Description

【発明の詳細な説明】[Detailed description of the invention]

(産業上の利用分野) 本発明は食品の風味を改良する動物のアキレス
腱を原料とした天然蛋白調味料の製造方法に関す
る。 (従来の技術) 従来畜肉、魚介類などから可溶性成分を抽出し
て製造した畜肉魚介類のエキス類は、化学調味料
では得られない複雑な呈味成分を多量に含有して
おり、呈味力の強い所謂うま味付与型の天然調味
料として一般に広く利用されている。 このようなエキス類の製造方法としては、原料
を常圧あるいは加圧下で熱水抽出し、残査部分と
エキス部分とに分離し、エキス部分を濃縮あるい
は乾燥して調味料とする方法や、家畜肉缶詰を製
造するときの煮汁を濃縮あるいは乾燥して調味料
とする方法、或いは大豆蛋白などを酵素及び/又
は酸等で加水分解して調味料とする方法等により
製造されていた。 これらの各方法に於て得られるエキス類はいず
れも各種アミノ酸、ペプチド、リボ核酸などのう
ま味成分を豊富に含み、コク味のあるうま味付与
型の天然調味料である。 ところが近年組立て食品などの加工食品の発達
に伴つて天然調味料が重要な食品素材の1つとな
つて来ており、しかも単にうま味を付与するのみ
でなく、それ自身は味がなくしかも食品に添加使
用時に豊かにコク味を与える天然調味料である所
謂風味向上型の天然調味料が必要となつて来てい
る。 上記した需要に応えるべく、種々の努力がなさ
れてきており、例えば、動物性蛋白質ないしは植
物蛋白質を分解した後分解液を限外濾過して得ら
れる分子量が20000より大きく且つ100000以下の
ポリペプチドを飲食品に添加し、飲食品の風味を
改良する方法(特開昭61−141858号公報)、また
動物性油脂、肉エキス、コラーゲン分解物、糖
類、醤油、野菜類を混合し、該混合物中の水分含
量を3〜30重量%に調整後90゜〜190℃で加熱する
食品風味改良剤の製造方法(特公昭58−46813号
公報)等が提案されている。 (発明が解決しようとする問題) しかしながら、上記特開昭61−141858号公報の
方法により得られる天然調味料は、調味力が弱
く、後記比較例に記載のように本発明により得ら
れる天然蛋白調味料と同等の調味効果を発揮する
のに、4倍乃至5倍量の添加を必要とするばかり
でなく製造コストが高く、且つ連続使用時に目詰
りによる濾過膜の性能低下が問題となる限外濾過
を必要とする欠点がある。 また特公昭56−46813号の方法では、風味改良
剤を製造するのに、コラーゲンの加水分解物を使
用しているが、肉エキス、糖類、野菜類と共に混
合した後90゜〜190℃で加熱することを主旨とする
ものであり、呈味の主体は肉エキス、野菜類であ
り、コラーゲンの加水分解物の果す役割効果につ
いては何等明確に記載していない。 (問題を解決するための手段) 本発明は上記の如き従来の問題にかんがみ、食
品の風味を改良し、豊かなコク味を与える風味向
上型の天然調味料の開発を目的としてなされたも
のである。かかる目的を達成するための本発明の
要旨とする構成は、動物のアキレス腱を水を添加
せずに蒸煮し、次いでその蒸煮したアキレス腱を
細断した後蛋白分解酵素を作用させて分解し、更
に酵素を加熱失活させた後に未分解物、異物を篩
別除去し、得られた蛋白分解液を均質化処理する
ことを特徴する天然蛋白調味料の製造方法に存す
る。 (作用) 動物のアキレス腱を蒸煮処理し、次いで蛋白分
解酵素で分解処理した後均質化処理する方法によ
り得られる天然調味料は、それ自身には殆ど呈味
力がないにもかかわらず、食品に少量添加した場
合に今迄の天然調味料では得られない豊かなコク
味を与え、香辛料、塩味などのカドをとり、風味
を改良する風味向上型の作用をなす。 (実施例) 次に本発明を実施例に基づいて詳細に説明す
る。 本発明で原料として使用するアキレス腱は牛、
豚、羊、馬など四足動物のもの等の、いずれでも
良く、衛生的に採取したものを使用する。通常ア
キレス腱は水分60〜70重量%、蛋白質20〜25重量
%、油分3重量%程度含有し、蛋白質の中の80〜
90%がコラーゲンであり、蛋白質含量の高い素材
である。就中牛のアキレス腱は流通が整つてお
り、安定して供給を受けられるので、本発明で使
用する原料として極めて好適である。 本発明では、まず第一工程として動物のアキレ
ス腱を蒸煮する。 この蒸煮方法としてはアキレス腱を容器中に入
れてレトルト内に収容し、アキレス腱の中心温度
を100゜〜120℃とし、30分間以上蒸煮する。即ち
通常はレトルト内温度を100゜〜120℃、好ましく
は115゜〜120℃とし1.5〜3.0時間好ましくは2時間
程度蒸煮する。 つまりレトルト内で湿熱により加熱を行うこと
により、水を添加することなく加熱でき、加えて
加圧することにより蒸煮時間が極めて短縮される
という利点がある。この時アキレス腱中に含まれ
る蛋白であるコラーゲンは加熱変性して水溶性の
ゼラチンとなり、他の蛋白は不溶性となる。しか
しながら不溶性となる蛋白はごく少量であり、実
際には収率的に余り問題とならない。この場合蒸
