イ 発明の目的
産業上の利用分野
本発明は新規β−ガラクトシダーゼ阻害物質ガ
ラクトスタチンに関し、さらに詳しくは下記一般

ラクトスタチン類、さらにそれらの塩およびその
製造法に関するものである。
従来の技術
RNAウイルスで癌化した3T3線維芽細胞では
グルコシダーゼ活性、特にβ−ガラクトシダーゼ
活性が上昇することがボスマン(H.B.Bosmann)
により報告されている[Biochem.Biophys.
Acta,264,339(1972)]。そこで、β−ガラクト
シダーゼ阻害物質が制癌剤として利用できる可能
性に注目して種々研究されており、たとえば特開
昭57−74090;特公昭53−31238;The Journal
of Antibiotics28,1006(1975);同32(3),212
(1979);および同32(3),217(1979)などに記
載がある。本発明のガラクトスタチン構造上類似
物質としてはたとえば、特公昭43−760;特開昭
51−151393;特開昭51−151394などに記載がある
が構造、生理活性とも異なる。
発明が解決しようとする問題点
β−ガラクトシダーゼ阻害物質としては、先に
本発明者らが発見した特開昭57−74090記載の
GT−2558−A及びGT−2558−Bが知られてい
るが、これらの物質は非常に不安定で取扱いが非
常に困難であつた。今回、以前発見されたβ−ガ
ラクトシダーゼ阻害物質と異なる安定な新規酵素
阻害物質を見い出すことに成功した。
ロ 発明の構成
問題点を解決するための手段
GT−2558−A及びGT−2558−Bを生産する
菌、ストレプトマイセス・ライデイツカスPA−
5726、微工研菌寄第5657号を詳細に検討した結
果、GT−2558−A,GT−2558−B以外にβ−
ガラクトシダーゼ阻害活性を有する弱塩基性糖類
似物質5−アミノ−5−デオキシ−D−ガラクト
ピラノースの存在することを発見し、ガラクトス
タチンと命名した。その後当菌株の培養条件の検
討をおこない、生産性を向上させる事に成功し、
ガラクトスタチンを大量に単離・精製した。更に
ガラクトスタチンよりそのラクタム体及びデオキ
シ体を調製した。これらの化合物は、いずれもβ
−ガラクトシダーゼに対して強い阻害作用を有す
る酵素阻害物質である。以下にガラクトスタチ
ン、ガラクトスタチン亜硫酸付加物、ガラクトス
タチンラクタム、1−デオキシガラクトスタチン
の性状を記載する。なお、本発明はこれら化合物
のみならず、可能な場合にはこれら化合物の製薬
上許容される塩をも包含する。

上記性状に示したβ−ガラクトシダーゼ阻害活
性及び阻害比活性は、o−ニトロフエニル・β−
D−ガラクトピラノースを基質としてβ−ガラク
トシダーゼを作用せしめ、加水分解され遊離する
o−ニトロフエノールを比色法で定量することに
より測定した。すなわち、0.1M酢酸緩衝液(PH
5.0)に溶かしたβ−ガラクトシダーゼ溶液0.25
mlに検体を含む溶液0.25mlを加え、30℃で10分間
加温した後、あらかじめ30℃に加温しておいた上
記緩衝液に溶かした基質0.01Mo−ニトロフエニ
ル・β−D−ガラクトピラノース溶液0.5mlと混
和する。30℃で15分間反応させたのち、1M炭酸
ナトリウム溶液1mlを加えて反応を停止させ、水
8mlを添加して全量10mlとした後、420nmにおけ
る吸光度(A)を測定した。同時に、上記検体を
含まない緩衝液のみを用いた対照の吸光度(B)
を測定し、阻害率を(B−A)/B×100により
計算した。なお、β−ガラクトシダーゼはタカジ
アスターゼより調製したものを用いた。前記条件
でβ−ガラクトシダーゼ活性を50%阻害(IC50)
する検体を1阻害単位量(1Iu)とした。
ガラクトスタチンは公知のβ−ガラクトシダー
ゼ阻害物質とは異なる性状を有しており、新規β
−ガラクトシダーゼ阻害物質であると判断され
る。かかるガラクトスタチンの産生菌は長崎県南
松浦群中通島にて採取された土壌試料より分離し
たPA−5726株が挙げられる。
本菌株は分類学状ストレプトマイセス・ライデ
イツカス(Streptomyces lydicus)の一菌株で
