地、ヒト・アルブミ
ンを0.1〜1%(w/v)程度添加したRITC56−
培地などが例示される。
低級アルコールとの接触時の培養条件は、例え
ば次の通りである:
通常細胞濃度5〜6×105cell/ml程度、培養
温度は30〜40℃、好ましくは37℃程度、培養時間
は1〜4日、好ましくは2〜3日程度である。こ
の際低級アルコールの最終濃度は、IFN−αの産
生を増強するに十分な量であればよく、それは通
常0.1〜3{v/v)%程度、好ましくは0.1〜1
(v/v)%程度である。一般に低級アルコール
の濃度の上昇に応じてIFN−α産生量は上昇する
が、3(v/v)%をこえるとアルコールの毒性
によつて逆にIFN−α産生量は低下する。
低級アルコールとしてはメタノール、エタノー
ル、n−プロパノールなどが例示されるが、エタ
ノールが特に好ましい。
また、アルコールに加えてグルココルチコイド
(例、デキサメサゾン、ハイドロコルチゾン等)
を添加することが好ましい。その際、低級アルコ
ール〔最終濃度0.1〜3(v/v)%〕を添加する
と同時にグルココルチコイドを最終濃度10-7〜
10-4M程度となるように添加すればよい。
本発明による低級アルコールとの接触処理を受
けたIFN−α産生性細胞は常法によつて誘発せし
め(たとえば、センダイウイルスなどのウイルス
による誘発)、斯くして当該細胞から産生される
目的とするIFN−αを分離・採取すればよい。そ
の際の培養条件としては、培地は前記と同様のも
のを使用すればよく、細胞濃度としては、たとえ
ば2×106cell/ml程度が好ましく、誘発剤は、
たとえばセンダイウイルスの場合100HAU/ml
程度が好ましい。培養時間は20〜24時間程度、培
養温度は25〜37℃程度である。
本発明によるIFN−α産生方法の一例の概念を
示せば次の通りである:
細胞濃度を5〜6×105細胞/mlとし、10(v/
v)%の割合に牛新生児血清を添加した
RPMI1640培地を用いて37℃程度で培養を開始す
る。
培養2〜3日目に低級アルコールを0.1〜3
(=/v)%程度添加し、さらに37℃程度で48〜
72時間培養を行う。
その後、培地を新鮮なRPMI1640培地に置き換
え、20×105細胞/mlに調整し、センダイウイル
スを最終100HAU/ml程度となるように添加し
37℃で20〜24時間IFN−αを誘発し、上清のIFN
−α力価をFL細胞−Sindbisウイルスの組合せに
よる細胞変性抑制法によつて測定する。
〔効果〕
本発明によれば、IFN−α産生細胞を培養して
IFN−αを産生させるに際して、IFN−αの産生
性が顕著に増され、IFN−αの工業的製造に有用
である。
実施例 1
エチルアルコール添加効果
Namalva細胞より、IFN−αを特に高率で産
生する株として分離・樹立した株であるGC8201
株を、10(v/v)%牛新生児血清加RPMI1640
培地を用い、250mlスピンナーフラスコ中で培養
する。培養開始後3日目細胞濃度が約150×104細
胞/mlとなつた時、エチルアルコールを最終濃度
0.1〜3(v/v)%となるように各々のスピンナ
ーフラスコに添加し、さらに48時間培養する。こ
の後遠心(1500rpm、5分間)により細胞を分
離、RPMI1640培地を用い、細胞沈渣を懸濁、細
胞濃度200×104細胞/mlとなるように調整する。
次ぎにIFN−αを誘導するためにセンダイウイル
スを最終100HAU/mlとなるように添加する。
これを37℃で20時間インキユベートし、遠心
(3000rpm、5分間)後の上清に含まれるIFN−
α力価を測定する。得られた結果は第1表の通り
である。
