キサール、50% DMSO、10mMりん酸ナトリウム緩衝液(pH
7.0))中で、1時間、50℃の処理によってグリオキサー
ル化を行なった後、2μlのアプライ用緩衝液(50% (w/
v) グリセロール、10mMりん酸緩衝液(pH7.0)、0.4%
(w/v)ブロムフェニルブルー)および1μlの1mg/ml臭化
エチジウム溶液を加え、10mMりん酸ナトリウム緩衝液
(pH7.0)、1%アガロースを含むゲル中で電気泳動を行
なった。電気泳動中は、ペリスタポンプを用いて、電気
泳動層中の緩衝液を常に循環させ、pHの勾配が生ずるこ
とを防いだ。ブロムフェニルブルーがゲルの長さの70%
程度まで達したときに電気泳動を中止し、紫外線トラン
スイルミネーターを用いて臭化エチジウムで染色された
ゲル中のRNAを観察し、リボゾーマルRNAを指標にRNAが
分解されていないことを確認した。その後に、ゲル中の
RNAを、その添付されたプロトコールに従って、ナイロ
ンメンブレンGenescreen-Plus(デュポン社)にブロッ
ティングし、RNAがブロッティングされたメンブレンに
対し、80℃、2時間のインキュベーション処理を行なっ
た。このメンブレンは、Genescreen-Plusに添付された
プロトコールに従ってノーザン分析に使用した。プロー

し、これを一晩、1 リットルのTE緩衝液中で透析し
た。こうして得られたDNA溶液を、Frischauf らの方
法(Mehtods in Enzymology, 152, p.183, Academic pre
ss1987)に準じて、制限酵素Sau3AIで部分分解し、再び1
0% 〜40% のショ糖密度勾配液にのせ、20℃、25000rp
m、22時間の遠心を行った。超遠心終了後、注射針を用
いて超遠心チューブの底に穴を開け、落下するサンプル
を約0.5ml ずつ分取し、各サンプルの一部をアガロース
ゲルを用いて電気泳動し、15〜20kbのDNAを含む画分
をエタノール沈殿によって回収した。この画分から1μ
gに相当するDNA溶液をとり、λ-EMBL3/BamHI vecto
r kit (ストラタジーン社)のλ-EMBL3ベクター1μg
と16℃で一晩、そのプロトコールに従ってライゲーショ
ンした。これをGIGAPACK GOLD (ストラタジーン社)を
用いて、そのプロトコールに従ってパッケージングを行
った。得られたパッケージング液50μl をλ-EMBL3/Bam
HIvector kit に添付されている宿主菌株P2392 に感染
させた。すなわち、P2392の1白金耳をTB培地(1%バ
クトトリプトン、0.5% NaCl 、0.2%麦芽糖、pH7.4)5ml
で一晩培養し、この培養液の1ml を50mlのTB培地に植
菌し、OD600=0.5 になるまで培養し、これを氷浴上で冷
却後、集菌し、15mlの氷冷した10mM硫酸マグネシウム液
に懸濁させた。この菌液1ml に、0.95mlのSM液(0.1M
塩化ナトリウム、10mM硫酸マグネシウム、50mMトリス塩
酸緩衝液(pH7.5) 、0.01% ゼラチン)と50μl のパッケ
ージング液を加えて軽く攪拌し、37℃で15分間保温し
た。この液を 200μl ずつ、あらかじめ 7mlずつ分注し
て47℃に保温しておいたBBL軟寒天培地(1% Triptic
ase peptone (BBL社) 、0.5% NaCl 、0.5%アガロース)
に加え、軽く攪拌した後、直径15cmのBBLプレート
(0.5%のアガロースのかわりに1.5%の寒天を加えた以外

37℃で8時間培養し、合計で約 30000個のサイズのファ
