経細胞が酸素欠乏状態(以下「脳ヒポキシア」という)
をきたすと、脳神経細胞膜の透過性に変化をもたらし、
細胞外液の浸入により浮腫がひき起こされる。脳浮腫が
ある程度以上に増大すると、脳圧が亢進し脳の循環障害
を起こす。そして、それによる脳ヒポキシアの増強とブ
ドウ糖欠乏及びその代謝物の蓄積が脳の浮腫を助長し、
脳浮腫,脳圧亢進が更に強くなり、脳幹の圧迫と髄液の
通過障害が起き、これらは更に脳ヒポキシアの増強,脳
浮腫の促進,脳圧の亢進という悪循環を形成する。した
がって、病巣が拡大し、健常脳組織までが脳ヒポキシア
をきたし、脳循環不全状態に陥り、障害は重篤となる。

限定されているのに対し、本発明の活性化合物及びその
アルカリ金属塩は催眠麻酔作用を示さないため、急性期
だけでなく、慢性期及び発作の再発を予防する目的での
投与が可能である。本発明の活性化合物及びそのアルカ
リ金属塩は毒性も低く、したがって高い安全性を有す

長させるのに必要な試験化合物の投与量(PD15)を用量

の0.5%トラガント溶液を経口投与した。対照群に比べ
て呼吸停止までの時間を30%延長させるのに必要な試験
化合物の投与量(PD30)を用量−作用の関係から算出し

試験例2 海馬CA1領域の遅発性神経細胞壊死に対する
保護作用
本試験は、Prain Res.,239,57〜69(1982)に記載の方

濁したものを、スナネズミの体重100g当り0.2mlの割合
で経口投与した。対照群には同容量の0.5%トラガント
溶液を投与した。投与1時間後に、スナネズミの頚部を
エーテル麻酔下に正中切開した後、左右の総頚動脈を露
出し、Zenクリップで両側の総頚動脈の血流を遮断し
た。血流を5分間遮断した後、クリップを解除すること
により血流を再開した。偽手術群として、同様の麻酔手
術後、血流遮断並びに再開通操作を行わない群を作製し
た。4日後に、スナネズミをペントバルビタールで麻酔
し、左心房より固定液[2.5%グルタールアルデヒドと
2%ホルムアルデヒドを含む0.1Mリン酸緩衝液(pH7.
4)]を120cm H2Oの潅流圧で潅流固定し、脳を取り出し
た。脳は固定液中に1日放置した後、背側海馬を含む組
織片を脱水透徹を経てパラフィンに包埋した。このパラ
フィンブロックから海馬の最先端より0.5〜1.0mm又は矢
状縫合の後方1.4〜1.9mmに相当する厚さ4μmの切片を
作製し、ヘマトキシリン−エオジンで染色した。この組
織標本について、海馬CA1領域に残存する錐体細胞の数
を200倍の倍率下に画像処理装置(イメージングリサー
チ社製,カナダ、MCID型)を用いて計測し、左右の海馬
錐体細胞層の面積1mm2当りの残存神経細胞密度を算出し
た。結果を表2に示す。表中の数値は、左及び右側海馬
の残存神経細胞数を計測後、左右の平均値を各固体の残
存神経細胞数として算出したものである。なお、統計学

る投与量(HD50)をプロビット法により算出した。結果

本発明の活性化合物及びそのアルカリ金属塩は、催眠麻
酔作用から十分に分離された強い抗脳ヒポキシア作用を
示すとともに、極めて強い細胞保護作用をも示し、かつ
その毒性は弱いので、脳虚値が誘因となる哺乳動物(ヒ
トを含む)の頭蓋内疾患又はそれに伴う各種症状の予防
及び治療に使用することができる。頭蓋内疾患の具体例
としては、脳梗塞(脳血栓,脳塞栓),脳内出血,クモ
膜下出血,頭部外傷,脳腫瘍,脳髄膜炎,脳浮腫が、頭
蓋内疾患に伴う症状の具体例としては、意識障害,神経
障害,痴呆,自覚症状(めまい,頭痛,片頭痛,嘔気,
嘔吐等),脳外科手術時あるいは手術後の頭蓋内圧亢進
が挙げられる。その投与経路としては、経口投与,非経

結晶セルロースを混和し、水に溶解したヒドロキシプロ
ピルセルロースを加えて練合したのち乾燥し顆粒状とす
る。これにステアリン酸マグネシウム及び軽質無水ケイ
酸を添加し、圧縮成形して1錠200mgの錠芯1000錠を調
製する。次いで、ヒドロキシプロピルメチルセルロー
ス,マクロゴール,酸化チタン,タルク及び軽質無水ケ
イ酸を用い、常法に従って剤皮を施しフィルムコーティ

μm)で濾過後10mlずつアンプルに充填、窒素置換し、
熔閉した後121℃で20分間高圧蒸気滅菌して注射剤1000
