【発明の詳細な説明】[Detailed description of the invention]

〔産業上の利用分野〕 本発明は、ラセミ体のN−カルバモイル−α−
アミノ酸を立体選択的に環化して生化学的に光学
活性ヒダントイン類に変換し光学分割する方法に
関し、医薬などとして有効な生理活性を示す光学
活性化合物ないしはその合成中間体を極めて有利
に製造することを目的とする。 〔従来技術〕 本発明の目的のひとつは、光学活性なヒダント
イン類を効率的に製造することにある。ヒダント
イン類のなかには、抗けいれん剤あるいは向精神
用薬などとして利用されているものも多く、種々
の生理活性を示す化合物が知られている。通常ラ
セミ体で用いられているが、分子内に不斉炭素原
子を有するヒダントイン類においては、一般に特
定の立体配置をもつもののみが生理活性に関与す
る場合が多く、薬効の向上あるいは副作用の減少
といつた観点から光学活性体の使用が望まれる。 従来、光学活性なヒダントイン類を製造する方
法としては、 1) 光学活性なα−アミノ酸にシアン酸アルカ
リ金属塩を反応させ、一旦、N−カルバモイル
−α−アミノ酸とした後、これを鉱酸中、加熱
環化させてヒダントイン類とする方法 2) 光学活性シアノ酢酸をイソシアネートに誘
導し、これをアミンと反応しN−カルバモイル
アミノニトリルを得、最後に鉱酸中、加熱環化
させてヒダントイン類とする方法 3) ラセミ体のヒダントイン類に光学活性ブル
シンを加え、ジアステレオマー塩を形成させ、
溶媒に対する溶解度差を利用して光学分割する
方法〔ジヤーナル・オブ・メデイカル・ケミス
トリー(J.Med.Chem.21(12)、1294、1978〕 4) ケトンに光学活性アミンを反応させケチミ
ンを得、これにシアン化水素を不斉的に付加さ
せた後、クロルスルホニルイソシアネートを反
応させ光学活性ヒダントインとする方法〔ジヤ
ーナル・オブ・オーガニツク・ケミストリー
(J.Org.Chem.)47、4081、1982〕などの方法
が知られている。 天然アミノ酸のように比較的容易に入手可能な
光学活性体を原料に使用できる場合は、1)の方
法が有利であるが、そうでない場合ラセミ体を酸
または塩基に誘導して光学分割するなど、通常、
原料を得るのに1)や2)の方法は複雑な操作が
あらかじめ必要である。また、ラセミ体のヒダン
トイン類を直接光学分割する3)の方法は、分割
剤に極めて強い毒性を有するブルシンを必要とす
ることから、その取り扱い及び製品中へのブルシ
ンの混入など操作性、安全性などの点で、工業的
規模での生産を目的とする場合、多くの問題が予
想される。不斉合成的に一挙に光学活性体を合成
しようとする4)の方法においては、高価な光学
活性アミンおよびクロルスルホニルイソシアネー
トを当モル以上消費する上、アミン由来の不要な
光学活性部位の除去が極めて困難であるなど、実
用上多くの難点を有している。 〔発明が解決しようとする課題〕 本発明らは、ヒダントイン類あるいはα−アミ
ノ酸から容易に誘導できるN−カルバモイル−α
−アミノ酸に着目し、光学活性ヒダントイン類の
効率的な合成法の開発を目的に、生化学的な手法
について鋭意検討を行なつた結果、微生物のなか
にはN−カルバモイル−α−アミノ酸を基質とし
立体選択的な環化反応により光学活性なヒダント
イン類を生成する能力を有する微生物が存在する
ことを見いだし、本反応が光学活性ヒダントイン
のみならず光学活性アミノ酸類の製造法としても
有効な、極めて適用範囲の広い光学分割手段とな
り得ることを明らかにして本発明を完成した。 〔課題を解決するための手段〕 本発明の概略は次の式で表わされる。 すなわち、本発明は一般式()で示されるN
