非ヌクレオチドを含有する酵素性核酸
発明の背景
本出願はアスマンらが1993年9月2日に出願した米国出願第08/116
,117号からの一部継続出願である1993年11月12日に出願した米国出
願第08/152,481の一部継続出願である。両出願共に「非ヌクレオチド
含有酵素的核酸」という発明の名称を有するものである。両出願の内容は本願に
参考として含まれる(図も含む)。
本発明は化学合成された非ヌクレオチドを含む酵素性核酸に関する。
酵素性RNA分子もしくはリボザイムの発見の歴史およびその活性について簡
単に述べる。この歴史は完全なものではなく、以下に説明する本発明の理解の補
助とするためのものである。このまとめは以下に示したすべてが特許請求の範囲
に対する従来技術であるということを意味するのではない。
1970年代以前はすべての遺伝子は、遺伝子によってコードされるタンパク
質と一対一で対応すると考えられていた。この単純な考えは、すべての遺伝子は
紙テープに書かれたメッセージのごときものであって、各三連のDNAの「文字
」は1つのタンパク質である「言葉」に翻訳されると考えられていた。タンパク
質合成は遺伝子のDNAからRNAへの書き換え(文字を異なる形態の文字とす
る)、次いでRNAをタンパク質へと翻訳する(1度に3文字)。1970年代
中盤において、遺伝子のうちのいくつかは、自身のコード化するタンパク質と正
確に一対一の対応をしないことが発見された。かかる遺伝子はコーディング配列
内にあって、タンパク質への翻訳前にRNAから除去すなわち「スプライス」さ
れる中断領域を有していることが見いだされた。このようなコーディング領域に
おける中断部には介在配列、すなわち「イントロン」という名前がつけられおり
、そしてこのイントロンがRNAから除去される過程には「スプライシング」と
名付けられている。少なくとも3つの異なる機構がRNAからイントロンを除去
する機構として見いだされている。かかるスプライシング機構のうちの2つには
様々なタ
ンパク質因子が結合し、その働きにより正しい位置でRNAが切断、連結される
ものである。第3の機構はRNAをイントロン自身により切断および連結させる
機構であり、これが触媒的RNAについての初めての発見である。
セッシュと共同研究者らは池に棲む単細胞生物であるテトラヒメナ・サーモフ
ィラ(Tetrahymena thermophila)においていかにしてRNAスプライシングが発
生するかにつき研究した。セッシュはRNAの介在配列が自身を周囲のRNAか
ら切り離すスプライシング因子として作用することを報告した。1980年代初
頭においてはテトラヒメナのイントロンの複雑な構造をはっきりさせ、セルフス
プライシングの機構を解読するためにさらなる研究が行われた。多くの研究グル
ープによりテトラヒメナのイントロンの特別な折り畳み構造が切断され、スプラ
イスされるRNA部分に伴ってあることを支持していた。スプライシングが終わ
った後でも、遊離したイントロンは触媒性構造を保持する。その結果、遊離した
イントロンはさらなる切断およびスプライシング反応を自身に対して行い、イン
トロン環を形成する。1986年までに、セッシュは短くされたテトラヒメナイ
ントロンが、RNAの他の切片に対して様々な切断および結合反応をもたらすこ
とを示すことができた。この証明によって、テトラヒメナイントロンが真の酵素
として働くことが導かれる:(i)各イントロン分子は多くの基質分子を切断す
ることができるが、イントロン自体は変化せず維持される、そして(ii)反応
は唯一の配列(CUCU)を含有するRNA分子に特異的であり、かかる配列に
よってイントロンがRNAを認識し、結合する。ザウとセッシュは「リボザイム
」という用語をすべての酵素様性質を有すRNAを表現する用語として新造した
。
同じ1986年に、セッシュはテトラヒメナのリボザイムによって認識される
RNA基質配列がリボザイム自身の内部の配列を変化させることによって変え得
ることを見いだした。この性質は他のRNA配列を切断するよう個々にデザイン
される多くの部位特異的リボザイムの開発につながった。
テトラヒメナのイントロンは最もよく研究されており、現在では大きな一群の
イントロン、すなわちグループIイントロンとして認識されている。いくつかの
配列要素を含有する完全に折り畳まれた構造はグループIのイントロン内で保存
されており、一般的なスプライシング機構に関与している。テトラヒメナのイン
トロンのごとく、グループIに含まれるイントロンのうちのいくつかは触媒活性
を有する、すなわちイントロン自身が自己スプライシング反応を生じる。他のグ
ループIのイントロンは他の(タンパク質)因子を、例えばイントロンの折り畳
みおよび/または活性構造の維持のために必要とすると考えられている。
リボヌクレアーゼP(RNアーゼP)はRNAおよびタンパク質成分の両方か
らなる酵素であり、前駆体tRNA分子の一方の末端から過剰のRNAを取り除
きその最終形態へと導く。RNアーゼPの活性は試験したすべての生物において
認められた。シドニー・アルトマンとその共同研究者らは、RNアーゼP中のR
NA成分はかかるプロセシング活性に必要であることを示した;しかしながら、
彼らは彼らの用いた条件においてはタンパク質成分もまたプロセシングに必要で
