For example, oxytocin and vasopressin consisting of 9 amino acids are known as important hormones, and angiotensin II consisting of 8 amino acids has physiological actions such as blood pressure increase. However, conventionally, there is little knowledge about cyclic oligopeptides, and the existence of cyclic pentapeptides consisting of 5 amino acids has been hardly known.

The following abbreviations are used in the examples. PyBOP: Benzotriazol-1-yl-oxy-tris (pyrrolidino) -phosphonium hexafluorophosphate HOBt: 1-hydroxybenzotriazole NMM: N-methylmorpholine Fmoc: 9-fluorenylmethoxycarbonyl The synthesis of cyclic pentapeptide is Using PyBOP-HOBt-NMM
It was synthesized by Fmoc Chemistry by the immobilization method using 2-chlorotrityl chloride-resin. Arg or T
For amino acids having a reactive side chain such as yr, amino acids to which a protecting group was introduced as needed were used. (1) Introducing the first amino acid For the substitution rate of 2-chlorotrityl chloride-resin
Dissolve 0.6 equivalents of Fmoc-amino acid in as little dry dimethylformamide (DMF) as possible, then
Dichloromethane (DCM) was added to 0 ml. 2-
After adding 1.0 g of chlorotrityl chloride-resin, diisopropylethylamine (DIEA) was added to the amino acid.
0.5 equivalent was added and stirred for 5 minutes. DIEA 1.0 equivalent and DCM
Add (1: 1) and stir for an additional 20 minutes, then add 0.8 ml of methanol.
Addition was continued for 10 minutes. Use a glass filter (N
After suction filtration at o.2), it was washed three times with 10 ml each of DCM, DMF, isopropanol, methanol and diethyl ether, and dried in a desiccator by a vacuum pump. (2) Coupling after the second amino acid Coupling after the second amino acid was performed in a 2.5 ml polypropylene reaction vessel, and stirring was performed by bubbling with N 2 gas. Add 100 ml of the amino acid-resin obtained in (1) above to the reaction vessel and add 1.5 ml of DMF.
Was added, and after bubbling for 3 minutes, the mixture was filtered. 1.5 ml of a DCM / DMF solution containing 5% piperidine was added, and after bubbling for 10 minutes, the mixture was filtered. DMF solution treatment with 30% piperidine (1.5 m
l, 2 minutes, 2 times), and after washing with DMF (1.5 ml, 1 minute, 5 times), Fmoc-amino acid, HOBt, and PyBOP were used in an amount of 600 μmol and NMM 1000 μmol, respectively, in DMF 1,5 ml. Coupling was performed. After reacting for 30 minutes,
DMF washing (1.5 ml, 1 minute, 5 times), DMF solution treatment containing 30% piperidine (1.5 ml, 2 minutes, 2 times), DMF washing (1.5 ml, 1 minute, 5 times) Fmoc-amino acid, HOBt, PyBOP and NMM were added and a coupling reaction was performed. In the same manner, after coupling 5 kinds of amino acids, treatment with DMF solution containing 30% piperidine (1.5 ml, 2 ml)
Washing was performed for 2 minutes) and DMF washing (1.5 ml, 1 minute, 5 times). (3) Cleavage of peptide from peptide-resin To a reaction vessel containing peptide-resin, 1.5 ml of acetic acid / trifluoroethanol / DCM (1: 1: 8) was added, and the mixture was reacted at room temperature for 60 minutes and filtered. Add the same solution to the reaction vessel again,
The resin was washed and then filtered. The filtrate and washings were combined, placed in a centrifuge tube, anhydrous diethyl ether was added, and the precipitated peptide was recovered by centrifugation (850 xg, 5 minutes), purged with N 2 gas and dried. (4) Cyclization of peptide Dissolve the peptide obtained in (1) to (3) above in DMF, add equimolar amounts of HOBt, PyBOP, and NMM to the peptide so that the final concentration is 1 mM. Add DMF at room temperature 1
The solution was allowed to stand for 0 hour to carry out a cyclization reaction. After the reaction,
(3) Similarly, the peptide was recovered by ether precipitation, washed with a small amount of ether, purged with N 2 gas and dried. (5) Deprotection Cyclic peptide 15 mg / ml trifluoroacetic acid
(TFA) / H 2 O / thioanisole / ethanediol (EDT) / ethyl methyl sulfide (EMS) / phenol (82: 5: 5: 3: 2: 3)
Was added, and the mixture was allowed to stand at room temperature for 6 hours to carry out a deprotection reaction. After completion of the reaction, the peptide was recovered by ether precipitation as in (3) above, washed with a small amount of ether, further washed with a small amount of distilled water, and then freeze-dried. (6) Confirmation of product The substance obtained by the above operation was analyzed by FAB-MS and amino acid analysis to confirm the synthesis of cyclic pentapeptide.
FAB-MS was measured by SX-102A (JEOL) using glycerin as a matrix. Amino acid analysis was performed by mixing 200 nmol of the synthesized peptide with 1 ml of 6N HCl at 110 ° C.
After acid hydrolysis for 0 hour, it was performed with an amino acid analyzer (Model Hitachi 835 amino acid analyzer). Production Example 1: A = D-Val; B = L-Leu; C = D-Leu; D = L-Phe;
Production of cyclic pentapeptide of E = D-Arg 16.93 mg of cyclic pentapeptide was obtained from 50 mg of Fmoc-D-Leu-resin by the above method. The obtained substance is FAB-M
The synthesis of the target substance was confirmed by analysis by S and amino acid analysis. FAB-MS analysis showed (M + H) + to be 629, and amino acid analysis showed Leu: Val: Arg: Phe = 1.8: 1.0: 1.0: 1.1.
