[Technical Field] The present invention relates to injectable preparations. [Prior art] Aspoxycycillin [Chemical name: (2R, 5R, 6R)
-6-{(2R)-2-[(2R)-2amino-3-(N
-methylcarbamoyl)propionamide]-2
-(p-hydroxyphenyl)acetamide}-3,
3-dimethyl-7-oxo-4-thia-1-azabicyclo[3.2.0]peptane-2-carboxylic acid]
is an excellent medicinal compound that has strong antibacterial activity against both Gram-positive and Gram-negative bacteria (Special Publication No. 43519/1983).A crude aspoxycillin-containing aqueous solution is purified using a non-polar porous adsorption resin and then freeze-dried. It is known that a highly pure powder can be obtained by doing so (Japanese Patent Application Laid-open No. 40686/1986). [Technical issues to be solved] However, aspoxycillin obtained by the method described in JP-A No. 56-40686 is an amorphous anhydride, has hygroscopic properties, and cannot be stored under conditions without moisture protection or light shielding. At times, it was difficult to say that it was sufficiently stable. In addition, this aspoxycillin is a new trihydrate crystal that has excellent properties as a raw material for pharmaceutical preparations, such as excellent stability over time, low crystal bulk density, and low chargeability when crystallized from an aqueous solution under weak acidity. This aspoxycillin 3
Hydrate has low solubility in water (2.8g/100
ml water, 0°C), it is difficult to use it as an injection preparation as it is. In other words, from the viewpoint of storage stability, it is desirable for injectable preparations to be in a solid state during storage and in a solution state during administration, but aspoxycillin trihydrate has low solubility as mentioned above, making it difficult to administer it to patients as is. In this case, a large amount of solvent is required, and it is unavoidable that administration takes a long time or that the patient is unnecessarily burdened. [Configuration and Effects of the Invention] The present invention relates to an injectable preparation containing aspoxycillin trihydrate and a pharmacologically acceptable basic substance. The above formulation has succeeded in using aspoxycillin trihydrate, which was difficult to use as an injection formulation due to its low solubility in water, as an injection formulation. For example, 1 mol of aspoxycillin trihydrate. About 0.4% of basic substances
~ When using 1 mole, aspoxycillin is about 5 ~
A solution containing 30 w/v% can be easily prepared. Furthermore, it has the excellent feature that a solution containing a desired amount of aspoxycillin can be obtained by appropriately adjusting the amount of basic substance. Examples of the pharmacologically acceptable basic substances (hereinafter simply referred to as basic substances) used together with aspoxycillin trihydrate in the injectable preparation of the present invention include alkali metal carbonates such as potassium carbonate and sodium carbonate. , lysine, arginine,
Examples include basic amino acids such as ornithine, and trishydroxymethylaminomethane. In the injectable preparation of the present invention, the ratio of aspoxycillin trihydrate and basic substance is as follows:
Most preferably about 0.4 to 1 mole, especially about 0.6 to 0.9 mole of basic material per mole of trihydrate. The injectable preparation of the present invention may be a single preparation in which aspoxycillin trihydrate and a basic substance are formulated together in crystal or powder form, or aspoxycillin trihydrate and a basic substance may be formulated together into a single preparation. It may also be a unit preparation in which each substance is formulated separately. These preparations can be produced by conventional methods,
For example, in the case of a single preparation, aspoxycillin trihydrate and a basic substance can be mixed in the form of crystals or powder, respectively, and this mixture is filled into a vial and the like and the stopper is closed. In the case of a unit preparation, aspoxycillin trihydrate and the basic substance are each filled as crystals or powder into vials separately, or aspoxycillin trihydrate is filled as crystals or powder into vials, etc. A basic substance can be prepared as a unit preparation by filling a vial as an aqueous solution and then sealing the vial to form a set of preparations containing each component. The thus obtained injectable preparation of the present invention dissolves rapidly to form a highly concentrated aspoxycillin solution as described above, and by appropriately adjusting the amount of basic substance, a solution containing the desired amount of aspoxycillin can be obtained. It has the excellent characteristics of being able to be used in a variety of ways, and also has good storage stability. In addition, aspoxycillin trihydrate crystals are prepared by, for example, aspoxycillin or its salt in an aqueous solvent (water,
water-containing lower alkanol, water-containing lower alkanone, etc.)
It can be produced by crystallizing it under weakly acidic conditions (PH 3 to 6). Hereinafter, the present invention will be explained in more detail with reference to Examples. Example 1 100 parts by weight of aspoxycillin trihydrate converted to aspoxycillin anhydride and sodium carbonate
Mix 17 parts by weight (0.79 molar ratio to aspoxycillin), fill this mixture into 10 ml vials so that each vial contains 1 g of aspoxycillin anhydride, and close the caps to prepare an injectable preparation. obtain. When 5 ml of distilled water for injection was added to this injection preparation, the contents of the vial were quickly dissolved and 20w/
The result is a clear injection solution (PH approximately 7.5) containing v% aspoxycillin. Example 2 100 parts by weight of aspoxycillin trihydrate converted to aspoxycillin anhydride and 21.3 parts of arginine
parts by weight (0.6 molar ratio to aspoxycillin), and add 25 ml of this mixture to 1 g of aspoxycillin anhydride per vial.
An injection preparation is obtained by filling a vial and capping it. When 20ml of distilled water for injection was added to this injection preparation, the contents of the vial were quickly dissolved and 5w/
The result is a clear injection solution (PH approximately 7.0) containing v% aspoxycillin. Example 3 1 g of aspoxycillin trihydrate as anhydrous aspoxycillin and 193 mg of sodium carbonate are filled into separate 10 ml vials, and the vials are capped to obtain an injectable preparation. 5 ml of distilled water for injection in a vial of sodium carbonate
was added to obtain an aqueous sodium carbonate solution, and when this aqueous solution was added to a vial of aspoxycillin trihydrate, the aspoxycillin trihydrate was rapidly dissolved to form a clear injection solution containing 20 w/v% aspoxycillin. (PH approximately 7.6). Examples 4 to 5 An injectable preparation of aspoxycillin is obtained in the same manner as in Example 1 except that 17 parts by weight of sodium carbonate is replaced with a basic substance in the amount shown in the table below. When 5 ml of distilled water for injection was added to these injection preparations, the contents of the vial were rapidly dissolved to form a clear injection solution containing aspoxycillin at the concentration shown in the table.