煮の温度条件が上記範囲以下の場合にはコラーゲ
ンが十分にゼラチン化せず、未変性コラーゲン等
の存在により蒸煮後のアキレス腱は非常にかたい
ものとなり、蒸煮後の細断及び酵素分解処理が上
首尾に行われない。次いで蒸煮によりコラーゲが
加熱変性してゼラチン化したアキレス腱を細断す
る。この細断にはミートチヨツパー、スライサ
ー、ボーンチヨツパー等のいずれを使用してもよ
く、後工程の酵素反応のために径5mm以下に細断
するのが好ましい。細断したアキレス腱を次いで
蛋白分解酵素で加水分解する。この蛋白分解酵素
による加水分解は中性付近で行うため、酵素の至
適温度があまり低いものは腐敗を防止する見地よ
り好ましくない。蛋白分解酵素の至適温度は細菌
繁殖を考慮して40゜〜70℃のものを使用するのが
好ましい。この蛋白分解酵素としてはパパイン
(例えば「プロナーゼ」、「ビオプラーゼ」として
市販のもの)、プロテアーゼ(例えば「アマノA」
として市販のもの)、フイシン、プロメリン(例
えば「タシナーゼ」として市販のもの)等の一種
或いは二種以上を使用する。蛋白分解処理は得ら
れる分解物が30℃で東京計器製B型粘度計No.2ス
ピンドル、20rpmで測定時の粘度が200〜600cps
になるように実施することが必要である。(試験
例の第1表参照) 酵素分解物の粘度が600cpsを越える場合には未
分解物が多く、収率面でよくなく又ほとんど調味
効果がない。また粘度が200cps未満の場合には分
解が進みすぎており苦味が生じ、且つ後工程で均
質化処理した後に均質化状態が壊れて油分が分離
してしまうという欠点がある。酵素の添加量は上
記粘度の分解物が得られる範囲内であれば良いの
であるが、操作性の観点より蒸煮後細断したアキ
レス腱に対して0.1〜0.5重量%とするのが好まし
い。例えば、細断した加熱処理アキレス腱にパパ
イン(天野製薬製)を0.1重量%添加し次いで60
℃で30分間分解する。次いでこの分解液を95゜〜
100℃で15〜20分間加熱してパパインを失活させ
る。次に得られた分解物を40〜60メツシユ(JIS
規格)の篩で篩別し、未分解物、異物等を除去す
る。次いで、PHを通過した分解液を均質化処理す
る。この均質化処理はアキレス腱に含まれる油分
を乳化しなお且つ分解液中に含有される粒子を細
かくすることを意図して行う。まずデイスパーミ
ル、マイコロイダー、ホモミキサー等通常使用さ
れる均質化機のうち一種或いは二種以上にて予備
均質化処理を行い、次いで高圧ホモゲナイザーを
使用して150〜200Kg/cm2の加圧下で均質化するの
が好ましい。この様にして均質化された分解液は
それ自身はほとんど呈味力はないものの、食品に
少量添加時に風味を著しく改良し、豊かなコク味
を付与するすぐれたものであつた。 代表的な応用例としては、例えばスープ類にお
いては0.3〜0.5重量%、煮物類においては1〜2
重量%添加時に好結果が得られた。なお上記の様
にして得られる均質化処理分解液は、必要により
更に濃縮し、濃縮エキスにすることもできる。 この場合、真空度45〜60mmHg、品温50℃〜70
℃の条件下で減圧濃縮することにより、固形分50
重量%のペースト状の天然蛋白調味料が得られ
る。このペースト状の天然蛋白調味料は常温で6
ケ月間の保存試験時に、何等品質に影響はなかつ
た。またこのペースト状天然蛋白調味料は油分を
含有しているため、通常、油の酸化が問題となる
のであるが、63℃のオーブンテスト時に酸化して
2ケ月間安定であることが判明した。 上記の方法で得られたアキレス腱分解ペースト
状調味液は、必要により更に乾燥して粉末化する
ことも可能である。この乾燥方法としては従来知
られている凍結乾燥、噴霧乾燥、真空乾燥等いず
れの乾燥方法でも良く、この場合デキストリン等
の賦形剤を適宜量添加して粉末化しても良い。こ
のようにして得られた粉末状調味料は、上記ペー
スト状天然蛋白調味料と同様に、常温で6ケ月間
密封保存した場合にも何等品質に影響はなかつ
た。また63℃のオーブンテスト時にも、油分の酸
化がなく2ケ月間安定であつた。 試験例 後記製造例1に記載の製造方法に従い、細断し
た加熱処理牛アキレス腱を調製した。蛋白分解酵
素としてはパパインW−40(天野製薬)、ビオプラ
ーゼSP−4(長瀬産業)、プロテアーゼ「アマノ
A」(天野製薬)、プロナーゼAS(科研化学)、プ
ロメリンF(天野製薬)を使用した。 上記加熱処理牛アキレス腱500gを1容ステ
ンレス製ビーカー内に収容し、品温を上記各酵素
の至適温度に調整した。 次いで各酵素0.4重量%を添加し、時々撹拌し
ながら酵素分解を2時間続行した。酵素添加時よ
り20分経過毎に、各試料を採取した後、95℃で10
分加熱失活処理した。 この酵素分解液は粘度測定後、官能試験に供し
た後、後記製造例1に記載の方法に従い濃縮して
分離試験に供した。 粘度は東京計器製B型粘度計を使いNo.2スピン
ドルで20rpm品温30℃で測定した。このアキレス
腱濃縮品100gを缶詰として室温で2ケ月間静置
した後、分離の有無を調べた結果を下記第1表に
示す。

