Description
本発明は、固定化生理活性物質及びその製造法
並びにその製造法に用いる担体に関し、更に詳し
くは、高いリガンド濃度を有し、かつ、安定であ
る固定化生理活性物質及びその製造法並びにその
製造法に用いる担体に関する。
生理活性物質を固定化して工業的に用いる際の
固定化法には種々の方法があるが、固定化物質の
安定性、固定化物質と反応物との非特異的反応性
等を考慮すると、リガンド(生理活性物質)を共
有結合により不溶性担体に固定化する方法が望ま
しい。
このような方法としては、臭化シアン法及びエ
ポキシ活性化法〔有機合成化学、第38巻、128〜
138頁(1980年)〕があるが、前者は非特異的吸着
が起こる場合が多く、また、一度固定化されたリ
ガンドの結合が不安定なために、リガンドが漏出
することが多いという欠点があり、リガンドの結
合量も満足とはいえない。一方、後者の方法によ
り調製された特異的吸着体(固定化生理活性物
質)は、優れた安定性を有し、かつ、非特異的吸
着が少ないという利点があるが、リガンドを結合
させる際に、高温の強アルカリ性条件下において
処理しなくてはならないため、不安定な生理活性
物質の固定化には不適当である。また、エポキシ
活性化担体を、更に、濃アンモニア水で処理し、
アミノ誘導体に変換した後、種々のリガンドを結
合させる方法も発明されているが、多くのリガン
ド、特に糖等の反応性の低いカルボニル基を有す
る化合物との反応に長時間を要するという欠点が
ある。
また、カルボジイミドを用いるアミノ基とカル
ボキシル基とのカツプリング反応を利用する方法
もあるが、この方法では、イソ尿素誘導体の形成
やチヤツプマン転位等の副反応の起こる可能性が
あり、また、最適酸度であるPH4.5においても、
さらに高い水素イオン濃度の条件下ではなおさ
ら、リガンドが重合する副反応が起こり易く、固
定化物の収率は満足なものとはいえず、より反応
性の高い官能基を有する担体が望まれている。
本発明者らは、前述の問題点を解消するため鋭
意研究を重ねた結果、エポキシ基を有する水不溶
性高分子を、ヒドラジン又はジヒドラジド化合物
と反応させて得たヒドラジン誘導体及び/又はジ
ヒドラジド誘導体を固定化用担体として用いるこ
とにより、糖等の反応性の低い生理活性物質も穏
和な条件下で固定化することができ、かつ、得ら
れる固定化物質が高いリガンド濃度を有するとと
もに安定であることを見出し、本発明を完成する
に至つた。
即ち、本発明の固定化生理活性物質及び不溶性
高分子(固定化用担体)は、

担体として用いる不溶性高分子は、目的に応じ
て種々の形態で用いられる。例えば、アフイニテ
イー吸着剤又は固定化酵素に用いる場合には、吸
着容量を高めたり、接触面積を大きくするため、
多孔質の粒状のものが好ましく、希薄な生理活性
物質溶液の処理に用いる場合には、多孔質の膜状
のものが好ましい。
ヒドラジン類としては、ヒドラジン水化物、ヒ
ドラジン二塩酸塩、ヒドラジン硫酸塩等の種々の
形態のものが用いられる。
ジヒドラジド化合物としては、シユウ酸ジヒド
ラジド、マロン酸ジヒドラジド、コハク酸ジヒド
ラジド、グルタル酸ジヒドラジド、アジピン酸ジ
ヒドラジド、ピメリン酸ジヒドラジド、スベリン
酸ジヒドラジド、アゼライン酸ジヒドラジド、セ
バシン酸ジヒドラジド等の炭素数2〜12の直鎖状
飽和ジカルボン酸ジヒドラジド;マレイン酸ジヒ
ドラジド、フマル酸ジヒドラジド等の不飽和ジカ
ルボン酸ジヒドラジド;及びフタル酸ジヒドラジ
ド、テレフタル酸ジヒドラジド等の芳香族ジカル
ボン酸ジヒドラジド等が挙げられる。
これらのヒドラジン類及びジヒドラジド化合物
は、単独で又は二種以上の混合物として用いられ
る。用いるヒドラジン類及びジヒドラジド化合物
の量は、前記エポキシ基を有する水不溶性高分子
又はエポキシ活性化不溶性高分子1g(乾燥重
量)当たり、ヒドラジン類では0.001〜0.5g、ジ
ヒドラジド化合物では、その種類によつて異なる
が、一般には、0.005〜1gである。溶媒として
は、一般には、水が用いられ、その量は前記不溶
性高分子1g(乾燥重量)当たり、5〜10mlであ
り、溶媒中には、必要に応じて、炭酸ナトリウ
ム、塩酸等を適宜添加してもよい。以上の条件
で、0〜100℃、好ましくは4〜50℃において、
0.5〜72時間、好ましくは1〜48時間反応させる
