Description
本発明は、プラスミノーゲン活性化剤に関し、
更に詳しくは適当な培地中でプロTPA生産細胞
培養物を培養し、ついでプロTPA(および要すれ
ばTPA)を回収することを特徴とするTPA/プ
ロTPAカツプルをベースとする精製プラスミノ
ーゲン活性化剤の製法に関する。TPA/プロ
TPAカツプルは薬理製剤に用いられる。
本明細書で用いる「TPA/プロTPAカツプ
ル」なる用語は、例えばボウズ(Bowes)メラ
ノマ細胞組織培養から回収可能であり、以下に規
定される組織プラスミノーゲン活性化剤(TPA)
または/および以下で規定されるその前駆物であ
るプロTPAのどちらかまたは両方の組み合せを
意味するものとする。
TPAは、組織プラスミノーゲン活性化剤
(tissus plasimnogen activator)に対して、当
該技術分野で用いられる略語であり、例えばボウ
ズ(Bowes)メラノーマ細胞において同定され
た二重鎖型プラスミノーゲン活性化剤について用
いられる。プロTPAはTPA前駆物(precursor
of TPA)の、略語であり、即ちTPAと同じ分
子量を有しかつある酵素により二重鎖TPAに転
換される一重鎖化合物である。従来技術文献によ
れば、一重および二重鎖形状の両方は実質上同一
の活性を有するとされている。本発明は、その観
点からは全く異なる知見を含んでいる。
プラスミノーゲン活性化剤は、プラスミノーゲ
ン(プラスミン前駆物)への作用によりプラスミ
ンを生成する物質である。そして、プラスミンは
フイブリン(または凝血)に作用してフイブリン
を液化または溶解させる。プラスミンはフイブリ
ン前駆物であるフイブリノーゲンの溶解現象も起
こす。
前述の効果は、自然のフイブリン溶解系におい
て大切な役割を果たす。プラスミノーゲン活性化
機構を通して局部的なフイブリン溶解または蛋白
溶解活性を生じさせることが望まれる血栓症また
は他の病状の処置のためのプラスミノーゲン活性
化剤の治療投与においてもプラスミノーゲン活性
化剤は用いられる。
プラスミノーゲン活性化剤は、生体外のフイブ
リン溶解を含む科学テスト、診断テストまたは病
理学テストのための試薬としても使用することが
できる。
2つのヒトプラスミノーゲン活性化剤が広範囲
に使用されている。第1の活性化剤は、非蛋白溶
解機構により作用するバクテリア蛋白、ストレプ
トキナーゼである。第二の活性化剤は、尿または
培養腎臓細胞から得られるプロテアーゼ、ウロキ
ナーゼであるが、ウロキナーゼは他のほとんどの
種類の哺乳類組織において生じることが知られて
いる。この2つ化合物は、その作用が凝血におけ
るフイブリンに関係したプラスミノーゲンに限定
されないという治療上の欠点を有する。これらの
化合物が一般に血液循環においてプラスミノーゲ
ンに作用し、結果としてフイブリンの前駆物であ
るフイブリノーゲンの広範囲の溶解に伴つて広範
にわたつてプラスミンを発生させる。これによ
り、血液の非効果的な通常凝集の結果として、出
血状態が導かれる。更に、ウロキナーゼは、単離
および製造に高くつくという不利を有し、現在用
いられている方法によつては低い収量でしか得ら
れない。この不利は、フイブリン溶解治療以外の
ウロキナーゼの貴重な用途にもあてはまり、この
ような用途での貴重な市販品であるウロキナーゼ
の入手可能性を限定する。ストレプトキナーゼ
は、異種蛋白であり、患者に免疫反応を誘引す
る。生ずる抗体はプラスミノーゲンに対するスト
レプトキナーゼの作用を中和し、よつて治療効能
を減少させる。
一般に「組織プラスミノーゲン活性化剤」。本
明細書では「TPA」と略称する第三のプラスミ
ノーゲン活性化剤も存在することが知られてい
る。この酵素はたいていのヒト組織に見られ、生
体外で培養されるヒトメラノーマ細胞の特性分泌
生成物でもある酵素と同一であるかまたは区別が
つかない。TPAはウロキナーゼが行なうのとほ
とんど同じようにプラスミノーゲンからのプラス
ミンの生成反応を触媒するが、多くの重要な点で
ウロキナーゼと異なつている。この2つの酵素
は、科学的に異なつている。分子量が異なつてお
り、一方の酵素は他方の酵素の抗体と反応しな
い。TPAの触媒作用はフイブリンにより高めら
れるが、ウロキナーゼはそうではない。TPAは
フイブリンに結合するという大切な性質を有する
が、ウロキナーゼは結合しない。これらの事実
は、C Dingeman C.Rijken等、“Purification
and characterisation of the plasminogen
activator secretedby human melanoma cells
in culture”(J.Biological Chem.,256,No.13,
7035−7041)という最近の論文に記されている。
この論文は、普通のヒト組織からと同様に培養ヒ