あることもまた示した。セッシュがテトラヒメナのイントロンによって自己スプ
ライシングを発見した後、RNアーゼP内のタンパク質とRNA成分が再調査さ
れた。1983年、アルトマンとパースがRNアーゼP複合体の酵素的成分がR
NAであることを発見した。このことは、あるRNA分子を実際の酵素として作
用し得、自分自身に変化を受けることなく数多くのtDNA分子を変化させるこ
とができることを示すものであった。
RNアーゼP RNAの折り畳み構造が調べられている。異なる生物間におい
て、RNAの配列は正確に保存されていないが、より高次の構造については保存
されていた。このことはRNアーゼP複合体のタンパク質成分が、インビボにお
いて折り畳まれたRNAの構造を安定化させているからであると考えられる。
シモンズとその共同研究者らは2種類の自己切断RNAを同定したが、これら
すでに報告されていた触媒性RNAの他の形態と異なっていた。シモンズは、ア
ボガド植物に感染するRNAウイルスであるアボカド・サンブロッチ・ウイロイ
ド(avocado sunblotch viroid(ASV))の繁殖について研究していた。シモンズは
55ヌクレオチドの小ささのASVのRNAが自己を2切片へと切断するよう、
折り畳まれ得ることを証明した。これらのRNAのインビボ自己切断によって、
ウイルスの増殖の間に、RNAが1ゲノム長の断片に切断されると考えられてい
る。シモンズはASVからのさまざまな最小触媒配列が他の植物病原性RNAに
も認められることを発見した。これらの配列を比較したところ、3本のステムと
多くの保存された(RNA間で変化しない)ヌクレオチドを有する中心ループで
接続されたループから構成される一般的な構造が判明した。この触媒性RNAに
対して予測される二次構造には、ハンマーヘッド型のの検索部位を有している;
従ってこれはハンマーヘッドと名付けられる。
ウーレンベック(Uhlenbek)はリボザイムの触媒領域を基質から単離することに
成功した。すなわち、2つ(または3つ)の小さな合成RNAから、ハンマーヘ
ッドリボザイムを組み立てることが可能となったのである。19個のヌクレオチ
ドからなる触媒領域および24個のヌクレオチドからなる基質が特定の切断を行
うには十分である。多くのハンマーヘッド型リボザイムの触媒性部位がウーレン
ベックのグループとシモンズのグループの両方によって研究されており、ここで
は特定の組み立ておよび触媒活性に必要なヌクレオチドの決定および様々な条件
下における切断速度の測定が行われている。
ヘーゼロフ(Haseloff)とゲルラック(Gerlach)はハンマーヘッドリボザイムの
ドメインを異なる方法によって分けることができることを示した。そうすること
によって、鎖中において必要とされる配列のほとんどを切断されない部位に置き
(リボザイム)、鎖中にUH(H=C,AまたはU)がある場合のみに切断され
る(基質)ようにデザインすることが可能となる。このことによって、触媒リボ
ザイムは長い「基質認識」配列の中にはめ込まれているどのUH RNA配列を
も切断するように設計できる。長いmRNAの特別な切断を、いくつかのハンマ
ーヘッドリボザイムを用いて予言した方法によって行うことが、1988年に報
告されている。
ある植物病原性RNA(タバコリングスポットウイルスのネガティブ配列から
得た)は自己切断を受けるが、上述のコンセンサスなハンマーヘッド構造に折り
畳まれない。ブルーニング(Bruening)とその共同研究者らは、このRNAに対し
て50ヌクレオチドの触媒ドメインを同定した。1990年ハンペル(Hampel)と
トリッツ(Tritz)は触媒性ドメインを2つの部分に分け、それぞれ基質とリボ
ザイムとして、多段階繰り返し切断反応を行わせることに成功した。ハンマーヘ
ッドリボザイムのごとく、触媒性部位には触媒活性に必要とされる配列のほとん
どを含んでおり、標的においては短い配列(この場合にはGUC)のみが必要と
される。ハンペルとトリッツはこのRNAの折り畳まれた構造を、シングルヘア
ピンを構成しているとして表現し、「ヘアピン」リボザイム(ブルーニングと共
同研究者らはこのリボザイムモチーフに対して「紙ばさみ」と名付けた)と命名
した。その後に研究によって、標的RNAの結合および切断の機構において、ヘ
アピンとハンマーヘッドリボザイムの間の類似性が増加することが示されている
。
肝炎デルタウイルス(HDV)は、ゲノムが一本鎖RNAであるウイルスであ
る。ゲノムRNAおよび相補的なアンチゲノムRNAの両方における小さな領域
(約80ヌクレオチド)があれば自己切断を行える。1991年にビーン(Been)
とペロッタ(Perrotta)はHDVのRNAの二次構造がゲノムRNAとアンチゲノ
ムRNA間で保存されており、これが触媒活性に必要であることを提案した。H
DVのRNAを「リボザイム」と「基質」部分へ分離することが近年になってビ
ーンにより達成された。ビーンはさらにHDVリボザイムの大きさを約60ヌク
レオチドにまで減少させることに成功した。

発明の要約
本発明は、二本鎖核酸(例えばRNAもしくはDNA)のステム(二重らせん
ステム)のベースまたは酵素核酸の一本鎖領域、触媒コア、ループもしくは認識