あると判断され、PA−5726株を微生物工業技術
研究所にストレプトマイセス・ライデイツカス
PA−5726、微工研条寄第61号(微工研菌寄第
5657号)として寄託してある。
同菌株の菌学的諸性状は下記のとおりである。
(a) 形態学的性状(ベンネツト寒天培地、28℃、
14日間培養)
胞子のう、鞭毛胞子、菌核はいずれも認められ
ず、また基底菌糸にはフラグメンテーシヨンによ
る分裂は認められない。同培地上での気中菌糸は
豊富に形成される。胞子着生菌糸は気中菌糸上に
着生し、その主軸より単純分枝して側枝となり、
その末端はら線状(スパイラル状)を呈する。電
子顕微鏡観察による胞子の表面構造は、平滑
(smooth type)である。本菌株は時間の経過に
ともない。コロニーの表面の一部が湿潤し、いわ
ゆるハイグロスコピツク(hygroscopic)状にな
り黒変化する。

5726株の諸性状とこれら文献中に記載されたスト
レプトマイセス・ライデイツカス
(Streptomyces lydicus)の諸性状とを比較した
結果、両者の性状が極めて良く一致した。従つて
PA−5726株はストレプトマイセス・ライデイツ
カスと同種に属する一菌株であると同定し、PA
−5726株はストレプトマイセス・ライデイツカス
PA−5726(Streptomyces lydicus PA−5726)
と命名した。
本発明においては、上記のPA−5726株ならび
にその天然および人工変異株は勿論のこと、スト
レプトマイセス属ライデイツカス種に属し、ガラ
クトスタチンを産生する菌株はすべて使用でき、
本発明の範囲内とする。
ガラクトスタチンの生産は、ガラクトスタチン
産生株を栄養培地に好気的条件下に培養し、培養
終了後培養物からガラクトスタチンを分離採取す
ることにより行なう。以下にガラクトスタチンの
一般的製造方法を記載する。
培地組成、培地条件などは抗生物質の生産に一
般に用いられているものを用いるとよい。培地は
原則として、炭素源、窒素源、無機塩などを含
む。必要に応じて、ビタミン類、先駆物質などを
加えてもよい。炭素源としては、例えば、グルコ
ース、澱粉、デキストリン、グリセリン、糖密、
有機酸などが単独でまた混合物として用いられ
る。窒素源としては、例えば、大豆紛、コーンス
チープリカー、肉エキス、酵母エキス、綿実粉、
ベブトン、小麦胚芽、硫酸アンモニウム、硝酸ア
ンモニウムなとが単独または、混合物として用い
られる。無機塩としては、例えば、炭酸カルシウ
ム、塩化ナトリウム、塩化カリウム、硫酸マグネ
シウム、塩化コバルト、各種リン酸塩などがあげ
られ、必要に応じて培地に添加する。
培養は一般に抗生物質の製造に用いられる方法
に準じて行なえばよく、好ましくは液体培養が、
大量生産を行なう場合は、深部通気培養がよい。
培地のPHが変動しうる場合には、培地に炭酸カル
シウムなどの緩衝剤を加えてもよい。培養は約20
〜40℃で行なうとよく、特に25〜32℃が好まし
い。培養時間は発酵の規模に大きく左右される
が、約20〜80時間が大量生産を行なう場合に要す
る培養時間である。培養中に厳しい発泡が起こる
場合には、培養前または培養中に例えば、植物
油、ラード、ポリプロピレングリコールなどの消
泡剤を適宜添加するとよい。
培養終了後、培養物からガラクトスタチンを分
離採取するには、通常の発酵生産物を分離採取す
る方法を適宜用いる。例えば濾過、遠心分離、各
種イオン交換樹脂やその他の活性吸着剤による吸
脱着やクロマトグラフイー、各種有機溶媒による
抽出などを適当に組み合わせるとよい。
ガラクトスタチンは分離操作のため、また医
薬、動物薬として使用する便宜上、塩とするのが
望ましいことがある。ガラクトスタチンと塩を形
成しうる酸としては、亜硫酸、塩酸、臭化水素
酸、ヨウ化水素酸、硫酸、硝酸、リン酸、炭酸な
どの無機酸および酢酸、フマル酸、リンゴ酸、酒
石酸、マレイン酸、クエン酸、マンデル酸、アス
コルビン酸、没食子酸などの有機酸がある。
さらに得られたガラクトスタチンから種々の誘
導体を調製することが可能である。例えばガラク