トメラノーマ細胞からTPAを製造することをも
記載している。精製方法は洗剤存在下で亜鉛キレ
ートアガロース、コンカナバリン−Aアガロース
およびセフアデツクスG−150のクロマトグラフ
イーから構成される。
フイブリンにTPAの結合する傾向、フイブリ
ンの存在下おおいに高められたTPAのフイブリ
ン溶解作用、およびウロキナーゼならびにストレ
プトキナーゼと比較した場合フイブリンの不在下
でプラスミノーゲン活性化剤として比較的劣る
TPAの作用が積み重なつて、TPAがヒトの血栓
治療のプラスミノーゲン活性化剤に選択される。
フイブリンとTPAの間のかなりの程度の相互作
用は、凝血部分にプラスミン生成を局在化させ、
ウロキナーゼまたはストレプトキナーゼを使用す
る場合に見られるような無差別なプラスミノーゲ
ン活性化の影響を静めるか、または回避する。更
に、ストレプトキナーゼは免疫化学的にヒトに対
して異質蛋白であるが、TPAはそうではない。
W.Weimar等、“Specific lysis of an
iliofemoral thrombis by administration of
extrinsic(tisse type)plasminogen activator”
(Lancet,November 7,1018(1981))という
最近の報告は、ヒトの血栓症の処置における
TPAの臨床的有用性を試験している。
P.Wallen等(Prog.Chem.Fibrinolysis
Thrombolysis5,16−23(1981))によれば、
TPAは2つ異なつた形状で生じ、1つは一重鎖
形状であり、この一重鎖形状はプラスミンまたは
トリプシンによりジスルフイドブリツジで結合す
る二重鎖に転化する。この著者らは、この二重鎖
を一重鎖の分解生成物と考えた。二重鎖形状は、
フイブリン溶解テスト系(生体外)において一重
鎖形状よりフイブリン溶解性について効果的であ
ると言える。アプロチニンは、唯一のまたは優先
する形状として一重鎖形状が回収させる様に、二
重鎖形状への一重鎖の転化を妨害する。両方の形
状は組織プラスミノーゲン活性化剤であると考え
られる。
この研究については、Dingeman,Rijken,
HoylaertsおよびCollenの更に最近の出版物(J.
Biol.Chem.,257.2920−2925(1982),ヨーロツパ
特許出願0041766号)が参照され、彼らは、両者
の「プラスミノーゲン活性化性質は……同じであ
る。」とプラスミノーゲン活性化剤として両方の
形状を記載している。一重鎖形状は、フイブリン
凝血に吸着された場合、フイブリン溶解が「主に
二重鎖誘導体を通して起こる」ようにすぐ二重鎖
形状に転化することを彼らは示唆した。彼らは、
「この転化は、フイブリン溶解の調節における役
割を果たしていると思えない。」と結論している。
彼らの発見によれば、「二つの分子形状は同じ活
性(溶解時間)を持ち」、かつ「両方の分子形状
の触媒効果は実質的に同じであるので、一重鎖形
状は活性が低い前駆物であるとか、二重鎖形状は
活性が低い分解生成物であるとは考えられない」。
前述の結論が全く正しいかそうでないにしても、
両方の「分子形状」が生体外の溶解と同じくらい
治療上の有用性を有することは明らかである。
実際に、本発明者らの最近の研究結果所見によ
ると、前述の結論がある限定を必要とすることは
明らかである。プロTPAは本当の意味でTPAの
前駆物であるが、全く不活性な前駆物ではない。
プロTPAの固有活性はTPAの約1/10であるの
で、これだけに基づけば、プロTPAはその最大
限の治療効果を発現するためには予めTPAに転
化せねばならないのでプロTPAよりもTPAを治
療目的で投与することの方が好ましいと即座に考
えられる。
実際、ヨーロツパ特許出願0041766号には、プ
ロTPAと同様にTPAを多分表わす2つの化合物
が記載されているが、後者の化合物だけがフイブ
リン溶解、フイブリノーゲン溶解および血栓溶解
効果に関して特色がある。プロTPAについての
このような記載および薬理組成物としてのプロ
TPA使用に関する特別な記載はない。化合物が
その特徴を発揮するような薬理的使用方法は、二
重鎖形状、即ちTPAに対してだけ特許請求され
ている。唯一の出版された実際の治療における物
質の使用において、二重ストランド形状TPAを
使用している(Weimer et al.,The Lancet,
Nov.7,1018−1020(1981))。
前述の従来技術によれば、商業的規模で二重鎖
TPAおよび本明細書中で「プロTPA」と言つて
いる一重鎖形状物を容易に製造する方法は知られ
ていなかつた。
本発明者らの最近の提案(南アフリカ特許出願
第82/9168号およびこれに基づく他国の出願であ
り、該出願は現時点で当該技術水準の一部ではな
い。)によると、商業的規模でたやすく製造する
方法が提出されている。
