Abstract
メラ遺伝子、及び植物に一般的な除草剤、特にホスホノ
メチルグリシン系除草剤に対する強い耐性を与えるため
の植物中でのそれらの使用、これらの遺伝子により形質
転換された植物細胞、これらの細胞から再生した形質転
換植物並びにこれらの形質転換植物を用いる交雑育種か
ら誘導された植物の提供。 【構成】転写方向に、プロモーター領域、トランジット
ペプチド領域、グリフォセート耐性に対するコーディン
グ配列及びポリアデニル化シグナル領域を含み、トラン
ジットペプチド領域は、翻訳方向に、プラスチド−局在
化酵素をコードする植物遺伝子の少なくとも1つのトラ
ンジットペプチド、プラスチド−局在化酵素をコードす
る植物遺伝子のN−末端成熟部分の部分配列及びプラス
チド−局在化酵素をコードする植物遺伝子の第2のトラ
ンジットペプチドを含むキメラ遺伝子、及び除草剤グリ
フォセート−耐性植物の産生。
Description
チドDNA配列、新規キメラ遺伝子、及び植物に一般的
な除草剤、特にホスホノメチルグリシン系除草剤に対す
る強い耐性を与えるための植物中でのそれらの使用に関
する。本発明は、さらにこれらの遺伝子により形質転換
された植物細胞、これらの細胞から再生した形質転換植
物並びにこれらの形質転換植物を用いる交雑育種から誘
導された植物にも関する。
ォセート(sulfosate)またはフォサメチン(fosametin
e)は、ホスホノメチルグリシン系の広スペクトルの移
行性除草剤である。これらは本質的に、PEP(ホスホエ
ノールピルベート)に対し5-(エノールピルビル)シキ
メート-3-ホスフェートシンターゼ(EC2.5.1.19)即
ち、EPSPSの拮抗阻害剤として作用する。これらの除草
剤を植物に適用すると、これらは植物内を移行し、すぐ
に発育する部分、特に茎や根の先端に集まり、敏感な植
物を劣化させたり死滅させてしまう。

する植物遺伝子のトランジットペプチド、プラスチド−
局在化酵素をコードする植物遺伝子のN−末端成熟部分
の部分配列及びプラスチド−局在化酵素をコードする植
物遺伝子の第2のトランジットペプチドからなる。

(BAP)を含む30g/l 蔗糖を補ったMS培地上に新芽(sho
ot)を15日間誘導することからなる。このステージの間
に形成した新芽を、30g/l蔗糖を補ったがホルモンは含
まないMS培地上で10日間培養することにより成長させ
た。成長した新芽を剥がし、半分の量の塩、ビタミン及
び糖を含むがホルモンは含まないMS栽培培地で培養し
た。約15日後、深く根を張った新芽を土壌に植えた。
ナマイシンを含む30g/l 蔗糖、0.05mg/l ナフタレン酢
酸及び2mg/l BAPを補ったMS培地上で30日間成長させた
後、直径0.5cmの100個の葉状ディスクに外挿したカルス
重量を測定することにより耐性を測定した。これらの条
件下では、本発明のpRPA BL 410のキメラ遺伝子により
改質したタバコ植物の場合、カルス重量が34gであった
のに対し、第2のトランジットペプチドの無いキメラ遺
伝子により改質した植物の場合は、たったの12gであっ
た。

の植物を温室に移し、5葉期で0.6kg/ha グリフォセー
ト(Round up)に対応する投与量の水性懸濁液を噴霧す
ることにより処理した。21日後、形質転換しなかった対
照植物に対し植物の表現型実験を実施した。これらの条
件下では、pRPA-BL 410を用いて形質転換した植物の植
物毒性は無視できる程度であったのに対し、対照植物は
完全に枯れてしまった。さらに、第2のトランジットペ
プチドを有さない点で先のキメラ遺伝子と異なるキメラ
遺伝子を用いて形質転換した植物は、30%以上の死滅の
植物毒性を有していた。

対し耐性を示した。この処理で、形質転換しなかった植
物は枯れた。

- 【請求項1】 転写方向に、プラスチド−局在化酵素を
コードする植物遺伝子のトランジットペプチドをコード
する配列、プラスチド−局在化酵素をコードする植物遺
伝子のN−末端成熟部分の部分配列及びプラスチド−局
在化酵素をコードする植物遺伝子の第2のトランジット
ペプチドをコードする配列を含む、トランジットペプチ
ド領域植物DNA配列。 - 【請求項2】 第2のトランジットペプチドをコードす
る配列が、第1のトランジットペプチドをコードする配
列と同一植物から誘導される請求項1に記載のトランジ
