- 【特許請求の範囲】 1.植物細胞を再生培地中で培養することを含む植物の形成方法であって、細 胞が気孔細胞であることを特徴とする方法。 2.気孔細胞を、該細胞を含有する無傷の器官内で再生させる、請求項1に記 載の方法。 3.気孔細胞を、該細胞を含有する植物組織から単離する、請求項1に記載の 方法。 4.気孔細胞が単離された葉の表皮の中にある、請求項1に記載の方法。 5.植物がビート(Beta vulgaris)である、請求項1−4のい ずれか1項に記載の方法。 6.植物がサトウダイコンである、請求項5に記載の方法。 7.再生がホルモンを含有しない培地上でのカルスの形成を含む、請求項1− 6のいずれか1項に記載の方法。 8.遺伝材料を植物の細胞に導入し、この形質転換細胞から全植物体を再生さ せることを含む、植物の遺伝子形質転換法であって、細胞が気孔細胞であること を特徴とする方法。 9.形質転換を、気孔を含む無傷の組織につき実施する、請求項8に記載の方 法。 10.組織が老化した葉の組織である、請求項9に記載の方法。 11.組織が葉の表皮である、請求項9または10に記載の方法。 12.形質転換した気孔組織をマセレートして細胞懸濁液を調製し、これから 植物を再生する、請求項9または10または11に記載の方法。 13.気孔細胞を再生前にプロトプラストに変換する、請求項8−12のいず れか1項に記載の方法。 14.遺伝材料が抗生物質に対する耐性を特定しない選択性マーカー遺伝子を 含むDNA構築体であり、かつ形質転換された組織を適切な選択試薬に暴露する 、請求項8−13のいずれか1項に記載の方法。 15.形質転換を、孔辺細胞を含有する細胞集団につき実施する、請求項8− 14のいずれか1項に記載の方法。 16.形質転換を、孔辺細胞濃度の高い細胞集団につき実施する、請求項15 に記載の方法。 17.形質転換を細胞懸濁液につき実施する、請求項8−16のいずれか1項 に記載の方法。 18.気孔細胞を形質転換の前または後に細胞壁の酵素消化によりプロトプラ ストに変換する、請求項8−17のいずれか1項に記載の方法。 19.形質転換法が、気孔細胞の懸濁液を微細繊維材料と混合し、これを遺伝 材料の存在下で撹拌することを含む、請求項8−18のいずれか1項に記載の方 法。 20.再生がホルモンを含有しない培地上でのカルスの形成を含む、請求項8 −18のいずれか1項に記載の方法。 21.ベータ・ブルガリス(Beta vulgaris)の形質転換方法で あって: (i)気孔細胞を含有する植物組織を用意し: (ii)遺伝材料を気孔細胞に直接DNA挿入により導入し;そして (iii)形質転換した気孔細胞から植物を再生させる 工程を含む方法。 22.導入された遺伝材料が抗生物質耐性を特定する遺伝子を含まないDNA 構築体である、請求項21に記載の方法。 23.植物組織が葉の表皮である、請求項21または22に記載の方法。