トスタチンラクタムはガラクトスタチンに次亜ヨ
ウ素酸ナトリウム溶液、ヨウ素ヨウ化カリウム溶
液、過酸化水素、オゾン、あるいは四酢酸鉛等の
酸化剤を作用させることにより得られる。また、
1−デオキシガラクトスタチンはガラクトスタチ
ンを水素気流中、二酸化白金、白金パラジウム等
の触媒を用いての接触還元、あるいは水素化ホウ
素ナトリウム等の還元剤を作用させる等して調製
することができる。以上の2例だけでなく他の誘
導体を調整することも可能である。これらの誘導
体も、ガラクトスタチンと塩を形成しうる前記の
酸と塩を形成しうる。
実施例
以下に本発明にかかるガラクトスタチンの一群
の化合物の製造例を示すが、これらの実施例は何
ら本発明を制限するものではない。
実施例 1
ガラクトスタチンの調製方法
グルコース1.0%、カザミノ酸0.2%、酵母エキ
ス0.2%、肉エキス0.1%を含む培地(PH7.0)を
500ml容量の振盪フラスコに100mlずつ分注し、
120℃、20分間滅菌したものを種用培地とする。
ストレプトマイセス・ライデイツカスPA−
5726株(Streptomyces lydicus PA−5726、微
工研条寄第61号(微工研菌寄第5657号))の斜面
培養から−白金耳量を上記種用培地に接種し、28
℃で毎分140往復、振巾7cmで2日間振盪培養し
て種母を調製する。
一方、グリセリン3.0%、肉エキス2.0%、ペプ
トン10%、塩化カリウム0.076%、塩化マグネシ
ウム・6水和物0.042%、塩化鉄・6水和物
0.0025%、硫酸亜鉛・7水和物0.0022%、塩化マ
ンガン・4水和物0.0018%を含む培地(PH7.0)
を調整する。
この培地100mlを500ml容量の振盪フラスコに分
注し、120℃、30分間滅菌したものを主発酵培地
として、前記の種母をこの主発酵培地に2%接種
し、28℃で毎分140往復、振巾7cmで5日間振盪
培養を行なう。
この培養液中にβ−ガラクトシダーゼ阻害物質
ガラクトスタチン43500IU/ml(IC50=
0.0057mcl/ml)が蓄積される。
実施例 2
実施例1と同様にして、主発酵培地20を用い
て培養を行ない、培養終了後直ちに遠心分離によ
り菌体を分離して清澄な濾液15.8を得る。
この濾液に塩酸を加えてPH4.5に調製し、活性
炭1.5%(237g)を加え30分間攪拌して色素及び
不純物を吸着させて、セライトを助剤として濾過
し、清澄な濾液15を得る。
この濾液を強塩基性陰イオン交換樹脂ダウエツ
クス−1×8(OH型)(商品名、ダウ・ケミカル
社)2に通過させて、陰イオン性物質を除く。
この溶液を塩酸で中和し、更に強酸性陽イオン交
換樹脂ダウエツクス−50W×8(H型)1を充
填したカラムに通液し、ガラクトスタチンを吸着
させ、樹脂を十分水洗いした後、0.5N塩酸で溶
出する。
溶出液のβ−ガラクトシダーゼ阻害活性区分を
分画採取し、1.5のガラクトスタチン溶液を得
る。この溶液のβ−ガラクトシダーゼ阻害活性は
238100IU/ml(IC50=0.0010mcl/ml)である。
実施例 3
実施例2で得られたガラクトスタチン溶液を減
圧下1/5容(300ml)に濃縮する。この濃縮液に2
倍容(600ml)のエタノールを添加し析出する不
溶物を濾過により除去する。得られた濾液を更に
減圧濃縮し、100mlとする。
この溶液を直ちに弱塩基性陰イオン交換樹脂ア
ンバーライトIRA−47(OH型)(商品名、ロー
ム・アンド・ハース社)1を充填したカラムに
注ぎ、水で展開溶出する。
溶出液のβ−ガラクトシダーゼ阻害活性区分を
分画採取し、1.1のガラクトスタチン溶液を得
る。この溶液を再度減圧下濃縮し、150mlとする。
この濃縮液を氷冷下、6%亜硫酸水(和光純薬
工業)70mlを添加し、更にエノタールを500ml添
加してガラクトスタチンの亜硫酸付加物を針状結
晶(5.54g)として析出させる。この結晶を60℃
の熱水100mlに溶解しエタノール400mlを添加して
再結晶をおこない、ガラクトスタチン亜硫酸付加
物の無色針状結晶4.85gを得る。この結晶のβ−