アームにおけるスペーサー要素としての非ヌクレオチド分子の使用に関する。二
重らせんステムは、酵素RNA分子中に遍在する構造要素である。上記ステムは
、通常一本鎖ヌクレオチド連鎖によって接続されているが、かようなステムの合
成を容易にするため、単一塩基もしくは塩基対の擬似体(ミメティック:mimeti
c)を使用して、このような分子を合成する際のヌクレオチドの必要数を減らし
、かつヌクレアーゼ耐性を付与する(これら擬似体は非核酸要素であるから)こ
とができる。またこのことは、リボザイムの触媒コアと認識アームの両者にも当
てはまる。
このような非ヌクレオチド擬似体の例を図4に示し、これらのハンマーヘッド
リボザイム中への組込みを図5に示す。これら非ヌクレオチドのリンカーは、ポ
リエーテル、ポリアミン、ポリアミドまたはポリ炭化水素の化合物でもよい。具

したがって本発明は、第一の態様で、一つ以上の非ヌクレオチド成分を有しか
つRNAまたはDNA分子を切断する酵素活性を有する酵素性核酸を提供するも
のである。
このような非ヌクレオチド擬似体の例を図4に示し、それらのハンマーヘッド
リボザイムへの組込みを図5に示す。これらの非ヌクレオチドリンカーはポリエ
ーテル、ポリアミン、ポリアミドまたはポリ炭化水素の化合物でもよい。
好ましい実施態様で、その酵素性核酸はRNAの一つ以上の領域(stretch)
を含有し、その領域は、分子の酵素活性を提供し、非ヌクレオチトの部分に連結
される。
「非ヌクレオチド」という用語は、糖および/またはホスフェートの置換を含
めて一つ以上のヌクレオチド単位の代わりに核酸連鎖中に組込むことができかつ
残りの塩基にその触媒活性を示させることができる基または化合物を意味する。
その基または化合物は、アデノシン、グアニン、シトシン、ウラシルまたはチミ
ンのような公知のヌクレオチド塩基を含有していない点で非塩基である。その基
または化合物は、当該技術分野で報告されているように2'もしくは3'のHもし
くはOHが置換されていてもよい(EckteinらおよびUsmanらの前記文献参照)。
好ましい実施態様で、その酵素性核酸はRNAの一つ以上の領域を含有し、そ
の領域は分子酵素活性を提供し、非ヌクレオチドの部分に連結される。必要なR
NA成分は当該技術分野で公知である(例えばUsmanの前記文献参照)。
「非ヌクレオチド含有酵素性核酸」という用語は、本願で用いる場合、リボザ
イムの一部、例えば限定されないが二本鎖ステム、一本鎖「触媒コア」配列、一
本鎖ループまたは一本鎖認識配列を置換する少なくとも一つの非ヌクレオチド要
素を含有する核酸分子を意味する。これらの分子は、別のRNAまたはDNAの
分子を、ヌクレオチド塩基配列に特異的な方式で切断(好ましくは繰返し切断)
することができる。またこのような分子は、所望に応じて分子内で切断するよう
作用させることができる。このような酵素分子は事実上、RNAの転写を標的に
することができる。またかような分子としては、エキソヌクレアーゼによる消化
を防止するキャップ形成基として有用な3'−または5'−に非ヌクレオチドを有
する核酸分子がある。
本発明の酵素分子は、まず標的のRNAまたはDNAに結合することによって
作用する。このような結合は、標的のRNAまたはDNAを切断する作用がある
分子の酵素部分のごく近くに保持されている酵素の標的結合部分を通じて起こる
。したがってその分子は、まず標的核酸を認識し次いで相補的塩基対形成によっ
て標的核酸を捕捉し、そして正しい部位に一旦結合されると酵素として作用して
標的を切断する。このような標的の効果的な切断によって、コードされているタ
ンパク質の合成を制御する標的核酸の性能が破壊される。本発明の酵素は、その
標的を捕捉して切断した後、その標的から放出され、別の標的を探して、新しい
標的を繰返し捕捉して切断することができる。
本発明の酵素の酵素特性は、他の方法、例えばアンチセンス法(この場合、核
酸分子は単に核酸標的に結合して核酸標的の翻訳を遮断するだけである)を超え
て有利である。というのは、治療処置を行うのに必要な酵素の有効濃度がアンチ
センスオリゴヌクレオチドの有効濃度より低いからである。この利点は本発明の
酵素が酵素として作用する性能を反映している。即ち、単一の酵素分子が多数の
標的RNA分子を切断することができる。その上本発明の酵素は、高度に特異的
な阻害剤であり、その阻害作用の特異性は、結合に際しての塩基対生成の機構の
みならず、その分子が結合するRNAの発現を阻害する機構に依存している。す
なわち、その阻害作用は標的の切断によって起こり、その特異性は、標的核酸の
切断率/非標的核酸の切断率の比率と定義する。この切断機構は、塩基対生成に
関与する因子の外に追加される因子に依存している。したがって本発明の酵素の
作用の特異性は、同じ標的部位を捕捉するアンチセンスオリゴヌクレオチドの作
用の特異性より大きいと考えられる。
「フレーズ酵素(phrase enzyme)」という用語は、基質結合領域に、指定の
核酸標的に対する相補性を有しかつその標的中のRNAまたはDNAを特異的に
切断する酵素活性も有する非クレオチド含有触媒核酸分子を意味する。すなわち
、
その酵素は、分子内または分子間が、RNAまたはDNAを切断して、標的のR
NAまたはDNAの分子を不活性化することができる。この相補性は、本発明の
