Claims (1)
- 【特許請求の範囲】 1.麦酒の腐敗の原因となる微生物のrRNAまたはrDNAに優先的にハイブ リダイズする、単離および精製された核酸。 2.微生物がLactobacillus属およびPediococcus属からなる群より選択される、 請求項1記載の核酸。 3.次の核酸: (a)P.damnosusと非Pediococcus種とを特異的に識別する核酸; (b)麦酒腐敗の原因となるPediococcus株の大半と他の種とを特異的に識別する 核酸; (c)L.brevisと非Lactobacillus種とを特異的に識別する核酸; (d)Lactobacillus種のクラスターと非クラスター種とを特異的に識別する核酸 ;ここで該クラスターは、L.fructivorans、L.casei、L.curvatus、L.brevi sおよびL.buchneriからなる; (e)P.damnosusおよびL.brevisからなる群と他の種とを特異的に識別する核 酸; (f)麦酒腐敗の原因となるPediococcus種およびLactobacillus種の大半と他の 種とを特異的に識別する核酸; (g)PediococcusおよびLactobacillusおよび関連する種の大半と他の種とを特 異的に識別する核酸; からなる群より選択される、請求項1記載の核酸。 4.16S rRNAに優先的にハイブリダイズする、請求項1記載の核酸。 5.23S rRNAに優先的にハイブリダイズする、請求項1記載の核酸。 6.プローブ2858、2861、2867、2876、2877、2868、 2869、2880、2891、2892、2895、2899、2904、2 896、2873、2881、2887、2875、2901、2854、28 79および2902からなるプローブの群により定義される配列の群より選択さ れる配列中のいずれか10個の連続したヌクレオチドを含む配列に相補的である かまたは少なくとも90%相同である、請求項1記載の核酸。 7.麦酒の腐敗の原因となる微生物のrRNAもしくはrDNAに優先的にハイ ブリダイズする核酸プローブ。 8.微生物がLactobacillus属およびPediococcus属からなる群より選択される、 請求項7記載のプローブ。 9.次のプローブ: (a)P.damnosusと非Pediococcus種とを特異的に識別するプローブ; (b)麦酒腐敗の原因となるPediococcus株の大半と他の種とを特異的に識別する プローブ; (c)L.brevisと非Lactobacillus種とを特異的に識別するプローブ; (d)Lactobacillus種のクラスターと非クラスター種とを特異的に識別するプロ ーブ;ここで該クラスターは、L.fructivorans、L.casei、L.curvatus、L.b revisおよびL.buchneriからなる; (e)P.damnosusおよびL.brevisからなる群と他の種とを特異的に識別するプ ローブ; (f)麦酒腐敗の原因となるPediococcus種およびLactobacillus種の大半と他の 種とを特異的に識別するプローブ; (g)PediococcusおよびLactobacillusおよび関連する種の大半と他の種とを特 異的に識別するプローブ; からなる群より選択される、請求項7記載のプローブ。 10.16S rRNAに優先的にハイブリダイズする、請求項7記載のプロー ブ。 11.23S rRNAに優先的にハイブリダイズする、請求項7記載のプロー ブ。 12.プローブ2858、2861、2867、2876、2877、2868 、2869、2880、2891、2892、2895、2899、2904、 2896、2873、2881、2887、2875、2901、2854、2 879および2902からなるプローブの群により定義される配列の群より選択 される配列中のいずれか10個の連続したヌクレオチドを含む配列に相補的であ るかまたは少なくとも90%相同である、請求項7記載のプローブ。 13.麦酒の腐敗の原因となる微生物の存在を検出する方法であって、 標的を含むと疑われる試料を、次の生物: (a)P.damnosusおよび非Pediococcus種; (b)他の種ではなく、麦酒腐敗の原因となるPediococcus株の大半; (c)他のLactobacillus種ではなく、L.brevis; (d)他の種ではなく、L.fructivorans、L.casei、L.curvatus、L.brevisお よびL.buchneriからなるLactobacillus種のクラスター; (e)他の種ではなく、P.damnosusおよびL.brevisの群; (f)他の種ではなく、麦酒腐敗の原因となるPediococcus種およびLactobacillu s種の大半;および (g)他の種ではなく、PediococcusおよびLactobacillusおよび関連する種の大 半; からなる群より選択される生物のrRNAもしくはrDNAに優先的にハイブリ ダイズする少なくとも1つの核酸と接触させ; 核酸が標的rRNAまたはrDNAと優先的に結合して核酸複合体を形成するよ うに試料にハイブリダイゼーション条件を付与し;そして 標的生物の存在を指示するものとして複合体を検出する の各工程を含む方法。 14.核酸が、プローブ2858、2861、2867、2876、2877、 2868、2869、2880、2891、2892、2895、2899、2 904、2896、2873、2881、2887、2875、2901、28 54、2879および2902により定義される配列からなる群より選択される 、いずれか10個の連続したヌクレオチドを含む配列と少なくとも90%相同で ある、請求項13記載の方法。 15.麦酒の腐敗の原因となる微生物の存在を検出するために用いられるキット であって、少なくとも2つの核酸を含み、各核酸が10から250ヌクレオチド を含みかつ異なる塩基配列組成を有し;ここで各核酸がプローブ2858、28 61、2867、2876、2877、2868、2869、2880、289 1、2892、2895、2899、2904、2896、2873、2881 、 2887、2875、2901、2854、2879および2902により定義 される配列の群より選択されるいずれか10個の連続したヌクレオチドを含む配 列に相補的であるかまたは少なくとも90%相同である核酸の組を含むキット。 16.さらに、試薬および指示書を含む、請求項15記載のキット。