−カルバモイル−α−アミノ酸()のラセミ体
の一方の立体配置のみを微生物の変換能力により
ヒダントイン類()に変換せしめたのち、未利
用対掌体のN−カルバモイル−α−アミノ酸
()を分取し、酸性下に加熱環化反応すること
により容易に立体配置を保持したまま光学活性ヒ
ダントイン類()に化学的に変換することを目
的とするものである。本発明方法はラセミ体のN
−カルバモイル−α−アミノ酸を実質的に()
と()の光学活性ヒダントイン類に光学分割す
る方法としても利用できる。更にまた、本発明方
法で得られる光学活性ヒダントイン類()およ
び対掌体N−カルバモイル−α−アミノ酸()
は、いずれもアルカリ加水分解して対応する光学
活性α−アミノ酸(,)に変換できることか
ら、本発明方法はラセミ体のN−カルバモイル−
α−アミノ酸を光学活性α−アミノ酸に光学分割
する方法としても利用できるなど広い適用性を有
している。 以下、本発明を詳細に説明する。 本発明で原料として用いられるラセミ体のN−
カルバモイル−α−アミノ酸は、ケトンを出発化
合物として、α−アミノ酸の一般的な合成法とし
て周知のストレツカーー(Strecker)法またはブ
ツフエラー(Bucherer)法によつて一旦α−ア
ミノ酸を合成し、これを更にシアン酸アルカリ金
属塩と反応させることにより容易に合成すること
ができる。またブツフエラー法の合成中間体であ
るヒダントイン類を、制御された条件下に加水分
解することにより直接N−カルバモイル−α−ア
ミノ酸を合成することも可能である。 本発明で使用する、N−カルバモイル−α−ア
ミノ酸を立体選択的に環化して光学活性ヒダント
イン類を生成する能力を有するこれら微生物の分
布は極めて広く各種の種属にわたつている。例え
ば細菌に属するものとしては、アエロバクター属
(Aerobacter)、アグロバクテリウム属
(Agrobacterium)、バシルス属(Bacillus)、コ
リネバクテリウム属(Corynebacterium)、など
がある。また放線菌に属するものとしてはノカル
デイア属(Nocardia)がある。 これら微生物の培養は通常液体栄養培地で行な
われる。培地には、通常資化し得る炭素源、窒素
源、各微生物の生育に必須の無機塩栄養素を含有
させるが、更に各種核酸塩基およびそれらの誘導
体その他の酵素誘導剤を少量添加して、必要な酵
素を適応的に増強させることが望ましい。培養時
の温度は20〜70℃、PHは4〜10の範囲が用いられ
る。通気撹拌により微生物の生育を促進すること
もできる。 N−カルバモイル−α−アミノ酸のヒダントイ
ン類への環化反応において、前記のようにして得
た微生物の培養液、菌体または菌体処理物などを
使用することができる。微生物の培養液をそのま
ま用いることもできるが、更に望ましくは培養液
から分離した菌体を使用する。菌体は生菌体のま
まで用い得ることは勿論であるが、貯蔵および取
扱いの便宜から凍結乾燥菌体のような乾燥菌体と
して用いることもできる。更に、全菌体の状態を
保つていなくとも菌体磨砕物または菌体抽出物な
ど、当該変換能力が発揮できる限り、いかなる態
様のものでも本反応に利用することができる。 反応基質であるN−カルバモイル−α−アミノ
酸の反応液中での濃度は、0.1%から30%程度の
高濃度まで用いることができる。好ましい反応PH
としては5〜8の範囲が用いられる。PHが5未満
では当該能力が失活しやすく、PH8をこえるとヒ
ダントイン類の溶解度が増加して反応が完結しに
くくなるので実用性に乏しい。最適PHは、反応基
質および当該能力の由来する微生物によつて異な
るが、おおむねPH5〜8の範囲に含まれている。
環化反応の進行に伴つてPHは次第にアルカリ側に