酵素分子を標的のRNAまたはDNAと充分にハイブリッドを形成させて切断を
起こす機能がある。100%の相補性が好ましいが、50〜75%と低い相補性でもこ
の発明に有用である。
本発明の好ましい実施態様では、本発明の酵素分子は一般にハンマーヘッドの
モチーフで形成されるが、ヘアピン、肝炎デルタウイルス、グループIイントロ
ンまたはRNアーゼP RNA(RNAガイド配列と関連する)のモチーフで形
成することもできる。このようなハンマーヘッドモチーフの例はRossiら、Acids
Research and Human Retroviruses、8巻、183頁、1992年に記載されており;
ヘアピンモチーフの例は、Hampelらの1988年9月20日付け出願の米国特許第07/2
47,100号の一部継続出願である1989年9月20日付け出願の「RNA Catalyst for C
leaving Specific RNA Sequences」;Hampel and Tritz、Biochemistry、28巻、
4929頁、1989年およびHampelら、Nucleic Acids Research、19巻、299頁、1990
年に記載されており;肝炎デルタウイルスモチーフの例はPerrottaおよびBeen、
Biochemistry、31巻、16頁、1992年に記載され;RNアーゼPのモチーフの例は
Guerrier-Takadaら、Cell、35巻、849頁、1983年に記載され;ならびにグループ
Iイントロンのモチーフの例はCechらの米国特許第4,987,071号に記載されてい
る。本発明はこれらの特異的モチーフに限定されるものではなく、当該技術分野
の当業者は、本発明の酵素分子において重要なことは、その酵素分子が少なくと
も一つの非ヌクレオチド部分および標的遺伝子のRNA領域の一つ以上に相補的
な特異的基質結合部位を有し、かつ核酸切断活性をその酵素分子に付与する基質
結合部位の中またはその部位を囲むヌクレオチド配列を有することであることが
分かるであろう。
本発明は、所望の標的の核酸に対して高い特異性を示す一クラスの切断酵素剤
の製造方法を提供するものである。本発明の酵素分子は、標的の高度に保存され
た配列領域を標的とすることが好ましく、その結果、疾患また症状の特異的な治
療を単一の酵素で行うことができる。このような酵素分子は、必要に応じて特定
の細胞に対して外因的に送達することができる。好ましいハンマーヘッドのモチ
ーフにおいて、小さい分子(長さが60個未満のヌクレオチド好ましくは30〜40個
のヌクレオチド)を用いれば、他のリボザイムのモチーフより治療費用が減少す
る。
長さが100ヌクレオチドを超える核酸分子は自動化された方法を用いて合成す
ることは困難なのでこのような分子の治療費用は法外なものになる。本発明では
、外因的送達に、小さな酵素のモチーフ(例えばハンマーヘッドのモチーフ)を
用いる。これらの分子は構造が単純なので、酵素がmRNA構造の標的領域に侵
入する性能が増大する。ハンマーヘッド構造が長い転写物内に含まれている場合
と異なり、酵素構造の正しい折畳みまたは相補的領域を妨害する非酵素の隣接配
列は存在しない。
本発明のその外の特徴と利点は、下記の、本発明の好ましい実施態様の説明と
特許請求の範囲から明らかになるであろう。

の二つのストランドを接続するループ(図5中に/ /で示す)を置換するこ
とができる。ステムII自体は1〜10個の塩基対である。後記の実施例1と2では
、化合物1と2が、長さが1〜5個の塩基対のステムIIを有する分子中に組込ま
れた。また化合物1、4および5は、ステムIとIIIの認識アームの中のヌクレ
オチド、またはステムII自体の中のヌクレオチドを置換する。

本発明に有用な非ヌクレオチド擬似体としては先に一般的に述べたものがある
。当該技術分野の当業者は、これらの擬似体は、標準の方法によって酵素分子中
に所望の位置に組込むことができることが分かるであろう。標準の実験を行い例
えば分子を合成してその酵素活性を試験することによってその最適位置を決定し
て、適切な選択を行うことができる。最適の分子は、酵素活性を得るのに必要な
公知のリボヌクレオチドを含有し、かつ分子の構造を簡単なものに変える非ヌク
レオチドを含有している。要求されることは、いくつかのヌクレオチドが一つの
非ヌクレオチドで置換され、酵素分子合成の合成ステップが節約されて、RNA
またはDNAにさえ比べて酵素分子の安定性が高められることである。実施例
以下に、非ヌクレオチド擬似体を含有する触媒核酸の非ヌクレオチドホスホル
アミダイトを用いる合成の実施例を示すが、本発明を限定するものではない。実施例1:非ヌクレオチド擬似体:ポリエステルスペーサーを含有するハンマー ヘッドリボザイムの合成
ポリエステルスペーサーである化合物1(図4;X=0、n=2または4)の
、n=2もしくは4のものを単独でまたはn=2のものを二重に、ハンマーヘッ
ドリボザイムのステムIIのベースにループ2を置換して組込んだところ触媒効率
が低いリボザイムが生成したことが分かった。利用した合成法は、Usmanら、J.A
m.Chem.Soc.、109巻、7845頁、1987年;およびScaringeら、Nucleic Acids Res.