ことにより、前記式()で示される固定化用担
体を得ることができる。
このようにして得た固定化用担体に生理活性物
質を固定化する方法は、生理活性物質の種類によ
り、適宜選択される。
生理活性物質が、乳糖、デキストラン硫酸、ヘ
パリン、N−アセチルグルコサミン、N−アセチ
ルムラミン酸、トリ−N−アセチルキトトリオー
ス、キチン、ブドウ糖、麦芽糖等の糖類;ムチ
ン、酸性α1−糖タンパク質、プロトロンビン、ハ
プトグロビン、ゴナドトロピン等の糖タンパク
質;フレノシン、ケラシン、ネルボン、オキシネ
ルボン、ガングリオシド等の糖脂質;グルコシ
ド、ガラクトシド、マンノシド、フルクトシド等
の各種配糖体;の他、カルボニル基、アルデヒド
基、カルボキシル基、アルコキシカルボニル基、
アミノカルボニル基、ハロホルミル基等の広義の
カルボニル基を有する化合物である場合には、以
下のようにして固定化することができる。
即ち、前記生理活性物質を、水素化シアノホウ
素ナトリウムの存在下において、前記式()で
示される固定化用担体と反応させることにより、
目的とする固定化生理活性物質を得ることができ
る。用いる水素化シアノホウ素ナトリウムの量
は、前記固定化用担体1g(乾燥重量)当たり、
10mg以上であればよいが、特に100〜500mgである
ことが好ましい。生理活性物質の量は、その種類
によつて異なるが、一般には、前記固定化用担体
1g(乾燥重量)当たり、50〜500mgである。溶
媒としては、一般には、リン酸水素二カリウム水
溶液、リン酸緩衝液、酢酸緩衝液、ホウ酸緩衝
液、炭酸緩衝液、重炭酸緩衝液等が用いられ、そ
のPHは8〜10であることが好ましく、その量は前
記固定化用担体1g(乾燥重量)当たり、3〜20
mlであることが好ましい。以上の条件で、0〜
100℃、好ましくは4〜80℃において1〜72時間、
好ましくは2〜48時間反応させることにより、目
的とする固定化生理活性物質を得ることができ
る。このようにして得た固定化生理活性物質は、
必要に応じて、無水酢酸で処理して、未反応部位
をアセチル化してもよい。
次に、生理活性物質が、コンカナバリンA(以
下、「ConA」という。)、ウシ血清アルブミン
(以下、「BSA」という。)、γ−グロブリン(抗
体)、酵素、植物性血球凝集素(PHA)、フイブ
ロネクチン、プロテインA等の蛋白質;キニン、
副腎皮質刺激ホルモン放出因子(CRF)、カルシ
トニン、グルカゴン等のペプチド;核タンパク
質、色素タンパク質、糖タンパク質、リンタンパ
ク質等の複合タンパク質;種々のアミノ酸;ヒス
タミン、ドパミン、グルコサミン、トリプタミン
等の生体アミン;等のアミノ基を有する化合物で
ある場合には、以下のようにして固定化すること
ができる。
即ち、前記式()で示される固定化用担体
を、水素化シアノホウ素ナトリウムの存在下にお
いて、

で示されるアルデヒド活性化不溶性高分子とす
る。用いる水素化シアノホウ素ナトリウムの量
は、担体の含水量によつて異なるが、前記固定化
用担体1g(乾燥重量)当たり、50mg以上であれ
ば一応の目的に達するが特に80〜1000mgであるこ
とが好ましい。直鎖状ジアルデヒド化合物として
は、一般には、炭素数2〜10のもの、例えば、グ
リオキサール、プロパンジアール、ブタンジアー
ル、グルタルアルデヒド(ペンタンジアール)、
デカンジアール等が単独で又は二種以上の混合物
として用いられる。溶媒としては、水が用いら
れ、その量は前記固定化用担体1g(乾燥重量)
当たり、5〜20mlであることが好ましい。以上の
条件で、0〜80℃、好ましくは20〜40℃におい
て、0.5〜10時間、好ましくは1〜4時間反応さ
せることにより、前記アルデヒド活性化不溶性高
分子を得ることができる。
次いで、このアルデヒド活性化不溶性高分子
を、水素化シアノホウ素ナトリウムの存在下にお
いて、前記生理活性物質と反応させることによ
り、目的とする固定化生理活性物質を得ることが
できる。この時、用いる溶媒としては、リン酸緩
衝液、酢酸緩衝液、ホウ酸緩衝液、炭酸緩衝液等
が挙げられ、そのPHは5〜8であることが好まし
く、その量はアルデヒド活性化不溶性高分子1g
(乾燥重量)当たり、5〜40mlであることが好ま
しい。用いる水素化シアノホウ素ナトリウムの量
は、アルデヒド活性化不溶性高分子1g(乾燥重