移行するので、適時中和剤を添加して最適PHに保
持することが望ましい。中和剤としては、塩酸、
硫酸など鉱酸が適当である。本反応に際しての反
応温度は、通常30〜60℃の範囲が用いられるが、
個々の場合に適した温度が採用される。これら反
応条件ならに当該能力の活性の程度を選択するこ
とにより、ほぼ定量的な変換率を得ることは比較
的容易であり、未反応N−カルバモイル−α−ア
ミノ酸も極めて高い光学純度を有する対掌体とし
て単離することが可能である。 この環化反応によつて生成した光学活性ヒダン
トイン類および未利用対掌体N−カルバモイル−
α−アミノ酸を単離するには、公知の方法が採用
できる。一般に本発明方法で生成するようなα−
炭素原子に水素原子を含まないヒダントイン類
は、PH8ないし9以下では水性媒体に対して極め
て離溶性であるため、環化反応の進行に伴つて生
成するヒダントイン類は通常ほとんど大部分が結
晶として析出した状態になる。ところがPH11以上
ではこれらのヒダントイン類はアルカリ金属塩と
なつて易溶性を示す。一方、反応基質であるN−
カルバモイル−α−アミノ酸は、本反応条件(PH
5〜8)ではアルカリ金属塩となり、極めて大き
な溶解度を示し、完全に溶解した状態となるが、
PH4以下ではカルボキシル基が遊離の状態となり
極端な不溶性を示す。このようなヒダントイン類
およびN−カルバモイル−α−アミノ酸の水性媒
体中でのPHに対する溶解度の変化を利用して、極
めて容易に生成ヒダントイン類と未利用対掌体N
−カルバモイル−α−アミノ酸を分別、単離する
ことも可能である。勿論、酢酸エチルの如き有機
溶剤への高い溶解度を利用した抽出方法によつて
も容易に分別、単離することができる。 未利用対掌体N−カルバモイル−α−アミノ酸
は、塩酸または硫酸のような鉱酸酸性下、あるい
は酢酸のような有機酸中で70〜100℃に加熱攪拌
するといつた公知の方法を採用することにより、
容易に化学的に環化して光学活性ヒダントイン類
に変換することも可能である。また、苛性ソー
ダ、苛性カリ、水酸化バリウムのようなアルカリ
金属塩を用いて加水分解するか、あるいは鉱酸酸
性下に当量の亜硝酸ソーダなどを用いてカルバモ
イル基を酸化するなどの方法によつて、このN−
カルバモイル−α−アミノ酸を光学活性α−アミ
ノ酸に変換することも可能である。 また、本発明により必然的に副生する、目的と
しない対掌体は、アルカリ加水分解などを通じ
て、一旦α−アミノ酸とした後、これを次亜ハロ
ゲン化水素酸などの酸化剤を用いる公知の方法に
よりケトンとして回収し、これを新たなラセミ体
のN−カルバミル−α−アミノ酸となして循環再
利用することにより、本発明の効率を一層高め得
ることは云うまでもない。 〔実施例〕 以下に、実施例を記載して本発明を説明する。 実施例 1 下記の組成からなる栄養液体培地を調製し、綿
栓をした500ml容肩付フラスコに100mlずつ分注
後、120℃で20分間蒸気殺菌を行なつた。 培地組成: 肉エキス 0.5%(重量%) 酵母エキス 0.5% ポリペプトン 1.0% NaCl 0.15% ウラシル 0.10% MnCl2・4H2O 20 ppm (PH7.0) 予め、ブイヨン寒天スラトンで30℃、24時間培
養した表−1に示す微生物を上記栄養培体培地に

価な微生物などの生体反応を用いる本発明は、極
めて実用性に富むこれら光学活性化合物の有効な
製造法を提供するものであるといえる。さらに本
発明は、従来予想もされていなかつたα−炭素上
に水素原子を持たないN−カルバモイル−α−ア
ミノ酸が環化反応を受けてヒダントイン類に生化
学的に変換しうるという全く新しい知見を提供す
るものである。