、18巻、5433頁、1990年に記載されているような通常のRNA合成法の手順にし
たがった合成法であり、通常の核酸保護基と核酸カップリング基が用いられ、例
えば5'末端ではジメトキシトリチルが3'末端ではホスホルアミダイトが用いら
れる。段階的カップリングの平均収率は>98%であった。これらの型の擬似体の
構造はまだ最適化されていないが、先に述べたように、このことは標準の試験法
を用いて容易に達成することができるであろう。これらの試験は、このような擬
似体が、触媒活性を維持しながらリボザイムのループと一部分を置換する可能性
を示している。これらの擬似体はハンマーヘッドリボザイム中に組込むことがで
きるだけでなく、ヘアピン、肝炎デルタウイルス、またはグループ1もしくはグ
ループ2のイントロン中にも組込むことができる。したがってこれら擬似体は任
意の核酸構造中に置換モチーフとして汎用することができる。このような擬似体
を用いると、非ヌクレオチド擬似体なしのオリゴヌクレオチドを使用する場合と
比べて、最終の核酸分子から約2〜10個のヌクレオチドを省略することができる
。実施例2:非ヌクレオチド擬似体:芳香族スペーサーを含有するハンマーヘッド リボザイムの合成
別の実施例で、ハンマーヘッドリボザイムのステムIIC−Gのベースに対する
特異的なリンカーを設計した。本発明の発明者は、C−Gの塩基対のC1'炭素
間の距離は約16オングストロームであると考えている。これら二つのRNAピー
スを、共有結合類似体のC−G塩基対で結合するため、GとCそれぞれの残基の
3'−OHと5'−OHの間にリンカーを含有する新しい型の二量体ホスホルアミ
ダイトを構築することができる。図4に示す二つの型の塩基対擬似体2または3
は剛性の芳香族スペーサーである。これらのスペーサーを、実施例1に記載され
ているようにして、ループ2を置換してハンマーヘッドリボザイムのステムIIの
ベースに組込んだところ、触媒効率が低いリボザイムが生成することが分かった
。他の擬似体は、Nielsenら、Science、254巻、1497頁、1991年に記載されてい
るポリアミド骨格に類似の可撓性のアルキルスペーサーである(図4の6または
その誘導体6a;Zuckermanら、J.Am.Chem.Soc.、114巻、10464頁、1992年参照
)。このような擬似体を使用すると、非ヌクレオチド擬似体なしのオリゴヌクレ
オチドを使用する場合と比べて、最終の核酸分子から約2〜10個のヌクレオチド
を省略することができる。実施例3:非ヌクレオチド擬似体の芳香族スペーサーのホスホルアミダイト2の 合成
この化合物は、Salunkheら、J.Am.Chem.Soc.、114巻、6324頁、1992年に初め
て報告された。その合成法は以下のように改変した。DMF(12ml)中のテレフ
タル酸(1.0g、6.0mmol)に、EDC(2.54g、13.2mmol)、アミノヘキサノール
(1.55g、13.2mmol)およびN−メチルモルホリン(1.45ml、13.2mmol)を添加
した。その反応混合物を一夜撹拌したがその時、溶液は濁っていた。その反応混
合物に水を添加して生成物を沈澱させた。固形分をろ取して水で洗浄し乾燥して
562mg(25.7%)のジオールを得た。
DMSO(40ml)中の上記ジオール(250mg、0.687mmol)に、トリエチルアミ
ン(287μl、2.06mmol)、ジメトキシトリチルクロリド(220mg、0.653mmol)お
よび触媒DMAPを添加した。その反応混合物を40℃まで加熱して一夜撹拌した
。得られた混合物を室温(約20〜25℃)まで冷却し、水で反応を停止させ、次い
でEtOAcで3回抽出した。固体の沈澱が有機層中に残留したので、これを単
離したところ出発物質のジオール(50mg、20%)であることが分かった。有機層
をNa2SO4で乾燥し次に蒸発させた。得られた油状物をフラッシュクロマトグ
ラ
フィー(ヘキサン中10%のEtOAc〜100%EtOAc)で精製して250mg(55
%)のモノトリチル化化合物を得た。
0℃のTHF(1ml)中の上記アルコール(193mg、0.29mmol)に、ジイソプ
ロピルエチルアミン(101μl、0.58mmol)を添加し、次に2−シアノエチルN,
N−ジイソプロピルアミノクロロホスホルアミダイト(78μl、0.35mmol)を滴
下した。得られた混合物を5分間撹拌し次に室温まで昇温させた。1時間後、反
応混合物の反応をメタノールで停止させ次に該混合物を蒸発させた。得られた油
状物をフラッシュクロマトグラフィー(1:1ヘキサン:EtOAc)で精製し
て158mg(63%)のホスホルアミダイトを得た。実施例4:非ヌクレオチド擬似体の芳香族スペーサーホスホルアミダイト3の合 成
図6に示すように、キノリン(10ml)中の3、4、9、10−ペリレンテトラカ
ルボン酸二無水物11(1.0g、2.55mmol)に、エタノールアミン(919μl、15.3mm
ol)とZnOAc・2.5H2O(140mg、0.638mmol)を添加した。反応混合物を19
0℃で8時間加熱した。得られた溶液を冷却し、1NHClを添加して生成物を
沈澱させ、次いでその混合物をろ過した。固形分を、ろ液がもはや淡緑色でなく