量)当たり、100mg以上であればよいが、特に200
〜400mgであることが好ましい。生理活性物質の
量は、その種類によつて異なるが、一般には、ア
ルデヒド活性化不溶性高分子1g(乾燥重量)当
たり、100〜1000mgである。以上の条件で、0〜
80℃、好ましくは4〜40℃において、5〜100時
間、好ましくは12〜72時間反応させることによ
り、目的とする固定化生理活性物質を得ることが
できる。
一方、生理活性物質が、前述した蛋白質、ペプ
チド、複合タンパク質、アミノ酸、ヘパリン、N
−アセチルムラミン酸;及びパルミチン酸、オレ
イン酸等の脂肪酸;ペニシリン、プロスタグラン
ジン;等のカルボキシル基を有する化合物である
場合には、以下のようにして固定化することがで
きる。
即ち、前記生理活性物質を、カルボジイミド化
合物の存在下において、前記式()で示される
固定化用担体と反応させることにより、目的とす
る固定化生理活性物質を得ることができる。用い
るカルボジイミド化合物としては、1−エチル−
3−(3−ジメチルアミノプロピル)カルボジイ
ミド・塩酸塩(以下、「EDC」という。)、1−シ
クロヘキシル−3−(2−モルホリノエチル)カ
ルボジイミド・メソ−p−トルエンスルホン酸
塩、ジシクロヘキシルカルボジイミド等が単独で
又は二種以上の混合物として用いられ、その量は
前記固定化用担体1g(乾燥重量)当たり、10〜
200mgであることが好ましい。生理活性物質の量
は、その種類によつて異なるが、一般には、前記
固定化用担体1g(乾燥重量)当たり、30〜100
mgである。溶媒としては、一般には、水が用いら
れ、その量は前記固定化用担体1g(乾燥重量)
当たり、2〜10mlであり、溶媒中には、必要に応
じて、塩化ナトリウム、塩化カリウム等を適宜添
加してもよい。以上の条件で、0〜80℃、好まし
くは4〜40℃において、10〜100時間、好ましく
は24〜72時間反応させることにより、目的とする
固定化生理活性物質を得ることができる。
本発明の製造法によれば、穏和な条件下で、か
つ、反応中のPH調整を行なうことなく、しかも、
従来法に比し短い反応時間で目的とする固定化生
理活性物質を得ることができる。さらに、得られ
た固定化生理活性物質は、従来のものに比し、高
いリガンド濃度を有している。
以下、実施例により本発明を更に詳細に説明す
る。
実施例 1
(1) ヒドラジンアガロースの調製
アガロースとしてフアルマシア社製のセフアロ
ース4Bを用いた。グラスフイルター上で水でよ
く洗浄後、吸引過したアガロース20gをフラス
コに入れ、水30ml、2M NaOH水溶液13ml及び
エピクロルヒドリン3mlを順次加えた。懸濁液を
40℃のインキユベーター中で2時間振盪後、グラ
スフイルター上で水で充分に洗浄してエポキシ活
性化アガロースを得た。このエポキシ活性化アガ
ロースに導入されたエポキシ基の量をNa2S2O3を
用いる中和滴定法により測定したところ、乾燥重
量1mg当たり、約500μmoleのエポキシ基が導入
されていた。
このエポキシ活性化アガロース1容(20g)に
80%ヒドラジン水化物水溶液1.5容(30ml)を加
え、40℃のインキユベーター中で1時間半振盪し
た。反応後、グラスフイルター上で水で充分に洗
浄してヒドラジンアガロースを得た。
(2) 固定化乳糖の調製
(1)で得たヒドラジンアガロース2g〔乾燥重量
(湿重量20g)〕に、乳糖520mgと水素化シアノホ
ウ素ナトリウム256mgとを0.2M K2HPO4水溶液
15mlに溶解したものを加え、室温で3日間振盪し
た。反応後、グラスフイルター上で水10ml及び
0.2M CH3COONa水溶液10mlで順次洗浄した。
次いで、無水酢酸5mlを加え、0℃で30分間反応
させた後、更に、無水酢酸5mlを加え、室温で30
分間処理して、未反応の(乳糖が固定化されてい
ない)ヒドラジン残基をアセチル化した。反応
後、水、0.1M NaOH水溶液、水、リン酸緩衝化
生理食塩水(PBS)で順次洗浄して、乳糖結合
アガロースを得た。
実施例 2
(1) アジピン酸ジヒドラジドアガロースの調製
アガロースとしてフアルマシア社製のセフアロ
ースCL6Bを用いた。実施例1(1)と同様に処理す
ることにより得たエポキシ活性化アガロース1
に、アジピン酸ジヒドラジド115gを0.1M
Na2CO3水溶液に溶解し塩酸でPHを9に調整した