本発明の目的は、特に精製プラスミノーゲン活
性化剤製造のために溶液からTPAおよび/また
はプロTPAの選択的可逆的吸着の新規で比較的
簡単な方法を提供することである。
本発明の特別な目的は、要すれば精製TPA、
しかし好ましくは精製プロTPAおよびプロTPA
が主であるTPAとプロTPAの精製混合物を製造
する新規な方法を提供することである。更なる目
的は、TPAおよびプロTPAをウロキナーゼから
分離するのを可能にする前述の方法および所望な
らウロキナーゼの分離回収に適用される方法を提
供することである。
要すればTPA、しかし好ましくはプロTPAま
たはプロTPAが主である両者の混合物の製剤を
提供することも目的である。
TPAを凌ぐプロTPAの以前には知られていな
かつた性質および有益性に基づいた薬理製剤であ
り、該製剤は血栓症および塞栓症の治療において
有益に使用されるだけでなく、このような疾病の
予防も行なう薬理製剤を提供することは特別な目
的である。
本発明の一つの要旨は、TPA/プロTPAカツ
プルをベースとする精製されたプラスミノーゲン
活性化剤の製法であつて、プロTPA生産細胞培
養物を適当な培地中において培養するにあたり、
細胞をプロTPAからTPAへの変換を触媒する酵
素の不存在下、または該酵素が存在した場合には
該酵素を阻害するのに適した阻害剤の存在下に培
養して、カツプルが50〜100%プロTPAおよび残
部のTPAで示される収穫液を生産し、溶解した
プロTPAおよび不純物を含む収穫液を、エリス
リナ ラテイツシマおよび他のエリスリナ種の種
子中に生成し、かつトリプシン、プラスミンおよ
びTPAの阻害剤であるが、ウロキナーゼには作
用しない型の固定化クーニツツ型阻害剤を含んで
成る親和試薬と密接に触媒させ、これにより選択
的にプロTPAおよび存在するTPAを媒体から吸
着し、プロTPAおよび存在するTPAを負荷した
親和試薬を水性媒体および非吸着不純物との接触
から除き、プロTPAおよび存在するTPAを親和
剤から脱着し、ついで脱着したプロTPAおよび
要すればTPAを回収することを特徴とする製法
に存する。
ウロキナーゼをも生産する細胞培養を使用する
ならば、収穫液中のプロTPAおよび存在する
TPAはウロキナーゼにより不純になつており、
ウロキナーゼは非吸着不純物とともに除かれてそ
れから要すれば貴重なる生成物として回収され
る。
エリスリナ ラテイツシマ(Erythrina
Latissima、広葉エリスリナ)および他のエリス
リナ種の種子は、2つの蛋白分解酵素阻害剤を含
む(Francois J Joubert et al.,Z.Physiol.
Chem.,362,531−538)。この2つの阻害剤は、
上記文献ではDE−1およびDE−3よ呼ばれてい
る。DE−1は牛のキモトリプシンを阻害し、牛
のトリプシンを阻害しないが、DE−3は牛のト
リプシンを阻害し、牛のキモトリプシンを阻害し
ない。DE−3はクーニツツ(Kunitz)型酵素阻
害剤としての性質およびトリプシン、プラスミン
ならびにTPAの阻害剤としての性質を有するが、
ウロキナーゼに効果を有さない。DE−3は、何
百もの酵素阻害剤のスクリーニングの後、TPA
とウロキナーゼを区別できる唯一の阻害剤として
選ばれた。この阻害剤が、例えば自体既知の方法
で固定化されるならば、以前は知られていなかつ
たTPAの前駆物質、プロTPAと同様にTPAを吸
着により選択的に抽出して親和試薬を形成する。
しかし、非吸着不純物は水溶液中に残る。DE−
3はプラスミンの阻害剤であるので、TPAおよ
び/またプロTPA供給源として適当な水溶液に
普通存在するプラスミン作用によるプロTPAの
TPAへの転化を阻害する。従つて、この固定化
された阻害剤は、プロTPAの吸着剤として用い
る場合、顕著なる性質を有する。DE−3の更に
完全は化学特性は、前述のJoubertらの文献に記
述されている。
ウロキナーゼは水溶液中に存在するとしても吸
着されない。従つて、TPAおよび/またはプロ
TPAがウロキナーゼにより汚染されている場合
にも本発明の方法において親和試薬を使用でき、
ウロキナーゼは非吸着不純物とともに除かれる。
このことは、TPAおよび/またはプロTPAがウ
ロキナーゼとの混合物として生じている種々の組
織のような多数の供給源からプロTPAおよび
TPAを回収するのにこの方法を適用できること
を意味している。
しかし、プロTPAおよび要すればTPAを水性
媒体から抽出するに用いられる特に好ましい水性
媒体は、細胞の分泌生成物におけるプロTPAお
よび0〜50%のTPAを含むヒトメラノーマ細胞
の組織培養から得られる収穫液である。
好ましい態様に従えば、プロTPAおよび存在
するTPAは、これらを含む水性媒体を親和試薬
層へ通過させることより吸着され、次いで層を洗
浄し、プロTPAおよび存在するTPAを脱着さ