なるまで熱10%CaCO3溶液で洗浄した。残留した鮮やかな赤色の沈澱12を乾
燥した。
得られたジオール12を、2について先に述べたようにして処理し、ホスホルア
ミダイト3を得た。実施例5:非ヌクレオチド擬似体:非塩基のヌクレオチド4と5を含有するハン マーヘッドリボザイムの合成
化合物4(R=H)をIyerら、Nuleic Acids Res.、18巻、2855頁、1990年に
したがって製造した。図7に示すように、化合物4と5(R=o−t−ブチルジ
メチルジリル)のホスホルアミダイトを以下のようにして製造した。
N,N−ジメチルホルムアミド(250ml)にD−リボース(20.0g、0.105mol)
を溶解して得た溶液に、2,2−ジメトキシプロパン(50ml)とp−トルエンス
ルホン酸一水和物(300mg)を添加した。その反応混合物を室温で16時間撹拌し
次いで蒸発乾固した。得られた組生成物をピリジン(2×150ml)とともに同時
蒸発させたのち乾燥ピリジン(300ml)に溶解し、次に4,4−ジメトキシトリ
チルクロリド(37.2g、0.110mol)を添加し、室温で24時間撹拌した。得られた
反応混合物をメタノール(50ml)で希釈し次いで蒸発乾固した。残留物をクロロ
ホルム(800ml)に溶解し、5%NaHCO3(2×200ml)、食塩水(300ml)で
洗浄し、乾燥し、蒸発させ、トルエン(2×100ml)とともに同時に蒸発させ次
にフラッシュクロマトグラフィー(CHCl3)で精製して40.7g(78.1%)の化
合物aを得た。
ジメトキシトリチル誘導体の(9.0g、18.3mmol)とDMAP(4.34g、36mmol
)を乾燥CH3CNに溶解して得た溶液に、クエノキシチオカルボニルクロリド
(3.47g、20.1mmol)をアルゴン雰囲気下滴下して加えた。得られた反応混合物
を室温で16時間放置し次いで蒸発乾固した。得られた残留物をクロロホルム(20
0ml)に溶解し、5%NaHCO3、食塩水で洗浄し、乾燥し、蒸発し、次いでフ
ラッシュクロマトグラフィー(CHCl3)で精製して、β−アノマーとして8.0
g(69.5%)の化合物bを得た。
中間体b(3.0g、4.77mmol)のトルエン(50ml)溶液に、AIBN(0.82g、5
.0mmol)とBu3SnH(1.74g、6.0mmol)をアルゴン雰囲気下で添加し、その
反応混合物を80℃で7時間保持した。その溶液を蒸発し、得られた残留物をフラ
ッシュクロマトグラフィー(CHCl3)で精製して、1.5g(66%)の保護され
たリビトールcを得た。
次にすべての保護基を酸処理によって除去し次いでトリチル化を行い保護され
たリビトールdを得た。このリビトールdを、Scaringtら、Nucleic Acids Res.
、18巻、5433頁、1990年に記載されている一般的な方法によって目的物のホスボ
ルアミダイト4と5に変換した。実施例6
図8aと8bを参照して、基質の切断を、8nMと40nMの濃度で各種修飾リ
ボザイムを未修飾リボザイムと比較することによって示す。具体的に述べると、
配列ucuccA UCU GAU GAG GCC GAA AGG CCG AAA Auc ccu(小文字は、2'−0−
メチル基を含有する)の対照リボザイムを、リボザイムA(ucu ccA UCU GAU GA
G GCC SGG CCG AAA Auc ccu)、B(ucu ccA UCU GAU GAG CSG CG AAA Auc ccu
)、C(ucu ccA UCU GAU GAG GCC bbb bGG CCG AAA Auc ccu)、およびD(ucu
ccA UCU GAU GAG Cbb bbG CGAA AAu ccc u)(S=ヘキサエチレングリコール
リンカー;およびb=非塩基のヌクレオチド4)と比較した。これらはすべて、
基質を切断する活性を有していた。リボザイムの投与
選択されたリボザイムは例えば、イオン導入法、電気穿孔法またはイオン対分
子もしくは共有結合付加物、およびその外の薬理学的に承認された送達法を用い
て、適切な送達ビヒクル例えばリポソーム、放出調節媒体により関連組織にリボ
ザイムを外因的に送達することによって、ウイルス感染患者または疾病患者に予
防的に投与することができる。投与経路としては、筋肉内、エアゾール、経口(
錠剤または丸剤の形態)、局所、全身、眼、腹腔内および/またはくも膜下の経
路がある。
任意の選択されたリボザイムの特定の送達経路は、リボザイムの用途によって
決まる。一般に各リボザイムの特定の送達プログラムは、細胞内局在に関する未
修飾リボザイムの取込み次いで効力の実証に集中している。あるいは、動物の臓
器または組織内のこれら同じ細胞への送達を追跡してもよい。取込みの試験法と
しては、送達ビヒクルまたは送達法にかかわらず細胞のリボザイム取込みを評価
する取込み検定法がある。このような検定法によって、取込みによるリボザイム
の細胞内局在性も測定され、結局、標的配列を含有している細胞コンパートメン
ト(細胞核および/または細胞質)内の定常状態濃度を維持するのに必要な条件
が確認される。次に効力と細胞毒性を測定する。毒性は細胞生存能力のみならず
細胞機能にも関連する。
使用できる送達法としては以下のものがある。
