Description

【発明の詳細な説明】
【0001】
【産業上の利用分野】本発明は電気噴霧イオン源、更に
言えば、ニュートラルノイズを減少させるイオン源に関
する。
【0002】
【従来の技術】電気噴霧処理では、付近の表面に比べて
高電圧に保持されたチューブ、即ち細管を通じてサンプ
ル液が噴出される。この液体は電気的に良好に荷電され
た小滴に、細管の先端における電圧こう配によって分散
される。イオン化メカニズムには、電気的に良好に荷電
された小滴からイオンを大気圧にて脱着させる処理が含
まれる。多くの場合には、電気噴霧の小滴の溶解を促進
するため、加熱ガスを電気噴霧に対して逆方向に流出さ
せる。その後、電気噴霧によって作り出されたイオン
は、質量分析計のような質量分析器により、その質量が
分析される。
【0003】適切な状況下において、電気噴霧は、小滴
(直径約1μm)の非常に微細な霧(即ち、フォッグ)
から成る均整のとれた円錐に類似する。微細な霧が作り
出された場合、優れた感度とイオン電流安定性が得られ
る。不幸にも、電気噴霧の「質」は、分析される溶液の
内部特性に非常に依存している。最も重要なことは、表
面張力と導電率である。質の低い電気噴霧は、より大き
な小滴(>直径10μm)あるいは非分散スチームを含
む。
【0004】高水性含有サンプル溶液を電気噴霧する場
合には、安定した噴霧作業を確実にするため、シース液
及び焦合ガスがときとして用いられる。特許第4,997,78
5 号、4,885,076 号及び継続中の出願第575,183 号には
シース液を含む電気噴霧イオン源が記述されている。こ
の継続中の出願に記述されている装置では、幅広い溶液
状態(例えばpH、導電率、及び水性含有物)にわたっ
てイオン電流を安定させるような多層フローニードルア
センブリが用いられる。この多層フローニードルは、サ
ンプルを導入するための中央チューブと、シース液を導
入するための第2同心チューブ、及びガスフローを付加
するための第3同心チューブからなる。液体シースの第
1の目的は、溶離剤スチームの表面張力を減少させ、電
気噴霧を高水性含有の溶液にも使用可能とすることにあ
る。同心ガスフローは、高誘電率の溶液を電気噴霧する
場合において、イオン電流の安定を改善させるという点
で重要である。このシースフローにより、非所望なニュ
ートラルノイズが質量分析計に作り出されてしまうこと
になる。
【0005】これらのニュートラルが、シースガスフロ
ーによってESI雰囲気/減圧インタフェースを通過す
る小滴によって引き起こされることは明かである。これ
らの小滴が減圧状態となった場合、それらの小滴は冷却
され、最終的には凍りつき、そして小さな氷ペレットを
形成することが分かっている。これらの粒子はフリージ
ェット膨張によって超音波速度にまで加速される。この
フリージェット膨張において、これらの粒子は、それら
が表面に衝突したときに第2イオンを作り出すに充分な
だけの運動エネルギーを得る。この結果、質量分析計の
検出器で非常に大きなノイズ、「スパイク」が生じてし
まう。
【0006】
【発明の概要及び目的】本発明の目的は、高速にされた
水滴あるいは粒子から生じる雑音を最小限にするような
イオン源及び方法を提供することにある。本発明の他の
目的は、水と混合することができるイオン表面張力を有
しまた水よりも高い沸点を備えるシース液を用いて粒状
形成を最小にするようなイオン源及び方法を提供するこ
とにある。
【0007】本発明のこれらの目的及び他の目的は、低
い表面張力を有し、水と混合でき、また水よりも高い沸
点を有するようなシース液を用いるシースフロー電気噴
霧源により達成される。
【0008】
【実施例】図1には、電気噴霧イオン源11が、分析チ
ャンバ12と共に示されている。このイオン源は、液体
サンプル14が導入される入力ニードル13を備える。
特に図2を参照すれば明かなように、ニードルは液体サ
ンプルが導入される内部チューブ16を備えている。こ
のチューブを取り囲むのは第2チューブである。この第
2チューブは第1チューブを用いて環状領域を形成し、
シース液はこの環状領域を通じてサンプル液との混合の
ため導入される。外部チューブ18は第2環状形態を形
成する。焦合ガスはこの第2環状形態を通じて導入さ
れ、細管チューブ19に向けてニードルから噴出すると
きに小滴を焦合させる。前に説明したように、ニードル
