(産業上の利用分野)
本発明は、4−フルオロフエノール
【式】を出発原料とし、これにアク
リロニトリルを第三級アミン存在下で反応させる
ことにより生成する式()
で表わされる3−(4−フルオロフエノキシ)プ
ロピオニトリルを合成中間体として用いる、式
()
で表わされる6−フルオロ−4−クロマノンの製
造方法に関するものである。
6−フルオロ−4−クロマノン()は、強い
アルド−クリダクターゼ阻害活性を有し、糖尿病
合併症の新規治療薬として期待されている(S)−
2,3−ジヒドロ−6−フルオロ−スピロ〔4H
−1−ベンゾピラン−4,4′−イミダゾリシン〕
−2′,5′−ジオン(VSAN:ソルビニル
(sorbinil));
式()
の合成中間体として重要な化合物であり、シアン
化ナトリウム及び炭酸アンモニウム存在下加熱反
応する、うわゆるBucherer合成により容易に
(RS)−2,3−ジヒドロ−6−フルオロースピ
ロ〔4H−1−ベンゾピラン−4,4′イミダゾリ
ジン〕−2′,5′−ジオンに誘導することができる。
(従来の技術)
従来、6−フルオロ−4−クロマノン()の
製造法としては、次式に示す如く、4−フルオロ
フエノールと3−クロロプロピオン酸とを、水酸
化ナトリウム存在下、いわゆるWilliamson合成
して3−(4−フルオロフエノキシ)プロピオン
酸()を形成せしめた後、このものをポリリン
酸中で加温することにより6−フルオロ−4−ク
ロマノン()を製造する方法が用いられている
(米国特許第4117230号、同第4130714号)。
しかしながら、この方法では4−フルオロフエ
ノールからの3−(4−フルオロフエノキシ)プ
ロピオン酸()の合成において、3−クロロプ
ロピオン酸の3−ヒドロキシプロピオン酸への分
解副反応が優先するため、エーテル結合形成が妨
げられ、大過剰の3−クロルプロピオン酸を用い
てもなお、化合物()の合成収率は極めて低く
〔J.Amer,Chem.Soc.、81、94(1959)〕、高価な
4−フルオロフエノールを出発原料として使用す
ることを考慮すれば、6−フルオロ−4−クロマ
ノン()の工業的製造法としては必ずしも有効
な方法とは言い難い。
(発明が解決しようとする課題)
そこで本発明者らは、経済的で、操作性及び安
全性に優れた6−フルオロ−4−クロマノン
()の工業的な新規製造法を確立すべく鋭意検
討した結果、4−フルオロフエノールにアクリロ
ニトリルを触媒として第3級アミンの存在下で作
用させる、いわゆるシアノエチル化反応によりエ
ーテル結合を効果的に形成せしめて、3−(4−
フルオロフエノキシ)プロピオニトリル()を
得、ついでこれを処理して3−(4−フルオロフ
エノキシ)プロピオン酸()とした後、これを
硫酸中、脱水閉環反応することにより効率的に6
−フルオロ−4−クロマノン()が合成できる
ことを見い出し、本発明を完成した。
(課題を解決するための手段及び作用効果)
本発明方法を反応式で示すと下記の如く表わす
ことができる。
すなわち、本発明は、4−フルオロフエノール
とアクリロニトリルを触媒存在下に反応させて得
られる3−(4−フルオロフエノキシ)プロピオ
ニトリル()を酸加水分解し、得られる3−
(4−フルオロフエノキシ)プロピオン酸()
を硫酸の存在下閉環反応ち付することを特徴とす
る6−フルオロ−4−クロマノン()の製造方
法に関するものである。
従来、フエノール類とアクリロニトリルとのシ
アノエチル化反応は、トリトンB(水酸化トリメ
チルベンジルアンモニウム)、金属ナトリウム、
ナトリウムメトキシドなどの強塩基存在下〔J.