せ、脱着したプロTPAおよび存在するTPAを層
から洗い出す。層はカラムの形状をしていてよ
い。
脱着は、好ましくは約1〜3モル/(例えば
1.4〜2モル/、好ましくは1.6モル/)のカ
リウムチオシアネート水溶液を用いて行う。この
場合、該化合物はTPAが安定であるPHにおいて、
好ましくはほぼ中性のリン酸緩衝塩水(例えば
NaClについては0.3〜1モル/)、好ましくは
約0.4モル/)に溶解され、好ましくは安定化
量の適当な海面活性剤(例えば0.1%のTritonX
−100またはTween)を存在させる。脱着は、カ
リウムチオシアネート以外のカオトロピツク
(chaotropic)脱着剤によつて行つてもよい。脱
着は、低いPH、例えば2〜3.5、好ましくは2.8〜
3で行われてもよい。他の適当な脱着条件は、高
濃度の塩水および/またはアルギニンの存在およ
び/またはベンズアミジンの存在である。このよ
うな脱着条件は、当該技術者には既知であるの
で、詳細には記述しない。
本発明に従つても、プロTPAを含まないTPA
を回収するのが所望される場合、プロTPAは、
前期の方法段階で作られていなくても、例えば水
溶液中のプロTPAをプラスミンまたはプロTPA
に等価効果を有する酵素にさらすことによつて、
容易に触媒的にTPAへ転化することができる。
まだ同定されていないそのような酵素は、TPA
へのプロTPAの転化がフイブリンの存在下で起
こるのと同じ理由で、例えばフイブリン中で生じ
る。カリクレインもまた、効果は低いが、この効
果を生じる。
本発明に従つて製造されたプロTPAは、これ
が少しのプラスミノーゲン活性化剤性質しか有さ
ない一重鎖化合物であるが、TPA(分子量72000
ドルトン)が二重鎖化合物であり、プロTPAの
約10倍の強さである著しいプラスミノーゲン活性
化性質を有するという点において、TPAと異な
つていることが見い出された。プロTPAがTPA
と同じ分子量を有する。
一重鎖形状における弱い活性の故および二重鎖
TPAがずつと活性であるという知見の故、生体
内で凝血溶解過程でより効果的なのはTPA形で
あるので、従来技術において薬理組成物中に
TPA形(即ち、二重ストランド分子形状)のプ
ラスミノーゲン活性化剤を使用する強い動機が通
常あつた。この考えは、プロTPAは非常に弱い
活性であるという知見により確固たるものにされ
る。
しかし、驚くべきことに、プラスミノーゲン活
性化剤のフイブリン溶解効果が利用される血栓症
の処置および他の治療のための薬理組成物の活性
成分として、TPAを使用するよりプロTPA形の
プラスミノーゲン活性化剤(好ましくは少なくと
も主としてプロTPAを含み、更に好ましくは全
部がプロTPAである。)を使用することに確信で
きる理由があることが実験証拠により立証され
た。この理由は、プロTPAがTPA形よりもフイ
ブリンへの吸着のより高い親和性を有すること、
およびプロTPAがフイブリンへ吸着すると非常
に早くTPAに転化され、次いでこれを必要とす
る正にその場所で、即ち凝血部分で最大限の所望
の活性が得られるということである。この発見か
ら下される結論を以下に示す。
(a) プロTPAは、より迅速にかつ完全に凝集フ
イブリン部分、すなわち活性化剤を実際上必要
とする場所に吸着される。その場所でプロ
TPAはフイブリンにある蛋白分解酵素により
TPAに迅速に転化され、TPAの最大限のプラ
スミノーゲン活性化剤活性は凝血の実際部分で
直ちに利用されるようになる。
(b) プロTPAは、フイブリン溶解をもたらすこ
とにおいてTPAより選択的である。なぜなら、
プロTPAはフイブリンに結合するまで比較的
不活性であり、その場でTPAに転化すること
によつて最大限に活性になるからである。
TPAには、フイブリン溶解効果を必要とする
血栓部分でなくて血流でのプラスミノーゲンを
活性化させる約10倍の可能性がある。このかな
りの可能性は、フイブリン部分へ吸着する
TPAの低い親和性によつて更に増加される。
(c) 血流において激しく減少された活性のため、
未来の血栓または塞栓が起こる危険性のある場
合の予防にプロTPAを用いることを確信をも
つて勧めることができる。
(d) 一方、急性の血栓閉塞において比較的多い投
薬量を用いる場合、副作用の危険性を減少させ
る。
(e) TPAは生体内で短い半減期を有する。プロ
TPAの性質は、プラスミノーゲン活性化剤の
利用をより効果的にすることである。一方では
プロTPAは血流にある時の無効な反応によつ
て消費される可能性が低く、他方ではプロ
TPAは、プロTPAが有用であり得る凝血部分
に迅速に「捕捉」される可能性が高い。
前期の知見における大切な概念に基づいて、本
発明は、プラスミノーゲン活性化機構を通して局
在的フイブリン溶解または蛋白溶解活性を生じさ
せることが望まれる血栓症、塞栓症または他の疾