a.リポソーム内への封入
b.レトロウイルスベクターによる形質導入
c.コレステロールとの接合
d.大部分のsnRNAまたは核タンパク質に見られる抗原結合部位または核
標的部位を利用する核コンパートメントへの局在化
e.ヌクレオチド誘導体を用いることによるリボザイムの電荷の中和、および
f.血液幹細胞を用いて行うリボザイムの全身体への分布、である。
本発明に有用な送達法には、リボザイムの修飾および粒子担体の医薬送達ビヒ
クルを含む。恐らく大部分が小分子の未修飾リボザイムは、ゆっくりではあるが
細胞によって吸収される。細胞の取込みを増大させるため、リボザイムはその電
荷を減らすが特定の官能基を維持する方式で、ほとんどランダムに修飾すること
ができる。その結果、細胞膜を横切って拡散できる分子が得られ、透過性にかか
るバリヤーが除かれる。
電荷を減らすために行うリボザイムの修飾は、これらの大きい分子の細胞取込
みを増大する一方法である。しかし上記のランダム法は、リボザイムが構造およ
び機能の面で、小さい医薬分子より複雑なので得策ではない。リボザイムの触媒
活性を維持するのに必要な構造上の必要条件は当該技術分野の当業者は充分理解
していることである。これらの必要条件は、細胞送達を強化するために修飾を計
画するときに考慮される。この修飾は、ヌクレアーゼによる分解に対する感受性
を低下させるためにも計画される。これら両者の特徴はリボザイムの効力を大き
く改善するはずである。細胞の取込みは、リボザイムの切断活性のために必要な
ホスホジエステル結合を変える必要なしでいく桁も増大させることができる。
ホスフェートの骨格の化学的修飾で、負の電荷が減少し、膜を横切って自由に
拡散できるようになる。この原理は、アンチセンスDNA法について実証され成
功している。DNAとRNAとは化学的組成が類似しているので、この方法は便
利な方法である。身体内では、修飾リボザイムを組織の細胞中に拡散させるには
外部の濃度(external concentration)を維持することが必要である。疾患組織
が一過性高濃度の医薬に暴露され、その医薬が全身吸収によってゆっくり分散さ
れる投与経路が好ましい。リボザイムの循環半減期を増大するために計画された
医薬担体とともに静脈内投与する方法も利用できる。医薬担体の大きさと組成に
よって、血液流からの迅速なクリアランス(clearance)が制限される。感染部
位に蓄積するように製造された担体は、リボザイムを分解プロセスから保護する
ことができる。
医薬送達ビヒクルは全身投与と局所投与の両者に対して有効である。これらの
媒体は、除放用容器として働くようにまたはその内容物を標的細胞に直接送達す
るように設計することができる。直接伝達医薬媒体を用いる利点は、1回の取込
み当たり多数の分子が送達されることである。このような媒体は、他の方法では
血液流から速やかに除去される医薬の循環半減期を増大させることができること
が示されている。この範疇に入るかような特別の医薬送達ビヒクルの例としては
、リポソーム類、ヒドロゲル類、シクロデキストリン類、生分解性ナノカプセル
類および生物接着性ミクロスフェア(bioadhesive microsphere)類がある。
この範疇の送達系のうちリポソームが好ましい。リポソームは細胞内安定性を
増大し、取込み効率を高め、そして生物活性を改善する。
リポソームは、細胞膜を構成している脂質と類似の方式で配列された脂質で構
成された中空の球状媒体である。リポソームは、水溶性化合物を保有する内部水
性空間を有し、大きさは直径が0.05〜数ミクロンの範囲内にある大きさである。
いくつかの試験の結果、リポソームはRNAを細胞に送達することができかつそ
のRNAは生物活性を保有したままであることが分かった。
例えば、研究用具として最初設計されたリポソーム送達ビヒクルのリポフェク
チン(Lipofectin)は、無傷のmRNA分子を細胞に送達して対応するタンパク
質を産生させ得るることが分かった。他の試験では、長さが3500ヌクレオチドで
かつHIVの構造タンパク質に対してアンチセンスのRNA分子を含有する抗体
標的(antibody targeted)リポソーム伝達系は、配列特異的にウイルスの増殖
を阻害した。その抗体はリポソームを目標の感染細胞に向けたのみならず、感染
細胞によるリポソームのインターナリゼーションをトリガーした。エンドサイト
ーシスをトリガーすることはウイルスを阻害するのに有用である。最終的に、リ
ポソームで送達された合成のリボザイムは、H9細胞(HIV感受性細胞の一例
)の核に集中して機能することが分かり、それらリボザイムが配列を細胞内で切
断することが実証された。一層小さいリボザイム(長さが142個未満のヌクレオ
チ
ド)を用いて他の細胞型にリポソームで伝達すると、異なる細胞内局在を示す。
リポソームはいくつもの利点を提供する。すなわちリポソームは組成が非毒性
でかつ生物分解性であり;リポソームは長い循環半減期を示し;そして認識分子
をその表面に容易に結合して、組織をターゲティングすることができる。最終的
に、液体懸濁物または凍結乾燥物の製品のリポソームベース医薬をコスト面で効
率的に製造できたので、この方法が容認可能な医薬伝達系として実施できること