溶液を加え、40℃で一夜振盪した。反応後、グラ
スフイルター上で0.2M NaCl水溶液で充分に洗
浄してアジピン酸ジヒドラジドアガロースを得
た。
(2) 固定化デキストラン硫酸の調製
(1)で得たアジピン酸ジヒドラジドアガロース
100g〔乾燥重量(湿容積1)〕に、デキストラ
ン硫酸(フアルマシア社製)26gと水素化シアノ
ホウ素ナトリウム12.8gとを0.2M K2HPO4水溶
液750mlに溶解したものを加え、室温で3日間振
盪した。反応後、グラスフイルター上で水、
0.2M CH3COONa水溶液で順次洗浄した。次い
で、無水酢酸250mlを加え、0℃で30分間振盪後、
更に、無水酢酸250mlを加え、室温で30分間振盪
して、未反応の(デキストラン硫酸が固定化され
ていない)アジピン酸ジヒドラジド残基をアセチ
ル化した。反応後、水、0.1M NaOH水溶液、
水、PBSで順次洗浄して、デキストラン硫酸結
合アガロースを得た。反応前の前記デキストラン
硫酸含有0.2M K2HPO4水溶液及び反応後の上澄
み液中のデキストラン硫酸をフエノール硫酸法に
従い測定したところ、得られたデキストラン硫酸
結合アガロースには、用いたデキストラン硫酸の
25%が結合していることが判明した。
実施例 3
(1) アルデヒド活性化アガロースの調製
実施例2(1)と同様に処理することにより得たア
ジピン酸ジヒドラジドアガロースに、25%グルタ
ルアルデヒド水溶液1ml及び水素化シアノホウ素
ナトリウム82mgを加えて、40℃のインキユベータ
ー中で2時間振盪後、グラスフイルター上で水で
充分に洗浄してアルデヒド活性化アガロースを得
た。
(2) 固定化ConAの調製
(1)で得たアルデヒド活性化アガロース1gに、
ConA100mg及びメチル−α−D−マンノシド110
mgを溶解した0.2Mリン酸緩衝液(PH7.0)2mlを
加えた後、水素化シアノホウ素ナトリウム24mgを
加えて、4℃のインキユベーター中で2日間振盪
した。反応後、上清を除き、0.2Mリン酸緩衝液
で充分に洗浄してConA結合アガロースを得た。
(3) リガンド濃度の測定
上清及び洗浄液中の非結合残存ConAの量を、
その280nmにおける吸収の測定値より算出するこ
とによつて、アガロースに結合したConAの量を
算出した。尚、対照として、CNBr法によりアガ
ロースに結合させた酵素〔千畑一郎 外著;“ア
フイニテイークロマトグラフイー”、40頁、東京、
講談社(1976年)〕(対照1)、過ヨウ素酸酸化法
により各種多糖に結合させたBSA
〔Immunology、第20巻、1061〜1065頁(1971
年)〕(対照2)及びヒドラジド誘導体に結合させ
たBSA〔Biochemistry、第8巻、4074〜4082頁
(1969年)〕(対照3)を用いた。結果を第1表に

上で0.5M NaCl水溶液8ml及び水100mlで順次洗
浄して、BSA結合アガロースを得た。洗浄液中
の非結合残存BSAの量を、その280nmにおける
吸収の測定値より算出することによつて、得られ
たBSA結合アガロースには、湿重量1g当たり
47mgのBSAが結合していることが判明した。
実施例 5
(1) アジピン酸ジヒドラジドアガロースの調製
実施例1(1)と同様に処理することにより得たエ
ポキシ活性化アガロース20g(湿重量)に、アジ
ピン酸ジヒドラジド2.3gを0.1M Na2CO3水溶液
に溶解し塩酸でPHを9に調整した溶液を加え、室
温で一夜振盪した。反応後、グラスフイルター上
で、洗浄液がトリニトロベンゼンスルホン酸
(TNBS)と反応を示さなくなるまで、0.2M
NaCl水溶液約500mlで洗浄してアジピン酸ジヒド
ラジドアガロースを得た。
(2) 固定化BSAの調製
(1)で得たアジピン酸ジヒドラジドアガロース1
gに、BSA50mgを溶解した0.5M NaCl水溶液2
mlを加えた後、EDC33mgを加えた。PHを5に調
整後、室温で一夜振盪した後、更に、EDC33mg
を加えて一夜振盪した。反応後、グラスフイルタ
ー上で0.5M NaCl水溶液8ml及び水100mlで順次
洗浄して、BSA結合アガロースを得た。洗浄液
中の非結合残存BSAの量を、その280nmにおけ
る吸収の測定値より算出することによつて、得ら
れたBSA結合アガロースには、湿重量1g当た
