Claims (1)
- 【特許請求の範囲】 1.配列番号1または配列番号1の相補体と選択的にハイブリダイズすることが 可能なヌクレオチドの連続配列を含む、実質的に単離された形態のポリヌクレオ チド。 2.DNAポリヌクレオチドである、請求項1に記載のポリヌクレオチド。 3.前記の連続配列が少なくとも20個のヌクレオチドを含む、請求項1または 2に記載のポリヌクレオチド。 4.配列番号1を含む、請求項3に記載のポリヌクレオチド。 5.請求項1から4のいずれか1つに記載のポリヌクレオチドおよびその相補体 を含む2本鎖ポリヌクレオチド。 6.顕示(revealing)標識をもつ、請求項1から5のいずれか1つに記載のポ リヌクレオチド。 7.請求項1から6のいずれか1つに記載のポリヌクレオチドを含む、複製可能 な組換えベクター。 8.請求項7に記載のベクターを含む宿主細胞。 9.(a)配列番号2のタンパク質;または (b)その対立遺伝子変異体または種相同体(species homologue);また は (c)(a)に少なくとも70%相同なタンパク質;または (d)E2F−1タンパク質またはその関連ファミリーのメンバーと複合体 を形成することが可能な、(a)から(c)のいずれかの断片;または (e)少なくとも15個のアミノ酸からなる、(a)から (c)のいずれかの断片 を含む、実質的に単離された形態のポリペプチド。 10.顕示標識をもつ、請求項9に記載のポリペプチド。 11.請求項9または10に記載のポリペプチドと共に担体または希釈剤を含む 組成物。 12.固相に固定された、請求項9または10に記載のポリペプチド。 13.請求項9に記載のポリペプチドをコードする2本鎖DNA。 14.請求項9に記載のポリペプチドをコードするコード配列を含む複製可能な 組換えベクター。 15.請求項14に記載の組換えベクターによって形質転換された宿主細胞。 16.前記のコード配列が、該配列の発現を宿主細胞によってもたらすことが可 能な制御配列に機能しうる形で連結されている、請求項14に記載の組換えベク ターによって形質転換された宿主細胞。 17.請求項9に記載のポリペプチドを調製する方法であって、請求項16に記 載の宿主細胞を組換えベクターによるコード配列の発現をもたらす条件下で培養 し、発現されたポリペプチドを回収することを含む調製方法。 18.(a)配列番号2のタンパク質;または (b)その対立遺伝子変異体または種相同体(species homologue);ま たは (c)(a)に少なくとも70%相同なタンパク質;または (d)E2F−1タンパク質またはその関連ファミリーのメンバーと複合 体を形成することが可能な、(a)から(c)のいずれかの断片:または (e)少なくとも15個のアミノ酸からなる、(a)から(c)のいずれ かの断片 と結合可能な抗体。 19.モノクローナル抗体である、請求項18に記載の抗体。 20.顕示標識をもつ、請求項18または19に記載の抗体。 21.固相に固定された、請求項18から20のいずれか1つに記載の抗体。 22.請求項19に記載のモノクローナル抗体を産生するハイブリドーマ細胞系 。 23.試料中の請求項9に記載のポリペプチドの存在または不在を検出するため のイムノアッセイであって、 (a)請求項18から21のいずれか1つに記載の抗体を供給し; (b)試料を該抗体と共に、抗体−抗原複合体の形成を可能とする条件下でイ ンキュベートし;そして (c)該抗体−抗原複合体を検出する ことを含むイムノアッセイ。 24.増殖性またはウイルス性疾患を治療するための推定上の化学療法剤を同定 するためのスクリーニング方法であって、 (A)以下の成分: (i)請求項9または10に記載のポリペプチド; (ii)(a)E2F−1タンパク質、または (b)その対立遺伝子変異体または種相同体、または (c)DNA結合領域において少なくとも70%の相同性を有するE 2F−1ファミリーのメンバー、または (d)配列番号2のタンパク質と機能性トランス作用複合体を形成す ることが可能な、(a)、(b)または(c)の断片;または (e)(a)、(b)、(c)または(d)を含合タンパク質;およ び (iii)推定上の化学療法剤; を成分(i)および(ii)が成分(iii)の不在下で複合体を形成する条件 下で接触させ、そして (B)成分(iii)がどの程度まで該複合体の活性を破壊または阻止できるかを 測定する ことを含むスクリーニング方法。 25.前記の(i)と(ii)との複合体が、E2F DNA結合部位とin vitro で結合するその能力によって測定される、請求項23に記載のアッセイ。 26.前記の(i)と(ii)との複合体が、レポーター遺伝子に連結したE2F 結合部位を含むプロモーターをin vivoで活性化するその能力によって測定され る、請求項23に記載のアッセイ。 27.アッセイが酵母細胞、昆虫細胞または哺乳動物細胞を用いて実施される、 請求項26に記載のアッセイ。 28.前記の推定上の化学療法剤が、 (a)配列番号2のタンパク質;または (b)その対立遺伝子変異体または哺乳動物の相同体;または (c)(a)に少なくとも70%相同なタンパク質 の10個以上のアミノ酸からなる断片である、請求項23から26のいずれか 1つに記載のアッセイ。 29.前記の断片が配列番号2の160から220の領域内に入るポリペプチド 断片である、請求項28に記載のアッセイ。 30.ヒトまたは動物の身体を治療する方法に使用するための、請求項9または 10に記載のポリペプチド、または請求項18から20のいずれか1つに記載の 抗体。 31.ヒトまたは動物の身体を治療する方法に使用するための、請求項7または 14に記載のベクター。 32.治療を必要とする患者における制御されない細胞増殖を治療する方法であ って、該患者に請求項7または14に記載のベクター、または請求項1に記載の ポリヌクレオチドを有効量投与することを含む方法。
