Description
【発明の詳細な説明】 本発明は、血小板凝集の治療を目的とする、ディルチア
ゼムとアスピリン、なかでもアセチルサリチル酸のリジ
ン塩を主成分とする医薬組成物に関するものである。
ゼムとアスピリン、なかでもアセチルサリチル酸のリジ
ン塩を主成分とする医薬組成物に関するものである。
式（Ｉ）： で示されるディルチアゼム（diltiazem）は、その抗ア
ンギナ、抗高血圧、カルシウムアンタゴニストおよび血
小板凝集阻害活性のために知られている。

式： で示されるアセチルサリチル酸のリジン塩（LAS）は、
その鎮痛剤、抗炎症および血小板凝集阻害特性のために
知られている。

本出願人は、驚くべきことに、これら２化合物を組み合
わせて使用する場合、これらの化合物の各々の血小板凝
集阻害特性が高められるということを見い出した。

本発明の医薬組成物を一連の薬理試験に付した。
以下の方法に従い、本発明組成物をイン・ビトロで試験
した： 250μセルの凝集測定器に入れた血小板と血漿の懸濁
液にこれらの化合物を単独でまたは組み合わせて加え、
２分後、血小板凝集を誘導する薬物であるアラキドン酸
10μを加える。ボーン（Born）の方法［ジャーナル・
オブ・フィジオロジー（Journal of Physiology）、16
8、178−195（1963）］に従い、２チャンネル経時（chr
onological）凝集測定器中で凝集を調べる。

得られた結果を表１および表２に示す。
以下の方法に従い、本発明の組成物をイン・ビボで試験
した： ディルチアゼム10mg/kg投与量とLAS1mg/kg投与量を単独
でまたは組み合わせて２匹のラットからなる５群に経口
投与する。１時間後に血液のサンプルを採取する。次い
で、血漿と血小板の懸濁液を調製し、250μセルの凝
集測定器に入れ、２分後にアラキドン酸（200−250μg/
ml）を加える。ボーンの方法に従い、２チャンネル経時
凝集測定器中で凝集を調べる。

結果を表３に示す。
これらの結果は、アセチルサリチル酸のリジン塩とディ
ルチアゼムの組み合わせ物は、各々の化合物を別個に投
与して得られる効果の和より有意に大きい血小板凝集阻
害効果を提供するということを示している。

本発明の医薬組成物は、単位投与量当たりディルチアゼ
ム３〜60mgとLAS10〜300mgを含有することができる。

口または非経口投与に好適な形態であってよい。

梢血管の症状、血小板凝集のトラブルおよび片頭痛の治
療に使用することができる。

- 【請求項１】ディルチアゼムとアセチルサリチル酸のリ
ジン塩を組み合わせて含有することを特徴とする血小板
凝集阻害剤。 - 【請求項２】単位投与量当たり、ディルチアゼム３〜60
mgと アセチルサリチル酸のリジン塩10〜300mgを含有
することを特徴とする第１項に記載の血小板凝集阻害
剤。