が実証された。
ナノカプセルおよびヒドロゲルのような他の徐放性医薬送達系をリボザイムの
送達ビヒクルとして用いてもよい。これらの担体は化学療法剤およびタンパク質
ベースの医薬用に開発されたものであるので、リボザイムの送達に適応させるこ
とができる。
リボザイムの局所投与は、投与部位に濃度が局在して全身吸収が最少になるの
で有利である。このことから疾患部位に対するリボザイムの送達法が簡単になり
、毒性の程度が低下する。さらに、塗布すべき物質の量が他の投与経路の場合に
必要な量よりはるかに少ない。有効な送達を行うにはリボザイムが感染細胞中に
拡散する必要がある。負の電荷を中和するためリボザイムを化学的に中和するこ
とが浸透させるのに必要である。しかし、電荷の中和が不充分な場合、修飾リボ
ザイムを、アゾーン(Azone)またはオレイン酸のような浸透エンハンサーをリ
ポソーム中に混合して調合してもよい。リポソームが、修飾リボザイムと浸透エ
ンハンサーがリポソームから感染細胞中に移動する場合に除放性を与える媒体で
あるか、または、リポソームのリン脂質が、修飾リボザイムと浸透エンハンサー
の細胞への送達を容易にすることとに直接参画する。場合によっては、リボザイ
ムと浸透エンハンサーの両者を、除放性を付与するため坐剤に配合してもよい。
またリボザイムは全身投与を行ってもよい。全身吸収とは、薬剤が血流中に蓄
積され次いで全身に分布されることを意味する。全身吸収をもたらす投与経路と
しては、静脈内、皮下、腹腔内、鼻腔内、くも膜下および眼の経路がある。これ
らの各投与経路によって、接近可能な疾患組織がリボザイムに対して暴露される
。
皮下投与の場合は局在リンパ節に注入されそこからリンパネットワークを通じて
循環が行われる。循環系に入る速度は分子量または大きさの関数であることが示
されている。リポソームなどの医薬担体を使用すると、リボザイムはリンパ節に
局在する。そのリボザイムは細胞中に拡散するよう修飾することができ、または
未修飾リボザイムまたは修飾リボザイムを細胞中に送達するのにリポソームは直
接参画することができる。この方法は、抗HIVリボザイムを用いてAIDSを
治療するのに特に有用である。
AIDS療法に好ましいのは、オリゴヌクレオチドをリンパ球とマクロファー
ジに送達できるリポソーム配合物を使用することである。このオリゴヌクレオチ
ド送達系は、感染した一次免疫細胞内でHIVの増殖を阻害する。この配合物は
37℃にて8時間後に、90%のリンパ球が吸収することを全血の試験結果が示して
いる。生体分布(biodistribution)および薬物動態学の予備試験では、静脈内
投与を行ってから1時間後に、脾臓組織の1g当たり注射された投与量の70%が
脾臓で得られた。この配合物は、二つの理由から、抗AIDSリボザイムに用い
る優れた送達ビヒクルを提供する。第一に、Tヘルパーリンパ球とマクロファー
ジがウイルスに感染した一次細胞であり、そして第二に皮下投与によって、リボ
ザイムが、リンパ節内に常在しているHIVに感染したリンパ球とマクロファー
ジに送達される。リポソームはリンパ系を出て循環系に入り、次いで脾臓内に蓄
積され、脾臓内でリボザイムが、常在しているリンパ球とマクロファージに送達
される。
腹腔内投与の場合もやはり循環系に入り、リボザイム送達ビヒクルの複合体の
分子量または大きさが循環系に入る速度を制御している。
静脈内に注射されたリポソームは、肝臓、肺および脾臓への蓄積を示す。その
組成と大きさを調節して、注射投与量の30〜40%が蓄積されるようにすることが
できる。残りの投与量は血流中を24時間まで循環する。
選択される送達方法は、感染細胞中の細胞質への蓄積が行われねばならずかつ
分子は最適の投与を行うためある種のヌクレアーゼ耐性を有していなければなら
ない。核への送達も利用できるが余り好ましくない。最も好ましい送達法として
は、リポソーム(10〜400nm)、ヒドロゲル、放出制御ポリマー、マイクロイン
ジェクションもしくは電気穿孔法(エキソビボ処置の場合に用いる)およびその
外の薬学的に許容される媒体が挙げられる。投与量は疾患の症状と投与経路によ
って決まるが、体重1kg当たり1日に100〜200mgである。治療期間は、疾患の症
状の過程によって定まり、通常少なくとも14〜16日間、可能であれば連続して行
う。局所投与、眼への投与および膣への投与の場合、毎日複数回投与することが
考えられる。その投与回数は、疾患送達ビヒクルおよび臨床試験による効力のデ
ータによって決まる。
リボザイムの細胞内治療濃度は、取込みと分解の速度によって決定される。分
解度が低下するとリボザイムの細胞内半減期が延長される。したがって、化学的
に修飾されたリボザイム、例えばホスフェート骨格の修飾またはリボザイムの5
'と3'の末端のヌクレオチド類似体によるキャッピングを行ったリボザイムでは
異なる投与量を投与する必要がある。有用な系の説明は上記技術中に提供されて
いるが、これらはすべて本明細書に援用するものである。
リボザイムの設計に関するさらに詳細な説明については1993年8月6日付けで
出願されたDraperの米国特許願第08/103,243号を参照のこと。なおこの文献は
すべて本明細書に援用するものである。