わかつた。 比較例 後記製造例1に記載の製造方法に従い、アキレ
ス腱調味料を調製して本発明品とした。 一方、対照品は、特開昭61−141858号公報に記
載されている方法に従い、下記のようにして調製
された。 即ち、5%ゼラチン溶液2に蛋白分解酵素
(ビオプラーゼSP−4)0.2gを添加して溶解さ
せ、この溶液のPHを7.2に調整した後に、50℃に
おいて1時間緩やかに撹拌することによりゼラチ
ンを分解させた。 分解終了後、80℃で、5分間加熱して酵素を失
活させて分解調味液を得た。この分解液をアミコ
ン社のホローフアイバーシステムDC4型により限
外濾過して分子量20000〜100000の画分を分取し、
凍結乾燥して約30gの粉末を得、該粉末を対照品
とした。 上記両試料を製造例1のおでんスープに添加溶
解し、専門家パネル30人による官能試験を行つ
た。即ち、本発明により得られる分解調味液の

較した。 判定はコク味があり風味良いものを1位としク
レーマーの検定法に従い危険率5%で実施した。
上記官能試験の結果を下記第3表に示す。

ス腱分解調味液は、特開昭61−141858号公報記載
の限外濾過法により製造したポリペプチドに比
べ、添加量が1/4乃至1/5量で同等の調味効
果を有することがわかつた。 製造例 1 コラーゲン含量22重量%、水分含量61重量%の
生の牛アキレス腱市販品40Kgを、10Kgづつ20容
ステンレス製容器に入れ、レトルト内に収容して
レトルト温度115℃で2時間加圧蒸煮した。この
ときのアキレス権の中心温度は110℃であつた。
次いで圧縮空気により加圧冷却を行いながらレト
ルト内温度を100℃以下にした後、柔軟化した蒸
煮アキレス腱をステンレス製品容器内より取り出
し、増幸産業製ボーンチヨツパーMKBC32型を
用いて孔径5mmにて細断し、そぼろ状アキレス腱
39.5Kgを得た。 次いで得られたそぼろ状アキレス腱を梶原工業
製真空レオニーダーKHU−1に投入し、品温60
℃まで冷却した後、天野製薬製パパインW−40、
40gを200mlの水に溶解したものを添加し、撹拌
翼を24rpmで撹拌させながら60℃で30分間保持し
て加水分解し液状のアキレス腱分解物39Kgを得
た。 得られたアキレス腱分解物39Kgを、95℃で15分
間保持しパパインを失活させた。次いで上記アキ
レス腱分解物38.4Kgを、クリアランス0.3mmのデ
イスパーミルに通し摩砕した。摩砕したアキレス
腱分解物を50メツシユのバケツト型遠心分離機
(2000rpm)で濾過した後品温70℃の濾過分解物
を、深尾精機製作所製高圧ホモゲナイザーを用い
て150Kg/cm2の加圧下で均質化処理し、均質化し
たアキレス腱エキス37Kgを得た。 得られたアキレス腱エキスを再度梶原工業製真
空レオニーダーに投入し、精製塩3.8Kgを添加し
撹拌して完全に溶解した。 次いで品温を95℃まで加熱し、撹拌しながら20
分間保持した後50mmHg、品温60℃条件下で減圧
濃縮し、固形分60重量%のペースト状加塩アキレ
ス腱エキス25Kgを得た。 このペースト状エキスは特有な香味を有し、常
温に6ケ月間保持しても風味にはなんら影響がな
かつた。 一方明治製菓製「おでんの素」20gを1の湯
に溶解し、次に上記ペースト状加塩アキレス腱エ
キスを各0.2、0.4、0.6重量%添加した。これら試
験品を、ペースト状加塩アキレス腱エキス無添加
のおでんスープを対照品とし、30人の専門家パネ
ルによる官能試験に供した。官能試験の評価とし
ては対照品を0点とし、「おでんの素」の風味と
調和し且つコクがある事を効果として、非常に効
果あり…3点、効果あり…2点、やや効果あり…
1点、効果なし…0点の4段階の評点をパネルに
記入させ、パネル30人の合計点を求めた。結果は
下記第4表に示す通りであつた。