Chem.Soc.、920(1945);F.Amer、Chem.Soc.、
70,599(1948);Bull.Soc.chim.France1288
(1957)など〕や銅化合物存在下(米国特許第
2974160号)あるいは、無水塩化アルミニウム−
乾燥HCI存在下に行なう〔J.Org.Chem.、22、=
1264(1957)〕などの種々の方法が用いられてい
た。
本発明者らは、4−フルオロフエノールとアク
リロニトリルとのシアノエチル化反応に有効な触
媒の探索を、上記触媒を含め、種々の化合物につ
いて検討した結果、銅化合物のなかでも水酸化第
2銅や酢酸銅一水和物などが触媒として優れてい
ることを見い出すとともに、従来報告例の無い有
機アミンとりわけトリエチルアミンの如き第3級
アミンが本反応に特異的に有効な触媒となり得る
ことを見い出し、更に、こうして得られる3−
(4−フルオロフエノキシ)プロピオニトリル
()の6−フルオロ−4−クロマノン()合
成への効率的な利用方法を確立した。以下に本発
明を具体的に説明する。
本発明方法を実施するにあたり、まず、4−フ
ルオロフエノールとアクリロニトリルを適当な鵜
触媒の存在下、シアノエチル化反応させエーテル
結合を形成せしめる。触媒としては、水酸化第2
銅、酢酸銅、酢酸銅一水和物、炭酸銅、酸化第2
銅などの銅化合物及び大粉末、あるいはトリメチ
ルアミン、トリエチルアミン、トリ−n−ブチル
アミンヘキシルアミン、トリ−n−オクチルアミ
ン、ジメチルシクロなどの第3級アミン、更には
水酸化カルシウム、水酸化バリウム、水酸化亜鉛
などが有効であるが、反応収率、反応後の触除去
など後処理の操作性及び経済性を考慮すれば、工
業的規模の生産においてはトリエチルアミンの如
き第3級アミンの使用が望ましい。
本反応の条件は、使用する触媒の種類により変
化するので一律に規定することはできないが、一
般に触媒は4−フルオロフエノールに対して0.01
倍モル量以上、好ましくは0.1〜0.5倍モル量程度
使用する。またアクリロニトリルは、4−フルオ
ロフエノールに対して過剰量用いる方が好結果を
与えるが、通常2〜3倍モル量で充分である。反
応は通常アクリロニトリルを溶媒を兼ねて使用す
るが、必要とあらばベンゼン、トルエンなどの非
極性溶媒や水、エタノールなどの極性溶媒を使用
することもできる。反応温度はアクリロニトリル
を溶媒とする場合、環流温度程度が望ましく、こ
の場合、反応は6〜16時間でほぼ完結するが、環
流温度以下であると反応速度が低下し、反応完結
までには更に長時間必要となる。反応終了後、使
用した過剰のアクリロニトリル、溶媒、第3級ア
ミンならびに若干の未反応の4−フルオロフエノ
ールは減圧蒸留で除くことができるが、固体触媒
を使用した場合は、それを別するなどの操作が
必要となる。このようにして得られた3−(4−
フルオロフエノキシ)プロピオニトリル()
は、そのまま次の反応に使用することもできる
が、更に適当な濃度の水酸化ナトリウム水溶液で
洗浄するなどすれば、ほぼ純粋な3−(4−フル
オロフエノキシ)プロピオニトリル()を高収
率で単離することも可能である。
3−(4−フルオロフエノール)プロピオニト
リル()を6−フルオロ−4−クロマノン
()に誘導するには、3−(4−フルオロフエノ
キシ)プロピオニトリル()を酸加水分解し
て、まず3−(4−フルオロフエノキシ)プロピ
オン酸()へ導く。この際、使用しうる酸とし
ては、塩酸、臭化水素酸、硫酸などを挙げること
ができる。加水分解の反応温度を増すため高濃度
の臭化水素酸や硫酸を用いると、それぞれエーテ
ル結合の開裂、芳香環のスルホン化など好ましく
ない副反応を伴う傾向があるが、塩酸の場合は、
こうした副反応は全く認められない。反応速度お
よび反応収率を考慮すれば、濃塩酸を使用するの
が好ましく、この場合、化合物()を濃塩酸中
約10時間還流することにより定量的な収率で化合
物()が得られる。
次に、3−(4−フルオロフエノキシ)プロピ
オン酸()の6−フルオロ−4−クロマノン
()への環化誘導は、化合物()をポリリン
酸中、100℃にて10分間攪拌して化合物()を
得る公知の方法(米国特許第4117230号、同第
4130714号)や化合物()を濃硫酸中、室温で
1時間攪拌するなどの方法により良好な収率で行
なうことができる。これら酸加水分解および閉環
反応の条件を整えれば、ほぼ定量的な収率で、3
−(4−フルオロフエノキシ)プロピオニトリル
()から6−フルオロ−4−クロマノン()
を得ることも可能である。
(実施例)
以下に実施例をあげて本発明を説明する。本発
明は、もとよりこれに限定されるものではない。
実施例 1
4−フルオロフエノール11.2g、アクリロニト
リル10.6g、トリエチルアミン2.02gからなる混
合液を攪拌しつつ16時間還流した後、減圧下でア
クリロニトリル、トリエチルアミン、4−フルオ
ロフエノールを除いた。得られた残物をエーテル
で希釈し、10%水酸化ナトリウム水溶液、ついで
2N塩酸で洗浄し、無水硫酸マグネシウムで脱水
した。エーテルを留去すると3−(4−フルオロ
フエノキシ)プロピオニトリルの無色液体11.51
gを得た(収率70%)。
実施例 2
4−フルオロフエノール1.12g、アクリロニト
リル2.65g及び触媒の混合物を、アクリロニトリ
ルの還流温度にて10時間攪拌下反応した。反応
後、3−(4−フルオロフエノキシ)プロピオニ
トリルの4−フルオロフエノールに対する生成収
率をGLCにて分析し、以下の結果を得た。

実施例 3
3−(4−フルオロフエノール)プロピオニト
リル45.50gと12N塩酸の混合液を約120℃にて10
時間攪拌した。冷後、酢酸エチル−水に分配した
後、酢酸エチル層を分離し、無水硫酸マグネシウ
ムで脱水した。酢酸エチルを留去すると3−(4
−フルオロフエノキシ)プロピオン酸の白色結晶
50.70gが得られた。
mp 84〜85℃(文献値86℃)
なお、IR、1H MNRは4−フルオロフエノー
ルと3−クロロプロピオン酸から、公知の方法で
合成した標品と一致した。
実施例 4
3−(4−フルオロフエノキシ)プロピオン酸
30.0g(163.0mmol)と濃硫酸180mlの混合物を
室温で1時間攪拌した後、氷700g中に注ぎ入れ
ると直ちに白色結晶が析出した。この白色結晶を
取、水洗した後、空気乾燥した。これをエタノ
ールから再結晶し、6−フルオロ−4−クロマノ
ンの白色結晶20.55gを得た(収率76%)。