病の治療または予防のための薬理組成物であり、
該薬理組成物は、50〜100%のプロTPAおよび0
〜50%のTPAから成るTPA/プロTPAカツプル
と表示されるプラスミノーゲン活性化剤成分を含
んで成る。
TPA/プロTPAカツプルは、好ましくは、プ
ロTPA30〜70%およびTPA0〜30%、更に好ま
しくはプロTPA80〜100%およびTPA0〜20%か
ら成る。
本発明の薬理組成物は、血栓閉塞がすでに生じ
た場合の血栓症、塞栓症の処置に適用されてよ
い。
本発明は、TPA/プロTPAカツプル1〜10mg
の投薬単位で患者に静脈投与するか、または
TPA/プロTPAカツプル注入速度0.5〜2mg/時
でプラスミノーゲン活性化組成物を静脈注入する
ことを特徴とする血栓閉塞を含む疾病の予防にも
特に適用できる。このような処置は、危険状態に
ありながら血栓症または塞栓症の徴候を全く示さ
ない患者に施されてよい。例として、静脈血行停
止のためにひどい静脈血栓症および塞栓症を特に
患いがちである寝たきり患者および衰弱患者が挙
げられる。他の例として、冠状動脈血栓閉鎖が起
こりそうな不安定な狭心症の患者がある。別の例
として、血栓部位が存在し、脳塞栓の危険性のあ
る心臓弁膜症患者がある。例として、心膜腔また
は大血管は、凝血が移動して心臓血管閉塞を起こ
す(発作)危険が存在する様な部位または損傷を
有する患者がある。例として、補綴血管弁膜、心
房細動およびやや急性のバクテリア性心内膜炎な
どを有する患者もある。本発明薬理組成物は、大
静脈へ静脈ラインを挿入する場合および刺激剤溶
液を注入する場合、静脈の血栓閉塞および/また
は塞栓を生じそうである静脈治療または非経口栄
養を受ける患者の予防処置に有用である。
投与のため連続静脈注入は好ましいが、断続的
な一気の静脈投与は2〜24時間毎に例えば1〜10
mgの前記投薬量で適用されてよい。
本発明は、前記用途に使用するために特に調製
および適用される種々の薬理組成物を提供する。
本発明により、精製TPAまたはプロTPAを分
離してまたは混合物として調剤できる。
本発明により、TPAおよびプラスミンおよ
び/またはプラスミノーゲンおよびプラスミノー
ゲン活性化剤を含む水性媒体からTPAを回収で
きる。このために、水性触媒体中のプラスミンを
阻害して、プラスミンが阻害されている間に水性
媒体からプロTPAを除去することが要求される。
これらのことから、存在するTPAは、阻害され
ねばならないプラスミンの形成を起こすプラスミ
ノーゲン活性化剤として作用することが指摘され
る。
本発明に従つて、前記方法において、プロ
TPAへの親和性を有するとともにプラスミンの
固定化阻害剤を含んで成る親和試薬によつて水性
媒体からプロTPAを吸着し、次いで親和試薬か
らTPAを脱着してプロTPAを精製状態で回収す
る。このように回収されたプロTPAは次に、殺
菌消毒され、生理的に適合可能な安定剤により安
定化され、静脈注入できる水性溶液の形にされ
る。
本発明に従つて親和試薬として固定化形状で使
用されるDE−3阻害剤は偶然にプラスミンの阻
害剤であり、しかもプラスミンはTPAおよびプ
ロTPAが回収される媒体中に普通存在し、プロ
TPAをTPAに転化する性質を有するので、プロ
TPAを同時に単離するためのDE−3阻害剤の使
用はプロTPAをプロTPA形状に保つ役目を果た
す。
本発明の他の要旨に従うと、両方のプラスミノ
ーゲン活性化剤(そのもの自体または前駆物とし
て)を含む水性媒体からウロキナーゼと分離して
またはとともにTPAおよび/またはプロTPAを
選択的に吸着することは可能である。これは、一
方でウロキナーゼ無含有精製TPAおよび/また
はプロTPAまたは他方でTPAおよびプロTPA無
含有精製ウロキナーゼまたは両方を含む精製プラ
スミノーゲン活性化剤の製造に適用される。これ
は、このような水性媒体を、エリスリナ ラテイ
ツシマおよび他のエリスリナ種の種子中に生成
し、かつトリプシン、プラスミンおよびTPAの
阻害剤であるが、ウロキナーゼには作用しない型
の固定化クーニツツ型阻害剤を含んで成る親和試
薬と接触させ、これにより選択的にTPAおよ
び/またはプロTPAを媒体から吸着し、TPAお
よび/またはプロTPAを負荷した親和試薬を、
TPAおよび/またはプロTPAがなくなつたが、
まだウロキナーゼを含む水性媒体との接触から除
去し、TPAおよび/またはプロTPAのなくなつ
た水性媒体からウロキナーゼを回収することによ
り実施される。この方法に適当な水性媒体は、媒
体の両方のプラスミノーゲン活性化剤を含むの
で、任意の哺乳類組織細胞の水性抽出物であつて
よく、またそのような細胞の分泌物を含む媒体で
あつてもよい。もう一度述べるが、吸着された
TPAおよび/またはプロTPAは脱着、回収さ
れ、もし所望なら選択性親和試薬に吸着される前
または後にプロTPAはTPAに転化してよい。
