Claims (1)
- 【特許請求の範囲】 1.(i)約20℃ないし約50℃の温度範囲の水溶解性、(ii)約0.001%ないし約10 %(重量/容量)の範囲の分離媒質における濃度、(iii)約5×103ないし約1×106 ダルトンの範囲の分子量、および(iv)約6ないし約9の範囲のpHを有する水 性媒質中の荷電基の不在を特徴とする1またはそれ以上の荷電していない水溶性 シリカ吸着ポリマーを含有する分離媒質を提供することからなる、キャピラリー 電気泳動において電気浸透流を抑制する方法。 2.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーが実質 的にヒドロキシル基がない請求項1記載の方法。 3.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がポリラクタムである請求項2記載の方法。 4.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がポリビニルピロリドンである請求項3記載の方法。 5.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がN,N−二置換ポリアクリルアミドまたはN−置換ポリアクリルアミ ドであり、前記窒素置換基がC1ないしC3アルキル;ハロ置換C1ないしC3アル キル;メトキシ置換C1ないしC3アルキル;およびヒドロキシル置換C1ないし C3アルキルからなる群から選ばれる請求項1記載の方法。 6.前記窒素置換基がC1ないしC3アルキル;ハロ置換C1ないしC3アルキル ;およびメトキシ置換C1ないしC3アルキルからなる群から選ばれる請求項5記 載の方法。 7.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がポリ(ジメチルアクリルアミド)である請求項6記載の方法。 8.前記電気浸透流が約2×10-5cm2/秒−ボルトよりも小さい請求項7記載の 方法。 9.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーが理論 プレートの数と、約100ないし約500のヌクレオチドの大きさの範囲のポリヌクレ オチドの大きさとの間で実質的に直線関係を与える請求項7記載の方法。 10.(i)約20℃ないし約50℃の温度範囲の水溶解性、(ii)約0.001%ないし約 10%(重量/容量)の範囲の分離媒質における濃度、(iii)約5×103ないし約1× 106ダルトンの範囲の分子量、および(iv)約6ないし約9の範囲のpHを有する 水性媒質中の荷電基の不在を特徴とする1またはそれ以上の荷電していない水溶 性シリカ吸着ポリマーを含有する分離媒質を提供することからなる、キャピラリ ー電気泳動において壁−分析物の相互作用を抑制する方法。 11.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーが実質 的にヒドロキシル基がない請求項10記載の方法。 12.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がポリラクタムである請求項11記載の方法。 13.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がポリビニルピロリドンである請求項12記載の方法。 14.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がN,N−二置換ポリアクリルアミドまたはN−置換ポリアクリルアミ ドであり、前記窒素置換基がC1ないしC3アルキル;ハロ置換C1ないしC3アル キル;メトキシ置換C1ないしC3アルキル;およびヒドロキシル置換C1ないし C2アルキルからなる群から選ばれる請求項10記載の方法。 15.前記窒素置換基がC1ないしC3アルキル;ハロ置換C1ないしC3アルキル ;およびメトキシ置換C1ないしC3アルキルからなる群から選ばれる請求項14 記載の方法。 16.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーの少な くとも1がポリ(ジメチルアクリルアミド)である請求項15記載の方法。 17.前記電気浸透流が約2×10-5cm2/秒−ボルトよりも小さい請求項16記載 の方法。 18.前記1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーが理論 プレートの数と、約100ないし約500のヌクレオチドの大きさの範囲のポリヌクレ オチドの大きさとの間で実質的に直線関係を与える請求項16記載の方法。 19.電荷運搬成分; 篩分け成分;および (i)約20℃ないし約50℃の温度範囲の水溶解性、(ii)約0.001%ないし約 10%(重量/容量)の範囲の分離媒質における濃度、(iii)約5×103ないし約1× 106ダルトンの範囲の分子量、および(iv)約6ないし約9の範囲のpHを有する 水性媒質中の荷電基の不在を特徴とする1またはそれ以上の荷電していない水溶 性シリカ吸着ポリマーからなる表面相互作用成分 からなる、キャピラリー電気泳動によってポリヌクレオチドを分離するための組 成物。 20.約5000センチポアズよりも小さい粘度を有する請求項19記載の組成物。 21.コーティングしていないシリカキャピラリーにおいて電気泳動によって異 なる大きさのポリヌクレオチドを分離する方法が: 第一端部と第二端部を有するコーティングしていないシリカキャピラリーを用 意し、このコーティングしていないシリカキャピラリーが(i)約20℃ないし約50 ℃の温度範囲の水溶解性、(ii)約0.001%ないし約10%(重量/容量)の範囲の 分離媒質における濃度、(iii)約5×103ないし約1×106ダルトンの範囲の分子量 、および(iv)約6ないし約9の範囲のpHを有する水性媒質中の荷電基の不在を 特徴とする1またはそれ以上の荷電していない水溶性シリカ吸着ポリマーを含み ; コーティングしていないシリカキャピラリー中に異なる大きさのポリヌクレオ チドの試料を装填し;そして コーティングしていないシリカキャピラリーを通って試料中の異なる大きさの ポリヌクレオチドが移動するように、コーティングしていないシリカキャピラリ ーの第一と第二の端部間に電場を印加する、 各工程からなる方法。