即ち細管は、イオンチャンバ21を形成する付近の表面
と比較して高電圧に保持されており、液体が分散される
際に、細管の先端における電圧こう配によって小滴ある
いは粒子が荷電されるのである。このイオン化メカニズ
ムでは、電気的に良好に荷電された粒子からイオンを大
気圧にて脱着させる。この脱着処理は矢印22で示され
ているような逆流するガスによって強化される。チャン
バ23を流れるガスは、細管19の端部を通過し、24
で示したようにイオンチャンバ21から出ていく。
【0009】チャンバ21の気圧よりも低圧に保持され
ているチャンバ26は、細管チューブ19を通じてイオ
ンチャンバと通じている。気圧差により、イオン及びガ
スは細管19を通じてチャンバ26中へと流れる。導電
スリーブ27と28の間に電圧を印加することにより、
絶縁細管にそって電圧こう配が形成される。細管の端部
は、低圧領域26を分析器12内の低圧領域から分離す
るスキマーと向き合っている。このスキマーは、一般に
は細管のボアの軸と整列された中央オリフィスあるいは
開口32を含む。スキマーの後ろには、第2スキマー3
3及びレンズ34を備えたイオンオプチックスが存在
し、イオンを分析チャンバ及び適当な分析器へと導く。
【0010】上で述べたように、脱溶媒されていない小
滴あるいは粒子が細管へ流れ込み、スキマー開口32を
通じてスキマー33及びレンズ34を備えるレンズ領域
に入りこむに充分なだけの運動エネルギーを得る。これ
らの小滴あるいは粒子は、スキマー33あるいはレンズ
34の表面に衝突して第2イオンを作り出す。これらの
イオンはでたらめであるが検出器へと入り込み、検出器
で発見されるノイズを引き起こす。これにより信号対雑
音レベルは減少し、質量分析器において電気スパイクが
作り出されてしまう。
【0011】本発明によれば、分析計の低圧分析領域へ
入り込む水滴の形成を最小にする特性を持つようにシー
ス液が選択される。液体クロマトグラフィー/質量分析
計の性能にとって重要なシース液の特性は、第1に、シ
ース液は、高いパーセンテージで水を含む溶液と調和で
きるよう、比較的小さな表面張力を有していなければな
らないということである。第2に、シース液は、メタノ
ールやアセトニトリル(一般にはLC溶剤が使用され
る)と同様、水と混合できなればならないということで
ある。最後に、シース液は、水よりも高い沸点を有して
いなければならないということである。この特性は、装
置によって作り出されるニュートラルノイズを最小にす
るために重要である。非常に揮発しやすい溶剤は、これ
らの小滴からあまりに早く蒸発してしまうため、高いパ
ーセンテージで水を含有する小滴を生じてしまう。なぜ
なら、これらの小滴は大きな表面張力を有し、より小さ
な小滴にするのは容易ではなく、ニュートラルノイズを
作り出しやすいからである。
【0012】溶剤2−メトキシエタノールは、上に掲げ
た全ての所望の特性を有する。3つの異なるシース液を
用いて得られるニュートラルノイズの比較を図3に示し
た。この図は、2−メトキシエタノールを用いることに
より、ノイズの強度と同様にその頻度も劇的に減少する
ことを示している。この溶剤をシース液として使用する
ことによる他の利点は、0.1%のトリフルオロ酢酸
(TFA)を含有するモービルフェーズが用いられた場
合には、信号応答が増大(2〜3回)するということで
ある。
【0013】このように、シース液を提供することによ
り、水滴によって引き起こされる電気噴霧におけるニュ
ートラルノイズは減少する。

Claims (2)

    【特許請求の範囲】
  1. 【請求項1】 シース液のサンプルが導入されるイオン
    化チャンバと、このイオン化チャンバと自身と通じさせ
    るような手段を備えることによって前記イオン化チャン
    バ内のイオン及びガスを自身の領域に流出させる隣接の
    低圧領域とを有する電気噴霧イオン源において、 低い表面張力を有し、高いパーセンテージで水を有する
    液体と調和でき、水、メタノール及びアセトニトリルと
    混合可能であり、水より高い沸点を有するようなシース
    液を用いるという改善を備えることを特徴とする電気噴
    霧イオン源。
  2. 【請求項2】 請求項1記載の電気噴霧イオン源におい
    て、前記シース液は2−メトキシエタノールである電気
    噴霧イオン源。