ウロキナーゼは水性媒体から適当な方法で、例
えばウロキナーゼに親和性を有する親和試薬を用
いて水性媒体から吸着されることにより、回収さ
れてよい。そのような親和試薬は、ウロキナーゼ
を上まわつてTPAに選択性を有さない固定化阻
害剤であつてよい。親和試薬として自体既知の方
法で適当に固定化された抗ウロキナーゼ抗体を使
用することも可能である。
本発明におるクーニツツ型阻害剤は、エリスリ
ナ ラテイツシマおよび他のエリスリナ種の種子
中に生成し、かつトリプシン、プラスミンおよび
TPAの阻害剤であるが、ウロキナーゼには作用
しない型であり、例えば担体に共有結合されるこ
とより固形担体に結合されて固定化されている。
例えば、阻害剤は、当業者に既知である方法で臭
化シアン活性化アガロースに結合されてよい。し
かし、阻害剤が結合する表面の性質の化学性質に
依存するが、他のカツプリング剤および他の固形
担体が使用されてよい。例としては、次のものが
挙げられる。
(a) 遊離カルボキシル基へ遊離アミノ基を結合さ
せるカルボジイミドカツプリング剤。
(b) 前もつてアミノアルキル形に転化したアガロ
ースへNH2基を結合させるグルタルアルデヒ
ドカツプリング剤。
(c) アルデヒド官能基を生成し、次にPH4〜6で
阻害剤のアミンと反応してシツフ塩基を形成
し、次いでナトリウムボロハイドライドまたは
ナトリウムシアノボロハイライドによつて還元
する過ヨウ素酸塩活性化アガロース。
(d) アガロースは、ヒドラジドスクシニル誘導体
に転化してよく、この為、カルボキシル配位子
はEDC、またはジアゾ化によつてアミンと結
合してよい。
(e) セルロース繊維または粒子はは、ヒドラジド
誘導体に転化され、Parikhらの方法
(Methods in emzymology,34,77〜102,
editors W B Jakobi and Wilcheck
Academic Press N Y)に従つて用いられ
てよい。
(f) ポリアクリルアミド粒子は、直接グルタルア
ルデヒド法またはp−アミノベンズアミドエチ
ル誘導体もしくはヒドラジド誘導体のジアゾ化
によつて結合されてよい。
(g) 蛋白質、例えばアルブミンまたはトリプシン
またはゼラチンは、例えばヒドラジドの形で結
合してよい。
(h) 最後に、蛋白質で被覆され得るガラス粒子
(または他の固形物)も使用されてよく、蛋白
質として、南アフリカ特許出願第81/4481号、
および海外対応特許である西ドイツ特許出願
P3224837.7号、特願昭57−116086号、イギリス
特許出願8219070号、アメリカ合衆国特許出願
392111号に記載されているカツプリング法に類
似した方法によつて阻害剤に結合し、架橋して
よいゼラチンが挙げられる。
前記の方法は、当該業者に既知であり、かつ/
または示した文献に記載されているので、更に詳
しくは述べない。
本発明の親和試薬は、例えば前に詳しく述べた
ような方法で、TPAおよび/またはプロTPAあ
るいはTPAおよび/またはプロTPAを含んで成
る製剤を製造するのに用いられてよい。しかし、
親和試薬も、診断または分析の試薬としての有用
性を有しうる。
本発明の範囲は、TPAとプロTPAの精製混合
物および特にプロTPAのTPAに対する比が1:
1を越える混合物と同様に、本発明に従つて精製
されたプロTPAを含むものである。
高い比、例えば9:1またはそれ以上の高い比
を得るために、プロTPAとTPAへの転化を接触
する酵素不在の適当な媒体中、またはこのような
酵素(これを含む媒体として血清がある)ならび
にプロTPAのTPAへの転化を触媒する酵素(特
にプラスミン)を阻害するのに適用される適当な
阻害剤を含む媒体中でプロTPA生産細胞培養
(例えばメラノーマ細胞)によりプロTPAを生産
することが提案される。適当な阻害剤は、大豆か
ら誘導されたトリプシン阻害剤またはα−l−ア
ンチプラスミンである。アプロチニンが代わりに
使用され得る(既知のように)。次いで、プロ

本発明の範囲は、生理的に適合し得る注入媒体
に溶解され、本発明の方法により調製または精製
されたTPAおよび/またはプロTPAを含んで成
る薬理組成物に及ぶ。本発明の範囲は、例えば診
断または病理的または一般科学の目的のため生体
外でフイブリン溶解を実施するフイブリン溶解製
剤にも及ぶ。
唯一の活性成分としてTPAもしくはプロ
TPA、または所望の比でTPAならびにプロTPA
を含んで成る前述の組成物を生産するのに実用的
に随意に適用され得ることは本発明の有益性と考
えられる。
以下に実施例を示すことにより、本発明を更に
詳しく説明する。
実施例 1
親和試薬の調製:−
DE−3阻害剤を以下のように調製した:
エルスリナ ラレイシマ種子をJoubertらの方
法に従つて採集し、加工した。種子をすりつぶ
し、脱脂し、0.5モル/NaCl水溶液により10℃
