Claims (1)
- 【特許請求の範囲】 1.野生型3−ホスフォグリセレートデヒドロゲナーゼ(PGD)と比較してセ リンによる阻害に対する感受性が減少したPGDをコードしてなる工学的DNA 。 2.野生型PGDが、微生物又は酵母PGDである請求の範囲第1項に記載の工 学的DNA。 3.前記DNAが、野生型PGDのC−末端25%に変化を含んでなるPGDを コードしている請求の範囲第1項に記載の工学的DNA。 4.前記DNAが、野生型PGDのC−末端アミノ酸50個に変化を含んでなる PGDをコードしている請求の範囲第3項に記載の工学的DNA。 5.前記DNAが、野生型PGDのC−末端欠失を含んでなるPGDをコードし ている請求の範囲第3項に記載の工学的DNA。 6.前記C−末端変性が、野生型PGD配列への挿入を含んでなる請求の範囲第 3項に記載の工学的DNA。 7.前記挿入が、野生型PGDのVAL363とASN364との間か、ALA 392とGLN394との間になされている請求の範囲第6項に記載の工学的D NA。 8.下記のDNAからなる群から選択される請求項1に記載の工学的DNA: i)serA1455vi)serA1510ii)serA1459vii) serA1511iii)serA1507viii)serA1512iv) serA1508ix)serA1530v)serA1509x)serA1 5319.請求の範囲第1項〜第8項のいずれかに記載の工学的DNAによりコ ードされたアミノ酸配列を有する3−ホスフォグリセレートデヒドロゲナーゼ。 10.請求の範囲第1項〜第8項のいずれかに記載の工学的DNAと、宿主発現 系において前記工学的DNAを発現するように配置し指向した調節DNAとを含 んでなる発現ベクター。 11.請求の範囲第1項〜第8項のいずれかに記載の工学的DNAと調節DNA とを含んでなる細胞であって、前記調節DNAが前記細胞において前記工学的D NAを発現するように配置し指向している細胞。 12.前記細胞が、野生型serAについて欠失している請求の範囲第11項に 記載の細胞。 13.セリン又はセリン由来である所望の生成物の製造方法であって、請求の範 囲第11項に記載の細胞を培養する工程と、前記所望の生成物を回収する工程と を含んでなる製造方法。 14.前記生成物がセリンである請求の範囲第13項に記載の方法。 15.変化3′端を有するPGDをコードする転写体を産生するように工学的処 理を施した細胞であって、前記転写体を野生型PGDと比較してセリンによる阻 害に対する感受性が減少したPGDに翻訳することができる細胞。
