- 【特許請求の範囲】 1.検査試料中のTrypanosomacruzi抗体の存在の確認アッセイ であって、 a.検査試料中の第1T.cruzi抗体の存在を判定するステップであって、 i.検査試料の第1アリコートを、固体支持体に接合させた第1T.cruzi 抗原と接触させて混合物を形成し、それをインキュベートして第1抗原/抗体複 合体を形成し、ii.前記第1抗原/抗体複合体を指示薬と、第1抗原/抗体/ 指示薬複合体を形成するのに十分な時間及び条件下で接触させ、更に iii.生成されたシグナルを測定することにより第1T.cruzi抗原の存 在を検出する ことからなるステップと; b.検査試料中の第2T.cruzi抗体の存在を判定するステップであって、 i.検査試料の第2アリコートを、固体支持体に接合させた第2T.cruzi 抗原と接触させて混合物を形成し、それをインキュベートして第2抗原/抗体複 合体を形成し、ii.前記第2抗原/抗体複合体を指示薬と、第2抗原/抗体/ 指示薬複合体を形成するのに十分な時間及び条件下で接触させ、更に iii.生成されたシグナルを測定することにより第2T.cruzi抗原の存 在を検出する ことからなるステップと: c.検査試料中の第3T.cruzi抗体の存在を判定するステップであって、 i.検査試料の第3アリコートを、固体支持体に接合させた第3T.cruzi 抗原と接触させて混合物を形成し、それをインキュベートして第3抗原/抗体複 合体を形成し、ii.前記第3抗原/抗体複合体を指示薬と、第3抗原/抗体/ 指示薬複合体を形成するのに十分な時間及び条件下で接触させ、更に iii.生成されたシグナルを測定することにより第3T.cruzi抗体の存 在を検出する ことからなるステップとを含んでおり、少なくとも2種の抗体の存在をもって検 査試料中のT.cruzi抗体の存在を認定する方法。 2.前記第1T.cruzi抗原を、Gp90、Gp60/50及びLPPGか らなる群から選択し、第1抗原として使用される抗原はステップ(b)の第2抗 原またはステップ(c)の第3抗原には使用されない請求項1に記載のアッセイ 。 3.前記第2T.cruzj抗原を、Gp90、Gp60/50及びLPPGか らなる群から選択し、第2抗原として使用される抗原はステップ(a)の第1抗 原またはステップ(c)の第3抗原には使用されない請求項2に記載のアッセイ 。 4.前記第3T.cruzi抗原を、Gp90、Gp60/50及びLPPGか らなる群から選択し、第3抗原として使用される抗原はステップ(a)の第1抗 原またはステップ(b)の第2抗原には使用されない請求項3に記載のアッセイ 。 5.前記ステップ(a)、ステップ(b)及びステップ(c)の指示薬が、色原 体、触媒、発光化合物、化学発光化合物、放射性元素及び直接可視ラベルからな る群から選択されるラベルからなる請求項1に記載のアッセイ。 6.前記ステップ(a)、ステップ(b)及びステップ(c)を実施する前に検 査試料を前試験し、反応性であることを確認する請求項1に記載のアッセイ。 7.前記ステップ(a)、ステップ(b)及びステップ(c)を同時に実施する 請求項1に記載のアッセイ。 8.T.cruziのGp60/50抗原を精製する方法であってa.T.cr uziの上鞭毛虫期の膜をドウンスホモジナイズすることにより単離するステッ プと; b.Gp60/50抗原を抽出するステップと:c.ステップ(b)で得られた 抽出物をGalanthusnivaIisレクチンアフィニティーカラムに添 加し、炭水化物を用いて溶出するステップと: d.溶出液を、Gp60/50に特異的なモノクローナル抗体を含むアフィニテ ィーカラムを用いて精製するステップとからなる方法。 9.前記ステップ(b)の抽出を非イオン性活性剤を用いて実施する請求項8に 記載の方法。 10.請求項8に記載の方法に従うGp60/50抗原。 11.T.cruziの抗原性糖脂質をタンパク質担体に結合する方法であって 、 a.T.cruziの上鞭毛虫期から糖脂質リポホスホノペプチドグリカン(L PPG)を得るステップと;b.ステップ(a)のLPPGをタンパク質にエチ ルジメチルアミノ−プロピルカルボジイミド(EDAC)を使用して結合するス テップであって、 i.LPPGをEDACと接触させて混合物を形成し、得られた混合物を活性化 するのに十分な時間及び条件下でインキュベートし、 ii.活性化した混合物をタンパク質と一緒に、LPPGをタンパク質に結合す るのに十分な時間及び条件下でインキュベートする ことからなるステップと; C.混合物を精製するステップと; d.LPPGを混合物から溶出するステップとからなる方法。 12.前記タンパク質がウシ血清アルブミン(BSA)である請求項11に記載 の方法。 13.前記ステップ(c)の精製を、混合物をブルーデキストランセファロース に通すことにより実施する請求項11に記載の方法。 14.請求項11に記載の方法に従うLPPG.15.T.cruziに対する 抗体のアッセイに使用するための診断試薬であって、Gp90、Gp60/50 及びLPPGからなる群から選択される抗原である試薬。 16.T.cruziに対する抗体の検出に有用な診断キットであって、Gp9 0、Gp60/50及びLPPGからなる群から選択される少なくとも1種のT .cruzi抗原を含む容器を具備する診断キット。 17.前記抗原が固相に結合されている請求項16に記載の診断キット。 18.前記LPPG抗原がタンパク質に固定されている請求項17に記載の診断 キット。