で一夜抽出した。抽出物を遠心分離し、DE−3
を硫酸アンモニウム沈澱により上澄みから回収
し、続いてセフアデツクスG50,DEA−セルロ
ースおよびDEA−セフアローズのクロマトグラ
フイーにかけた。最終精製物は、0.1%ドデシル
硫酸ナトリウム(SDS)含有15%ポリアクリルア
ミドゲルの電気泳動に付した場合、みかけ分子量
22000ドルトンの単一バンドとして移動した。
精製DE−3(26mg)を市販臭化シアン活性化ア
ガロース5mlに通常方法で結合させた(調製物の
使用方法も教示するPharmaciaにより市販され
ているセフアローズ4b)。親和試薬、NaCl0.4モ
ル/,0.1%TritonX−100(当該技術において
普通使用される市販界面活性剤)および0.02%ナ
トリウムアジド安定剤を含むPH7.4のリン酸緩衝
液に対して平衡化された。この親和試薬を使い捨
てプラスチツクシリンジの円筒から造られた5ml
のカラムに充填した。
実施例 2
TPAおよびプロTPAの調製および精製:−
TPAおよびプロTPAを含む媒体を以下のように
して製造した。ボウス(Bowes)細胞RPMI−
7272(コロラド、デンバー在、デンバー病院から
入手可能。J.Biol.Chem.,256,7035−7041に記
述されている。)として知られる細胞系列のヒト
メラノーマ細胞を、10%熱不活性(56℃,30分
間)退治牛血清(FCS)および抗生物質(ペニシ
リン300μg/ml、ストレプトマイシン200mg/ml
およびチロシン10μg/ml)を補充したRPMI−
1640組織培養媒体に付着単一層として成長させ
た。集合している(約4×105細胞/cm2)細胞を
トリプシン分解に付し、5×105細胞(100mm皿で
再植する。媒体を吸い出した後、細胞をトリスダ
ルベツコ塩水(Na2HPO40.1ミリモル/,
KCl5ミリモル/,NaCl0.14モル/、トリス
HCl24.8ミリモル/を含むPH7.4の液)中の0.25
%トリプシン液中で37℃で5分間培養した。分離
した細胞を穏やかにピペツトで移動させることよ
り分散させ、プロテアーゼを中和するために分散
液をFCSを含む媒体を含む等体積の媒体に加え
た。350Gで5分間の遠心分離により細胞を洗い、
新しい皿に再植した。
この原培養物から、75cm2または150cm2の組織培
養フラスコ中で付着単一層として細胞を育成し、
10%FCSに入つた媒体中で細胞をほぼ融合するま
で育てた。プロTPA:TPAの最終比を1:1以
上にするために、血清の蛋白質溶解酵素(即ちプ
ラスミン)を実施例5で更に詳しく記述している
ようにもしくはアプロチニンで阻害するか、また
は除去した。培養物を一度洗い、血清を含まない
媒体20mlで被覆した。24時間後、媒体を集め、新
しい媒体を加えた。
全細胞および細胞片を除くため、収穫液を
2000rpmで5分間遠心分離した。溶液をTritonX
−100(0.1%)またはTween(0.1%)で安定化し、
氷酢酸で酸性化し(PH5.5〜6.0)、−20℃で貯蔵し
た。
収穫液2を、NaClに関して0.4モル/に
し、0.45μm膜で濾過した。濾過収穫液を室温に
おいて流速45ml/時で親和カラムに適用した。4
℃で流出液を集め。プラスミノーゲン依存フイブ
リン溶解活性の存在を測定した。TPAまたはプ
ロTPAは検出されなかつた。
全収穫液をカラムに通過させた後、NaCl0.4モ
ル/および0.1%TritonX−100を含む6倍カラ
ム容量のリン酸緩衝塩水(PBS)でカラムを洗
つた。この方法により、TPAまたはプロTPAは
溶出しなかつた。
吸着した蛋白質は、カリウムチオシアネート
1.6モル/、NaCl0.4モル/および0.1%の
TritonX−100を含むPBSを用いて溶出された。
280nmの吸収を読むことより、蛋白質含量を測
定した。
溶出緩衝液にカリウムチオシアネートを加える
場合、全てのプラスミノーゲン依存フイブリン溶
解活性剤は溶出し、鋭いピークとして見られ、こ
のピークは蛋白質の小さなピークに一致する。
最も高い活性を有する分画を貯留して、−20℃
で貯蔵される6〜8mlの溶液を得た。この分画は
カラムに適用された活性の70〜80%を示す。低い
活性を有する分画は、別に貯留される。この両方
の貯留中の全回収活性は通常、90〜100%になる。
カリウムチオシアネート溶出液は、純粋なTPA
およびプロTPAの混合物に一致する分子量72000
ドルトンの蛋白質の単一バンドを含み、別のラン
における後者は全体の約39〜50%またはそれ以上
になる。
同一カラムをくり返して何度も使用すると、収
量は向上する。カラムは何度も再使用するのに効

実施例 3
TPAおよび/またはプロTPAを含む薬理製
剤:−
TPAおよび/またはプロTPAを含む貯留分画
を0.01容量%のTween80を含むNaCl0.3モル/
水溶液で透析し、−80℃で貯蔵する。投与前に濃