ガラクトシダーゼ阻害活性は51900IU/mg(IC50
=0.0048mcg/ml)である。
実施例 4
ガラクトスタチン亜硫酸付加物1gを水20mlに
懸濁し、更に強塩性陰イオン交換樹脂ダウエツク
ス−2×8(OH型)(商品名 ダウケミカル社)
10ml添加して攪拌する。
この溶液全量を前記と同じ樹脂ダウエツクス−
2×8(OH型)200mlを充填したカラムに注ぎ水
で展開溶出する。溶出液のβ−ガラクトシダーゼ
阻害活性区分を分画採取し、減圧濃縮し濃縮液2
mlに対してエタノール20ml添加して更に濃縮をお
こない白色沈澱を生成させる。この沈澱を濾取し
乾燥してガラクトスタチン遊離塩基の白色粉末
628mgを得る。この粉末のβ−ガラクトシダーゼ
阻害活性は71200IU/mg(IC50=0.0035mcg/ml)
である。
実施例 5
ガラクトスタチンラクタムの調製方法
ガラクトスタチン200mgを水5mlに溶解し、攪
拌しながら、4%ヨウ化カリウムを含む0.2Nヨ
ウ素溶液2ml添加して5分間反応する。反応液に
0.2N水酸化ナトリウム溶液3mlを添加して5分
間攪拌した後、再びヨウ素溶液2mlを添加し上述
の操作を6回くり返し、ヨウ素溶液計12ml、水酸
化ナトリウム溶液計18ml添加終了後、更に50分攪
拌し反応をおこなう。
この反応液全量を強酸性陽イオン交換樹脂ダウ
エツクス−50W×8(H型)20mlを充填したカラ
ムに注ぎ水30mlで展開溶出する。
この溶出液を攪拌しながら炭酸銀2gを添加
し、30分後、生成した沈澱を濾過により除き清澄
なる濾液を得る。
この濾液を更に強酸性陽イオン交換樹脂ダウエ
ツクス−50W×8(H型)10mlを充填したカラム
に注ぎ水20mlで展開溶出する。この溶出液は微酸
性を程する為、弱塩基性陰イオン交換樹脂アンバ
ーライトIRA−47(OH型)を添加して中和する。
樹脂は濾過により除き、清澄なる中性濾液を得
る。
この濾液を減圧下、濃縮し、エタノールを添加
してガラクトスタチンラクタムを針状結晶(105
mg)として析出させる。この結晶を1mlの水に溶
解し、エタノール15ml添加して再結晶をおこな
い、ガラクトスタチンラクタムの無色針状結晶
95.1mgを得る。この結晶のβ−ガラクトシダーゼ
阻害活性は43.5IU/mg(IC50=5.75mcg/ml)で
ある。
実施例 6
1−デオキシガラクトスタチンの調製方法
ガラクトスタチン200mgを50%メタノール5ml
に溶解し、二酸化白金40mg及び酢酸0.2mlを添加
して攪拌しながら常圧下水素気流を通じて接触還
元を5時間おこなう。反応終了後、遠心分離によ
り得られる上清を濾過により清澄な濾液を得る。
この濾液を減圧下濃縮した後、強塩基性陰イオ
ン交換樹脂ダウエツクス−2×8(OH型)25ml
を充填したカラムに注ぎ水で展開溶出する。溶出
液のβ−ガラクトシダーゼ阻害活性区分を分画採
取し、減圧濃縮して油状物質を得る。
この油状物質をメノタール1mlに溶解し、アセ
トン2mlエーテル10ml添加して白色沈澱を生成さ
せる。この沈澱を濾取し、乾燥して1−デオキシ
ガラクトスタチンの白色粉末107mgを得る。この
粉末のβ−ガラクトシダーゼ阻害活性は
2170IU/mg(IU50=0.115mcg/ml)である。
発明の効果
ガラクトスタチンは酸性及び中性溶液中では非
常に安定でありほとんど分解変化せず、GT−
2558も酸性および中性溶液では比較的安定であ
る。しかし塩基性溶液中においてGT−2558は急
速に褐変分解し、シリカゲル薄層クロマトグラフ
イーでGT−2558と異なる原点からのテーリング
物質の増加が認められるほど非常に不安定である
のに対してガラクトスタチンは、わずかにテーリ
ングが認められる程度である。ガラクトスタチン
の粉末を冷所4℃に保存する限り変色せずβ−ガ
ラクトシダーゼ阻害活性の低下もおこらず、長期
間安定であるのに対して、GT−2558はわずかで
あるが、黄着色しながら変化して、β−ガラクト
シダーゼ阻害活性も徐々に低下する。さらにガラ
クトスタチン類とGT−2558の安定性の差は、放
線菌の培養濾液からの回収率の差にも現れる。以