0.6重量%添加で非常に効果ありと判定された。 製造例 2 コラーゲン含量23重量%、水分含量63重量%の
市販冷凍アキレス腱15Kgを、湘南産業製フローズ
ンカツターMZ型にて、スライス幅1.5mmで細断
し、得られた削り節状の細断された冷凍アキレス
腱13Kgを製造例1に記載の真空レオニーダーに投
入し、13Kgの熱水を加えたのち、品温100℃まで
加熱し、撹拌翼を24rpmで撹拌しながら1.5時間
保持した。 次に煮沸されて柔軟化した細断アキレス腱18Kg
を品温50℃まで冷却し、300mlの水に溶解した長
瀬産業製ビオプラーゼSP−4を75g添加し、撹
拌翼をを24rpmで撹拌しながら、50℃で30分間保
持して加水分解を行い、液状のアキレス腱分解物
15Kgを得た。得られた分解物15Kgを95℃に15分間
保持し、ビオプラーゼを失活させた。 上記分解物を10メツシユの篩に通した後クリア
ランス0.2mmのデイスパーミルに通し摩砕した。
得られた摩砕品13.5Kgを50メツシユのバケツト型
遠心分離機(2000rpm)に通し品温約70℃の濾液
を得た。得られた濾液を製造例1に記載の真空レ
オニーダーに投入し撹拌翼で24rpmで撹拌しなが
ら95℃に加熱後20分間保持し、固形分量22重量%
のアキレス腱エキス12.2Kgを得た。 上記アキレス腱エキス12.2Kgを入口温度140℃、
出口温度80℃の条件下で大川原化工機製スプレー
ドライヤーL−8型にて噴霧乾燥し、水分含量約
3重量%のアキレス腱エキスパウダー2.1Kgを得
た。 本アキレス腱エキスパウダーは常温に6ケ月保
存しても風味、品質に何等影響はなかつた。 製造例1に記載の「おでんの素」に上記アキレ
ス腱エキス粉末を各0.1、0.2、0.3重量%添加し、
専門家パネル30名による官能試験に供した。これ
等に対しアキレス腱エキス粉末無添加品を対照品
とした。対照品を0点とし「おでんの素」と風味
が調和し且つコクがある事を効果として、非常に
効果あり3点、効果あり2点、やや効果あり1
点、効果なし0点の4段階の評点をパネルに記入
させ、30人の合計点を求めた。結果は下記第5表
に示す通りであつた。

有しないが、食品に添加時に調味し、豊かなコク
味を付与する効果を有しており、例えば従来知ら
れている動植物蛋白分解液を限外濾過して得られ
るポリペプチドに比べ、1/4〜1/5量の添加
で同等の調味効果を発揮するすぐれたものであ
る。 本発明によるアキレス腱調味料は、ラーメン、
チヤンポン、チヤーハン等を始め、スープ、カレ
ー、おでん、コロツケ等の調理食品に調味料とし
て添加使用できる他、一般加工飲食品の製造にも
利用出来る汎用性を有するものである。 更に今迄廃棄処分に近い利用方法しかなかつた
アキレス腱より有効調味成分をほほぼ完全に採取
する本発明方法は、動植物蛋白資源の有効利用の
見地からも画期的な技術と言える。

に、高圧ホモナゲイザーを使用して150〜200Kg/
cm2の加圧下で均質化処理することを特徴とする天
然蛋白調味料の製造方法。 2 アキレス腱を容器に入れ、レトルト内に収容
し、水を添加せずに蒸煮する特許請求の範囲第1