度をTPAとプロTPA75μg/ml(治療目的での
TPAの投与速度はプロTPAと同じである)およ
びNaCl0.3モル/に調節した。0.22μm濾過膜に
よる濾過により溶液を殺菌する。
溶液を投与速度3〜15mg/24時間、典型的には
7.5mg/24時間で注入し、静脈投与する。理想的
には投与速度は、処置の間、血中の全プラスミノ
ーゲン活性(前駆物を含む)レベルが0.6〜1、
好ましくは約0.8国際単位/mlになる様でなくて
はならない。そのような濃度での処置は血栓をき
れいにするのに効果的であるのがわかつており、
全身的フイブリン溶解は観測されない。その他の
毒性も特に観測されない。
しかし、プロTPAはフイブリンに結合してか
らTPAに転化するのみなので、血流中にある時
の実際的な活性は、全プラスミノーゲン活性化剤
が最大限に活性なTPA形にある場合に存在する
活性の10%程度の小さいものであることがわか
る。さらにプロTPAはTPAより早く、強くフイ
ブリンによつて吸着されるからである。無差別反
応が更に減少される。
実施例 4
メラノーマ細胞の大量培養:−
Rosswell Park Memorial Institute 培地
1640および10%FCSをつめたスピナーフラスコに
濃度5×105細胞/mlでメラノーマ細胞を接種し
た。フラスコを37℃に保ち、30rpmでマグネツト
撹拌した。フラスコ容積の1%/時の速度で媒体
をフラスコからポンプで外へ出し、5%CO2を含
む空気と平衡にある新鮮な媒体を同速度で入れ
る。ガラスウールで出口ガラス管を詰めた培養容
器中に細胞を保つ。回収した媒体をミリポアフイ
ルターで濾過する。
集めた液を低温殺菌し、実施例2に従つて培養
することができる。
実施例 5
プロTPA高生産メラノーマ細胞の培養:−
十分な大豆トリプシン阻害剤または血清プラス
ミンを阻害するα−l−アンチプラスミンを培養
媒体に加えて、実施例2を変形した。他の点で
は、実施例2をくり返し、最終生成物のプロ
TPAのTPAに対する比はほぼ9:1に増加し
た。実験の詳細は次の通りである。
ボウズ(Bowes)細胞培地は0.1%のTritonお
よび10KIU Trasylol(プラスミン阻害剤)を含
んでいた。元の収穫液は体積1960mlであつた。カ
リウムチオシアネート溶出液は体積20.8mlであつ
た。TPA+プロTPAの回収は100%であり、そ
のうち78%がプロTPAであつた。Trasylolを用
いない比較実験を実施すると、回収は98%であ
り、これは全部TPAであり、プロTPAは検出さ
れなかつた。
実施例 6
プロTPAとTPAの混合物で残存のTPAの除
去:−
TPAおよびプロTPAの溶液のジイソプロピル
フルオロリン酸を10ミリモル/にする。PHを8
に調節する。4時間後、TPAは永久に阻害され
る。それから残存する阻害剤を透析により除去す
る。
実施例 7
プロTPAのTPAへの転化:−
蛋白質100μg/ml含有の水溶液をプラスミン
5μg/ml含有の同体積のリン酸緩衝塩水と混合
し、20℃で16時間培養する。トデシル硫酸ナトリ
ウムを最終濃度0.1%になる様に加え、6%トリ
クロロ酢酸で蛋白質を沈澱させる。沈澱物をアセ
トンで洗い、1%ドデシル硫酸ナトリウムおよび
10%グリセロールを含むトリスHCl0.06モル/
のPH6.8の液に溶解する。
この処理の後、プロTPAは活性TPAへ完全に
転化したことが電気泳動により示された。
実施例 8
不溶化フイブリンへのプロTPAの選択性:−
種々の割合のプロTPA(残りはTPA)含有
TPA/プロTPAカツプルのサンプルについて、
各々の場合にフイブリンへ結合する物質の割合を
確証するために同一条件下で試験した。
TPA/プロTPAカツプルのサンプル中のプロ
TPAの割合(残りはTPA)を、プラスミンによ
る処理(プロTPAをTPAに転化するため)の前
後におけるサンプルの活性を測定することにより
決定した。その後、プロTPAはTPAの10%の活
性を有するという事実があるので、補正するため
に補正係数10%を適用した。0.1%TritonX−100
含有トリスHCl0.1モル/のPH8.1の液にサンプ
ルを希釈すると、プラスミノーゲン数μgの存在
下で、ヨウ素125でラベルされ、かつリンブロウ
エルズ(Limbro wells)の底を被覆していたフ
イブリンの約30〜50%が1時間のうちに0.2mlで
解放された。放射性同位ヨウ素でラベルされたフ
イブリンで被覆された4つのリンブロウエルズに
少量(0.2ml)の試料を4回加えた。0℃で1時
間培養後、そのうち2つのウエルをトリスHClで
3回処理し、結合していないTPAを除去した。
その後結合活性を決定した。この結果を洗浄して
いないウエルで得られた結果と比較した。結果を
第1表に示す。
結果は、プロTPAが実質的にTPAより完全に
