2. Description of the Related Art Clothes that are worn daily vary considerably depending on the type, individual differences, environmental differences, etc., but are formed in association with sweat and sebum secreted in large amounts from the skin surface and keratinization of cells on the skin surface layer. In addition to the dirt on the skin and dust from the outside, it is also contaminated by microorganisms such as bacteria and mold.
In particular, natural fibers such as cotton, wool, and silk, which have high hygroscopicity and have a well-developed fiber structure and have many voids, have significantly more bacteria attached than chemically synthesized fibers. From a hygienic point of view, it would be highly desirable if many bacteria that adhered to the worn clothing could be easily removed, along with other common stains, by regular washing. It is one of the original purposes of cleaning. The removal of bacteria adhering to the fabric by washing varies considerably depending on the type of bacteria and the growth state of the bacteria, but simply increasing the detergent concentration or the washing time does not substantially increase the disinfection efficiency of the fabric. On the other hand, by increasing the washing temperature, increasing the number of rinses, or drying the fabric with hot air, the sterilizing or sterilizing effect can be remarkably enhanced, and the effect can be further enhanced by combining these conditions. Is recognized (Minagawa; Antibacterial and antifungal, Vol.4, p.15)
(1976)). However, unlike the Western countries (washing temperature of 60 to 100 ° C), the most common method of washing clothes in Japan is low-temperature washing (room temperature to 40 ° C). Is difficult and leaves many problems from a hygienic standpoint. As a countermeasure from the viewpoint of cleaning contaminated bacteria adhering to clothes, a method of decomposing and removing microorganisms by adding a lytic enzyme to detergent can be considered. As the lytic enzyme, egg white-derived lysozyme is widely known, but the optimum pH is neutral to weakly acidic, and the bacterial species to lyse are limited (the lytic spectrum is narrow), and the surface activity is also high. It is not suitable as an enzyme to be added to detergents because it has low activity in the agent. In addition, properties different from lysozyme
Although some lytic enzymes having a lytic spectrum, an optimum pH, etc. have been reported, none of them satisfy the properties as a detergent enzyme. Enzymes of the genus Nocardiopsis (PCT Int.Appl.WO 91 10723) show lytic activity against a limited number of strains in detergents, but the optimum pH is around neutral and Considering the medium pH (around 10), it has disadvantages. For this reason,
It has an optimum pH on the alkaline side, and is also used as a detergent.
The appearance of a lytic enzyme having improved properties as a detergent enzyme, such as stability against various surfactants and a broader lytic spectrum, has been strongly desired.

[Means for Solving the Problems] The present inventors are capable of producing a lytic enzyme having novel properties, particularly an enzyme having a wide range of activity from Gram-positive bacteria to negative bacteria and having an optimum pH on the alkaline side. In order to find such a microorganism, as a result of intensive research, a bacterium belonging to the alkalophilic Bacillus genus, an alkalophilic Nocardiopsis genus, and a lytic enzyme derived from an actinomycete belonging to the alkalophilic Streptomyces genus,
We found that it has an optimum pH on the alkaline side and has new properties such as being stable in various surfactants, and that it can lyse a wide range of bacteria from Gram positive bacteria to negative bacteria in the presence of surfactants. The present invention has been completed by taking out the target enzyme from the culture solution of the strain and elucidating its properties. The bacterium producing the lytic enzyme of the present invention was newly isolated by the present inventors, and each of them has NSE-11.
4, there are 2 strains named NSE-565, but they have the following mycological properties. For testing and classification of mycological properties, "Classification and Identification of Microorganisms" (edited by Hasegawa) and Berg
ey's Manual of Systematic Bacteriology (1986). Since both strains hardly grow in a neutral medium, a medium adjusted to pH 9 with 0.2% sodium carbonate was used in the test described below.

Next, a method for culturing the above-mentioned microorganism and collecting a novel lytic enzyme from the culture solution will be described. Usable medium is a medium used for culturing ordinary microorganisms, and is not particularly limited as long as the strain of the present invention can grow. For example, gluco- is used as an organic carbon source and a nitrogen source.
Sodium, starch, dextrin, peptone, yeast extract and the like are suitable, and addition of potassium phosphate, sodium phosphate, magnesium sulfate, salt and the like is effective as the inorganic salt. In addition, various organic and inorganic substances necessary for the growth of bacteria and the production of enzymes can be added as required. Although it is desirable to add bacterial cells to the medium, it is not always necessary for enzyme production. As a method for culturing in the above-mentioned medium, shaking culture or aeration stirring culture using a fermenter or the like is suitable. The culture temperature is not particularly limited as long as it can grow, but is preferably around 30 ° C. Further, since the optimum growth pH of the microorganism used in the present invention is within the basic range, it is necessary to adjust the pH value of the medium using an appropriate alkaline reagent. Therefore, 0.2% sodium carbonate can be mentioned as a typical example, but needless to say, sodium hydroxide,
Alkaline reagents such as potassium hydroxide, sodium hydrogen carbonate, sodium phosphate and calcium hydroxide can also be used.

After culturing as described above, the lysing enzyme, which is the target substance, can be collected and purified from the culture medium according to a general method for collecting and purifying the enzyme. That is, solids such as bacterial cells are first removed from the culture solution by a method such as filtration or centrifugation to obtain a crude enzyme solution. The crude enzyme solution thus obtained can be used as it is, but this crude enzyme solution is subjected to a conventional enzyme purification method, that is, salting out with ammonium sulfate, precipitation with an organic solvent such as acetone or alcohol, ultrafiltration, ion By using techniques such as an exchange resin and a gel filtration method alone or in combination, it is possible to obtain a lysing enzyme with higher purity. The activity of lytic enzyme is measured by the following method. That is, heat-treated freeze-dried cells of Corynebacterium aquaticum IFO 12154 were added to 25 mM CHES buffer (pH 9) at OD.
Suspend to 660 = 1.0. To 3 ml of this cell suspension,
Add 0.2 ml of the enzyme solution, react at 37 ° C for 30 minutes, and measure the decrease in absorbance at 660 nm. Regarding the display of the enzyme activity, the amount of enzyme that reduces the absorbance of 0.001 per minute under the above conditions was defined as 1 unit.
